The detection of bacterial pathogen such as Staphylococcus aureus(S.aureus) is essential for the regulation of food hygiene and disease diagnosis.Herein,we developed a simple one-step fluorescence resonance energy transfer(FRET)-based sensor for specific and sensitive detection of S.aureus in food and serum samples,in which aptamer-modified quantum dots(aptamer-QDs) was employed as the energy donor and antibiotic of teicoplanin functionalized-gold nanoparticles(Teico-AuNPs) was chosen as the energy acceptor.Within 1 h,the FRET-based sensor showed a linear range of from 10 cfu/mL to 5 × 10~8 cfu/mL,with the low limit of detection(LOD,2 cfu/mL) for S.aureus in buffer.When further applied to assay S.aureus in real samples,the FRET-based sensor showed good recoveries ranging from 84.5% to 110.0%,with relative standard derivations(RSDs) of 0.01%-0.44% and a LOD of 100 cfu/mL in milk,orange juice and human serum.  相似文献

A series of diazenyl schiff bases have been synthesized by reaction of salicylaldehyde containing azo dyes with various substituted aniline derivatives in the presence of acetic acid as catalyst. The structures of diazenyl derivatives were determined by FTIR, UV–vis, 1H NMR, 13C NMR, CHN analysis, fluorimetric and mass spectroscopic studies. The synthesized derivatives were screened for their in vitro antimicrobial activity against various Gram-positive (S. aureus, B. subtilis, B. cereus), Gram-negative (S. typhi, S. enterica, E. coli, P. aeruginosa) bacterial and fungal (C. albicans, A. niger and A. fumigatus) strains, using cefadroxil (antibacterial) and fluconazole (antifungal) as standard drugs. The diazenyl schiff bases were also screened for their cytotoxicity against human colorectal carcinoma cell line (HCT-116) using 5-fluorouracil as standard drug by Sulforhodamine-B Stain (SRB) assay. The schiff bases exhibited significant activity toward both Gram-positive, Gram-negative bacterial and fungal strains. Most of the synthesized derivatives showed high activity against S. enterica. 4-((2,5-Dichlorophenyl)diazenyl)-2-((3-bromophenylimino)methyl)phenol (SBN-40) was found to be very active against S. aureus, B. cereus and E. coli, with MIC = 0.69 (µM/ml × 102). The compound 4-((2-bromophenyl)diazenyl)-2-((4-nitrophenylimino)methyl)phenol (SBN-13) possessed comparable activity (IC50 = 7.5 µg/ml) to the standard drug 5-fluorouracil (IC50 = 3.0 µg/ml) against human colorectal carcinoma cell line (HCT-116).  相似文献

The radiosensitivities of Escherichia coli and Staphylococcus aureus on poached chicken meal (PCM) and minced chicken substrate (MCS) were determined. Effect of irradiation (0, 1, 2 kGy) on total viable cells (TVC) of PCM components was determined under chilled (3–5 °C) storage (0, 9, 14, 21 days) and challenge testing of the bacterial isolates with irradiation (0, 2, 3 kGy) was also conducted on PCM under chilled storage (0,7, 14, 21, 28 days). Additionally, sensory evaluation of the PCM components was assessed with irradiation (0, 2, 3 kGy) during chilled storage (0, 7, 14, 21 days). D10 of E. coli on PCM and MCS were 0.18 and 0.25 kGy whiles those of S. aureus were 0.27 and 0.29 kGy, respectively. D10 values for PCM<MCS and values for S. aureus>E. coli. 2 kGy controlled TVC and extended the shelf life of meals to ⩾14 days but 3 kGy was required to eliminate E. coli and S. aureus. Sensory qualities of the meal were not affected by an irradiation dose of 3 kGy.  相似文献

This study evaluated effect of gamma irradiation on survival of Salmonella Typhimurium and Staphylococcus aureus on lettuce and damage of cell envelope. S. Typhimurium and S. aureus were inoculated on red leaf lettuce, and they were irradiated at 0, 0.5, 1, 1.5, 2, 2.5, and 3 kGy, and the samples were then stored at 7 and 25 °C for 7 days. Survival of S. Typhimurium and S. aureus were enumerated on xylose lysine deoxycholate agar and Baird–Parker agar, respectively. D10 value (dose required to reduce 1 log CFU/leaf) was calculated, and kinetic parameters (maximum specific growth rate; μmax and lag phase duration; LPD) were calculated by the modified Gompertz model. In addition, cell envelope damage of the pathogens was observed by scanning electron microscope (SEM) and transmission electron microscope (TEM). D10 values were 0.35 and 0.33 kGy for S. Typhimurium and S. aureus, respectively. During storage at 7 °C, S. Typhimurium and S. aureus had significant (P<0.05) growth only on non-irradiated samples up to about 2.5 and 4 log CFU/leaf at 0.42 and 1.28 log CFU/leaf/day of μmax, respectively. At 25 °C, cell counts of S. Typhimurium and S. aureus on the samples irradiated at 0 and 0.5 kGy increased (P<0.05) up to 3–6 log CFU/leaf. The μmax of both pathogens were higher in 0 kGy (1.08–2.27 log CFU/leaf/day) and 0.5 kGy (0.58–0.92 log CFU/leaf/day), and LPDs ranged from 1.53 to 3.14 day. SEM and TEM observations showed that cells irradiated at 1.5 and 3 kGy showed disrupted cell membrane. These results indicate that gamma irradiation could be a useful decontamination technology to improve food safety of lettuce by destroying cells of S. Typhimurium and S. aureus.  相似文献

Predominant monoacylation of 1,n-linear disulfonamides took place in the presence of pyrrolidinopyridine-type organocatalysts when the chain length of the linear disulfonamides was n = 3, 4, or 5 (monoacylate/diacylate = up to 44). The chemoselectivity of the competitive acylation between N,N′-ditosyl-1,5-pentanediamine (n = 5) and N,N′-ditosyl-1,3-propanediamine (n = 3) was found to be 36, favoring the former substrate. Different chain length by only one carbon atom was discriminated in the competitive acylation between N,N′-ditosyl-1,5-pentanediamine (n = 5) and of N,N′-ditosyl-1,4-butanediamine (n = 4) with the relative acylation rate of 16 in the presence of the organocatalyst.  相似文献

Two new tetrahydrofuran derivatives, mucorinic acids A (1) and B (2) as well as the three known secondary metabolites dehydroabietic acid (3), Δ8-dihydroabietic acid (4) and 8-pimarenic acid (5) were isolated from a solid culture of the fungus Mucor spp. isolated on insect Acalymma bivittula. The structure of these compounds was elucidated by analysis of NMR and MS spectroscopic data. Compounds were tested in antimicrobial and insecticidal assays. Dehydroabietic acid exhibited moderate larvicidal activity on Aedes aegypti larvae with 65% mortality at 10 ppm. Both new compounds 1 and 2 showed interesting antibacterial activity on Staphylococcus aureus and methicillin resistant S. aureus with MIC values in the 8–16 μg/ml.  相似文献

A nafion covered carbon nanotubes-paste electrode modified with poly(m-ferrocenylaniline), (Nf/p(FcAni)-CNTsPE), provides a novel voltammetric sensor for the selective determination of dopamine (DA) and uric acid (UA) in the presence of ascorbic acid (AA). We studied the electrochemical activity of Nf/p(FcAni)-CNTsPE toward DA, UA, and AA by differential pulse voltammetry (DPV). DA and UA anodic peaks appear at 0.30 and 0.45 V, respectively while an anodic peak for AA was not observed. DPV oxidation peak values are linearly dependent on DA concentration over the range 1–150 μM (r2 = 0.992), and on UA concentration over the range 5–250 μM (r2 = 0.997). DA and UA detection limits are estimated to be 0.21 and 0.58 μM, respectively. The modified electrode shows both good selectivity and reproducibility for the selective determination of DA and UA in real samples. Finally, the modified electrode was successfully applied for the determination of DA and UA in pharmaceutical or biological sample fluids.  相似文献

An ESIPT-based fluorescent probe (Probe 1) using acrylate as recognition group for the selective and sensitive detection of cysteine/homocysteine (Cys/Hcy) has been developed. In the presence of Cys/Hcy, this probe was transformed into 1,3-bis(bispyridin-2ylimino)isoindolin-4-ol (dye 4) which displayed red fluorescence with a large Stokes shift (217 nm) when excited. The detection limits are as low as 5.4 nM and 7.0 nM for Cys and Hcy respectively (based on S/N = 3). Importantly, this probe has been successfully demonstrated for the detection of intracellular Cys/Hcy in living cells.  相似文献

Tellurium (IV) complexes with pyridine-2,6-dicarboxylate ligand were synthesized by slow evaporation from aqueous solutions yielding a new compound: [(C7H6NO4)2TeBr6·4H2O]. The structure of this compound was solved and refined by single-crystal X-ray diffraction. The compound is centrosymmetric P21/c (N°: 14) with the parameters a = 8.875(5) Å, b = 15.174(5) Å, c = 10.199(5) Å, β = 94.271° (5) and Z = 2. The structure consists of isolated H2O, isolated [TeBr6]2? octahedral anions and (pyridine-2,6-dicarboxylate) [C7H6NO4]+ cations. The stability of the structure was ensured by ionic and hydrogen bonding contacts (N–H?Br and O–H?Br) and Van-Der Walls interaction. The thermal decomposition of the compound was studied by thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). The FTIR and Raman spectroscopy at different temperatures confirm the existence of vibrational modes that correspond to the organic, inorganic and water molecular groups. Additionally, the UV–Vis diffuse reflectance spectrum was recorded in order to investigate the band gap nature. The measurements show that this compound exhibits a semiconducting behavior with an optical band gap of 2.66 eV.  相似文献

In an effort to develop novel antiamoebic scaffolds having better efficacy than the standard drug metronidazole (IC50 = 1.80 μM) used against Entamoeba histolytica, quinazolin-4(3H)-one Schiff base conjugates were synthesized and evaluated against HM1: IMSS strain of E. histolytica. Out of the thirteen compounds (S2-S14), six compounds (S2, S3, S4, S5, S6 and S11) were found to be better inhibitors than metronidazole and showed low cytotoxicity on HeLa cells, a cervical cancer cell line. The structure of intermediate compound S1 was confirmed by crystal structure studies.  相似文献

Anti-inflammatory and antibacterial activities of some novel quinazolinones were determined. Evaluation of anti-inflammatory activity of test compounds was performed using carrageenan induced paw edema in rats. Oral administration of test compounds 25 mg/kg and 50 mg/kg reduced the paw edema significantly (P < 0.05) in a dose dependent manner compared to carrageenan induced rats. The test compounds were also screened for their antibacterial activity against the strains of Staphylococcus aureus and Escherichia coli at the concentrations of 200 μg/ml and 1 mg/ml. The test compounds showed better activity as that of the standard lincomycin at the tested higher concentration against S. aureus. None of the compounds exhibit comparable activity to that of the standard ceftazidime against E. coli.  相似文献

We have synthesized and characterized Ag NPs decked GO composite and studied its role as reusable catalyst for the ‘ON WATER’ chemoselective synthesis of pyranodipyrazolones via the reaction of different carbonyl compounds with 3-methyl-1-phenyl-1H-pyrazol-5(4H)-one. This method illustrates significant selectivity for pyranodipyrazolones over arylmethylene bispyrazolols and arylmethylenepyrazolones. Synergistic effect of heterogenic nature of water with reactants and Ag NPs/GO had profuse outcome on reaction as indicated by high TOF (18.03 × 10?5 mol g?1 min?1). Since the copper ions (Cu2+) play a fatal role in many foundational physiological processes, it is important to develop a simple, highly sensitive and selective sensor for Cu2+ detection in living systems. Herein, an intramolecular charge transfer (ICT) and dansyl-based fluorescent chemosensor 1 was designed, synthesized and characterized for the sensitive and selective quantification of Cu2+. It exhibited remarkable fluorescence quenching upon addition of Cu2+ over other selected metal ions, attributed to the complex formation between 1 and Cu2+ with the association constant 6.7 × 105 M?1. The sensor 1 showed a fast and linear response towards Cu2+ in the concentration range from 0 to 12.5 × 10?6 mol L?1 with the detection limit of 2.5 × 10?7 mol L?1. This detection could be carried out in a wide pH range of 5.0–14. Furthermore, sensor 1 can be used for detecting Cu2+ in living cells.  相似文献

In this research, the effect of gamma irradiation on the inactivation of Escherichia coli O157:H7 (ATCC 33150), Staphylococcus aureus (ATCC 2392) and Salmonella typhimurium (NRRL 4463) inoculated into Tekirdag meatballs was investigated. The meatball samples were inoculated with pathogens and irradiated at the absorbed doses of 1, 2.2, 3.2, 4.5 and 5.2 kGy. E. coli O157:H7 count in 1 kGy irradiated meatballs stored in the refrigerator for 7 days was detected to be 4 log cfu/g lower than the count in nonirradiated samples (p<0.05). S. aureus counts were decreased to 4 log cfu/g after being exposed to irradiation at a dose of 1 kGy. Although it was ineffective on elimination of S. typhimurium, irradiation at a dose of 3.2 kGy reduced E. coli O157:H7 and S. aureus counts under detectable values in the meatballs. However, none of the test organisms were detected in the samples after irradiation with 4.5 kGy doses.  相似文献

We presented the design, synthesis and preliminary evaluation of the 2,4-dinitrobenzenesulfonate (DNBS) of imidazo[1,5-α]pyridine derivative, NIPY-DNBS, as a turn-on fluorescent probe for the detection of thiols in aqueous solution. The reaction mechanism was confirmed by means of fluorescence, absorption and HRMS. The large Stokes shift (201 nm), high sensitivity (the detection limit for Cys was calculated to be as low as 0.17 μM) and fast response (10 min) of NIPY-DNBS make it a practical and reliable method for fluorescence imaging. Furthermore, application of NIPY-DNBS for the selective detection of intracellular thiols has been successfully demonstrated in living A549 cells.  相似文献
