We are proposing for the first time the successful immobilization of DNA at glassy carbon electrodes (GCE) modified with carbon nanotubes (CNT) dispersed in glucose oxidase (GOx) (GCE/CNT‐GOx) either by direct adsorption or by layer‐by‐layer self‐assembling using polydiallyldimethylamine (PDDA). The presence of GOx allows an efficient dispersion of CNT and gives a most favorable environment that promotes the adsorption and makes possible a more sensitive electrooxidation of DNA. The PDDA incorporated in the self‐assembled architecture largely facilitates the adsorption and electrooxidation of dsDNA and the adsorbed layer can be successfully used for evaluating the interaction of DNA with methylene blue.  相似文献

The negatively charged (at pH 8.2) glucose oxidase (GOx, pI ca. 4.2) was assembled onto the surface of single-walled carbon nanotubes (SWNT), which was covered (or wrapped) by a layer of positively charged polyelectrolyte poly(dimethyldiallylammonium chloride) (PDDA), via the electrostatic interaction forming GOx-PDDASWNT nanocomposites. Fourier transform infrared (FTIR), UV-Vis and electrochemical impedance spectroscopy (EIS) were used to characterize the growth processes of the nanocomposites. The results indicated that GOx retained its native secondary conformational structure after it was immobilized on the surface of PDDA-SWNT. A biosensor (Nafion-GOx-PDDA-SWNT/GC) was developed by immobilization of GOx-PDDA-SWNT nanocomposites on the surface of glassy carbon (GC) electrode using Nafion (5%) as a binder. The biosensor showed the electrocatalytic activity toward the oxidation of glucose under the presence of ferrocene monocarboxylic acid (FcM) as an electroactive mediator with a good stability, reproducibility and higher biological affinity. Under an optimal condition, the biosensor could be used to detection of glucose, presenting a typical characteristic of Michaelis-Menten kinetics with the apparent Michaelis-Menten constant of KM^app ca. 4.5 mmol/L, with a linear range of the concentration of glucose from 0.5 to 5.5 mmol/L (with correlation coefficient of 0.999) and the detection limit of ca. 83 μmol/L (at a signal-to-noise ratio of 3). Thus the biosensor was useful in sensing the glucose concentration in serum since the normal glucose concentration in blood serum was around 4.6 mmol/L. The facile procedure of immobilizing GOx used in present work would promote the developments of electrochemical research for enzymes (proteins), biosensors, biofuel cells and other bioelectrochemical devices.  相似文献

We report a flow injection amperometric choline biosensor based on the electrostatic assembly of the choline oxidase (ChO) enzyme and a bienzyme of ChO and horseradish peroxidase (HRP) onto multi-wall carbon nanotubes (MWCNT) modified glassy carbon (GC) electrodes. These choline biosensors were fabricated by immobilization of enzymes on the negatively charged MWCNT surface through alternately assembling a cationic poly(diallydimethylammonium chloride) (PDDA) layer and an enzyme layer. Using this layer-by-layer assembling approach, a bioactive nanocomposite film of PDDA/ChO/PDDA/HRP/PDDA/CNT (ChO/HRP/CNT) and PDDA/ChO/PDDA/CNT (ChO/CNT) was fabricated on the GC surface. Owing to the electrocatalytic effect of carbon nanotubes, the measurement of faradic responses resulting from enzymatic reactions has been realized at low potential with acceptable sensitivity. The ChO/HRP/CNT biosensor is more sensitive than the ChO/CNT one. Experimental parameters affecting the sensitivity of biosensors, e.g., applied potential, flow rate, etc., were optimized and potential interference was examined. The response time for this choline biosensor is fast (few seconds). The linear range of detection for the choline biosensor is from 5.0 x 10(-5) to 5.0 x 10(-3) M and the detection limit is about 1.0 x 10(-5) M.  相似文献

In this paper, a novel biosensor was prepared by immobilizing glucose oxidase (GOx) on carbon nanotube-gold-titania nanocomposites (CNT/Au/TiO2) modified glassy carbon electrode (GCE). SEM was initially used to investigate the surface morphology of CNT/Au/TiO2 nanocomposites modified GCE, indicating the formation of the nano-porous structure which could readily facilitate the attachment of GOx on the electrode surface. Cyclic voltammogram (CV) and electrochemical impedance spectrum (EIS) were further utilized to explore relevant electrochemical activity on CNT]Au/TiO2 nanocomposites modified GCE. The observations demonstrated that the immobilized GOx could efficiently execute its bioelectrocatalytic activity for the oxidation of glucose. The biosensor exhibited a wider linearity range from 0.1 mmol L-1 to 8 mmol L^-1 glucose with a detection limit of 0.077 mmol L^- 1.  相似文献

The interaction between multiwalled carbon nanotubes (MWCNTs) and aqueous poly(diallyl dimethylammonium) chloride (PDDA) was studied by X-ray photoelectron (XPS) and photoacoustic Fourier transform infrared (PA-FTIR) spectroscopies. We have found that the mild sonication of MWCNTs in aqueous PDDA results in a significant improvement of CNT dispersibility and greatly enhances their adhesion to Au and Si substrates. The MWCNT-PDDA interaction is due to the presence of an unsaturated contaminant in the PDDA chain, as confirmed by both XPS and PA-FTIR, which enters into a pi-pi interaction with the CNTs. Electrostatic group repulsions of the coated CNTs then provide the dispersibility and adhesion.  相似文献

An electrochemical DNA sensing film was constructed based on the multilayers comprising of poly‐L ‐lysine (pLys) and Au‐carbon nanotube (Au‐CNT) hybrid. A precursor film of mercaptopropionic acid (MPA) was firstly self‐assembled on the Au electrode surface. pLys and Au‐CNT hybrid layer‐by‐layer assembly films were fabricated by alternately immersing the MPA‐modified electrode into the pLys solution and Au‐CNT hybrid solution. Cyclic voltammetry was used to monitor the consecutive growth of the multilayer films by utilizing [Fe(CN)6]3?/4? and [Co(phen)3]3+/2+ as the redox indicators. The outer layer of the multilayer film was the positively charged pLys, on which the DNA probe was easily linked due to the strong electrostatic affinity. The hybridization detection of DNA was accomplished by using methylene blue (MB) as the indicator, which possesses different affinities to dsDNA and ssDNA. Differential pulse voltammetry was employed to record the signal response of MB and determine the amount of the target DNA sequence. The established biosensor has high sensitivity, a relatively wide linear range from 1.0×10?10 mol/L to 1.0×10?6 mol/L and the ability to discriminate the fully complementary target DNA from single or double base‐mismatched DNA. The sequence‐specific DNA related to phosphinothricin acetyltransferase gene from the transgenically modified plants was successfully detected.  相似文献

Sodium alginate (AlgNa) and poly(diallyldimethylammonium chloride) (PDDA) were mixed to obtain an interpenetrating polymer composite via electrostatic interaction and then cast on an Au electrode surface, followed by incorporation of metal ions (e.g. Fe3+ or Ca2+, to form AlgFe or AlgCa hydrogel) and glucose oxidase (GOx) (or lactate oxidase (LOx)), to prepare amperometric enzyme electrodes. The interactions of PDDA, Alg, and Fe3+ are studied by visual inspection as well as microscopic and electrochemical methods. Under optimized conditions, the PDDA-AlgFe-enzyme/Au and PDDA-AlgCa-enzyme/Au electrodes can give good analytical performance (e.g. nM-scale limit of detection of glucose or lactate, and sensitivities > 50 μA cm−2 mM−1) in the first-generation biosensing mode, which are better than the reported analogs using typical polysaccharide biopolymers as enzyme-immobilization matrices. The enzyme electrodes also worked well in the second-generation biosensing mode in the coexistence of p-benzoquione or ferrocene monocarboxylic acid artificial mediator. Biofuel cells (BFCs) with the enzyme electrodes as the bioanodes and glucose (or lactate) as the biofuel were also fabricated with satisfactory results. The proposed protocols for preparation of high performance Alg-based biocomposites may find wide applications in bioanalysis.  相似文献

An amperometric glucose biosensor is developed that is based on immobilization of glucose oxidase (GOD) in a composite film of poly(o-aminophenol) (POAP) and carbon nanotubes (CNT), which are electrochemically co-polymerized at a gold (Au) electrode. Because of the high surface per volume ratio and excellent electrical conductivity of CNT, the biosensor based on an Au/POAP/CNT/GOD electrode has lower detection limit (0.01 mM), larger maximum response current (0.24 mA cm(-2)) and higher sensitivity (11.4 mA M(-1) cm(-2)) than the values of the biosensor based on an Au/POAP/GOD electrode. Additionally, the biosensor shows fast response time, large response current, and good anti-interferent ability for ascorbic acid, uric acid and acetaminophen. Good reproducibility and stability of the biosensor are also observed.  相似文献

Acid functionalized multi‐walled carbon nanotubes (f‐MWCNTs) were decorated with Au and Fe2O3 nanoparticles (FeONPs) and deposited on glassy carbon electrode (GCE). The resulting hybrid Au/Fe2O3/f‐MWCNTs/GCE electrode and the one further modified by glucose oxidase were compared for detection of glucose. FeONPs and Au were deposited on the f‐MWCNTs by sonication‐assisted precipitation and deposition‐precipitation methods, respectively. The morphology and structure of the samples were characterized by transmission electron microscopy, scanning electron microscopy, X‐ray diffraction and Raman spectroscopy. A uniform distribution of FeONPs with an average size of 5 nm increased the surface area of functionalized nanotubes from 39 to 50 m2/g. The electrocatalytic glucose detection on the modified electrodes was evaluated using cyclic voltammetry and chronoamperometry in 0.1 M phosphate buffer solution at pH 7.0. The non‐enzymatic and enzymatic electrodes show sensitivity of 512.4 and 921.4 mA/mM.cm2 and detection limit of 1.7 and 0.9 mM, respectively. The enzymatic and enzymeless electrodes retained more than 70 % and 80 % of their cathodic faradic current after 70 days, respectively. The sensing mechanism of the non‐enzymatic biosensor is described through the reaction of glucose with iron (III) ions, while in the case of enzymatic electrode, glucose is oxidized by glucose oxidase.  相似文献

Titanium dioxide nanoparticle/gold nanoparticle/carbon nanotube (TiO2/Au/CNT) nanocomposites were synthesized, and then characterized using scanning electron microscopy (SEM), transmission electron microscopy (TEM), and energy dispersive X-ray spectroscopy (EDX). A TiO2/Au/CNT nanocomposite-modified glassy carbon (GC) electrode was prepared using the drop coating method and was investigated using electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV), differential pulse voltammetry (DPV), and amperometric current–time response (I-T). The modified material is redox-active. The nonenzymatically detected amount of ascorbic acid (AA) on the TiO2/Au/CNT electrode showed a linear relationship with the AA concentration, for concentrations from 0.01 to 0.08 μM; the sensitivity was 117,776.36 μA?·?cm?2?·?(mM)?1, and the detection limit was 0.01 μM (S/N?=?3). The results indicated that the TiO2/Au/CNT nanocomposite-modified GC electrode exhibited high electrocatalytic activity toward AA. This paper describes materials consisting of a network of TiO2, Au, and MWCNTs, and the investigation of their synergistic effects in the detection of AA.  相似文献

A photoelectrochemical immunosensor based on multi‐electrode array was developed for simultaneous and sensitive determination of veterinary drug residues. In this system, poly(dimethyldiallylammonium chloride) (PDDA), Au nanoparticles (Au NPs) and thioglycolic acid (TGA)‐capped CdS quantum dots (QDs) were layer‐by‐layer assembled onto the home‐made Au electrode array. The assembling process of the (CdS/PDDA/Au NPs/PDDA)n multilayer was characterized by electrochemical impedance spectroscopy. And then the antibodies for clenbuterol (CB), ractopamine (RAC) and chloramphenicol (CAP) were covalently immobilized onto the Au electrode array by 1‐ethyl‐3‐(3‐dimethylaminopropyl) carbodiimide (EDC) coupling reaction, respectively. The concentrations of CB, RAC and CAP were measured based on the photoelectrochemical effects of CdS QDs. Under the optimal conditions, the limits of detection (LOD) for CB, RAC and CAP were 25, 50 and 2.2 pg/mL (3Δ), respectively, with acceptable recovery over the range of 95.40%–105.5% in pig liver samples. All results indicate that the immunosensor array system has potential application for practical, effective and high throughput analysis of veterinary drugs residues.  相似文献

A promising electrochemical sensor based nickel‐carbon nanotube (Ni‐CNT) modified on glassy carbon (GC) electrode had been developed and the properties of the modified electrode were characterized by multispectroscopic analysis. The fabricated sensor (GC/Ni‐CNT) electrode was utilized to determine the catecholamines such as epinephrine and dopamine simultaneously. Differential pulse voltammetry and amperometry were used to verify the electrochemical behavior of the studied compounds. The GC/Ni‐CNT based amperometric sensor showed a wide linear range and low detection limit with high analytical sensitivity of 8.31 and 6.61 μA μM?1 for EP and DA, respectively which demonstrates better characteristics compared to other electrodes reported in the literature. Further, no significant change in amperometric current response was observed in presence of biological interference species such as glucose, cysteine, citric acid, uric acid and ascorbic acid in the detection of EP and DA. The utility of this GC/Ni‐CNT electrode was well established for the determination of EP and DA in human urine samples.  相似文献

An electrochemical noise (ECN) device was utilized for the first time to study and characterize a glucose/O2 membraneless biofuel cell (BFC) and a monopolar glucose BFC. In the glucose/O2 membraneless BFC, ferrocene (Fc) and glucose oxidase (GOD) were immobilized on a multiwalled carbon nanotubes (MWCNTs)/Au electrode with a gelatin film at the anode; and laccase (Lac) and an electron mediator, 2,2′‐azinobis (3‐ethylbenzothiazoline‐6‐sulfonate) diammonium salt (ABTS), were immobilized on a MWCNTs/Au electrode with polypyrrole at the cathode. This BFC was performed in a stirred acetate buffer solution (pH 5.0) containing 40 mmol/L glucose in air, with a maximum power density of 8 μW/cm2, an open‐circuit cell voltage of 0.29 V, and a short‐circuit current density of 85 μA/cm2, respectively. The cell current at the load of 100 kΩ retained 78.9% of the initial value after continuous discharging for 15 h in a stirred acetate buffer solution (pH 5.0) containing 40 mmol/L glucose in air. The performance decrease of the BFC resulted mainly from the leakage of the ABTS mediator immobilized at the cathode, as revealed by the two‐channel quartz crystal microbalance technique. In addition, a monopolar glucose BFC was performed with the same anode as that in the glucose/O2 membraneless BFC in a stirred phosphate buffer solution (pH 7.0) containing 40 mmol/L glucose, and a carbon cathode in Nafion‐membrane‐isolated acidic KMnO4, with a maximum power density of 115 μW/cm2, an open‐circuit cell voltage of 1.24 V, and a short‐circuit current density of 202 μA/cm2, respectively, which are superior to those of the glucose/O2 membraneless BFC. A modification of the anode with MWCNTs for the monopolar glucose BFC increased the maximum power density by a factor of 1.8. The ECN device is highly recommended as a convenient, real‐time and sensitive technique for BFC studies.  相似文献

Two carbon nanotube supported palladium catalysts were prepared using a chemical reduction technique (Pd/CR‐CNT) and a conventional impregnation method (Pd/CNT) respectively, and their catalytic performances for Heck reaction were investigated. The catalysts were characterized by TEM and XPS techniques and the products were characterized by 1H NMR. Research results showed that the (Pd/CR‐CNT) catalyst showed a better catalytic activity than the (Pd/CNT) catalyst, owing to better dispersion of palladium nanoparticles and stronger interaction between the active palladium species and carbon nanotube. Meanwhile, the product yield maintained 99.93% of its initial value at five‐times re‐use, compared with that at the first time use. The catalyst prepared with the chemical reduction method represented a better reusing performance.  相似文献

Abstract

In this report, we describe the finding that bamboo‐structured carbon nanotubes (CNT) showed superior electrocatalytic activity toward hydrogen peroxide. The slope of the calibration curve for hydrogen peroxide obtained with the bamboo‐structured CNT paste electrode was more than 20 times as large as the slopes obtained with hollow‐structured CNT paste and glassy carbon electrodes at an operating potential of ?0.1 V, with no interfering reactions. Incorporation of glucose oxidase within the bamboo‐structured CNT paste electrode allows the selective detection of glucose in the presence of common interferents without using any permselective membranes. This excellent ability of the bamboo‐structured CNT paste electrode toward hydrogen peroxide is applicable to the development of other enzymatic biosensors.  相似文献

Multiwalled carbon nanotube (CNT) modified glassy carbon electrode immobilized with horseradish peroxidase (HRP) in Nafion coating showed direct electron transfer between HRP enzyme and the CNT‐modified electrode. A mediator‐free bienzyme glucose biosensor based on horseradish peroxidase and glucose oxidase was constructed. The bienzyme biosensor exhibited a high sensitivity for glucose detection at zero applied potential.  相似文献

By using carbon nanotubes (CNTs) as a shape template and glucose as a carbon precursor and structure‐directing agent, CNT@Fe3O4@C porous core/sheath coaxial nanocables have been synthesized by a simple one‐pot hydrothermal process. Neither a surfactant/ligand nor a CNT pretreatment is needed in the synthetic process. A possible growth mechanism governing the formation of this nanostructure is discussed. When used as an anode material of lithium‐ion batteries, the CNT@Fe3O4@C nanocables show significantly enhanced cycling performance, high rate capability, and high Coulombic efficiency compared with pure Fe2O3 particles and Fe3O4/CNT composites. The CNT@Fe3O4@C nanocables deliver a reversible capacity of 1290 mA h g?1 after 80 cycles at a current density of 200 mA g?1, and maintain a reversible capacity of 690 mA h g?1 after 200 cycles at a current density of 2000 mA g?1. The improved lithium storage behavior can be attributed to the synergistic effect of the high electronic conductivity support and the inner CNT/outer carbon buffering matrix.  相似文献

The reagentless and oxygen‐independent biosensors for ethanol were developed based on the covalent immobilization of alcohol dehydrogenase (ADH) and its cofactor nicotinamide adenine dinucleotide (NAD+) on chitosan (CHIT) chains. The CHIT‐NAD+‐ADH structures were adsorbed onto carbon nanotubes (CNT) in order to provide a signal transduction based on the recycling of redox states of NAD cofactor at CNT (detection limit, 8–30 µM ethanol; dynamic range up to 20 mM). The CHIT‐NAD+‐dehydrogenase/CNT hybrid material represents a general approach to the development of dehydrogenases‐based electrochemical biosensors. Interestingly, the CHIT‐NAD+ solutions preserved their enzymatic activity even after five years of storage at 4 °C.  相似文献

The electrochemical quartz crystal impedance (EQCI) technique has been applied to investigate glucose oxidation on bare and Ni(OH)2‐modified Au electrodes in 0.2 mol L?1 KOH aqueous solution. The EQCI responses suggest different contributions of H+‐release and OH?‐incorporation reactions of the Ni(OH)2‐film redox process in 0.2 mol L?1 aqueous KOH at different potentials. Glucose adsorption on the Ni(OH)2‐modified Au electrode was studied. A mechanism for potential cyclic redox process of glucose at Ni(OH)2‐modified Au electrode is suggested, mainly based on a comparative EQCI analysis with direct glucose oxidation on bare gold and glucose ad‐/desorption on Ni(OH)2 film.  相似文献

Poly(diallydimethylammonium chloride) functionalized graphene/double‐walled carbon nanotube composite (PDDA‐GN/DWCNT) was synthesized by a facile microwave‐assisted reduction reaction. PDDA‐GN/DWCNT exhibits enhanced electrocatalytic activity towards nitrite oxidation. The direct electrooxidation behaviors of nitrite on PDDA‐GN/DWCNT were investigated by cyclic voltammetry, amperometric i‐t curve, and rotating disk electrode experiments. Under the optimum conditions, PDDA‐GN/DWCNT was applied to the quantitative analysis of nitrite with a wide linear range covering 0.07–3000 µM and a low detection limit of 0.03 µM (S/N=3). Moreover, PDDA‐GN/DWCNT exhibits satisfied results in the determination of nitrite that released from breast cancer cells.  相似文献
