A reinvestigation of the monoterpene chromane ester enriched fraction from Peperomia obtusifolia using chiral chromatography led to the identification of a minor peak, which was elucidated by NMR and HRMS as fenchyl-3,4-dihydro-5-hydroxy-2,7-dimethyl-8-(3″-methyl-2″-butenyl)-2-(4′-methyl-1′,3′-pentadienyl)-2H-1-benzopyran-6-carboxylate, the same structure assigned to two other fenchyl esters described previously, pointing out a stereoisomeric relationship among them. Further NMR analysis revealed that it was actually a mixture of two compounds, whose absolute configurations were determined by VCD measurements. Although, almost no vibrational transitions could be assigned to the chiral chromane, the experimental VCD spectrum was largely opposite to that obtained for the average experimental VCD [(2S,1?R,2?R,4?S + 2R,1?R,2?R,4?S)/2] for fenchol derivatives. These results allowed us to assign the putative compounds as a racemic mixture of the chiral chromane esterified with the monoterpene (1S,2S,4R)-fenchol, which had not been identified in our early work.  相似文献

New chiral fluorescent sensors derived from tetraphenylethylene and proline hydrazide were synthesized and applied in the chiral recognition of various chiral compounds, including unprotected amino acids, acidic compounds, chiral amines and even neutral alcohols. These results demonstrated that the excellent enantioselective response ability to various chiral substrates could be attributed to the –NH moieties of pyrrolidine ring and thiourea unit which acted as hydrogen-bonding donors. This result is of potential significance in enantiomeric discrimination and high-throughput analysis of the enantiomeric purity of chiral guests.  相似文献

We report the synthesis and chiroptical properties of novel chiral carbon nanorings S p-/Rp-[12]PCPP containing a planar chiral [2.2]PCP unit, and demonstrate that S p-/Rp-[12]PCPP can not only host crown ether 18-Crown-6 to form ring-in-ring complexes with a binding constant 3.35×103 M−1, but also accommodate the complexes of 18-Crown-6 and S/R-protonated amines to form homochiral S @Sp -/ R @Rp - and heterochiral S @Rp -/ R @Sp - ternary complexes, displaying significantly larger binding constants of up to 3.31×105 M−1 depending on the chiral guests. Importantly, homochiral S @Sp -/ R @Rp - ternary complexes exhibit an enhanced CD signal, while the heterochiral S @Rp -/ R @Sp - ones have a constant CD signal compared with the chiral carbon nanorings, respectively, which suggests that homochiral S @Sp -/ R @Rp - ternary complexes display a highly narcissistic chiral self-recognition for S/R-protonated chiral amines, respectively. Finally, the chiral ternary complexes can be further applied to determine the ee values of chiral guests. The findings highlight a new application of carbon nanorings in supramolecular sensors, beyond the common recognition of π-conjugated molecules.  相似文献

The chiral shift reagents Sm-(R)-pdta and Sm-(S)-pdta, which are based on (R)- or (S)-1,2-diaminopropane-N,N,N′,N′-tetraacetic acid were synthesized from easily accessible compounds in three simple steps, which makes the method suitable for laboratory-scale production. In addition, a new and efficient method for the preparation of pure anhydrous (R)- or (S)-1,2-diaminopropane-N,N,N′,N′-tetraacetic acid was developed.  相似文献

A chiral fluorescent sensor (Ra,S,l)-3 incorporating (R)-BINOL and l-prolinamide is found efficient in enantioselective recognition of N-Cbz-protected phenylglycine. It is observed that one enantiomer of N-Cbz-protected phenylglycine can obviously increase the fluorescence intensity of (Ra,S,l)-3, while the other enantiomer does not cause much fluorescence enhancement. Such highly enantioselective response makes the sensor (Ra,S,l)-3 useful for the enantioselective fluorescence recognition of other N-Cbz-protected amino acids.  相似文献

The asymmetric aza-Diels-Alder reaction of the (1R)-8-phenylmenthyl or (1R)-8-phenylisoneomenthyl glyoxylate-derived N-benzylimine with cyclopentadiene resulted in the enantioselective synthesis of the corresponding pure [(1S,3-exo)-2-benzyl-2-azabicyclo[2.2.1]hept-5-ene]-3-carboxylates (80 or 69% yield, respectively). Reduction of these cycloadducts with LiAlH4 afforded pure (−)-[(1S,3-exo)-2-benzyl-2-azabicyclo[2.2.1]hept-5-en-3-yl]methanol. Furthermore, a reaction sequence based on Barbier-Wieland degradation of both (1S,3-exo)-adducts afforded pure (+)-(1R)-2-benzoyl-2-azabicyclo[2.2.1]heptan-3-one. In the course of the two transformation sequences referred, the chiral auxiliaries were recovered in a virtually quantitative way.  相似文献

Using the optical measurement technique reflectometric interference spectroscopy (RIfS), the interaction of molecularly imprinted polymers (imprinted with either (R,R)- or (S,S)-2,3-di-O-benzoyltartraric acid) with the corresponding templates and template antipodes were investigated. With these sensors chiral separation with a separation factor of 1.2 could be achieved whereas a reference polymer resulted in no separation. RIfS signals were of opposite sign for imprinted polymer layers containing phenylboronic acid binding site monomers.  相似文献

Several new chiral urea and thiourea ligands have been prepared by reaction of (1R,2R)-1,2-diaminocyclohexane with various organic isocyanates and isothiocyanates. These were used as ligands in the ruthenium catalysed enantioselective reduction of aromatic ketones by isopropanol. The reduction proceeded at room temperature using 2 mol% of ruthenium catalyst to give good yields of the (R)-alcohol with enantiomeric excesses of up to 83%. By contrast, the use of bis-urea ligands gave much lower enantioselectivities. Amino-thiourea ligands led to the (S)-alcohol with low enantiomeric excess.  相似文献

A diastereoselective and enantioselective synthesis of (Z)-1-benzoylamino-2-tritylsulfanylcyclopropanecarboxylic acid derivatives 8a,b and 9a,b was achieved starting from (−)- or (+)-menthyl 2-benzoylamino-3-tritylsulfanylacrylates 3a,b. Compounds 3 were reacted with diazomethane giving the corresponding pyrazolines 4a,b and 5a,b. These compounds, on melting, were transformed, under steric control, into the cyclopropaneamino acid derivatives (R,R)-8a,b and (S,S)-9a,b. The synthesis of a large class of chiral 2-S-alkyl-1-aminocyclopropanecarboxylic acid derivatives is possible after removing the trityl protecting group and subsequent alkylation reactions.  相似文献

Starting from (1S,4R) chiral ketone (+)-6, we developed a synthetic route to the synthesis of new chiral γ-amino alcohols (+)- and (−)-syn-2-amino-7-hydroxy norbornane derivatives with excellent yields and enantiomeric excesses (up to 99%). These compounds were tested as chiral catalysts in the enantioselective addition of diethylzinc to benzaldehyde presenting moderate results. The results obtained, compared with others previously reported, showed that the relative disposition of the amino and hydroxyl groups on C(2) and C(7) positions, play an important role in the catalytic activity.  相似文献

A catalytic enantioselective desymmetrization of meso-N-(heteroarenesulfonyl)aziridines with TMSN3 using chiral Lewis acids afforded products with high enantioselectivity. As proof of the utility of this procedure, the precursor of selective κ-opioid agonist (1S,2S)-(−)-U-50,488 was synthesized.  相似文献

Two series of allylamines/benzylamines have been synthesised and evaluated for their antifungal activity towards Cryptococcus neoformans. All compounds are chiral derivatives of Butenafine and Terbinafine, having additional substituents at the carbon connected to the central nitrogen atom. In both series, the antifungal activity was strongly dependent on both the steric bulk and the electronic nature of the substituents. Compared to the parent compounds (Butenafine and Terbinafine), the activity was maintained when the hydrogen was replaced with a methyl group. Lower activity was observed for ethyl, whereas introduction of -CH2F, -CHF2, -CF3 or -CN substituents removed all antifungal activity. Testing of (R)- and (S)-N-(4-tert-butylbenzyl)-N-methyl-1-(naphthalen-1-yl)ethanamine against C. neoformans, Cryptococcus diffluens and Trichosporon cutaneum revealed that most of the activity resides in the (R)-enantiomer. The (R)-enantiomer performed as well as, or better (lower MIC values) than Butenafine against each test strain, suggesting that antimycotics based on this compound might be an improvement of existing Butenafine-based formulations.  相似文献

The chiral functionalization of a simple heterocycle, 1,3-dihydro-2-imidazolone, was achieved by the highly enantioselective monodeacylation of meso-1,3-diacetyl-2-imidazolidinones via an oxazaborolidine-catalyzed borane reduction. This kinetically controlled dissymmetrization is sufficiently effective to provide a synthetic route to either enantiomer of (4S, 5S)- or (4R, 5R)-4,5-dimethoxy-2-imidazolidinone derivatives, which serve as chiral synthons for threo-1,2-diamines.  相似文献

The catalytic potency of a series of perhydro-1,3-benzoxazines prepared from (?)-8-aminomenthol was examined in the enantioselective addition of diethylzinc to aldehydes. When (2R,3S,3aS,4aR,6R,8aS)-2-isopropyl-6,9,9-trimethyl-3-phenyldecahydro-4aH-pyrrolo[2,1-b][1,3]benzoxazin-3-ol 7b was used as a chiral ligand, 1-substituted propanols with an (R)-configuration were obtained in high yields and enantiomeric excesses up to >99%. Total synthesis of (4R,5S,6E,14S)- and (4R,5S,6E,14R)-cystothiazoles F 3 was achieved from the chiral bithiazole-type primary alcohols [(S)- and (R)-4-ethoxycarbonyl-2′-(1-hydroxymethylethyl)-2,4′-bithiazoles 8], which were obtained based on the enzymatic resolution of racemic alcohol 8 and its acetate 9. From a direct comparison by means of chiral HPLC between natural cystothiazole F 3 and synthetic compounds [(4R,5S,6E,14S)- and (4R,5S,6E,14R)-cystothiazoles 3], natural cystothiazole F 3 was found to be a 33:67 diastereomeric mixture [(4R,5S,6E,14S)-3:(4R,5S,6E,14R)-3 = 33:67].  相似文献

In a previous study we reported the identification of the first ketone found in the Pentatomidae family, the sex attractant pheromone of the male stink bug Pallantia macunaima, (6R,10S)-6,10,13-trimethyltetradecan-2-one. Here we describe an efficient enantioselective route for the synthesis of the four stereoisomers of this pheromone. The synthesis was conceived as the connection of two chiral building blocks, employing (R)- or (S)-citronellol and methyl (S)-3-hydroxy-2-methylpropionate as the source of chirality.  相似文献

Four optically active amino alcohols were synthesized via the ring opening of (R)-N-(2,3-epoxypropyl)phthalimide with (R)-2-phenyl glycinol, (1R,2S)-cis-1-amino-2-indanol, (R)-2-amino-1-butanol and (S)-phenyl ethylamine in 73-93% yields. The enantioselective recognition of these receptors towards the enantiomers of racemic carboxylic acids was studied by 1H NMR spectroscopy. The molar ratio and the association constants of the chiral compounds with each of the enantiomers of the guests were determined by using Job plots and a non-linear least-squares fitting method, respectively. Large non-equivalent chemical shifts (up to 30.0 Hz) can be achieved in the presence of chiral amino alcohols 2 and 5. Amongst the chiral receptors used, compound 5 was found to be the best chiral shift reagent, and was effective in the determination of the enantiomeric excess of chiral carboxylic acids.  相似文献

New enantiopure amines (R,R)-1 and (S,S)-1 were obtained from (R)- or (S)-2,2′-diamino-1,1′-binaphthyl and 2,6-diformylpyridine in a synthesis templated by lead(II) or lanthanide(III) ions, reduction with NaBH4 and subsequent demetallation. Similarly new amines (R,R,R,R)-2 and (S,S,S,S)-2 were obtained from (1R, 2R)- or (1S, 2S)-1,2-diphenylethylenediamine. The X-ray crystal structure of the Pb(II) complex with macrocyclic Schiff base precursor of (R,R)-1 indicates helical twisted conformation of this macrocycle, while the ROESY spectrum of R,R-1 suggests less twisted conformation. (R,R)-1 and (R,R,R,R)-2 were tested as chiral shift reagents (chiral solvating agents) for various α-substituted carboxylic acids, including non steroidal anti-inflammatory drugs. Enantiodiscrimination of carboxylate 1H NMR signals was observed with ΔΔδ values up to 0.1 ppm.  相似文献

Chiral metabolites are found in a wide variety of living organisms and some of them are understood to be physiologically active compounds and biomarkers. However, the overall analysis of chiral metabolomics is quite difficult due to the high number of metabolites, the significant diversity in their physicochemical properties, and concentration range from metabolite-to-metabolite. To solve this difficulty, we developed a novel approach for chiral metabolomics fingerprinting and chiral metabolomics extraction, which is based on the labeling of a pair of enantiomers of chiral derivatization reagents (i.e., DMT-(S,R)-Pro-OSu and DMT-3(S,R)-Apy) and precursor ion scan chromatography of the derivatives. The multivariate statistics is also required for this strategy. The proposed procedures were evaluated by the detection of a diagnostic marker (i.e., d-lactic acid) using the saliva of diabetic patients. This method was used for the determination of biomarker candidates of chiral amines and carboxyls in Alzheimer's disease (AD) brain homogenates. As the results, l-phenylalanine (L-Phe) and l-lactic acid (L-LA) were identified as the decreased and increased biomarker candidates in the AD brain, respectively. Therefore, the proposed approach seems to be helpful for the determination of non-target chiral metabolomics possessing amines and carboxyls.  相似文献
