An octyl-functionalized hybrid silica monolithic column was developed for in-tube solid-phase microextraction (SPME) to perform on-line preconcentration coupled to capillary high-performance liquid chromatography (microHPLC) analysis. A hybrid silica monolithic column functionalized with octyl groups was conveniently synthesized by a two-step acid/base-catalyzed hydrolysis/co-condensation of tetraethoxysilane (TEOS) and n-octyltriethoxysilane (C8-TEOS). The size of through-pores as well as the carbon content can be adjusted by changing the ratio of TEOS to C8-TEOS in the polymerization mixture. The extraction characteristics of the monolithic column prepared under optimized fabrication conditions were studied by using polycyclic aromatic hydrocarbons (PAHs) as the analytes. The sample volume that could be injected into the system was increased up to 1mL with simultaneous increase of column efficiency, when hybrid silica monolithic column was used as a precolumn. Good linear calibration curves (R>0.999) were obtained, and the limits of detection (signal-to-noise ratio, S/N=3) for the analytes were found to be between 2.4 and 8.1ng/mL with a UV absorbance detector, which are 299-456 times lower than those obtained without preconcentration. The column-to-column RSD values were 1.3-8.0% for recoveries of PAHs investigated.  相似文献

以苯基三乙氧基硅烷(PTES)和1,4-双三乙氧基硅基苯(BTEB)为反应单体,分别采用盐酸和十二胺为催化剂,通过两步酸碱催化的溶胶-凝胶法制备了新型双苯基杂化硅胶毛细管电色谱整体柱。分别采用扫描电镜、红外光谱和压汞法对材料的结构特性进行了表征。结果表明,所制备的材料具有特定的网络结构特征,孔径主要分布在3.4μm左右,材料的总孔容为3.5 cm3/g,比表面积为145 m2/g。新型整体柱能够很好地分离烷基取代苯、稠环芳烃、取代苯胺和硝基苯酚异构体。整体柱重复性实验表明,6种苯的同系物保留时间和容量因子的相对标准偏差分别小于0.61%和0.30%(n=8)。不同批次的整体柱的保留时间和容量因子的相对标准偏差分别小于7.2%和5.6%(n=3)。  相似文献

Monolithic silica columns of 2.7 mm ID were prepared and derivatized with C8-TEOS and TEOS by on-column sol-gel reaction. These C8 large diameter monolithic silica columns gave 21 000 theoretical plates for aromatic hydrocarbons in 60% acetonitrile and 40% Tris-HCI buffer. The surface areas as well as the separation reproducibility were improved on coating by the sol-gel approach. Joule heating was greatly reduced by using monolithic columns to which fine quartz sand had been added during column preparation. Since this is a preliminary investigation on a monolithic column with such a large inner diameter, the separation efficiency was not so high as that presently achieved in normal capillary electrochromatography (CEC). However, use of the columns improved sample loadability and concentration detectability of electrochromatography, and semi-preparative separations could be performed.  相似文献

An organic–inorganic hybrid silica monolithic column with octyl and sulfonic acid groups has been prepared by sol–gel technique for capillary electrochromatograhpy. The structure of hybrid monolith was optimized by changing the composition of tetraethoxysilane (TEOS), octyltriethoxysilane (C8-TEOS) and 3-mercaptopropyltrimethoxysilane (MPTMS) in the mixture of precursors. Then, the obtained hybrid monolith was oxidized using hydrogen peroxide (30%, w/w) to yield sulfonic acid groups. The sulfonic acid group, which served as strong cation-exchanger, dominated the charge on the surface of the capillary column and generated stable electroosmotic flow (EOF) in a wide range of pH. The monolithic column was characterized by scanning electron microscopy (SEM), Fourier transform infrared (FT-IR) spectroscopy and elemental analysis (EA), and the performance of column was evaluated in detail by separating different kinds of compounds with column efficiency up to 155,000 plates/m for thiourea. In addition, this monolithic column was also applied in the analysis of theophylline (TP) and caffeine (CA) in beverages. The detection limits were 0.39 and 0.48 μg/mL for theophylline and caffeine, respectively. The method reproducibility was tested by evaluating the intra- and inter-day precisions, and relative standard deviations of less than 3.9 and 8.4%, respectively, were obtained. Recoveries of compounds from spiked beverage samples ranged from 87.2 to 105.2%.  相似文献

A silica-based CEC monolithic column with mixed modes of RP and weak anion-exchange (WAX) was successfully prepared by using the sol-gel technique at mild temperature. The synthesizing procedure was optimized by changing the ratios of tetraethoxysilane (TEOS), aminopropyltriethoxysilane (APTES), and octyltriethoxysilane (C(8)-TEOS) in the mixture. While serving as WAX group, the amino group dominated the charge on the surface of the capillary column and generated an EOF from cathode to anode at low pH. At pH above 7.5, a cathodic EOF was observed due to the full ionization of silanol group and the suppression in the ionization of amino group. The morphology of monolithic columns was examined by SEM, and the performance of column was evaluated in detail by separating different kinds of compounds. As expected, the monolithic column exhibited RP chromatographic behavior for neutral solutes. Fast and efficient separation of six aromatic acids was obtained using acidic mobile phase with column efficiency up to 160,000 plates/m. Symmetrical peaks can be obtained for aromatic amines because positively charged amino groups on the surface can effectively minimize the adsorption of positively charged analytes to the stationary phase.  相似文献

采用溶胶-凝胶法,以四乙氧基硅烷和苯基三乙氧基硅烷作为反应单体,通过酸碱两步催化在毛细管中进行原位缩聚反应,制备了新型有机-无机杂化硅胶基质毛细管整体柱,制备过程简单。整体柱基质中均匀分布的苯基基团可直接用于反相毛细管电色谱的分离,因而不需要对基质再进行衍生化。优化了整体柱的制备条件,采用扫描电镜和压汞法对整体柱的微观结构和孔径分布进行了表征。分别考察了溶胶-凝胶初始反应液中水的用量对柱床结构的影响和两种单体的配比对材料孔径分布的影响。研究了稠环芳烃类化合物在整体柱上的保留行为,用所制备的整体柱分离了7种苯酚类化合物,平均柱效达100000塔板/m。  相似文献

A novel monolithic silica column that has a polar‐embedded amide‐secondary amine group linking with C16 functionality for RP‐CEC is described. The amide‐secondary aminealkyloxysilane was synthesized by the reaction of 3‐(2‐aminoethylamino) propyltrimethoxysilane with hexadecanoyl chloride. Then, the silylant agent was bonded to the silica monolith matrix to produce hexadecanamide‐secondary amine bonded silica (HDAIS) monolithic column. The electrochromatographic performance of HDAIS monolithic column for the separation of neutral, basic and polar solutes was studied, which was compared to that using the hexadecyl bonded silica monolithic column. The HDAIS monolithic column displayed reduced hydrophobic retention characteristics in the separation of five hydrophobic n‐alkylbenzenes and four polar phenols when compared to the hexadecyl bonded silica monolithic column. A very much reduced silanol activity of HDAIS monolithic column was observed in the separation of test basic mixture including four aromatic amines, atenolol and metoprolol with 10 mM borate buffer (pH 7.5) containing 30% v/v ACN as the mobile phase. The comparison results indicate good performance for both polar and basic mixtures on HDAIS monolithic column in RP‐CEC, and also show promising results for further applications.  相似文献

A hybrid organic–inorganic silica monolith with hydrophobic and strong cation-exchange functional groups was prepared and used as a sorbent for micro-solid phase extraction (micro-SPE). The hybrid silica monolith functionalized with octyl and thiol groups was conveniently synthesized by hydrolysis and polycondensation of a mixture of tetraethoxysilane (TEOS), n-octyltriethoxysilane (C8-TEOS) and 3-mercaptopropyltrimethoxysilane (MPTMS) via a two-step catalytic sol–gel process. Due to the favorable chemical reactivity of mercapto pendant moieties, the obtained hybrid monolith was oxidized using hydrogen peroxide (30%, w/w) to yield sulfonic acid groups, which provided strong cation-exchange sites. The obtained hybrid monolith was characterized by diffused infrared spectroscopy, elemental analysis, scanning electron microscopy and mercury intrusion porosimetry. The results show that the resulting monolith contains much higher carbon (31.6%) and sulfur (4.8%) contents than traditionally bonded silica materials. The extraction performance of the hybrid monolith was evaluated using sulfonamides as testing analytes by micro-SPE on-line coupled to HPLC. The results show that the hybrid monolith with hydrophobic and strong cation-exchange functional groups exhibits high extraction efficiency towards the testing analytes. The column-to-column RSD values were 1.3–9.8% for the extraction of SAs investigated. The extraction performance of the hybrid silica monolith remained practically unchanged after treated with acid (pH 1.0) and basic solutions (pH 10.5). Finally, the application of the hybrid monolith was demonstrated by micro-SPE of sulfonamide residues from milk followed by HPLC–UV analysis. The limits of detection (S/N = 3) for eight SAs were found to be 1.0–3.0 ng/mL in milk. The recoveries of eight SAs spiked in milk sample ranged from 80.2% to 115.6%, with relative standard deviations less than 11.8%.  相似文献

Hydrophilic C18 monolithic polymer sorbents were synthesized for use in solid phase extraction (SPE) and in capillary liquid chromatography (LC). The approach involved incorporating both hydrophobic and hydrophilic monomers into a monolithic material, by copolymerization of stearyl methacrylate (SMA), poly(ethylene glycol) methyl ether methacrylate (PEGMEMA) and ethylene dimethacrylate (EDMA) in the presence of selected porogens, to produce translucent mesoporous monolithic materials in bulk (SPE) or white macroporous monoliths inside fused silica capillary columns (capillary LC). A capillary column containing one of the hydrophilic C18 monoliths (i.e. poly(SMA-co-PEGMEMA-co-EDMA) with 15% (w/w) PEGMEMA) demonstrated nearly 35% reduction in retention of polycyclic aromatic compounds and greater than 40% increase in retention of phenols compared to a hydrophobic C18 monolithic column. In addition, the hydrophilic monolith demonstrated significantly improved resolution of phenols. Similar monolithic materials prepared in bulk were ground and sieved to obtain 45-65 μm particles with desired rigidity for SPE. To achieve optimum extraction performance for phenols, several parameters, including sample pH and volume, and eluent type and volume, were investigated. Under optimized experimental conditions, the method demonstrated good sensitivity (1.6 ng/mL LOD) and linearity (R(2)>0.97 for 10-200 ng/mL). Again, incorporation of 15% (w/w) PEGMEMA in the monolith increased the extraction efficiency of phenols in water from approximately 20% to 67-92% compared to a hydrophobic C18 monolithic material. Increased wettability of the sorbent by the aqueous sample matrix and the presence of hydrogen-bonding interactions are responsible for the improved retention of polar compounds.  相似文献

A novel mercaptotetrazole‐silica hybrid monolithic column was prepared for capillary liquid chromatography, in which the thiol‐end mercaptotetrazole was mixed with hydrolyzed γ‐methacryloxypropyltrimethoxysilane and tetramethyloxysilane for the co‐polycondensation and thiol‐ene click reaction in a one‐pot process. The effects of the molar ratio of silanes, the amount of mercaptotetrazole, and the volume of porogen on the morphology, permeability and pore properties of the as‐prepared mercaptotetrazole‐silica hybrid monoliths were investigated in detail. A series of test compounds including alkylbenzenes, amides and anilines were employed for evaluating the retention behaviors of the mercaptotetrazole‐silica hybrid monolithic columns. The results demonstrated that the mercaptotetrazole‐silica hybrid monoliths exhibited hydrophobic, hydrophilic as well as ion‐exchange interaction. The run‐to‐run, column‐to‐column and batch‐to‐batch reproducibilities of the mercaptotetrazole‐silica hybrid monoliths were satisfactory with the relative standard deviations less than 1.4 (= 5), 3.9 (= 3) and 4.0% (= 5), respectively. In addition, the mercaptotetrazole‐silica hybrid monolith was further applied to the separation of sulfonamides, nucleobases and protein tryptic digests. These successful applications confirmed the promising potential of the mercaptotetrazole‐silica hybrid monolith in the separation of complex samples.  相似文献

利用点击反应对含叠氮基的溶胶-凝胶整体柱进行了表面修饰,制备了C6-硅胶杂化整体萃取柱。实验以多环芳烃为分析对象,考察了制备和修饰条件对萃取效率的影响,在优化的条件下,新制备的整体柱对萘、菲、芘和苯并[a]芘的萃取富集倍数分别达到95.9、114.2、103.2和57.8。萃取实验的日内和日间精密度(RSD)分别小于5.5%(n=8)和7.3%(n=10)。建立的管内固相微萃取-高效液相色谱检测16种常见多环芳烃方法的检出限(S/N=3)达到0.08~3.72 μg/L,定量限(S/N=10)达到0.26~12.40 μg/L。土壤中多环芳烃分析的加标回收率为82.4%~110.6%, RSD为2.6%~7.9%(n=3)。与美国国家环境保护局检测土壤中多环芳烃的方法比较,结果一致,准确性高。实验表明,该方法萃取效率高,灵敏可靠,操作简便,重现性好,可满足土壤等样品中痕量多环芳烃检测的要求。  相似文献

A simple “one‐pot” approach for the preparation of a new vinyl‐functionalized organic–inorganic hybrid monolithic column is described. In this improved method, the hydrolyzed alkoxysilanes of tetramethoxysilane and triethoxyvinylsilane were used as precursors for the synthesis of a silica‐based monolith, while 1‐hexadecene and sodium ethylenesulfonate were used as vinyl functional monomers along with azobisisobutyronitrile as an initiator. The effects of reaction temperature, urea content, and composition of organic monomers on the column properties (e.g. morphology, mechanical stability, and chromatographic performance) were investigated. The monolithic column was used for the separation of neutral solutes by reversed‐phase pressurized capillary. Furthermore, the monolith can separate various aromatic amines, which indicated its excellent cation‐exchange capability and hydrophobic interactions. The baseline separation of the aromatic amines was obtained with a column efficiency of up to 78 000 plates/m.  相似文献

Analytical scale silica monoliths are commercially limited to three column selectivities (bare silica, C8 and C18). An in situ modification is reported in detail to overcome this barrier and allow for any functionality of choice to be bonded to the silica surface of the monolithic stationary phase support. The modification method was conducted on a commercial bare silica column to bond the C18 moiety to the silica surface through a silylation reaction. The C18 type of stationary phase was chosen, as this is the most commonly bonded functionality for the majority of stationary phases used for high-performance liquid chromatography (HPLC) separations. The C18-modified monolith’s performance was compared to a commercial C18 monolithic and a particle packed column of the same analytical scale column dimensions (100 × 4.6 mm). The modified C18 monolith proved to be of high quality with an efficiency of 73,267 N m?1, fast analysis times (operated at flow rates up to 3 mL min?1 using a conventional 400 bar HPLC system) and improved resolution of a set of polar and non-polar substituted aromatics in comparison to a commercial C18 monolith.  相似文献

The synthesis and chromatographic behavior of an analytical size mixed‐mode bonded silica monolith was investigated. The monolith was functionalized by an in situ modification process of a bare silica rod with chloro(3‐cyanopropyl)dimethyl silane and chlorodimethyl propyl phenyl silane solutions. These ligands were selected in order to combine both resonance and nonresonance π‐type bonding within a single separation environment. Selectivity studies were undertaken using n‐alkyl benzenes and polycyclic aromatic hydrocarbons in aqueous methanol and acetonitrile mobile phases to assess the methylene and aromatic selectivities of the column. The results fit with the linear solvent strength theory suggesting excellent selectivity of the column was achieved. Comparison studies were performed on monolithic columns that were functionalized separately with cyano and phenyl ligands, suggesting highly conjugated molecules were able to successfully exploit both of the π‐type selectivities afforded by the two different ligands on the mixed‐mode column.  相似文献

Reproducible fabrication of the hierarchically porous monolithic silica in a large volume exceeding 1000 mL has been established. By the hydrothermal enlargement of the fully accessible small pores to exceed 50 nm in diameter, the capillary force emerged on solvent evaporation was dramatically reduced, which allowed the preparation of crack‐free monoliths with evaporative solvent removal under an ambient pressure. The local temperature inhomogeneity within a reaction vessel in a large volume was precisely controlled to cancel the heat evolved by the hydrolysis reaction of tetramethoxysilane and that consumed to melt ice cubes dispersed in the solution, resulting in large monolithic silica pieces with improved structural homogeneity. Homogeneity of the pore structure was confirmed, both on macro‐ and mesoscales, using SEM, mercury intrusion, and nitrogen adsorption/desorption measurements. Furthermore, the deviations in chromatographic performance were examined by evaluating multiple smaller monolithic columns prepared from the monolithic silica pieces cut from different parts of a large monolith. All the daughter columns thus prepared exhibited comparable performances to each other to prove the overall homogeneity of the mother monolith. Preliminary results on high‐speed separation of peptides and proteins by the octadecylsilylated silica monolith of the above production have also been demonstrated.  相似文献

We here report a new type of stationary phase for microcolumns. C18 modified silica monolith particles were prepared by grinding and sieving the silica monolith followed by C18 modification and end-capping, and were used as packing material. Ground silica monolith particles were not spherical but irregular with some residual monolithic network structure. The separation efficiency of the stationary phase made of sieved monolith particles (5-10 microm) was better than that of the stationary phase made of unsieved particles. The microcolumn packed with the sieved C18 ground monolith particles (5-10 microm) showed quite good separation efficiency (height equivalent to theoretical plate, HETP, as low as 15 microm) and it was even superior to the microcolumn packed with a commercial spherical 5 microm C18 stationary phase. The column pressure drop of C18 monolith particles was about two-third of that of the commercial spherical C18 phase. The preparation method of C18 stationary phase with ground and sieved silica monolith particles presumably suggests advantages of simplicity and convenience in modification and washing procedures compared to bulk silica monolith. It also showed both improved separation efficiency and low back pressure.  相似文献

This article describes the synthesis, chromatographic characterization, and performance evaluation of analytical (100 x 4.6 mm id) and semipreparative (100 x 10 mm id) monolithic silica columns with mixed-mode RP/weak anion-exchange (RP/WAX) surface modification. The monolithic RP/WAX columns were obtained by immobilization of N-(10-undecenoyl)-3-aminoquinuclidine onto thiol-modified monolithic silica columns (Chromolith) by a radical addition reaction. Their chromatographic characterization by Engelhardt and Tanaka tests revealed slightly lower hydrophobic selectivities than C-8 phases, as well as higher polarity and also improved shape selectivity than RP-18e silica rods. The surface modification enabled separation by both RP and anion-exchange chromatography principles, and thus showed complementary selectivities to the RP-18e monoliths. The mixed-mode monoliths have been tested for the separation of peptides and turned out to be particularly useful for hydrophilic acidic peptides, which are usually insufficiently retained on RP-18e monolithic columns. Compared to a corresponding particulate RP/WAX column (5 microm, 10 nm pore diameter), the analytical RP/WAX monolith caused lower system pressure drops and showed, as expected, higher efficiency (e.g. by a factor of about 2.5 lower C-term for a tetrapeptide). The upscaling from the analytical to semipreparative column dimension was also successful.  相似文献

A phenylboronic acid-silica hybrid monolithic column for capillary liquid chromatography (cLC) was prepared through one-pot process by using 4-vinylphenylboronic acid (VPBA) and alkoxysilanes simultaneously. The effects of the molar ratio of tetramethyloxysilane/γ-methacryloxypropyltrimethoxysilane (TMOS/γ-MAPS), amount of VPBA, and the volume of diethylene glycol (DEG) on the morphologies, permeabilities and pore properties of the prepared VPBA-silica hybrid monolithic columns were studied in detail. A relatively uniform monolithic structure with high porosity was obtained with optimized ingredients. A series of cis-diol-containing compounds, alkylbenzenes, amides, and anilines were utilized to evaluate the retention behaviors of the VPBA-silica hybrid monolithic column. The result demonstrated that the prepared VPBA-silica hybrid monolithic column exhibited multiple interactions including hydrophobicity, hydrophilicity, as well as cation exchange apart from the expected affinity interaction. The run-to-run, column-to-column and batch-to-batch reproducibility of the VPBA-silica hybrid monolith were satisfactory with the relative standard deviations (RSDs) less than 1.63% (n = 5), 2.02% (n = 3) and 2.90% (n = 5), respectively, indicating the effectiveness and practicability of the proposed method. In addition, the VPBA-silica hybrid monolithic column was further applied to the separation of proteins and tryptic digest of bovine serum albumin (BSA), respectively. The successful applications suggested the potential of the VPBA-silica hybrid monolith in proteome analysis.  相似文献

A 10 cm silica monolith has been modified with iminodiacetic acid (IDA) groups and characterised for its selectivity toward alkali, alkaline earth, and selected transition metal cations. Physical characterisation of the modified monolith found non-homogeneous modification along the length of the monolith, although sufficient capacity was achieved to facilitate significant retention of alkaline earth and transition/heavy metal ions over a range of eluent pH and ionic strength conditions. For alkaline earth and transition/heavy metal ions, selectivity of the 10 cm IDA monolith closely matched that seen with a 25 cm IDA modified silica gel particle packed column, although the separation of alkali metal ions was noticeably poorer on the monolithic column. Peak efficiencies for most metal ions were of a similar order for both column types, except for Zn(II), which showed significant peak broadening on the IDA monolithic column.  相似文献

A novel type of macroporous molecularly imprinted hybrid silica monolithic column was first developed for recognition of proteins. The macroporous silica-based monolithic skeleton was synthesized in a 4.6 mm i.d. stainless steel column by a mild sol–gel process with methyltrimethoxysilane (MTMS) as a sole precursor, and then vinyl groups were introduced onto the surface of the silica skeleton by chemical modification of γ-methacryloxypropyltrimethoxysilane (γ-MAPS). Subsequently, the molecularly imprinted polymer (MIP) coating was copolymerized and anchored onto the surface of the silica monolith. Bovine serum albumin (BSA) and lysozyme (Lyz), which differ greatly in molecular size, isoelectric point, and charge, were representatively selected for imprinted templates to evaluate recognition property of the hybrid silica-based MIP monolith. Some important factors, such as template–monomer molar ratio, total monomer concentration and crosslinking density, were systematically investigated. Under the optimum conditions, the obtained hybrid silica-based MIP monolith showed higher binding affinity for template than its corresponding non-imprinted (NIP) monolith. The imprinted factor (IF) for BSA and Lyz reached 9.07 and 6.52, respectively. Moreover, the hybrid silica-based MIP monolith displayed favorable binding characteristics for template over competitive protein. Compared with the imprinted silica beads for stationary phase and in situ organic polymer-based hydrogel MIP monolith, the hybrid silica MIP monolith exhibited higher recognition, stability and lifetime.  相似文献
