Ordered assembly of collagen molecules on flat substrates has potential for various applications and serves as a model system for studying the assembly process. While previous studies demonstrated self-assembly of collagen on muscovite mica into highly ordered layers, the mechanism by which different conditions affect the resulting morphology remains to be elucidated. Using atomic force microscopy, we follow the assembly of collagen on muscovite mica at a concentration lower than the critical fibrillogenesis concentration in bulk. Initially, individual collagen molecules adsorb to mica and subsequently nucleate into fibrils possessing the 67 nm D-periodic bands. Emergence of fibrils aligned in parallel despite large interfibril distances agrees with an alignment mechanism guided by the underlying mica. The epitaxial growth was further confirmed by the formation of novel triangular networks of collagen fibrils on phlogopite mica, whose surface lattice is known to have a hexagonal symmetry, whereas the more widely used muscovite does not. Comparing collagen assembly on the two types of mica at different potassium concentrations revealed that potassium binds to the negatively charged mica surface and neutralizes it, thereby reducing the binding affinity of collagen and enhancing surface diffusion. These results suggest that collagen assembly on mica follows the surface adsorption, diffusion, nucleation, and growth pathway, where the growth direction is determined at the nucleation step. Comparison with other molecules that assemble similarly on mica supports generality of the proposed assembly mechanism, the knowledge of which will be useful for controlling the resulting surface morphologies.  相似文献

Supramolecular nanotube hosts with precisely controlled inner or outer diameters have been synthesized by self-assembly of unsymmetrical bolaamphiphilic monomers or glucopyranosylamide lipids, respectively. Time-resolved fluorescent measurement using 8-anilinonaphthalene-1-sulfonate (ANS) as a probe revealed that the water confined in a cardanyl-β-D -glucopyranoside lipid nanotube has relatively lower solvent polarity corresponding to that of propanol than bulk water. Extensively developed hydrogen bond networks also characterize the confined water in comparison to the case in bulk water. Encapsulation ability of the glucopyranosylamide lipid nanotube has been examined by filling the lyophilized LNTs with gold or silver nanoparticles, ferritin, or magnetic crystals. Filling the unsymmetrical bolaamphiphile nanotube possessing positively charged inner surfaces with negatively charged polymer beads or ferritin proved to be successful without depending on capillary action. © 2006 Wiley Periodicals, Inc. The adsorption and assembly of B18 peptide on various solid surfaces were studied by reflectometry techniques and atomic force microscopy. B18 is the minimal membrane binding and fusogenic motif of the sea urchin protein bindin, which mediates the fertilization process. Silicon substrates were modified to obtain hydrophilic charged surfaces (oxide layer and polyelectrolyte multilayers) and hydrophobic surfaces (octadecyltrichlorosilane). B18 does not adsorb on hydrophilic positively charged surfaces, which was attributed to electrostatic repulsion since the peptide is positively charged. In contrast, the peptide irreversibly adsorbs on negatively charged hydrophilic as well as on hydrophobic surfaces. B18 showed higher affinity for hydrophobic surfaces than for hydrophilic negatively charged surfaces, which must be due to the presence of hydrophobic side chains at both ends of the molecule. Atomic force microscopy provided the indication that lateral diffusion on the surface affects the adsorption process of B18 on hydrophobic surfaces. The adsorption of the peptide on negatively charged surfaces was characterized by the formation of globular clusters.  相似文献

We present a novel and simple method to fabricate two-dimensional (2D) poly(styrene sulfate) (PSS, negatively charged) colloidal crystals on a positively charged substrate. Our strategy contains two separate steps: one is the three-dimensional (3D) assembly of PSS particles in ethanol, and the other is electrostatic adsorption in water. First, 3D assembly in ethanol phase eliminates electrostatic attractions between colloids and the substrate. As a result, high-quality colloidal crystals are easily generated, for electrostatic attractions are unfavorable for the movement of colloidal particles during convective self-assembly. Subsequently, top layers of colloidal spheres are washed away in the water phase, whereas well-packed PSS colloids that are in contact with the substrate are tightly linked due to electrostatic interactions, resulting in the formation of ordered arrays of 2D colloidal spheres. Cycling these processes leads to the layer-by-layer assembly of 3D colloidal crystals with controllable layers. In addition, this strategy can be extended to the fabrication of patterned 2D colloidal crystals on patterned polyelectrolyte surfaces, not only on planar substrates but also on nonplanar substrates. This straightforward method may open up new possibilities for practical use of colloidal crystals of excellent quality, various patterns, and controllable fashions.  相似文献

The supramolecular dimerization of tetraanionic corannulene is utilized as a self-assembly motif for the construction of highly charged, buckybowl-based supramolecular oligomers. Oligomers of up to 7 units (3900 g mol(-1)) of reduced dicorannulenic monomers are identified and characterized by various NMR techniques, and the reversibility of their assembly is established through the concentration dependence of their molecular weight and the effect of monofunctional chain-stoppers.  相似文献

Aggrecan is a bottlebrush shaped macromolecule found in the extracellular matrix of cartilage. The negatively charged glycosaminoglycan (GAG) chains attached to its protein backbone give aggrecan molecules a high charge density, which is essential for exerting high osmotic swelling pressure and resisting compression under external load. In solution, aggrecan assemblies are insensitive to the presence of calcium ions, and show distinct osmotic pressure versus concentration regimes. The aim of this study is to investigate the effect of ionic environment on the structure of aggrecan molecules adsorbed onto well‐controlled mica surfaces. The conformation of the aggrecan was visualized using Atomic Force Microscopy. On positively charged APS mica the GAG chains of the aggrecan molecules are distinguishable, and their average dimensions are practically unaffected by the presence of salt ions. With increasing aggrecan concentration they form clusters, and at higher concentrations they form a continuous monolayer of conforming molecules. On negatively charged mica, the extent of aggrecan adsorption varies with salt composition. Understanding aggrecan adsorption onto a charged surface provides insight into its interactions with bone and implant surfaces in the biological milieu. © 2010 Wiley Periodicals, Inc. In this work, surface-enhanced Raman scattering (SERS) was applied to probe the orientation of cytochrome c (Cyt-c) on gold nanohole arrays functionalized with self-assembled monolayers (SAMs) of alkane thiols with positively (-NH2) and negatively (-COOH) charged terminal groups. Square grid gold nanohole arrays with a nanohole diameter of 270 nm and a grating of 350 nm were fabricated by electron beam lithography (EBL) and were used as the SERS substrates. The SERS intensities of the nontotally symmetric mode (B(1g) mode nu(11)) and the totally symmetric mode (A(1g) mode nu(4)) and their ratios were used to determine the orientation of Cyt-c on surfaces. The results indicate that the heme group is close and perpendicular to the negatively charged surface but is far from and oriented at an angle to the positively charged surface. Cyt-c has a random or more flat orientation on the bare Au nanoholes surface.  相似文献

Colloid particle deposition was applied to characterize fibrinogen (Fb) monolayers on mica, which were produced by controlled adsorption under diffusion transport. By adjusting the time of adsorption and the bulk Fb concentration, monolayers of desired surface concentration were obtained. The surface concentration of Fb was determined directly by AFM enumeration of single molecules adsorbed over the substrate surface. It was proven that Fb adsorbed irreversibly on mica both at pH 3.5 and at pH 7.4 with the rate governed by bulk transport. The electrokinetic properties of Fb monolayers produced in this way were studied using the streaming potential method. The dependence of the apparent zeta potential of Fb monolayers was determined as a function of the coverage. It was shown that for pH 3.5 the initial negative zeta potential of the mica substrate was converted to positive for Fb coverage exceeding 0.16. On the other hand, for pH 7.4, the zeta potential of a Fb-covered mica remained negative for the entire coverage range. The charge distribution in Fb monolayers was additionally studied using the colloid deposition method, in which negatively and positively charged polystyrene latex particles (ca. 800 nm in diameter) were used. An anomalous deposition of negative latex particles on substrates exhibiting a negative zeta potential was observed. Results of these experiments were quantitatively interpreted in terms of the fluctuation theory assuming that adsorption sites consisted of two and three Fb molecules, for pH 3.5 and 7.4, respectively. These results suggested that for pH 7.4, the distribution of charge on Fb molecules was heterogeneous, characterized by the presence of positive patches, whereas the average zeta potential was negative, equal to -19 mV. The utility of the colloid deposition method for studying Fb monolayers was further demonstrated in deposition experiments involving positive latex particles. It was shown that for a rather broad range of fibrinogen coverage, both the positive and the negative latex particles can adsorb on surfaces covered by Fb, which behaved, therefore, as superadsorbing surfaces. It was also concluded that the colloid deposition method can be used to determine the Fb bulk concentration for the range inaccessible for other methods.  相似文献

Surfaces coated with poly(ethylene oxide) containing nonionic polymers are of interest in medical applications due to, among other things, the low adsorption of proteins on such surfaces. In this paper we have studied the interfacial properties of surfaces coated with PEO by measuring the forces acting between two such surfaces in water and across a protein solution as well as between one such surface and a surface carrying adsorbed proteins. One type of surface coating was a graft copolymer of poly(ethylene imine) and poly(ethylene oxide) where the cationic poly(ethylene imine) group anchored the polymer to negatively charged mica surfaces. Three different ways to prepare this coating was used and compared. It was found that this coating was not stable in the presence of lysozyme, a small positively charged protein, when the PEO graft density was low. The other type of coating was obtained by adsorbing ethyl(hydroxyethyl)-cellulose onto hydrophobised mica surfaces. The driving force for adsorption is in this case the hydrophobic interaction between nonpolar segments of the polymer and the surface. The EHEC coating was stable in the presence of lysozyme and the interactions between adsorbed layers of lysozyme and EHEC coated surfaces are purely repulsive due to long-range steric forces.  相似文献

Heme protein hemoglobin (Hb) or myoglobin (Mb) and silica nanoparticles in a variety of charge states were assembled layer-by-layer into films on solid surfaces to investigate the driving forces for film assembly. Cyclic voltammetry (CV), quartz crystal microbalance (QCM), X-ray photoelectron spectroscopy (XPS), and UV-vis and reflectance absorption infrared (RAIR) spectroscopy were used to characterize the different [SiO2/protein]n films. Even when the proteins and silica were both negatively charged, stable layer-by-layer [SiO2/protein]n films were successfully fabricated, although amounts of protein were smaller than when nanoparticles and proteins had opposite charges. Results suggest the importance of localized Coulombic attractions between the negative nanoparticle surface and positively charged amino acid residues on the Mb or Hb surfaces in the assembly and for the stability of [SiO2/protein]n films.  相似文献

We have examined the adsorption of DNA-wrapped single-walled carbon nanotubes (DNA-SWNTs) on hydrophobic, hydrophilic, and charged surfaces of alkylthiol self-assembled monolayers (SAMs) on gold. Our goal is to understand how DNA-SWNTs interact with surfaces of varying chemical functionality. These samples were characterized using reflection absorption FTIR (RAIRS), X-ray photoelectron spectroscopy (XPS), and Raman spectroscopy. We have found that DNA-SWNTs preferentially adsorb to positively charged amine-terminated SAMs and to bare gold surfaces versus hydrophobic methyl-terminated or negatively charged carboxylic acid-terminated SAMs. Examination of the adsorption on gold of single-strand DNA (ssDNA) of the same sequence used to wrap the SWNTs suggests that the DNA wrapping plays a role in the adsorption behavior of DNA-SWNTs.  相似文献

Electrostatic assembly of cationic nanoparticles onto the negatively charged backbone of double-stranded DNA has been shown to produce one-dimensional chains with potential use as nanoelectronic components. In this paper, micron long DNA templates stretched on aminosilane- and hexamethyldisilazane-modified silicon surfaces are used to assemble 3.5 nm gold nanoparticles passivated with cationic thiocholine. Atomic force microscopy is used to analyze the density and defects along the approximately 5 nm high structures, with comparison between positively charged and neutral surfaces. Low background adsorption of nanoparticles is facilitated by both these surface chemistries, while the neutral surface yields a more densely packed assembly.  相似文献

A method to create assembly of polydiacetylene vesicles on solid substrates in a novel fashion is described. The formation of the assembly is based on electrostatic layer-by-layer deposition using negatively charged 10,12-pentacosadiynoic acid (PCDA) vesicles and polyelectrolyte polyethylenimine, or positively charged PCDA-2(')-aminoethylamide (PCDANH2) vesicles. This is an efficient method for preparing the chromatic sensor films of polydiacetylene vesicles after ultraviolet light irradiation. The strategy would be useful in the development of polydiacetylene-based chemosensors and biosensors.  相似文献

The adsorption of a biologically important glycoprotein, mucin, and mucin-chitosan complex layer formation on negatively charged surfaces, silica and mica, have been investigated employing ellipsometry, the interferometric surface apparatus, and atomic force microscopy techniques. Particular attention has been paid to the effect of an anionic surfactant sodium, dodecyl sulfate (SDS), with respect to the stability of the adsorption layers. It has been shown that mucin adsorbs on negatively charged surfaces to form highly hydrated layers. Such mucin layers readily associate with surfactants and are easily removed from the surfaces by rinsing with solutions of SDS at concentrations > or =0.2 cmc (1 cmc SDS in 30 mM NaCl is equal to 3.3 mM). The mucin adsorption layer is negatively charged, and we show how a positively charged polyelectrolyte, chitosan, associates with the preadsorbed mucin to form mucin-chitosan complexes that resist desorption by SDS even at SDS concentrations as high as 1 cmc. Thus, a method of mucin layer protection against removal by surfactants is offered. Further, we show how mucin-chitosan multilayers can be formed.  相似文献

Previously reported studies indicate that aluminum nanostructured substrates can potentially find widespread use in metal-enhanced fluorescence (MEF) applications particularly in the UV or near-UV spectral region toward label-free detection of biomolecules. MEF largely depends on several factors, such as chemical nature, size, shape of the nanostructure and its distance from the fluorophore. A detailed understanding of the MEF and its distance-dependence are important for its potential application in biomedical sensing. Our goal is to utilize intrinsic protein fluorescence for label-free binding assays. This is made possible by the use of metallic nanostructures which provide localized excitation and enhanced fluorescence of UV fluorophores and will also provide a way to separate the surface-bound proteins from the bulk samples. We evaluated varied probe distances from plasmonic nanostructures by the well-established layer-by-layer (LbL) technique. The investigated proteins were adsorbed on different numbers of alternate layers of poly(styrene sulfonate) (PSS) and poly(allylamine hydrochloride) (PAH). Bovine serum albumin (BSA) was electrostatically attached to the positively charged PAH layer, and goat and rabbit IgG were attached to negatively charged PSS layer. We obtained a maximum of a ~ 9 fold increase in fluorescence intensity from BSA at a distance of ~9 nm from the Al nanostructured surface. Approximately 6- and 7- fold increases were observed from goat and rabbit IgG at a distance of ~8 nm, respectively. The minimum lifetimes were about 3-fold shorter than those on bare control quartz slides for all three proteins. The time-resolved intensity decays were analyzed with a lifetime distribution model to understand the distance effect on the metal-fluorophore interaction in detail. The present study indicates the distance dependence nature of metal-enhanced intrinsic fluorescence of proteins and potential of LbL assembly to control the metal-to-fluorophore distance in the UV wavelength region.  相似文献

The self-assembly of surfactant-protected gold nanorods (aspect ratio 3.3 +/- 0.3, 20.6 +/- 5.5 nm width, and 67.5 +/- 9.0 nm length) into ordered structures using adipic acid is presented. As made, the gold nanorods are coated with cationic surfactant, which gives them a net positive charge in aqueous solution. The pH-dependent assembly is directed by electrostatic interactions between the positively charged nanorods and negatively charged, deprotonated adipic acid. Absorption spectra and light scattering measurements of these nanorods suggest that aggregation is initiated in solution in the presence of adipic acid at pH 7-8, but not at pH 3, to form small assemblies of nanorods. Zeta potential measurements show that the assembly is significantly less positively charged in the presence of deprotonated adipic acid than when adipic acid is fully protonated.  相似文献

The adsorption of amyloid beta-peptide at hydrophilic and hydrophobic modified silicon-liquid interfaces was characterized by neutron reflectometry. Distinct polymeric films were used to obtain noncharged (Formvar), negatively (sodium poly(styrene sulfonate)) and positively charged (poly(allylamine hydrochloride)) hydrophilic as well as hydrophobic surfaces (polystyrene and a polysiloxane-dodecanoic acid complex). Amyloid beta-peptide was found to adsorb at positively charged hydrophilic and hydrophobic surfaces, whereas no adsorbed layer was detected on hydrophilic noncharged and negatively charged films. The peptide adsorbed at the positively charged film as patches, which were dispersed on the surface, whereas a uniform layer was observed at hydrophobic surfaces. The thickness of the adsorbed peptide layer was estimated to be approximately 20 A. The peptide formed a tightly packed layer, which did not contain water. These studies provide information about the affinity of the amyloid beta-peptide to different substrates in aqueous solution and suggest that the amyloid fibril formation may be driven by interactions with surfaces.  相似文献

The Monte Carlo method is employed to study the conditions for nanoconductor formation on a DNA molecule in aqueous solution via electrostatic interactions between negatively charged groups of the matrix molecule and positively charged functionalized gold nanoparticles. A model is developed that explicitly takes into account low-molecular-mass counterions resulting from the dissociation of DNA molecule and surface functional groups of nanoparticles. It is shown that the explicit regard to the counterions is of great importance for investigating conditions of nanoaggregate formation. The model parameters are estimated at which continuous metal coatings are formed via self-assembly.  相似文献
