SiO2 nanosheets (SNS) have been prepared by a chemical method using montmorillonite as raw material and were characterized by scanning electron microscopy and X-ray diffraction. SiO2 nanosheet–Nafion nanocomposites with excellent conductivity, catalytic activity, and biocompatibility provided an extremely hydrophilic surface for biomolecule adhesion. Chitosan was used as a cross-linker to immobilize acetylcholinesterase (AChE), and Nafion was used as a protective membrane to efficiently improve the stability of the AChE biosensor. The AChE biosensor showed favorable affinity for acetylthiocholine chloride and catalyzed the hydrolysis of acetylthiocholine chloride with an apparent Michaelis–Menten constant of 134 μM to form thiocholine, which was then oxidized to produce a detectable and fast response. Based on the inhibition by pesticides of the enzymatic activity of AChE, detection of the amperometric response from thiocholine on the biosensor is a simple and effective way to biomonitor exposure to pesticides. Under optimum conditions, the biosensor detected methyl parathion, chlorpyrifos, and carbofuran at concentrations ranging from 1.0?×?10?12 to 1?×?10?10?M and from 1.0?×?10?10 to 1?×?10?8?M. The detection limits for methyl parathion, chlorpyrifos, and carbofuran were 5?×?10?13?M. The biosensor developed exhibited good sensitivity, stability, reproducibility, and low cost, thus providing a new promising tool for analysis of enzyme inhibitors.

Boron-doped diamond electrodes covered with a nanostructured Pt nanoparticle-polyaniline composite have been fabricated and employed as sensitive amperometric sensors with low detection limit. A highly conductive boron-doped diamond thin film (BDD) was prepared by chemical vapor deposition, and its morphology was characterized by scanning electron microscopy and transmission electron microscopy. The nanostructured composite layer was grown on the BDD electrode by electrochemical deposition of polyaniline and Pt nanoparticles. Glucose oxidase (GOx) was then adsorptively immobilized on the modified BDD electrode. The biosensor displays a large surface area, high catalytic activity of the Pt nanoparticles, efficient electron mediation through the conducting polymer, and low background current of the electrode. The biosensor exhibits an excellent response to glucose, with a broad linear range from 5.9 μM to 0.51 mM, a sensitivity of 5.5 μA·mM?1, a correlation coefficient (R) of 0.9947, and a detection limit of 0.10 μM. The apparent Michaelis-Menten constant (K M app ) and the maximum current density of the electrode are 4.1 mM and 0.021 mA, respectively. This suggests that the immobilized GOx possesses a higher affinity for glucose at the lower K M app , and that the enzymatic reaction rate constitutes the rate-limiting step of the response.  相似文献

Titanium dioxide nanorods (TNR) were grown on a titanium electrode by a hydrothermal route and further employed as a supporting matrix for the immobilization of nafion-coated horseradish peroxidase (HRP). The strong electrostatic interaction between HRP and TNR favors the adsorption of HRP and facilitates direct electron transfer on the electrode. The electrocatalytic activity towards hydrogen peroxide (H2O2) was investigated via cyclic voltammetry and amperometry. The biosensor exhibits fast response, a high sensitivity (416.9 μA·mM?1), a wide linear response range (2.5 nM to 0.46 mM), a detection limit as low as 12 nM, and a small apparent Michaelis-Menten constant (33.6 μM). The results indicate that this method is a promising technique for enzyme immobilization and for the fabrication of electrochemical biosensors.

We have developed an ultra-sensitive electrochemical DNA biosensor by assembling probe ssDNA on a glassy carbon electrode modified with a composite made from molybdenum disulfide, graphene, chitosan and gold nanoparticles. A thiol-tagged DNA strand coupled to horseradish peroxidase conjugated to AuNP served as a tracer. The nanocomposite on the surface acts as relatively good electrical conductor for accelerating the electron transfer, while the enzyme tagged gold nanoparticles provide signal amplification. Hybridization with the target DNA was studied by measuring the electrochemical signal response of horseradish peroxidase using differential pulse voltammetry. The calibration plot is linear in the 5.0?×?10?14 and 5.0?×?10?9 M concentration range, and the limit of detection is 2.2?×?10?15 M. The biosensor displays high selectivity and can differentiate between single-base mismatched and three-base mismatched sequences of DNA. The approach is deemed to provide a sensitive and reliable tool for highly specific detection of DNA.

A biosensor for hydrogen peroxide was constructed by immobilizing horseradish peroxidase on chitosan-wrapped NiFe2O4 nanoparticles on a glassy carbon electrode (GCE). The electron mediator carboxyferrocene was also immobilized on the surface of the GCE. UV?Cvis spectra, Fourier transform IR spectra, scanning electron microscopy, and electrochemical impedance spectra were acquired to characterize the biosensor. The experimental conditions were studied and optimized. The biosensor responds linearly to H2O2 in the range from 1.0?×?10?5 to 2.0?×?10?3?M and with a detection limit of 2.0?×?10?6?M (at S/N?=?3).

A novel glucose biosensor is presented as that based on a glassy carbon electrode modified with hollow gold nanoparticles (HGNs) and glucose oxidase. The sensor exhibits a better differential pulse voltammetric response towards glucose than the one based on conventional gold nanoparticles of the same size. This is attributed to the good biological conductivity and biocompatibility of HGNs. Under the optimal conditions, the sensor displays a linear range from 2.0?×?10?6 to 4.6?×?10?5?M of glucose, with a detection limit of 1.6?×?10?6?M (S/N?=?3). Good reproducibility, stability and no interference make this biosensor applicable to the determination of glucose in samples such as sports drinks.

We have developed a biosensor for highly sensitive and selective determination of the endocrinic disruptor bisphenol A (BPA). It is based on glassy carbon electrode modified with calf thymus DNA and a composited prepared from single walled carbon nanotubes (SWNT) and Nafion. The interaction between BPA and DNA was studied by voltammetry. The binding constant was determined to be 3.55?×?103 M?1, and the binding site has a length of 4.3 base pairs. These electrochemical studies provide further information for a better understanding of the toxicity and carcinogenicity of BPA. Under optimal conditions, the biosensor displays a linear electrochemical response to BPA in the 10 nM to 20 μM concentration range, with a detection limit as low as 5.0 nM (at an S/N of 3). The method was successfully applied to the quantification of BPA in leachates from plastic baby bottles. Recoveries range from 94.0 % to 106.0 % which underpins the excellent performance of this SWNT-based DNA sensor.

We have prepared a nanocomposite consisting of single-walled carbon nanotubes and polylysine. It was characterized by transmission electron microscopy, X-ray photoelectron spectroscopy, and by UV/vis and FTIR spectroscopy. Tyrosinase was covalently immobilized on the nanocomposite, and the resulting bioconjugate deposited on a glassy carbon electrode to form a biosensor for bisphenol A. The biosensor was characterized by scanning electron microscopy and electrochemical impedance spectroscopy. Under optimized experimental conditions, the biosensor gives a linear response to bisphenol A in the 4.00 nM to 11.5 μM concentration range. Its sensitivity is 788 mA M?1 cm?2, and the lower detection limit is 0.97 nM (at an S/N of 3). The biosensor shows good repeatability, reproducibility and long-term stability. In a preliminary practical application, it was successfully applied to the determination of bisphenol A in leachates of plastic spoons.

A novel enzyme-free electrochemical sensor for H2O2 was fabricated by modifying an indium tin oxide (ITO) support with (3-aminopropyl) trimethoxysilane to yield an interface for the assembly of colloidal gold. Gold nanoparticles (AuNPs) were then immobilized on the substrate via self-assembly. Atomic force microscopy showed the presence of a monolayer of well-dispersed AuNPs with an average size of ~4 nm. The electrochemical behavior of the resultant AuNP/ITO-modified electrode and its response to hydrogen peroxide were studied by cyclic voltammetry. This non-enzymatic and mediator-free electrode exhibits a linear response in the range from 3.0?×?10?5 M to 1.0?×?10?3 M (M?=?mol?·?L?1) with a correlation coefficient of 0.999. The limit of detection is as low as 10 nM (for S/N?=?3). The sensor is stable, gives well reproducible results, and is deemed to represent a promising tool for electrochemical sensing.

A glucose biosensor has been fabricated by immobilizing glucose oxidase (GOx) on unhybridized titanium dioxide nanotube arrays using an optimized cross-linking technique. The TiO2 nanotube arrays were synthesized directly on a titanium substrate by anodic oxidation. The structure and morphology of electrode material were characterized by X-ray diffraction and scanning electron microscopy. The electrochemical performances of the glucose biosensor were conducted by cyclic voltammetry and chronoamperometry measurements. It gives a linear response to glucose in the 0.05 to 0.65 mM concentration range, with a correlation coefficient of 0.9981, a sensitivity of 199.6 μA mM?1 cm?2, and a detection limit as low as 3.8 µM. This glucose biosensor exhibited high selectivity for glucose determination in the presence of ascorbic acid, sucrose and other common interfering substances. This glucose biosensor also performed good reproducibility and long-time storage stability. This optimized cross-linking technique could open a new avenue for other enzyme biosensors fabrication.

We have developed an enzymatic glucose biosensor that is based on a flat platinum electrode which was covered with electrophoretically deposited rhodium (Rh) nanoparticles and then sintered to form a large surface area. The biosensor was obtained by depositing glucose oxidase (GOx), Nafion, and gold nanoparticles (AuNPs) on the Rh electrode. The electrical potential and the fractions of Nafion and GOx were optimized. The resulting biosensor has a very high sensitivity (68.1 μA mM?1 cm?2) and good linearity in the range from 0.05 to 15 mM (r?=?0.989). The limit of detection is as low as 0.03 mM (at an SNR of 3). The glucose biosensor also is quite selective and is not interfered by electroactive substances including ascorbic acid, uric acid and acetaminophen. The lifespan is up to 90 days. It was applied to the determination of glucose in blood serum, and the results compare very well with those obtained with a clinical analyzer.

A new conductometric enzyme-based biosensor was developed for the determination of formaldehyde (FA) in aqueous solutions. The biosensor was prepared by cross-linking formaldehyde dehydrogenase from Pseudomonas putida with bovine serum albumin in saturated glutaraldehyde vapours (GA) at the surface of interdigitated gold microelectrodes. Nicotinamide adenine dinucleotide cofactor (NAD+) was added in solution at each measurement to maintain enzyme activity. Addition of a Nafion layer over the enzyme modified electrode resulted in a significant increase of biosensor signal due to enhanced accumulation of protons generated by enzymatic reaction at the electrode surface. Different parameters affecting enzyme activity or playing a role in ionic transfer through the Nafion membrane were optimised. In optimal conditions (0.045 mg enzyme, 30 min exposure to GA, 0.3 μL of a 1 % (v/v) Nafion solution deposit, measurement in 5 mM phosphate buffer pH 7 containing 20 μM NAD+), the biosensor signal was linear up to 10 mM FA, and the detection limit was 18 μM. Relative standard deviations calculated from five consecutive replicates of FA solutions were lower than 5 % in the 1–10 mM range. The biosensor was successfully applied to the determination of FA in spiked water samples (tap water and Rhone river water), with recoveries in the 95–110 % range.

We have developed screen–printed carbon electrodes for the determination of tyramine (Tyr) via plasma amine oxidase. The enzyme was immobilized on the carbon working electrode by cross–linking it with bovine serum albumin using glutaraldehyde. The employment of the mediator hydroxymethylferrocene lowers the working potential to +260 mV (vs. a screen–printed Ag/AgCl reference electrode). The effects of pH, potential and mediator concentration were optimized and resulted in reproducibility and repeatability values of 8.6 % and 8.7 %, respectively. Response is linear in the range from 2 to 164 μM, and the limit of detection is 2.0?±?0.18 μM. The effects of potentially interfering biogenic amines such as putrescine, cadaverine, histamine, spermine, spermidine and tryptamine were also evaluated. The biosensor was successfully applied to the determination of Tyr in cheese.

We have constructed a new electrochemical biosensor by immobilization of hemoglobin (Hb) and ZnWO4 nanorods in a thin film of chitosan (CTS) on the surface of carbon ionic liquid electrode. UV–vis and FT-IR spectra reveal that Hb remains in its native conformation in the film. The modified electrode was characterized by scanning electron microscopy, electrochemical impedance spectroscopy and cyclic voltammetry. A pair of well-defined redox peaks appears which indicates direct electron transfer from the electrode. The presence of CTS also warrants biocompatibility. The electron transfer coefficient and the apparent heterogeneous electron transfer rate constant were calculated to be 0.35 and 0.757 s?1, respectively. The modified electrode displays good electrocatalytic activity for the reduction of trichloroacetic acid with the detection limit of 0.613 mmol L?1 (3σ). The results extend the protein electrochemistry based on the use of ZnWO4 nanorods.

Human mammaglobin (MG) has been found to be the most specific molecular marker for the hematogenous spread of breast cancer cells. In our study, an electrochemical impedance spectroscopic DNA biosensor was established for the detection of MG in breast cancer patients. The working conditions for the biosensor, such as immobilization time, rinse process, and hybridization process, were optimized. Under the optimal conditions, the charge transfer resistance of the proposed DNA biosensor shows excellent correlation with the amount of the complementary oligonucleotides in the range from 1.0?×?10?9 to 2.0?×?10?8?M. The detection limit is 5.0?×?10?10?M. The proposed biosensor was used to detect the polymerase chain reaction amplification products of actual clinical breast cancer samples. The results were compared with that obtained by conventional gel electrophoresis. The results indicate that the electrochemical impedance spectroscopic assay is significantly sensitive and time-saving. The simple strategy described here is expected to be used in clinical application for early diagnosis of breast cancer.

In this work, an electrochemical dihydronicotinamide adenine dinucleotide (NADH) sensor based on the catalytic growth of Au nanoparticles (Au NPs) on glassy carbon electrode was developed. Catalyzed by Au NPs immobilized on pretreated glassy carbon electrode, the reduction of AuCl4 ? in the presence of hydroquinone and cetyltrimethyl ammonium chloride led to the formation of enlarged Au NPs on the electrode surface. Spectrophotometry and high-resolution scanning electronic microscope (SEM) analysis of the sensor morphologies before and after biocatalytic reaction revealed a diameter growth of the nanoparticles. The catalytic growth of Au NPs on electrode surface remarkably facilitated the electron transfer and improved the performance of the sensor. Under optimal conditions, NADH could be detected in the range from 1.25?×?10?6 to 3.08?×?10?4 M, and the detection limit was 2.5?×?10?7 M. The advantages of the proposed sensor, such as high precision and sensitivity, fast response, low cost, and good storage stability, made it suitable for on-line detection of NADH in complex biological systems and contaminant degradation processes.

We describe the use of individual zinc oxide (ZnO) micro/nanowires in an electrochemical biosensor for uric acid. The wires were synthesized by chemical vapor deposition and possess uniform morphology and high crystallinity as revealed by scanning electron microscopy, X-ray diffraction, and photoluminescence studies. The enzyme uricase was then immobilized on the surface of the ZnO micro/nanowires by physical adsorption, and this was proven by Raman spectroscopy and fluorescence microscopy. The resulting uric acid biosensor undergoes fast electron transfer between the active site of the enzyme and the surface of the electrode. It displays high sensitivity (89.74 μA cm?2 mM?1) and a wide linear analytical range (between 0.1 mM and 0.59 mM concentrations of uric acid). This study also demonstrates the potential of the use of individual ZnO micro/nanowires for the construction of highly sensitive nano-sized biosensors.

We have developed a lactate biosensor based on a bionanocomposite (BNC) composed of titanium dioxide nanoparticles (TiO2-NPs), photocatalytically reduced graphene, and lactate oxidase. Graphene oxide was photochemically reduced (without using any chemical reagents) in the presence of TiO2-NPs to give graphene nanosheets that were characterized by atomic force microscopy, Raman and X-ray photoelectron spectroscopy. The results show the nanosheets to possess few oxygen functionalities only and to be decorated with TiO2-NPs. These nanosheets typically are at least 1 μm long and have a thickness of 4.2 nm. A BNC was obtained by mixing lactate oxidase with the nanosheets and immobilized on the surface of a glassy carbon electrode. The resulting biosensor was applied to the determination of lactate. Compared to a sensor without TiO2-NPs, the sensor exhibits higher sensitivity (6.0 μA mM?1), a better detection limit (0.6 μM), a wider linear response (2.0 μM to 0.40 mM), and better reproducibility (3.2 %).

We report on a simple and reliable method for the determination of trace cadmium ion using a glassy carbon electrode (GCE) modified with cupferron, ß-naphthol and MWCNTs. The operational mechanism consists of several steps: first, the ligand cupferron on the modified electrode reacts with Cd2+ ion to form a chelate compound. Next, this chelate is adsorbed by the carrier ß-naphthol following the principle of organic co-precipitation. Finally, the coprecipitated complex is detected by the GCE. This scheme is interesting because it combines preconcentration and electrochemical detection. Two linear responses are obtained, one in the concentration range of 5.0?×?10?11 to 1.6?×?10?8 M, the other in the range of 1.6?×?10?8 to 1.42?×?10?6 M, with a lower detection limit of 1.6?×?10?11 M. This modified GCE does not suffer from significant interferences by Cu(II), Hg(II), Ag(I), Fe(III), Pb(II), Cr(III), Zn(II), NO3?, Cl?, SO 4 2? ions and EDTA. The response of the electrode remained constant for at least 3 weeks of successive operation. The method presented here provides a new way for the simultaneous separation, enrichment, and electrochemical detection of trace cadmium ion.

The potentiometric properties of all-solid-state nitrate-selective electrodes based on plasticized PVC and containing different types of nanosized carbon black were investigated. The use of a carbon black interlayer is shown to significantly improve the potentiometric response. The electrodes display a close-to-Nernstian slope in the range from 10?1 to 10?6 M, highly stable potentials and low membrane resistance. However, different analytical features were found depending on the type of carbon black used. The best long-term potential stability was observed for the electrode with Printex XE2-B carbon black that has a relatively high BET surface area (1000 m2?·?g?1) and an average particle size of 30 nm. Nevertheless, the electrodes with the Vulcan XC-72 (BET surface: 240 m2?·?g?1; average size: 55 nm) showed the most repeatable and reproducible standard potential. The lowest detection limit for nitrate (2.5·10?7 M) is observed for an electrode containing Vulcan XC-72.
