The synthesis and characterization of a new photocleavable crosslinker is presented here. Dual stimuli‐responsive P(VCL‐co‐NHMA) microgels were prepared by precipitation polymerization of vinylcaprolactam (VCL) with N‐hydroxymethyl acrylamide (NHMA) and the new crosslinker. The microgels had distinct temperature sensitivity as observed in the case of PVCL‐based particles and their volume phase transition temperature (VPTT) shifted to higher temperature with increasing NHMA content. Photolytic degradation experiments were investigated by irradiation with UV light, which led to microgel disintegration caused by cleavage of the photolabile crosslinking points. The degradation behavior of the microgels was conducted with respect to degradation rates by means of the relative turbidity changes. Hence, the microgels could totally degrade into short linear polymers by UV light, thus representing a great potential as new light and temperature dual responsive nanoscale materials. © 2014 Wiley Periodicals, Inc. Using an advanced functional photoresist we introduce direct‐laser‐written (DLW) 3D microstructures capable of complete degradation on demand. The networks consist exclusively of reversible bonds, formed by irradiation of a phenacyl sulfide linker, giving disulfide bonds in a radical‐free step‐growth polymerization via a reactive thioaldehyde. The bond formation was verified in solution by ESI‐MS. To induce cleavage, dithiothreitol causes a thiol–disulfide exchange, erasing the written structure. The mild cleavage of the disulfide network is highly orthogonal to other, for example, acrylate‐based DLW structures. To emphasize this aspect, DLW structures were prepared incorporating reversible structural elements into a non‐reversible acrylate‐based standard scaffold, confirming subsequent selective cleavage. The high lateral resolution achievable was verified by the preparation of well‐defined line gratings with line separations of down to 300 nm.  相似文献

Herein, the synthesis and potential application as cargo delivery systems of thermo‐responsive poly(N‐vinylcaprolactam) (PVCL)‐based, pH‐responsive poly(2‐(diethylamino)ethyl) methacrylate (PDEAEMA)‐based, and thermo‐, and pH‐responsive PDEAEMA/PVCL‐based core–shell nanogels are reported. All the nanogels have been synthesized using different dextran‐methacrylates (Dex‐MAs) as macro‐cross‐linkers. Doxorubicin hydrochloride (DOXO), an anticancer drug, has been effectively loaded into nanogels via hydrogen‐bonding interactions between ? OH groups of DOXO and ? OH groups of Dex‐MA chains. Drug‐release profiles at various pHs, and the cytocompatibility of the DOXO‐loaded nanogels have been assessed in vitro using cervical cancer HeLa and breast cancer MDA‐MB‐231 cell lines. In all the cases, the DOXO release is controlled by Fickian diffusion and case‐II transport, being the diffusional process dominant. In addition, DOXO‐loaded nanogels are efficiently internalized by HeLa and MDA‐MB‐231 cells and DOXO is progressively released in time. This article reports the synthesis of biohybrid nanogels by crosslinking six‐arm acrylate functionalized star shaped P(EO‐stat‐PO) with the amine groups of various amino acids of a hen egg ovalbumin in W/O emulsion. PEG Poly(ethylene glycol)‐diamines of different molecular weights (368.5 g/mol, 897.1 g/mol, 3000 g/mol, and 6000 g/mol) were used to optimize the crosslinking reactions in aqueous droplets. The increase of molecular weight of the PEG‐diamine led to decrease of the nanogel size due to better stabilization by longer PEG chains. The size of the ovalbumin nanogels was independent of the acrylate‐sP(EO‐stat‐PO):ovalbumin ratio in the reaction mixture. The bicinchoninic acid (BCA) assay proved that the ovalbumin has been effectively crosslinked by reactive prepolymers. From the results of BCA assay, it can also be established that there exists a limiting amount of ovalbumin which can be incorporated into the nanogels. Both Diamine and ovalbumin‐based nanogels exhibit amphoteric behavior and display a positive charge in acidic and a negative charge in basic environment. © 2012 Wiley Periodicals, Inc. A facile synthetic strategy was developed for the preparation of thermoresponsive nanocomposite hydrogels comprising crosslinked chitosan (CS) networks and poly(N‐isopropylacrylamide) [p(NIPAAm)] nanogels. First, thermoresponsive p(NIPAAm) nanogels were synthesized via emulsion polymerization. The p(NIPAAm) nanogels were introduced into methacrylamide CS (MC) solution and the free‐radical initiated crosslinking reaction of MC produced nanogel‐embedded hydrogels. The last step involves the loading of the antibacterial model drug levofloxacin (LFX) into the prepared nanocomposite hydrogels by allowing the preformed hydrogels to swell to equilibrium in the drug's aqueous solution. The integration of p(NIPAAm) nanogel into CS networks facilitates thermoresponsive release of LFX with an enhancement of the drug‐loading capacity within the hydrogel. Notably, thermoresponsive drug‐release was achieved without unwarranted modification of the hydrogel's dimension and shape, although an increase in temperature caused the collapse of the p(NIPAAm) nanogels. The thermoresponsive property of the investigated nanocomposite hydrogel is beneficial and may offer broad opportunities for drug temperature‐triggered release for clinical applications. © 2018 Wiley Periodicals, Inc. We present novel redox‐responsive hydrogels based on poly(N‐isopropylacrylamide) or poly(acrylamide), consisting of a reversible disulfide crosslinking agent N,N′‐bis(acryloyl)cystamine and a permanent crosslinking agent N,N′‐methylenebisacrylamide for microfluidic applications. The mechanism of swelling/deswelling behavior starts with the cleavage and reformation of disulfide bonds, leading to a change of crosslinking density and crosslinking points. Raman and ultraviolet‐visible spectroscopy confirm that conversion efficiency of thiol–disulfide interchange up to 99%. Rheological analysis reveals that the E modulus of hydrogel is dependent on the crosslinking density and can be repeatedly manipulated between high‐ and low‐stiffness states over at least 5 cycles without significant decrease. Kinetic studies showed that the mechanical strength of the gels changes as the redox reaction proceeds. This process is much faster than the autonomous diffusion in the hydrogel. Moreover, cooperative diffusion coefficient (Dcoop) indicates that the swelling process of the hydrogel is affected by the reduction reaction. Finally, this reversibly switchable redox behavior of bulky hydrogel could be proven in microstructured hydrogel dots through short‐term photopatterning process. These hydrogel dots on glass substrates also showed the desired short response time on cyclic swelling and shrinking processes known from downsized hydrogel shapes. Such stimuli‐responsive hydrogels with redox‐sensitive crosslinkers open a new pathway in exchanging analytes for sensing and separating in microfluidics applications. © 2019 Wiley Periodicals, Inc. Various polymerization mechanisms have been developed to prepare peptide‐immobilized poly(ethylene glycol) (PEG) hydrogels, a class of biomaterials suitable for studying cell biology in vitro. Here, a visible light mediated thiol‐acrylate photopolymerization scheme is reported to synthesize dually degradable PEG‐peptide hydrogels with controllable crosslinking and degradability. The influence of immobilized monothiol pendant peptide is systematically evaluated on the crosslinking of these hydrogels. Further, methods are proposed to modulate hydrogel crosslinking, including adjusting concentration of comonomer or altering the design of multifunctional peptide crosslinker. Due to the formation of thioether ester bonds, these hydrogels are hydrolytically degradable. If the dithiol peptide linkers used are susceptible to protease cleavage, these thiol‐acrylate hydrogels can be designed to undergo partial proteolysis. The differences between linear and multiarm PEG‐acrylate (i.e., PEGDA vs PEG4A) are also evaluated. Finally, the use of the mixed‐mode thiol‐acrylate PEG4A‐peptide hydrogels is explored for in situ encapsulation of hepatocellular carcinoma cells (Huh7). The effects of matrix stiffness and integrin binding motif (e.g., RGDS) on Huh7 cell growth and HIPPO pathway activation are studied using PEG4A‐peptide hydrogels. This visible light poly­merized thiol‐acrylate hydrogel system represents an alternative to existing light‐cured hydrogel platforms and shall be useful in many biomedical applications.  相似文献

New poly(ethylene oxide)‐based block copolymers (ssBCs) with a random copolymer block consisting of a reduction‐responsive disulfide‐labeled methacrylate (HMssEt) and a thermoresponsive di(ethylene glycol)‐containing methacrylate (MEO2MA) units were synthesized. The ratio of HMssEt/MEO2MA units in the random P(MEO2MA‐co‐HMssEt) copolymer block enables the characteristics of well‐defined ssBCs to be amphiphilic or thermoresponsive and double hydrophilic. Their amphiphilicity or temperature‐induced self‐assembly results in nanoaggregates with hydrophobic cores having different densities of pendant disulfide linkages. The effect of disulfide crosslinking density on morphological variation of disulfide‐crosslinked nanogels is investigated. In response to reductive reactions, the partial cleavage of pendant disulfide linkages in the hydrophobic cores converts the physically associated aggregates to disulfide‐crosslinked nanogels. The use of aliphatic polyesters for biomedical applications is limited by the lack of functionality of their backbones. The aim of the following study was to develop a novel elastic scaffold material containing functional groups to be used for future derivatization to tether peptide ligands to support cell adhesion, migration, and differentiation. The elastomer was based on three‐arm star copolymers composed of ε‐caprolactone and a functionalized ε‐caprolactone, 2‐oxepane‐1,5‐dione, and end‐terminated with acrylate groups. The elastomer thus contains a ketone and two approaches were examined for obtaining a photocrosslinkable elastomer containing functional groups: crosslinking followed by ketone reduction using sodium borohydride to generate pendant hydroxyl groups, and reaction of the ketone with hydrazines. Reduction of the ketone lead to degradation of the elastomer through transesterification and ethanolate mediated cleavage of the polymer backbone. Reaction with hydrazines did not degrade the polymer and resulted in efficient functionalization of the elastomer. © 2008 Wiley Periodicals, Inc. Water‐swellable biodegradable materials exhibiting mechanically tenacious and tough characters in the wet state were prepared by a simple blend of two enantiomeric polylactide‐grafted dextran copolymers (Dex‐g‐PLLA and Dex‐g‐PDLA). DSC and WAXD analyses demonstrated the formation of SC crystals in the copolymer blend films. SC blend films showed lamellar‐type microphase‐separated structures. When swollen with water, these blend films showed the same level of tensile strengths and Young's modulus as the films in the dry state. SC blend films degraded gradually over a month under physiological conditions with a degradation rate faster than the corresponding Dex‐g‐PLLA films. The SC‐forming enantiomeric mixture of polylactide‐grafted polysaccharides should be a good candidate for an implantable biocompatible material exhibiting favorable mechanical properties and degradation behavior. © 2012 Wiley Periodicals, Inc. Temperature sensitive poly{N‐[3‐(dimethylaminopropyl)]methacrylamide‐co‐acrylamide} [P(DMAPMA‐co‐AAm)] hydrogels were prepared by the free‐radical crosslinking copolymerization of corresponding monomers in water with N,N‐methylenebisacrylamide as the crosslinker, ammonium persulfate as the initiator, and N,N,N′,N′‐tetramethylethylenediamine as the activator. The swelling equilibrium of the P(DMAPMA‐co‐AAm) hydrogels was investigated as a function of temperature in aqueous solutions of the anionic surfactant sodium dodecyl sulfate and the cationic surfactant dodecyltrimethylammonium bromide. In pure water, regardless of the amount of N,N‐methylenebisacrylamide, the P(DMAPMA‐co‐AAm) hydrogels showed a discontinuous phase transition between 30 and 36 °C. However, the transition temperature changed from discontinuous to continuous with the addition of surfactants; this was ascribed to the conversion of nonionic P(DMAPMA‐co‐AAm) hydrogels into polyelectrolyte hydrogels due to the binding of surfactants through hydrophobic interactions. Additionally, the concentrations of free sodium dodecyl sulfate and dodecyltrimethylammonium bromide ions were measured at different temperatures by conductometry, and it was found that the electric conductivity of the P(DMAPMA‐co‐AAm)–surfactant systems depended strongly on the swelling ratio; most notably, it changed drastically near the phase‐transition temperature of the P(DMAPMA‐co‐AAm) hydrogel. © 2006 Wiley Periodicals, Inc. pH‐sensitive poly(acrylamide‐co‐itaconic acid) [P(AAm/IA)] hydrogels were prepared by radiation induced copolymerization of acrylamide (AAm) and itaconic acid (IA) at various ratios. Swelling and shrinking behaviors of these hydrogels were found greatly dependent on the composition of the hydrogel and pH of the buffer solution. To develop novel biomedical soft materials with degradability, amphiphilic poly(L ‐lactide)‐grafted dextrans (Dex‐g‐PLLAs) of relatively high sugar unit contents were synthesized with the trimethylsilyl protection method. The characteristic properties of solution‐cast films prepared from the obtained Dex‐g‐PLLAs were investigated. The water absorption and degradation rate of the Dex‐g‐PLLA films increased with increasing sugar unit content. The morphology of the bulk phase and top surface of the Dex‐g‐PLLA films was evaluated with transmission electron microscopy and atomic force microscopy, respectively. The bulk phase of the Dex‐g‐PLLA films with a sugar unit content of 16–25 wt % was found by transmission electron microscopy to form a lamellar type of phase‐separated structure composed of approximately 80–100‐nm‐wide nanodomains because of their amphiphilic and branched structures. The hydrophobic top surface for a Dex‐g‐PLLA film with a sugar unit content of 25 wt % covered with PLLA segments was confirmed by atomic force microscopy phase images to be easily converted to a wettable top surface covered with hydrophilic dextran aggregates showing an 8–10‐nm‐wide honeycomb pattern by means of annealing in water. © 2006 Wiley Periodicals, Inc. A surfactant‐free emulsion‐based approach is developed for preparation of nanogels. A water‐in‐oil emulsion is generated feasibly from a mixture of water and a solution of disulfide‐containing hyperbranched PEGylated poly(amido amine)s, poly(BAC2‐AMPD1)‐PEG, in chloroform. The water droplets in the emulsion are stabilized and filled with poly(BAC2‐AMPD1)‐PEG, and the crosslinked poly(amido amine)s nanogels are formed via the intermolecular disulfide exchange reaction. FITC‐dextran is loaded within the nanogels by dissolving the compound in water before emulsification. Transmission electron microscopy and dynamic light scattering are applied to characterize the emulsion and the nanogels. The effects of the homogenization rate and the ratio of water/polymer are investigated. Redox‐induced degradation and FITC‐dextran release profile of the nanogels are monitored, and the results show efficient loading and redox‐responsive release of FITC‐dextran. This is a promising approach for the preparation of nanogels for drug delivery, especially for neutral charged carbohydrate‐based drugs.

The dextran‐allyl isocyanate/poly(N‐isopropylacrylamide) (Dex‐AI/PNIPAAm) hydrogel was designed and prepared by copolymerization of the modified dextran with N‐isopropylacrylamide (NIPAAm). This novel Dex‐AI/PNIPAAm hydrogel is biodegradable and intelligent due to its biodegradable dextran linkage and thermosensitive PNIPAAm moiety. With an increase in dextran content, it exhibits the increased lower critical solution temperature (LCST) and decreased porous microstructure. Also, the thermosensitivity of this hydrogel is also controllable and adjustable depending on the different compositions.

Acrylamide (AAm)‐2‐acrylamide‐2‐methylpropanesulfonic acid sodium salt (AMPSNa) hydrogel and AAm‐AMPSNa/clay hydrogel nanocomposite having 10 w% clay was prepared by in situ copolymerization in aqueous solution in the presence of a crosslinking agent (N,N′‐methylenebisacrylamide (NMBA)). Swelling properties and kinetics of the hydrogel samples were investigated in water and aqueous solutions of the Safranine‐T (ST) and Brilliant Cresyl Blue (BCB) dyes. The swelling and diffusion parameters were also calculated in water and dye solutions. It was observed that the AAm‐AMPSNa/clay hydrogel nanocomposite exhibits improved swelling capacity compared with the AAm‐AMPSNa hydrogel. It was also found that the diffusion mechanisms show non‐Fickian character. Adsorption properties of the hydrogel samples in the aqueous solution of ST and BCB dyes were also investigated. Clay incorporation into the hydrogel structure increased not only the adsorption capacity but also the adsorption rate. Adsorption capacity values of the hydrogel nanocomposite were found to be 484.2 and 494.2 mg g?1 for the ST and BCB dyes, respectively. It was seen that the adsorption of dyes by the hydrogel nanocomposite completed in 10 min while the AAm‐AMPSNa hydrogel adsorbed dyes approximately in 90 min. Adsorption data of the samples were modelled by the pseudo‐first‐order and pseudo‐second‐order kinetic equations in order to investigate dye adsorption mechanism. It was found that the adsorption kinetics of hydrogel nanocomposite followed a pseudo‐second‐order model. Equilibrium isotherms were analyzed using the Langmuir and Freundlich isotherms. A biodegradable polymer network hydrogel system with both hydrophilic and hydrophobic components was synthesized and characterized. The hydrophilic and hydrophobic components were dextran and poly(D,L )lactic acid (PDLLA), respectively. These two polymers were chemically modified for incorporating unsaturated groups for subsequent UV crosslinking to generate a hydrogel with a three‐dimensional network structure. The effects of the reaction conditions on the synthesis of a dextran derivative of allyl isocyanate (dex‐AI) were studied. All newly synthesized materials were characterized by Fourier transform infrared and NMR. The swelling property of the hydrogels was studied in buffer solutions of different pHs. The results of this study showed that a wide‐range swelling property was obtained by changes in the dex‐AI/PDLLA composition ratio, the type and degree of unsaturated groups incorporated into dextran, and the UV photocrosslinking time. New isobutylene‐rich elastomers bearing multiple pendant styrenic, acrylic, maleimidic, vinylic, and allylic functional groups have been prepared and examined in the context of peroxide‐initiated crosslinking. Halide displacement from brominated poly(isobutylene‐co‐isoprene) (BIIR) by the requisite carboxylate nucleophiles in homogeneous toluene solutions provide the desired esters in quantitative yield without complications from dehydrohalogenation or premature crosslinking. Heating the resulting macromonomers with dicumyl peroxide to 160 °C under solvent‐free conditions gives thermoset derivatives, with reaction rates and yields depending markedly on functional group structure. In general, high cure extents can only be achieved using highly reactive pendant functional groups, owing to the competitive balance between crosslinking through C?C oligomerization, and degradation through β‐scission of backbone macroradical intermediates. Independent control of crosslinking rates and cure extents is gained through the use of nitroxyl radical traps bearing acrylate functionality. © 2014 Wiley Periodicals, Inc. J. Polym. Polymer nanoporous materials with periodic cylindrical holes were fabricated from microphase‐separated structure of diblock copolymers consisting of a radiation‐crosslinking polymer and a radiation‐degrading polymer through simultaneous crosslinking and degradation by γ‐irradiation. A polybutadiene‐block‐poly(methyl methacrylate) (PB‐b‐PMMA) diblock copolymer film that self‐assembles into hexagonally packed poly(methyl methacrylate) cylinders in polybutadiene matrix was irradiated with γ‐rays. Solubility test, IR spectroscopy, and TEM and SEM observations for this copolymer film in comparison with a polystyrene‐block‐poly(methyl methacrylate) diblock copolymer film revealed that poly(methyl methacrylate) domains were removed by γ‐irradiation and succeeding solvent washing to form cylindrical holes within polybutadiene matrix, which was rigidified by radiation crosslinking. Thus, it was demonstrated that nanoporous materials can be prepared by γ‐irradiation, maintaining the original structure of PB‐b‐PMMA diblock copolymer film. © 2007 Wiley Periodicals, Inc. Anionic hydrogen‐transfer homopolymerization of N‐isopropylacrylamide (NIPAAm) was carried out using t‐BuOK as an initiator in DMF under microwave irradiation. After 100 W of microwave was irradiated to the reaction mixture at 140°C for 6 h in the temperature control mode, corresponding polymer was obtained in 10% yield. In the case of conventional oil bath heating, by contrast, corresponding polymer was not obtained in similar anionic polymerization conditions. With 100 W and 2.45 GHz of microwave irradiation, formation of the polymer was obtained. Microwave‐assisted anionic hydrogen‐transfer copolymerization of NIPPAm and acrylamide (AAm) led to the formation of thermo‐sensitive copolymers whose thermo‐sensitivity was controlled by the NIPAAm/AAm unit ratio. © 2019 Wiley Periodicals, Inc.
