The liposidomycins comprise a family of complex nucleoside antibiotics that inhibit bacterial peptidoglycan synthesis. Their structures (1, 2) feature nucleoside, ribofuranoside, diazepanone, and lipid regions. Several stereogenic centers remain unassigned, including three within the diazepanone region: C-6', C-2'", and C-3'". An intramolecular reductive amination reaction has been used to prepare model diazepanones. Analysis of 40 and two of its diastereomers by NMR spectroscopy, X-ray crystallography, and molecular modeling indicates a close relative configurational and conformational match between 40 and the liposidomycin diazepanone degradation product 43 and allows the assignment of stereochemistry of the natural products as either [C-6'(R), C-2'"(R), C-3'"(R)] or [C-6'(S), C-2'"(S), C-3'"(S)].  相似文献

(29)Si-(13)C spin-spin couplings over one, two, and three bonds as well as other NMR parameters [delta((29)Si), delta((13)C), delta((1)H), (1)J((13)C-(1)H), and (2)J((29)Si-C-(1)H)] were calculated and measured for a series of trimethylsilylated alcohols of the types Me(3)Si-O-(CH(2))(n)CH(3) and Me(3)Si-O-CH(3-n)R(n)(n = 0-3; R = Me, Ph, or Vi). The signs of the coupling constants determined for selected compounds can likely be extended to all such compounds, as supported by theoretical calculations. Similar to couplings between other pairs of nuclei, the 2-bond and 3-bond (29)Si-O-(13)C couplings are of opposite signs ((2)J > 0 and (3)J < 0), and their relative magnitudes depend on the extent of branching at the alpha-carbon.  相似文献

A new 8.O.6'-neolignan, threo-(7R,?8R)-7-acetoxy-3',4'-dimethoxy-3,4-dimethoxy-Δ-(8')-8.O.6'-neolignan (1) along with two known lignans (2) and (3), was isolated from the leaves and stems of Taxillus theifer. Structural elucidation was carried out by 1-D and 2-D-NMR spectroscopic methods, and the absolute configurations of C-7 and C-8 in 1 were determined on the basis of circular dichroism. Compound 2 is threo-7-acetoxy-3'-methoxy-3,4-dimethoxy-Δ-(7')-8.O.4'-neolignan obtained from a natural source for the first time, previously derived from a synthesis study of 8,4'-oxyneolignans.  相似文献

Three new lignan glycosides (1-3) were isolated from the stems of Akebia trifoliata. Their structures were elucidated as (7R,8R,7'R,8'R)3,3',5,5'tetramethoxy-4,4'dihydroxy-7,9':7',9-diepoxylignan-4-O-beta-D-glucopyranoside (1), (7S,8S,8'R)-4,4',9-trihydroxy-3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (2), (7R,8R,8'S)-4,4',9-trihydroxy3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (3) by spectral analyses, primarily NMR, MS and CD. The NMR assignments for the compounds were carried out using 1H, 13C, DEPT, COSY, HSQC, HMBC and ROESY NMR experiments.  相似文献

Two new steroids, named 4α,22-dimethyl-Cholest-22-en-3β-ol (1) and 4α-methyl-Cholest-7,22-dien-3β-ol (2), along with two known steroids, were isolated from the soft coral Sinularia brassica. The structures of the new compounds were determined on the basis of extensive spectroscopic data (MS, (1)H and (13)C NMR, (1)H-(1)H COSY, (13)C-(1)H COSY, HMBC and NOESY) analysis.  相似文献

Intramolecular dipolar cycloaddition of bicyclic münchnones, 5H,7H-thiazolo[3,4-c]oxazol-4-ium-1-olates, derived from cyclodehydration of 2-substituted-N-acylthiazolidine-4-carboxylic acids are reported. A range of new pyrrolo[1,2-c]thiazole derivatives (7, 14, 15, 20, 23, and 26) were obtained as single enantiomers from 2-phenylthiazolidines, 2-benzoylthiazolidines, and 2-methylthiazolidine-4-carboxylates. Pyrrolo[1,2-c][1,4]thiazine derivative 27 was also obtained from pyrrolo[1,2-c]thiazole derivative 26. The structures of methyl (2R,4R)-2-(p-methoxybenzoyl)thiazolidine-4-carboxylate (17a), methyl (2R,4R)-2-(p-methoxybenzoyl)-N-(prop-2-ynyloxyacetyl)thiazolidine- 4-carboxylate (18), and 3-oxo-4-phenyl-3,4,6,8-tetrahydro-1H-furo[3',4':2,3]pyrrolo[1,2-c][1,4]thiazine (27) were determined by X-ray crystallography. Chirooptical studies of the pyrrolo[1,2-c]thiazoles were done by confirming the absolute configuration at the chiral center C-3.  相似文献

The application of mass spectrometry in drug discovery, especially in drug metabolites, is very important. This present paper is at first focused on the elucidation of fragmentation patterns of the phenolic bisbenzyltetrahydroisoquinoline alkaloid, neferine, together with its analogues isoliensinine and liensinine with anti-HIV activities using electrospray ionization tandem mass spectrometry (ESI-MS/MS) and hydrogen/deuterium (H/D) exchange. All title compounds displayed major diagnostic fragments that formed by the cleavage of the C1'--C9' bond resulting in positive group CD, and the loss of 4-ethyl-1-phenol or 4-ethyl-1-methoxybenzene following rearrangements. Their ESI-MS/MS spectra also showed the relatively stable fragment ions formed by the elimination of H2O, CH3NH2, CH3OH, and CH3-N==CH2. Secondly, the metabolites of neferine from dog hepatic microsomal incubations were analyzed and characterized by high-performance liquid chromatography (HPLC) and data-dependent ESI-MS/MS. Based on fragmentation patterns and compared with their retention times in LC, molecular weights and ultraviolet (UV) absorbances with standard compounds, six metabolites were identified as isoliensinine, liensinine and four novel bisbenzyltetrahydroisoquinoline alkaloids named as 6-O-desmethylneferine, 2'-N-desmethylneferine, 2'-N-6-O-didesmethylneferine, and 6,13-O-didesmethylneferine. All metabolites were desmethyl or didesmethyl products of neferine. The possible metabolic pathways for neferine have been proposed. The results suggest that N-demethylation and O-demethylation are two important metabolic pathways of neferine in dog hepatic microsomal incubations. This is critical for screening and development of phenolic bisbenzyltetrahydroisoquinoline alkaloids with anti-HIV activities such as neferine and its analogues isoliensinine and liensinine.  相似文献

Leucanthoside A, a new allose-containing triterpenoid saponin (1), was isolated from the aerial parts of Cephalaria leucantha L. Its structure was determined by electrospray ionization mass spectrometry and NMR spectroscopy. Complete assignments of the 1H and 13C NMR chemical shifts were achieved by two-dimensional NMR experiments: DQF-COSY, NOESY, TOCSY, HSQC, DINE-HSQC, HMBC, 13C-1H 2D-J-resolved spectroscopy, and 1,1-ADEQUATE.  相似文献

The complete assignments of the (1)H and (13)C NMR spectra of the hallucinogenic neoclerodane diterpenoid salvinorin A were determined in three different NMR solvents using HSQC, HMBC and COSY. Solvent systems are described that allow the resolution of all (1)H signals. Virtual coupling was observed for the protons at C-2, C-3 and C-4 in the 600 MHz (1)H spectrum in CDCl(3). The complete assignments of the (1)H and (13)C NMR spectra of salvinorin B are also reported.  相似文献

We show that (13)C-(1)H dipolar couplings in fully protonated organic solids can be measured by applying a Symmetry-based Resonance-Echo DOuble-Resonance (S-REDOR) experiment at ultra-fast Magic-Angle Spinning (MAS). The (13)C-(1)H dipolar couplings are recovered by using the R12 recoupling scheme, while the interference of (1)H-(1)H dipolar couplings are suppressed by the symmetry properties of this sequence and the use of high MAS frequency (65 kHz). The R12 method is especially advantageous for large (13)C-(1)H dipolar interactions, since the dipolar recoupling time can be incremented by steps as short as one rotor period. This allows a fine sampling for the rising part of the dipolar dephasing curve. We demonstrate experimentally that one-bond (13)C-(1)H dipolar coupling in the order of 22 kHz can be accurately determined. Furthermore, the proposed method allows a rapid evaluation of the dipolar coupling by fitting the S-REDOR dipolar dephasing curve with an analytical expression.  相似文献

The enantioselective synthesis of the previously proposed structure of malyngamide U (1) was accomplished in 18 steps from (S)-(+)-carvone. The key steps involved a hydroxymethylation of (S)-(+)-carvone and an asymmetric Henry reaction of aldehyde (+)-5, as well as condensation with the acid 3. The 1H and 13C NMR data of the synthetic compound 1 were not consistent with the data of the reported malyngamide U. The C-2' epimer of compound 1 was therefore synthesized by a similar reaction sequence. While the NMR data of C-2' epimer 23 were in full agreement with those of the reported product, the discrepancy in the specific rotation data suggested the correct structure of malyngamide U should be structure 2, in which the absolute configuration of the amine part was enantiomeric with that in compound 23. Then the correct absolute configuration of revised malyngamide U (2) was confirmed by the similar synthesis from (R)-(-)-carvone.  相似文献

[structure: see text] 2-Ureido-4-[1H]-pyrimidinones have been reported to dimerize via quadruple hydrogen bonding systems with dimerization constants >10(6) M(-1) in CDCl3. The dimerization constant, K(dim), is dependent on the solvent as well as the ring-substituents present, where previously alkyl (e.g., R1 = Me) and aromatic moieties (e.g., R1 = p-NO2C6H4, R1 = C6H2(OC13H27)3) have been incorporated at the C-6 position. To assess the influence of alternative, functionalizable, electron-donating groups on the dimerization motif and tautomeric distribution of isomers, the synthesis of compounds possessing aminophenyl functionality at the C-6 position has been achieved. NMR spectroscopy chemical shift analysis revealed that compound 2 (R1 = p-NH2C6H4, R2 = C6H13) existed as the 2-ureido-4-pyrimidinol dimeric DADA array in DMSO-d6, where a dimerization constant of 46 M(-1) was determined. This is the first time that a ureidopyrimidinone quadruple hydrogen bonding DADA array has been observed in pure DMSO, a highly polar solvent. The azo-derivative 5 of compound 2 was prepared which also adopted the pyrimidin-4-ol form in DMSO-d6. Compounds 7, 10 and 11 were then synthesized containing a more hydrophilic PEG unit in the lateral chain and the tautomeric distributions were determined.  相似文献

Methyl ester of abscisic acid (ABA), a plant hormone, gives a dehydrated ion at m/z 260 in electron ionization mass spectrometry (EI-MS). This dehydrated ion had been considered to be derived only from the elimination of the tertiary hydroxyl group at C-1'. We found that 34% of the dehydrated ion was formed by elimination of the oxygen atom at the 4'-carbonyl group, and the remaining 66% by elimination of the 1'-hydroxyl group. This unusual elimination of the carbonyl oxygen was shown with [4'-(18)O]ABA methyl ester. Involvement of the 4'-carbonyl oxygen in dehydration was observed in methyl ester of phaseic acid (PA), a natural metabolite of ABA, but not in 1'-deoxy-ABA methyl ester or isophorone. This suggested that the 1'-hydroxyl group was necessary for the elimination of the 4'-carbonyl oxygen. ABA methyl esters labeled with stable isotopes showed that hydrogen atoms at the 1'-hydroxyl group and at C-4 or -5 or -3' or - 5' or -7' were eliminated with the 4'-carbonyl oxygen. These results allow us to propose a formation mechanism of the dehydrated ion derived from the elimination of 4'-carbonyl oxygen and hydrogen atoms at C-4 and 1'-oxygen in ABA methyl ester as follows: first, ionization at the 1'-hydroxyl group occurs to give an ion radical, and the proton at the 1'-oxygen migrates to the 4'-carbonyl oxygen after the bond fission between C-1'-C-6'; second, migration of the proton at C-4 to the 1'-oxygen is followed by migration of the protons at C-5 and C-7' to C-4 and C-5, respectively; finally, the proton at the 1'-oxygen migrates to the 4'-hydroxyl group, and H(2)O at C-4' is eliminated to give the dehydrated ion. Our findings point out that a dehydrated ion is not always derived from the elimination of a hydroxyl group.  相似文献

At a glance: The stereochemical configuration of the diglycerophosphate backbone of the endosome-specific lipid bis(monoacylglycero)phosphate (BMP, see picture) was determined by (1)H?NMR spectroscopy. Enantiomeric discrimination was facilitated by introduction of D-camphor ketals as chiral shift reagents, and enantiopure synthetic BMP analogues were prepared as reference materials. Two new prenylated isoflavones, named flemiphilippinins E and F, were isolated from the roots of Flemingia philippinensis, together with six known ones. The structures of the above compounds were established by spectroscopic methods. Flemiphilippinins E and F were identified as 5,4'-dihydroxy-8-(3,3-dimethylallyl)-6',6'-dimethyl-5'-(1,1-dimethylallyl) pyrano(2',3':7,6)isoflavone and 5,4'-dihydroxy-4',4',5'-trimethyl-4',5'-dihydrofurano(2',3':7,8) isoflavone, respectively, by a combination of HR-EI-MS, (1)H NMR, (13)C NMR, HMQC, HMBC, and NOESY spectra.  相似文献
