A simple and sensitive high-performance liquid chromatography (HPLC) method is proposed for the analysis of some carboxylic acids in food samples and the environment. The use of 9-chloromethyl anthracene as a fluorescence-labeling reagent has been investigated. The derivatization reagent reacts with unitary carboxylic acids and tetrabutylammonium bromide as a catalyst within 50 min in acetonitrile to give esters, which can be separated by HPLC employing fluorescence detection at λ(ex) = 365 and λ(em) = 410 nm. The optimum conditions for derivatization, fluorescence detection and chromatographic separation are established. The method shows good sensitivity, with a detection limit from 0.18 to 2.53 pmol, and good linearity between 1-250 nmol/mL of each analyte. The practical applicability of the method was demonstrated by analyzing samples that were spiked with the acid standards, environment and food samples.  相似文献

Summary A simple and rapid derivatization method for C24-C6 fatty acids on HPTLC RP-18 phases is described. The prechromatographic reaction with monodansylpiperazine and monodansylcadaverine takes place at the start. Linear calibration curves are obtained after chromatography (e.g. dansyl palmityl piperazide: r=0.9998), which can be employed for quantitative analysis in the lower nanogram range (determination limit <10 ng). All 10 fatty acids can be separated excellently (R usually 1.5). Stepwise and gradient developments are employed (AMD system).This is the first time that in situ derivatization techniques have been employed for fatty acids leading to fluorescent products in the picomole range and a gradient method has been described which also leads to precise separations on RP phases.

A high-performance liquid chromatographic method for the analysis of the fatty acid composition of human serum lipids with fluorescence detection was examined. Both free and total fatty acids extracted from serum were derivatized with 9-anthryldiazomethane and were analysed using methanol-water (94.7:5.3) as mobile phase. Twelve kinds of fatty acid were detected, both in the free and total fatty acids, and were well separated. Concentrations of individual fatty acids of serum lipids were estimated from an internal standard, heptadecanoic acid. The results correlated well with those from two other quantitative analyses. These results indicate that the high-performance liquid chromatographic analysis of fatty acids is a reliable method for determining individual fatty acids of human serum lipids. The compositions of free fatty acids and total fatty acids of serum lipids were analysed and compared in 27 normal subjects, 27 diabetics, and 20 angina pectoris patients by this method.  相似文献

A new in-capillary derivatization method with naphtalene-2,3-dicarboxyaldehyde (NDA)/CN(-) has been developed for capillary electrophoresis with laser-induced fluorescence detection of brain microdialysate amino acids. Samples are sandwiched between two plugs of reagent mixture at the capillary inlet and subsequently separated. Highest derivatization yields are obtained by using a reagent to sample plug length ratio equal to 4, performing a first electrophoretic mixing followed by a zero potential amplification step before applying the separation voltage and using a NaCN to NDA concentration ratio equal to 1. This new single-step methodology allows the analysis of amino acid neurotransmitters in rat brain microdialysis samples.  相似文献

利用新型荧光试剂1,2 苯并 3,4 二氢咔唑 9 乙基对甲苯磺酸酯(BDETS)对19种游离脂肪酸(FFAs)进行柱前衍生,在EclipseXDB C8反相色谱柱上,采用梯度洗脱优化分离.90℃下在DMF溶剂中以K2CO3作催化剂,衍生反应30min获得稳定的荧光产物.激发和发射波长分别为λex=333nm,λem=390nm,采用大气压化学电离源(APCI)正离子模式进行柱后在线质谱定性.多数脂肪酸的线性回归系数大于0.9989,检测限为24.80~80.37fmol.实现了人体血清中长链脂肪酸的定性及相应含量测定.  相似文献

A highly sensitive and simple reversed phase high performance liquid chromatographic (HPLC) method for the quantitative determination of free fatty acids in human serum is presented. The method is based on the direct derivatization of serum fatty acids with 6,7-dimethoxy-1-methyl-2(1H)-quinoxalinone-3-propionylcarboxylic acid hydrazide. The derivatization reaction proceeds in aqueous solution in the presence of pyridine and 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide at 37 degrees C. The resulting derivatives are separated within 75 min on a reversed phase column (YMC Pack C8) with a gradient elution of aqueous acetonitrile and detected fluorimetrically. The detection limits are 2.5-5 fmol in a 10 microL injection volume. The sensitivity permits precise determination of free fatty acids in 5 microL serum. The method is simple and is without the conventional liquid-liquid extraction steps of serum fatty acids.  相似文献

The derivatization regents for carboxylic acids, DAABD-AE (4-[2-(N,N-dimethylamino)ethylaminosulfonyl]7-(2-aminoethylamino)-2,1,3-benzoxadiazole), MePZBD-AE ([4-(4-N-methyl)piperazinosulfonyl]-7-(2-aminoethylamino)-2,1,3-benzoxadiazole) and APZBD-NHMe ([4-(4-N-aminoethyl)piperazinosulfonyl]-7-methylamino-2,1,3-benzoxadiazole) were developed for electrospray ionization-mass spectrometry (ESI-MS). The derivatization reaction with fatty acids was completed at 60 degrees C within 30 min. The derivatives of fatty acids were separated on a reversed-phase column and detected with ESI-MS. The detection limits attained for fatty acids were femtomol range and the calibration curves were linear over the range from 0.1 to 100 pmol (r2 > 0.992) for DAABD-AE and MePZBD-AE. DAABD-NHMe was applied to the analysis of fatty acids in rat plasma samples.  相似文献

A study attempted to characterize natural waxes used in pictorial works of art was carried out by means of gas chromatography. The analytical treatment requires prior hydrolysis of the waxes to release the fatty acids (FA) (myristic (myr), palmitic (pal), oleic (ole), stearic (ste), araquidic (ara), behenic (beh), lignoceric (lig), cerotic (cer)) from the main esters of the waxes. The formation of volatile derivatives of the fatty acids was carried out by derivatization with ethyl choroformate (ECF). This derivatization reagent was chosen due to the speed, safety and quantitativity of the reaction. The analyzed hydrocarbons were n-eicosane, n-heneicosane, n-docosane, n-tricosane, n-tetracosane, n-pentacosane, n-hexacosane, n-heptacosane, n-octacosane, n-nonacosane, n-tricontane n-hentriacontane, n-dotriacontane, n-tritriacontane, n-tetratriacontane, n-pentatriacontane, main constituents of the waxes. No derivatization is needed to analyze the hydrocarbons. Ethyl ester derivatives and hydrocarbons are adequately separated by gas chromatography, identified by flame ionization detection and confirmed by mass spectrometry. To characterize natural waxes, peak area ratios of each fatty acids with respect to the palmitic acid and peak area ratios of each hydrocarbons with respect to n-heptacosane were calculated. The proposed method provides a good characterization of different waxes most frequently used in artworks, such as beeswax, carnauba wax and ceresin, and has been successfully applied to real samples. This is the first report on the application of ECF to the analysis of fatty acids in wax.  相似文献

The development of a new derivative, the dimethylaminoethyl ester, for the analysis of fatty acids by electrospray tandem mass spectrometry is described. Qualitative and quantitative analyses of long to very long chain fatty acids in plasma, blood, urine and wax were performed. Branched chain, unsaturated, dicarboxylic, hydroxy, amino and keto acids were studied. The quantitative analysis method using the new derivative is simple, rapid and precise with small sample size. A HPLC-fluorescence method for identification of drying oils from binding media or protective film used in pictorial works of art prior to conservation or restoration is proposed. Fluorescence derivatization of fatty acids released by hydrolysis of structural drying oils is studied. The derivatization reagent was 4-(bromomethyl)-7-methoxycoumarin with 18-crown-6 as catalyst. Mobile phase was programmed from methanol-water (90:10 v/v) to methanol-water (100:0 v/v) in 25 min. The excitation and emission wavelengths were 325 and 395 nm, respectively. Under these chromatographic conditions, coumarin derivatives of myristic, palmitic, oleic and stearic acids were satisfactorily resolved. The method shows good sensitivity, with a detection limit of 6.0 x 10(-8) mmol, and good linearity between 1.0 x 10(-7) and 1.8 x 10(-4) mmol of each analyte. Peak area ratios among fatty acids derivatives, especially the stearic acid/palmitic acid peak area ratio, are useful to identify the drying oils. The proposed method has been successfully applied to artistic samples from items of the cultural heritage of Valencia (Spain).  相似文献

以2-(2-(10-蒽基)-萘[2,3-d]咪唑)-乙基-对甲苯磺酸酯(ANITS)作为柱前衍生化试剂,在Eclipse XDB-C8色谱柱上,梯度洗脱实现了20种游离脂肪酸(FFA)衍生物的完全基线分离。90℃下在DMF溶剂中以K2CO3作催化剂,选取衍生试剂摩尔数为脂肪酸的7倍,衍生反应40min可获得稳定的荧光产物。激发和发射波长分别为250nm和512nm。采用大气压化学电离源(APCI)正离子模式,实现了油菜蜂花粉中游离脂肪酸的质谱鉴定。所有脂肪酸的线性相关系数均大于0.9999,检出限为24.76~98.79fmol。  相似文献

The organic basic solution, 1 M tetramethylammonium hydroxide (TMAH) in methanol, was employed for the hydrolysis of extremely small amounts of lipids compared to the classical inorganic basic solution, 1 M KOH in ethanol. The hydrolysed fatty acids were derivatized as N(O)-tert.-butyldimethylsilyl (tBDMSi) esters with N-methyl-N-(tert.-butyldimethylsilyl) trifluoroacetamide (MTBSTFA) and compared with the classical derivatives, the methyl esters, made by the BF3-methanol method. Recoveries of fatty acids determined on the standard fatty acids and soybean oil hydrolysed with TMAH were high: about 1.1-2.1- and 2.0-5.4-times, respectively, in all fatty acids compared with the hydrolysis by KOH regardless of derivatization method. The relative standard deviations (RSDs) on the recoveries of standard fatty acids were less than 5% when hydrolysed with TMAH, regardless of derivatives, but when hydrolysed with KOH, RSDs were more than 5% for most fatty acids, especially for long-chain fatty acids. The RSDs on the recoveries of fatty acids on the soybean oil were also very high in the KOH hydrolysis. Fatty acid compositions of soybean oil were similar in the main fatty acids regardless of hydrolysis methods, but showed slightly different values, depending on the methods of derivatization. RSDs were also very high in the KOH hydrolysis. In view of these results, precision of analysis by KOH hydrolysis was very poor, so we could not rely on the data. On the other hand, the reliability of data by TMAH hydrolysis method was very high, so it is a useful new hydrolysis method for extremely small amounts of lipid samples. Both derivatives of 35 standard fatty acids were successfully separated on a HP-1 nonpolar capillary column. tBDMSi derivatives were completely resolved in 70 min by 295 degrees C. In the methyl ester derivatives it took about 80 min to get satisfying resolution, but these derivatives were completely resolved by 250 degrees C. The sensitivity of tBDMSi derivatives was about 1.5-6.3-times higher than that with methyl ester derivatives. The stability of tBDMSi derivatives was constant for about 144 h except arachidic, docosahexanoic, behenic and heneicosanoic acids, which were stable for only 86 h.  相似文献

This study describes an application of on-line preconcentration by large-volume stacking in combination with in-capillary derivatization for enhancing spectrophotometric detection sensitivity in capillary electrophoresis. The method is illustrated by an example dealing with the determination of amino acids with 1,2-naphthoquinone-4-sulfonate as a labelling agent. Samples are dissolved in water in order to create a stacking process based on differences in the conductivity between this medium and a concentrated running buffer. The in-capillary derivatization is accomplished following a sandwich procedure in which the sample is inserted between two segments of reagent. Amino acid derivatives are obtained and separated in a fused-silica capillary with a sodium borate electrolyte buffer using 2-propanol as an organic modifier. The method is applied to the analysis of amino acids in pharmaceutical and feed samples. A good concordance between the predicted values and those obtained with the standard method is observed, with overall quantification error below 5%. The proposed procedure allows the detection limits sensitivity to be enhanced in 1000-fold with respect to conventional precapillary derivatization.  相似文献

合成了新的荧光衍生试剂1-[2-(对甲苯磺酸酯)乙基]-2-苯基咪唑[4,5-f]9,10-菲(TSEPIP),并将其作为柱前衍生化试剂,在Eclipse XDB-C8色谱柱上采用梯度洗脱实现了11种长链(C20~C30)游离脂肪酸(FFA)衍生物的基线分离。利用柱后在线的串联质谱并以大气压化学电离源(APCI)的正离子模式实现了各组分的质谱定性。对土壤及3种苔藓(东亚毛灰藓、锦丝藓、羽平藓)中FFA组分的定量结果表明,苔藓植物从土壤中富集了大量的长链游离脂肪酸。荧光检测的激发波长和发射波长分别为260 nm和380 nm。线性回归系数大于0.9996,检测限为26.19~76.67 fmol。所建立的方法具有良好的重现性,对实际样品的测定结果令人满意。  相似文献

The use of reversed-phase liquid chromatography (LC) with pre-column derivatization for the analysis of amino acid mixtures is becoming established as a possible cheaper alternative to commercial amino acid analysers. The available derivatization procedures all have disadvantages when applied to clinical samples, partly due to the interferences found with body fluids when ultraviolet or fluorescence detection is used. An LC method is described for the separation of amino acids in blood or urine, using pre-column derivatization with phenylisothiocyanate (PITC), gradient elution and electrochemical detection. The use of electrochemical detection of PITC derivatives virtually eliminates interferences and enables the secondary amino acids to be measured. Examples are shown of normal urine and plasma and samples from patients with cystinuria and maple syrup urine disease.  相似文献

A hydrophilic fluorescent derivatization reagent for fatty acids, 4-N-(4-N-aminoethyl)piperazino-7-nitro-2,1,3-benzoxadiazole (NBD-PZ-NH(2)), was designed and synthesized. NBD-PZ-NH(2) possesses not only a fluorophore and a reacting group but also a positive charge group and, thus, was hydrophilic and suitable for application to capillary electrophoresis. NBD-PZ-NH(2) reacted with fatty acids in the presence of triphenylphosphine (TPP) and 2,2'-dipyridyl disulfide (DPDS) at room temperature within 10 min. The derivatives were strongly fluoresced and were positively charged at pH below 3. The derivatives of C4-C20 fatty acids were separated within 10 min in 50% acetonitrile in water containing 30 mM ammonium acetate and 1.0 M acetic acid by capillary electrophoresis with laser-induced fluorescence (CE-LIF) detection. The detection limits attained were 6.5 nM (signal-to-noise ratio of 3). It is proposed that NBD-PZ-NH(2) is a prominent derivatization reagent for fatty acids which is suitable for CE-LIF application.  相似文献

A new high-performance liquid chromatographic method is described for the determination of fatty acids in seed oils. The method was based on precolumn derivatization with 9-fluorenylmethyl chloroformate as a labeling agent and fluorescence detection. Fatty acids were extracted from the samples and subjected to derivatization with the reagent at 60°C for 10?min. The chromatographic separation of 14 fatty acids (C10–C22) was achieved on a combined loading compression octadecyl sulfate (CLC-ODS) column with a run time of 30?min. Three-step gradient elution of a mobile phase consisted of acetonitrile and water was used, and the signal was monitored at excitation and emission wavelengths of 265 and 315?nm, respectively. The method indicated favorable sensitivity and reproducibility for fatty acids’ derivatives. The detection limits, at a signal-to-noise ratio of 3, were 0.01–0.05?µg/ml and relative standard deviations (RSDs) were less than 0.27%. Excellent linear responses were observed with coefficients of 0.9995. This method was applied to quantify fatty acids in white, brown, and black sesame seeds’ oil.  相似文献
