Present work demonstrates the fabrication of new and facile sandwich‐type electrochemical immunosensor based on palladium nanoparticles (PdNPs), polyaniline (PANI) and fullerene‐C60 nanocomposite film modified glassy carbon electrode (PdNP@PANI‐C60/GCE) for ultrasensitive detection of Prostate‐specific antigen (PSA) biomarker. PdNP@PANI‐C60 was electrochemically synthesized on GCE and used as an electroactive substrate. PdNP@PANI‐C60 was characterized by scanning electron microscopy (SEM), energy‐dispersive X‐ray spectroscopy (EDS), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Primary antibody anti‐PSA (Ab1) was covalently immobilized on PdNP@PANI‐C60/GCE using NHS/EDC linkers. In the presence of PSA antigen, horseradish peroxidase secondary antibody (HRP‐Ab2) was brought into the surface of the electrode, developing stable amplified signals of H2O2 reduction. Under the optimal conditions, a linear curve for determination of PSA at the proposed immunosensor was 1.6×10?4 ng.mL?1 to 38 ng.mL?1 with a limit of detection (LOD) of 1.95×10?5 ng.mL?1. The proposed immunosensor was successfully validated in serum and urine samples towards PSA detection with satisfactory and acceptable results.  相似文献

In this paper a novel potentiometric immunosensor for direct and rapid detection of diphtherotoxin (D‐Ag) has been developed by means of self‐assembly of monoclonal diphtheria antibody (D‐Ab) onto a platinum electrode based on nanoparticles mixture (containing gold nanoparticles and silica nanoparticles) and polyvinyl butyral (PVB) as matrixes. At first, D‐Ab was absorbed onto the surface of nanoparticles mixture, and then they were entrapped into polyvinyl butyral sol‐gel network on a platinum electrode. The detection is based on the change in the potentiometric response before and after the antigen‐antibody reaction in a phosphate buffer solution (pH 7.0). The immobilized D‐Ab exhibited direct potentiometric response toward D‐Ag. In comparison to the conventional applied methods, this strategy could allow antibodies immobilized with higher loading amount and better retained immunoactivity, as demonstrated by potentiometric response, cyclic voltammetry and electrochemical impedance spectroscopy of the immunosensor. The immunosensor with nanoparticles mixture exhibited much higher sensitivity, better reproducibility, and long‐term stability than that with gold nanoparticles or silica nanoparticles alone. The linear range was from 5.0×10?3 to 1.2 μg?mL?1 with a detection limit of 1.1×10?3 μg?mL?1. Up to 16 successive assay cycles with retentive sensitivity were achieved for the probes regenerated with in 0.2 mol?L?1 glycine‐hydrochloric acid (Gly‐HCl) buffer solution and 0.25 mol?L?1 NaCl. Moreover, the immunosensor with nanoparticles mixture was applied to evaluate a number of practical specimens with potentiometric results in acceptable agreement with those given by the ELISA method, implying a promising alternative approach for detecting diphtherotoxin in the clinical diagnosis.  相似文献

β‐Cyclodextrin functionalized graphene/Ag nanocomposite (β‐CD/GN/Ag) was prepared via a one‐step microwave treatment of a mixture of graphene oxide and AgNO3. β‐CD/GN/Ag was employed as an enhanced element for the sensitive determination of 4‐nitrophenol. A wide linear response to 4‐nitrophenol in the concentration ranges of 1.0×10?8–1.0×10?7 mol/L, and 1.0×10?7–1.5×10?3 mol/L was achieved, with a low detection limit of 8.9×10?10 mol/L (S/N=3). The mechanism and the heterogeneous electron transfer kinetics of the 4‐nitrophenol reduction were discussed according to the rotating disk electrode experiments. Furthermore, the sensing platform has been applied to the determination of 4‐nitrophenol in real samples.  相似文献

A new Cu(II) ion‐selective PVC membrane sensor based on 6‐methyl‐4‐(1‐phenylmethylidene)amino‐3‐thioxo‐1,2,4‐triazin‐5‐one (MATTO) as an excellent sensing material was developed. The electrode exhibits a Nernstian slope of 29.2±0.4 mV per decade over a very wide concentration range between 1.0×10?1 and 1.0×10?6 M, with a detection limit of 4.8×10?7 M (30.5 ng/mL). The sensor possesses the advantages of short conditioning time, fast response time (<10 s), and especially, very good selectivity towards transition and heavy metal, and some mono, di and trivalent cations. The proposed electrode was successfully applied to the determination of copper in wastewater of copper electroplating samples and as an indicator electrode in potentiometric titration of Cu(II) ions with EDTA.  相似文献

A poly(vinylchloride) (PVC) membrane based potentiometric immunosensor for the direct detection of alpha‐fetoprotein (AFP) has been developed. First, Au colloid particle was chemisorbed upon amino groups of o‐phenylenediamine, which were dissolved in plasticized PVC membrane. Then alpha‐fetoprotein antibody (anti‐AFP) was immobilized upon the surface of the Au colloid particle to prepare a potentiometric AFP immunosensor. The Au colloid particle modified PVC membrane was characterized by digital photo and transmission electron microscope (TEM). The immunosensor exhibited fast potentiometric response (≤4 min) and showed specific response to AFP in the range of 4.9 to 158.5 ng/mL with a correlation coefficient of 0.9971 and a detection limit of 1.6 ng/mL. The factors influencing the performance of the immunosensor were also studied in detail. Moreover, the proposed method is economical and efficient as well as potentially attractive for clinical immunoassays.  相似文献

A sensitive immunosensor for the detection of pregnancy marker, human chorionic gonadotropin hormone (hCG), was developed using the direct electrical detection of Au nanoparticles. We utilized disposable screen‐printed carbon strips (SPCSs) for the development of our immunosensor, which provided cost‐effective tests with the required antigen sample volume as small as 2 μL. After the recognition reaction between the surface‐immobilized primary antibody and hCG, the captured antigen was sandwiched with a secondary antibody that was labeled with Au nanoparticles. Au nanoparticles were exposed to a preoxidation process at 1.2 V for 40 s, which was subsequently followed with a reduction scan on the same surface using differential pulse voltammetry (DPV). We could observe Au nanoparticle‐labeled antigen‐antibody complexes immobilized on the surface of SPCS using scanning electron microscopy (SEM). Additionally, the number of Au nanoparticles on the immunosensor was determined using SEM images, and showed a linear relationship with the current intensity obtained from the DPV measurements with a detection limit of 36 pg/mL hCG (612 fM, 3.6×10?4 IU/mL). Our immunosensor system, a combination of the screen‐printing technology with Au nanoparticles provides a promising biosensor for various applications in life sciences.  相似文献

In this study, a bimetallic nanomaterial‐based electrochemical immunosensor was developed for the detection of carcinoembryonic antigen (CEA) and vascular endothelial growth factor (VEGF) cancer biomarkers at the same time. CEA and VEGF biomarkers are indicators for colon and breast cancers and stomach cancers, respectively. During the study, gold nanoparticle (AuNp), lead nanoparticle (PbNp), copper nanoparticle (CuNp) and magnetic gamma iron(III)oxide (γFe2O3 Np) were synthesized, characterized and used together for the first time in the structure of an electrochemical biosensor based on anti‐CEA and anti‐VEGF. For this purpose, Au SPE based sandwich immunosensor was fabricated by using labeled anti‐CEA (labeled with Pb+2) and labeled anti‐VEGF (labeled with Cu+2). As a result, CEA and VEGF biomarkers were detected following the oxidation peaks of label metals (Pb+2 and Cu+2) by using differential pulse voltammetry. After the experimental parameters were optimized, the linear range was found in the concentration range between 25 ng/mL and 600 ng/mL with the relative standard deviation (RSD) value of (n=3 for 600 ng/mL) 3.33 % and limit of detection (LOD) value of 4.31 ng/mL for CEA biomarker. On the other hand, the linear range was found in the concentration range between 0.2 ng/mL and 12.5 ng/mL with the RSD value of (n=3 for 12.5 ng/mL) 5.31 % and LOD value of 0.014 ng/mL for VEGF biomarker. Lastly, sample application studies for synthetic plasma sample and interference studies with dopamine, ascorbic acid, BSA, cysteine and IgG were carried out.  相似文献

A new sensor, gold‐6‐amino‐2‐mercaptobenzothiazole (6A2MBT), was fabricated via a self‐assembly procedure. Electrochemical properties of the monolayer were investigated by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The modified electrode showed excellent antifouling property against the oxidation products of DA, allowed us to construct a dynamic calibration curve with two linear parts, 1.00×10?6 to 3.72×10?4 and 3.72×10?4 to 6.42×10?4 M DA, with correlation coefficients of 0.997 and 0.992 and a detection limit of 1.57×10?7 M DA by using differential pulse voltammetry (DPV), respectively. Finally, the performance of the Au‐6A2MBT modified electrode was successfully tested for electrochemical detection of DA in a pharmaceutical sample.  相似文献

A signal‐enhanced immunosensor has been developed by self‐assembling Au NPs onto a ferrocene‐branched poly(allylamine)/multiwalled carbon nanotubes (PAA‐Fc/MWNTs) modified electrode for the sensitive determination of hepatitis B surface antigen (HBsAg) as a model protein. The formation of PAA‐Fc/MWNTs composite not only effectively avoided the leakage of Fc and retained its electrochemical activity, but also enhanced the conductivity and charge‐transport properties of the composite. Further adsorption of Au NPs into the PAA matrix provided both the interactive sites for the immobilization of hepatitis B surface antibody (HBsAb) and a favorable microenvironment to maintain its activity. Tests performed with this immunosensor showed a specific response to HBsAg in the range of 0.1–350.0 ng mL?1 with a detection limit of 0.03 ng mL?1.  相似文献

A potentiometric immunosensor for the label‐free detection of bisphenol A (BPA) was developed by covalently immobilizing a polyclonal antibody (PAb) onto the surface of a carboxylated poly(vinyl chloride) (PVC‐COOH) membrane. The immunosensor was characterized using scanning electron microscopy (SEM), X‐ray photoelectron spectroscopy (XPS), and electrochemical impedance spectroscopy. The immobilization of PAb and its interaction with antigen (Ag) and BPA were also confirmed by quartz crystal microbalance (QCM) studies. Experimental parameters affecting the immuno‐interaction between PAb and its Ag or BPA were examined in terms of pH, antibody concentration, and temperature. The immunosensor showed a specific recognition of BPA with less interference than 4.8% from other common phenolic compounds. A calibration plot was obtained between 1.0 and 30.0 ng/mL and the detection limit was determined to be 0.6 ng/mL. The proposed immunosensor was applied for a real water sample spiked with BPA and the recoveries were in the range between 102.5 and 105.7%.  相似文献

目的:建立刺激胰岛素分泌的新型降糖药物(-)-2 (S)-苄基-4-酮-4-(顺式-全氢化异吲哚-2-基)丁酸钙对映体的HPLC拆分方法。方法:采用Sumichiral OA-3300手性柱(250 × 4.6 mm I.D., 5 μm), 柱温35℃,以0.05 mol·L-1醋酸铵的甲醇溶液为流动相,检测波长为210 nm。结果:本品两对映体在22分钟内实现良好分离,分离度达3以上,S-异构体分别在0.028 ~ 5.6 μg mL-1和0.03 ~ 6.0 μg mL-1范围内线性关系良好,回归方程分别为:Y=1.32×103x-2.54 (r=0.9997)和Y=1.15×103x-1.78 (r=0.9998),最低检测限分别为0.15 ng和0.10 ng,方法精密度RSD低于1.0% (n=5)。结论:建立的对映体分离方法可用于本品光学异构体的质量控制。  相似文献

In the present study, we developed a novel label‐free capacitance impedimetric immunosensor based on the immobilization of the human monoclonal antibody anti‐interleukin‐10 (anti‐IL‐10 mAb) onto polypyrrole (PPy)‐modified silicon nitride (Si3N4) substrates. The immunosensor was used for the detection of the recombinant interleukin‐10 antigen (rh IL‐10) that may be secreted in patients at the early stage of inflammation. The immunosensor was created by chemical deposition of PPy conducting layer on pyrrole?silane (SPy)‐treated Si/SiO2/Si3N4 substrates (Si/SiO2/Si3N4?SPy), followed by anti‐IL‐10 mAb immobilization through carboxyl‐functionalized diazonium (CMA) protocol and carbodiimide chemistry. The surface characterization and the biofunctionalization steps were characterized by SEM, FTIR and cyclic voltammetry (CV) while the detection process was carried out by using electrochemical impedance spectroscopy (EIS) analyses. The created immunosensor showed two linear fittings (R2=0.999) for the detection of rh IL‐10 within the concentration range from 1–50 pg/mL. It exhibited high sensitivity (0.1128 (pg/mL)?1) with a very low limit of detection (LOD)=0.347 pg/mL, more particularly, at the low concentration range (1–10 pg/mL). Thus, this developed polypyrrole‐based immunosensor represents a promising strategy for creation of miniaturized label‐free, fast and highly sensitive biosensors for diagnosis of inflammation biomarkers at very low concentrations with reduced cost.  相似文献

A new, simple, rapid, sensitive, efficient and low‐cost spectrophotometric procedure for the determination of gold was developed. The method is based on the reaction of [AuCl4]? with 2‐[2‐(4‐dimethylaminophenyl)‐vinyl]‐1,3,3‐trimethyl‐3H‐indolium reagent to form a colored ion associate extractable by various organic solvents. The molar absorptivity of the ion associates is in the range (5.7–9.2) × 104 L mol?1 cm?1 depending on the extractant. Butyl acetate was chosen as the extractant. The optimum reaction conditions were established: pH 2–4, concentration of the dye reagent (0.8–1.5) × 10?4 mol L?1. The determination of gold is not hindered even by a 1000‐fold concentration of Ni and Co; a 500‐fold concentration of Pb and Zn; a 100‐fold concentration of Bi, Cu, Cd, Pt, Rh and Ru; or a 20‐fold concentration of Ag. The established method was applied to the determination of gold in model samples and enriched polymetallic ores.  相似文献

A signal‐enhanced label‐free electrochemical immunosensor was constructed by the employment of Prussian blue doped silica dioxide (PB‐SiO2) nanocomposite. At first, PB‐SiO2 nanocomposite which was produced by using a microemulsion method was used to obtain a nanostructural monolayer on a glassy carbon electrode (GCE) surface. Next amino‐functionalized interface were prepared by self‐assembling 3‐aminopropyltriethoxy silane (APTES) on the PB‐SiO2 nanoparticle surface. Then chitosan stabled gold nanoparticle (CS‐nanoAu) was subsequently attached, while the entire surface was finally loaded with neuron‐specific enolase antibody (anti‐NSE) via the adsorption of gold nanoparticle. The sensitivity of the proposed immunosensor has greatly improved as the PB‐SiO2 nanostructural sensing film provides plenty of active sites which might catalyze the reduction of H2O2. The immunosensor exhibited good linear behavior in the concentration range from 0.25–5.0 and 5.0–75 ng/mL for the quantitative analysis of neuron‐specific enolase (NSE), a putative serum marker of small‐cell lung carcinoma (SCLC), with a limit of detection of 0.08 ng/mL. The resulting NSE immunosensor showed high sensitivity and long‐term lifetime which can be attributed to the extremely high catalytic activity and biocompatibility of CS‐nanoAu/APTES/PB‐SiO2 nanostructural multilayers.  相似文献

A approach was successfully employed for constructing a solid‐state electrochemiluminescence (ECL) immunosensor by layer‐by‐layer self‐assembly of multiwall carbon nanotubes (MWCNTs)‐Nafion composite film, Ru(bpy)32+/nano‐Pt aggregates (Ru‐PtNPs) and Pt nanoparticles (PtNPs). The influence of Pt nanoparticles on the ECL intensity was quantitatively evaluated by calculating the electroactive surface area of different electrodes with or without PtNPs to immobilize Ru(bpy)32+. The principle of ECL detection for target α‐fetoprotein antigen (AFP) was based on the increment of resistance after immunoreaction, which led to a decrease in ECL intensity. The linear response range was 0.01–10 ng mL?1 with the detection limit of 3.3 pg mL?1. The immunosensor exhibited advantages of simple preparation and operation, high sensitivity and good selectivity.  相似文献

Various sensor‐based immunoassay methods have been extensively developed for the detection of cancer antigen 15‐3 (CA 15‐3), but most often exhibit low detection signals and low detection sensitivity, and are unsuitable for routine use. The aim of this work is to develop a simple and sensitive electrochemical immunoassay for CA 15‐3 in human serum by using nanogold and DNA‐modified immunosensors. Prussian blue (PB), as a good mediator, was initially electrodeposited on a gold electrode surface, then double‐layer nanogold particles and double‐strand DNA (dsDNA) with the sandwich‐type architecture were constructed on the PB‐modified surface in turn, and then anti‐CA 15‐3 antibodies were adsorbed onto the surface of nanogold particles. The double‐layer nanogold particles provided a good microenvironment for the immobilization of biomolecules. The presence of dsDNA enhanced the surface coverage of protein, and improved the sensitivity of the immunosensor. The performance and factors influencing the performance of the immunosensor were evaluated. Under optimal conditions, the proposed immunosensor exhibited a wide linear range from 1.0 to 240 ng/mL with a relatively low detection limit of 0.6 ng/mL (S/N=3) towards CA 15‐3. The stability, reproducibility and precision of the as‐prepared immunosensor were acceptable. 57 serum specimens were assayed by the developed immunosensor and standard enzyme‐linked immunosorbent assay (ELISA), respectively, and the results obtained were almost consistent. More importantly, the proposed methodology could be further developed for the immobilization of other proteins and biocompounds.  相似文献

This paper describes a highly sensitive and label‐free electrochemical immunosensor for the detection of 1‐pyrenebutyric acid (PBA) which is based on a graphene (GS), chitosan (CS), and ionic liquid (IL) composite modified glassy carbon electrode (GS‐CS‐IL/GCE). The modification process was monitored by transmission electron microscopy (TEM) and cyclic voltammetry (CV). Due to the synergistic effects of GS, CS, and IL, the biosensor exhibits excellent selectivity to PBA. The current response of the proposed immunosensor decreases linearly at two concentration ranges from 0.01 to 5 and from 5 to 150 ng mL?1 with a detection limit of 0.01 ng mL?1.  相似文献

In this study, an impedimetric immunosensor based on polymer poly(thiophene)‐graft‐poly(methacrylamide) polymer (P(Thi‐g‐MAm)) modified indium tin oxide (ITO) electrode is developed for the detection of the Neuron Specific Enolase (NSE) cancer biomarker. First, the P(Thi‐g‐MAm) polymer is synthesized and coated on the ITO electrode by using a spin‐coating technique. P(Thi‐g‐MAm) polymer acts as an immobilization platform for immobilization of NSE‐specific monoclonal antibodies. Anti‐NSE antibodies are utilized as biosensing molecules and they bind to the amino groups of P(Thi‐g‐Mam) polymer via glutaraldehyde cross‐linking. Spin‐coating technique is employed for bioelectrode fabrication and this technique provides a thin and uniform film on the ITO electrode surface. This bioelectrode fabrication technique is simple and it generates a suitable platform for large‐scale loadings of anti‐NSE antibodies. This immunosensor exhibits a wide linear detection range from 0.02 to 4 pg mL?1 and with an ultralow detection limit of 6.1 fg mL?1. It reveals a good long‐term stability (after 8 weeks, 78% of its initial activity), an excellent reproducibility (1.29% of relative standard deviation (RSD)), a good repeatability (5.55% of RSD), and a high selectivity. In addition, the developed immunosensor is proposed as a robust diagnostic tool for the clinical detection of NSE and other cancer biomarkers.  相似文献

A novel and highly sensitive electrochemical immunosensor was developed for the detection of protein biomarker tumor necrosis factor‐alpha (TNF‐α) based on immobilization of TNF‐α‐antibody (anti‐TNF‐α) onto robust nanocomposite containing gold nanoparticles (AuNP), multiwalled carbon nanotubes (MWCNTs) and ionic liquid (1‐buthyl‐3‐methylimidazolium bis (trifluoromethyl sulfonyl)imide). Functionalized MWCNT‐gold nanoparticle was produced by one‐step synthesis based on the direct redox reaction. The electrochemical properties of nanocomposite were characterized by electrochemical impedance spectroscopy and cyclic voltammetry. The anti‐TNF‐α was immobilized or entrapped in the nanocomposite and used in a sandwich type complex immunoassay with anti‐TNF‐α labeled with horseradish peroxidase as secondary antibody. Under optimum conditions, the immunosensor could detect TNF‐α in a linear range from 6.0 to 100 pg mL?1 with a low detection limit of 2.0 pg mL?1. The simple fabrication method, high sensitivity, good reproducibility, stability, as well as acceptable accuracy for TNF‐α detection in human serum samples are the main advantages of this immunosensor, which might have broad applications in protein diagnostics and bioassay.  相似文献

In present study electrografting of the in situ generated 3‐carboxy‐1,2,4‐triazoldiazonium chloride on the Au disk electrode have been studied. The electrode film thickness differences between electrodeposited due to the aryl radical structure 3‐carboxy‐1,2,4‐triazoldiazonium chloride and 4‐carboxyphenyldiazonium salt was shown. The mechanism of 3‐carboxy‐1,2,4‐triazoldiazonium chloride electrografting has been proposed. 4‐nitroanilin was used to investigate the carbodiimide crosslinking capacity. It was established that electrodeposited films are suitable for carbodiimide crosslinking but the reaction proceed only on the “external” electrografted layer. Under the chosen optimal parameters, the label‐free electrochemical immunosensor have been developed. 3‐carboxy‐1,2,4‐triazoldiazonium chloride electrografting provided improvement of analytical characteristics in respect to electrodeposited 4‐carboxyphenyldiazonium salt. The linear range for carcinoembryonic antigen (CEA) detection is 10–104 ng ? ml?1, the limit of detection estimated as 0.2 ng ? ml?1. The developed immunosensor is stable during 30 day′s storage and selective against excess of bovine serum albumin as an interfering reagent.  相似文献
