The development of an immunoaffinity chromatography (IAC) procedure for the selective extraction of the anti-fouling agent Irgarol 1051 [2-(tert.-butylamino)-4-(cyclopropylamino)-6-(methylthio)-1,3,5-triazine] from seawater is described. The anti-Irgarol 1051 antibodies were covalently bound to agarose-based beads support. IAC column capacities were higher than 400 ng and ethanol-water (70:30) was selected as eluting mixture. After percolation of 250 ml of water sample containing Irgarol 1051 at environmental levels (ng l(-1) ), the breakthrough volume was still not achieved. Other triazine herbicides percolated through the IAC column showed good recoveries. Thus, this IAC procedure may be useful to extract related compounds. The developed IAC column was applied to real seawater samples and compared with RP-C18 cartridges. The limit of detection (LOD) reached by using the IAC procedure was twenty times lower than the LOD achieved by the RP-C 18 cartridges using the same detection system. Irgarol 1051 was detected at ng l(-1) levels in the Barcelona marina (northwestern Mediterranean Sea). An acceptable correlation between enzyme-linked immunosorbent assay and gas chromatography with nitrogen-phosphorus detection was observed, thus analysis of Irgarol 1051 can be performed by either one of the methods. In this work, further confirmation of the analyte identity for real samples was accomplished by gas chromatography-electron impact mass spectrometry.  相似文献

A highly sensitive reversed-phase high-performance liquid chromatographic assay for ethanol and methanol in plasma, using a post-column enzymic reactor with electrochemical detection, has been developed. The alcohols, separated on the column, were converted by immobilized alcohol oxidase into their respective aldehydes with formation of stoichiometric amounts of hydrogen peroxide, detected via oxidation at a platinum electrode. As the chromatographic column, two glass cartridges (150 mm x 3 mm I.D.) in series, packed with 10 microns HEMA-S 1000 packing, were used. Alcohol oxidase from Candida boidinii was immobilized onto HEMA-BIO 1000 VS-L (10 microns), packed in a 30 mm x 3 mm I.D. glass cartridge. The reaction product, hydrogen peroxide, was detected with an amperometric detector with a platinum electrode, operated at +500 mV vs. an Ag/AgCl reference electrode. A 20-microliters volume of ten-fold diluted plasma was injected without any pre-treatment. Under the described conditions, methanol and ethanol were well resolved from each other and from the "front" of the chromatogram. The limit of detection was ca. 2.5 nmol for ethanol and 0.6 nmol for methanol in plasma, at a signal-to-noise ratio of 3. Excellent linearity was observed for ethanol, in the range 0.125-4 micrograms injected (r = 0.9999). In contrast, the response for methanol was markedly non-linear above 500 micrograms injected, presumably owing to progressive saturation of the reactor. The precision and accuracy of the assay were satisfactory, as was the reactor life (one month).  相似文献

A liquid chromatography with diode array or electrospray ionisation mass spectrometry detection (LC-DAD-ESI-MS) method for the determination of tiamulin residues in honey is presented. The procedure employs a solid-phase extraction (SPE) on polymeric cartridges for the isolation of tiamulin from honey samples diluted in aqueous solution of tartaric acid. Chromatographic separation of the tiamulin is performed, in isocratic mode, on a C18 column using methanol and ammonium carbonate 0.1% in water, in proportion (30:70, v/v). Average analyte recoveries were from 88 to 106% in replica sets of fortified honey samples. The LC-ESI-MS method detection limits differ from 0.5 microg kg(-1) for clear honeys to 1.2 microg kg(-1) for dark honeys. The developed method has been applied to the analysis of tiamulin residues in multifloral honey samples collected from veterinary treated beehives.  相似文献

建立了串联双柱固相萃取-高效液相色谱-电喷雾多级质谱(HPLC-ESI-MSn)确认及测定盐酸二甲双胍制剂中的盐酸二甲双胍及残留物三聚氰胺和双氰胺的方法。盐酸二甲双胍制剂样品在超声辅助下,采用含0.1%(v/v)乙酸的无水乙醇溶液提取,选择Cleanert PCX-C18串联固相萃取双柱净化富集,以5%(v/v)氨水甲醇溶液为洗脱剂进行洗脱。采用Kromasil-C18色谱柱(100 mm×4.6 mm,3.5 μm)分离,梯度洗脱,在电喷雾多级质谱、选择离子模式下检测,外标法定量。通过比较制剂残留物在不同萃取溶剂、固相萃取柱及洗脱液等条件下的回收率,优化了前处理方法。结果表明,在各自的线性范围内,3种目标物的峰面积与质量浓度呈良好的线性关系,相关系数(r2)≥0.9992;方法的检出限为1.48~13.61 μg/kg,定量限为5.96~45.67 μg/kg。3种目标物在低、中、高加标水平下的平均回收率为65.02%~118.33%,相对标准偏差(RSD)≤13.41%(n=5)。该法操作简单,净化效果好,准确度高,重现性好,适用于检测和分析不同盐酸二甲双胍制剂中盐酸二甲双胍及残留物三聚氰胺和双氰胺。  相似文献

The wet effluent diffusion denuder (WEDD) preconcentration technique for the determination of methanol, ethanol, 1-propanol, 2-propanol, 1-butanol, 1-pentanol, acetone, methyl ethyl ketone, diethyl ketone and methyl n-propyl ketone in air is discussed. The compounds are continuously collected into a thin film of deionized water flowing down the inner wall of the cylindrical wet effluent diffusion denuder. The concentrate is analysed by gas chromatography. Detection limits of alcohols and ketones are as low as 1 microg/l (GC-flame ionization detection) and/or 1 ng/l (GC-MS). This technique could be applicable for the continuous monitoring of ppb (v/v) levels of both alcohols and ketones in the air.  相似文献

Gas chromatography (GC) with electron-capture detection (ECD), mass spectrometry (MS) and tandem mass spectrometry (MS-MS) were employed for the identification of 12 pesticides in water samples. For this purpose, a solid-phase extraction procedure with C18 cartridges was used, optimising the breakthrough volume and the saturation concentration. In GC-MS-MS, the lowest detectable concentrations for the pesticides were between 2 and 26 ng l(-1), recoveries ranged from 70 to 133% in water samples spiked at 100 ng l(-1) and the relative standard deviations were in the range 5.3 to 17.4%. The proposed analytical methodology was applied to analyse pesticides in wetland samples from Almería (Spain).  相似文献

A simple and sensitive method for the determination of liquid methanol and ethanol at trace levels by an alkyl nitrite formation reaction has been established. Alcohol was allowed to react with nitrous acid, which was yielded from sulfuric acid and sodium nitrite in the solution, to form the corresponding alkyl nitrite in the hexane organic phase. Alkyl nitrites in hexane were analyzed by a gas chromatograph with an electron capture detector (GC-ECD). The detection limits, which were determined at a signal-to-noise ratio of 3, were 1.1 and 0.7 micrograms/L for methanol and ethanol, respectively, by 1 microL injection. The relative standard deviations for n = 8 were 4.0 and 3.3% for methanol and ethanol, respectively. The method was applied to determine the alcohol concentration in a rice paddy, pond water, tap water, and well water. Those aqueous samples were also spiked with standard alcohols; the average recoveries of spiked methanol and ethanol were 98 and 91% with relative standard deviations of 6.1 and 4.0%, respectively.  相似文献

Low molecular weight alcohols including fusel oil were determined using diethyl ether extraction and capillary gas chromatography. Twelve kinds of alcohols were successfully resolved on the HP-FFAP (polyethylene glycol) capillary column. The diethyl ether extraction method was very useful for the analysis of alcohols in alcoholic beverages and biological samples with excellent cleanliness of the resulting chromatograms and high sensitivity compared to the direct injection method. Calibration graphs for all standard alcohols showed good linearity in the concentration range used, 0.001-2% (w/v) for all alcohols. Salting out effects were significant (p < 0.01) for the low molecular weight alcohols methanol, isopropanol, propanol, 2-butanol, n-butanol and ethanol, but not for the relatively high molecular weight alcohols amyl alcohol, isoamyl alcohol, and heptanol. The coefficients of variation of the relative molar responses were less than 5% for all of the alcohols. The limits of detection and quantitation were 1-5 and 10-60 microg/L for the diethyl ether extraction method, and 10-50 and 100-350 microg/L for the direct injection method, respectively. The retention times and relative retention times of standard alcohols were significantly shifted in the direct injection method when the injection volumes were changed, even with the same analysis conditions, but they were not influenced in the diethyl ether extraction method. The recoveries by the diethyl ether extraction method were greater than 95% for all samples and greater than 97% for biological samples.  相似文献

The analysis of nitrogen containing amino alcohols, which are the precursors and degradation products of nitrogen mustards and nerve agent VX, constitutes an important aspect for verifying the compliance to the CWC (Chemical Weapons Convention). This work devotes on the development of solid-phase extraction method using silica- and polymer-based SCX (strong cation-exchange) and MCX (mixed-mode strong cation-exchange) cartridges for N,N-dialkylaminoethane-2-ols and alkyl N,N-diethanolamines, from water. The extracted analytes were analyzed by GC-MS (gas chromatography-mass spectrometry) in the full scan and selected ion monitoring modes. The extraction efficiencies of SCX and MCX cartridges were compared, and results revealed that SCX performed better. Extraction parameters, such as loading capacity, extraction solvent, its volume, and washing solvent were optimized. Best recoveries were obtained using 2 mL methanol containing 10% NH(4)OH and limits of detection could be achieved up to 5 x 10(-3) microg mL(-1) in the selected ion monitoring mode and 0.01 microg mL(-1) in full scan mode. The method was successfully employed for the detection and identification of amino alcohol present in water sample sent by Organization for Prohibition of Chemical Weapons (OPCW) in the official proficiency tests. The method was also applied to extract the analytes from human plasma. The SCX cartridge showed good recoveries of amino alcohols from human plasma after protein precipitation.  相似文献

A method using liquid chromatography-tandem mass spectrometry has been developed for determination of trace levels of tetracycline antibiotics in ground water and confined animal feeding operation waste water. Oxytetracycline (OTC), tetracycline (TC), and chlortetracycline (CTC) were extracted from water samples using both polymeric and C18 extraction cartridges. The addition of a buffer containing potassium phosphate and citric acid improved tetracycline recoveries in lagoon water. Method detection limits determined in reagent water fortified with 1 microg l(-1) OTC, TC, and CTC were 0.21, 0.20, and 0.28 microg l(-1). Method detection limits in lagoon water samples fortified at 20 microg l(-1) for OTC, TC, and CTC were 3.6, 3.1, and 3.8 microg l(-1). Variability in recovery from laboratory fortified blanks ranged from 86 to 110% during routine analysis.  相似文献

采用自主设计的连续流动气封壁超临界水氧化反应装置,研究了典型醇类物质甲醇、乙醇和异丙醇在超临界水中氧化的反应途径,并归纳了醇类物质超临界水氧化反应的规律及特点。研究结果表明,甲醇超临界水氧化反应的主要中间产物为甲醛,同样条件下转化率较乙醇和异丙醇低;乙醇和异丙醇超临界水氧化反应的主要中间产物为丙酮、乙酸、乙醛和甲醇等。三种醇超临界水氧化过程中均涉及到大量活性自由基的相互作用,表现为脱氢、裂解和聚合等反应形式;产物包括碳链增长、不变、降低三种类型。总体来看,醇类物质超临界水氧化反应的趋势是向碳链降低的方向进行,即通过一系列中间产物最后生成CO2和水。  相似文献

Anti-microcystin LR immunnoaffinity cartridges were evaluated for their ability to selectively remove microcystins from extracts of blue-green algae, fish and water samples for subsequent analysis by liquid chromatography with UV absorbance detection at 238 nm. Blue-green algae and fish samples were extracted with 75% methanol in water. A portion of the extract was diluted and passed through an immunoaffinity cartridge. Water samples were applied directly to the cartridge. The cartridge was rinsed with water and 25% methanol in water. The microcystins were eluted with 80% methanol in water containing 4% acetic acid. It was found that the cartridges were effective in isolating the microcystins from blue-green algae, fish and water samples, resulting in extracts that were clean enough to enable direct LC-UV detection down to approximately 0.03 microg/g in the blue-green algae and fish samples, and as low as 0.02 ng/ml for water samples. The cartridges were found to have a capacity of approximately 200 ng each for a mixture of microcystins RR, YR, LR and LA, or as much as 525-800 ng for individual compounds. Recoveries trough the complete analytical procedure ranged from 64 to 115% (all values) with an overall average of approximately 80% at spiking levels of 0.5-4.0 microg/g for the microcystins in blue-green algae. The average recoveries (n=8) from spiked (0.1-0.5 microg/g) fish samples were 73% for RR, 79% for YR, 81% for LR and 77% for LA, while from the spiked (2.0-0.04 ng/g) tap and river water samples (n=6), recoveries were 78% for RR, 86% for YR, 94% for LR and 89% for LA.  相似文献

A sensitive and selective gas chromatography with mass spectrometry method was developed for the simultaneous determination of three organophosphorus pesticides, namely, chlorpyrifos, malathion, and diazinon in three different food commodities (milk, apples, and drinking water) employing solid‐phase extraction for sample pretreatment. Pesticide extraction from different sample matrices was carried out on Chromabond C18 cartridges using 3.0 mL of methanol and 3.0 mL of a mixture of dichloromethane/acetonitrile (1:1 v/v) as the eluting solvent. Analysis was carried out by gas chromatography coupled with mass spectrometry using selected‐ion monitoring mode. Good linear relationships were obtained in the range of 0.1–50 μg/L for chlorpyrifos, and 0.05–50 μg/L for both malathion and diazinon pesticides. Good repeatability and recoveries were obtained in the range of 78.54–86.73% for three pesticides under the optimized experimental conditions. The limit of detection ranged from 0.02 to 0.03 μg/L, and the limit of quantification ranged from 0.05 to 0.1 μg/L for all three pesticides. Finally, the developed method was successfully applied for the determination of three targeted pesticides in milk, apples, and drinking water samples each in triplicate. No pesticide was found in apple and milk samples, but chlorpyrifos was found in one drinking water sample below the quantification level.  相似文献

We developed a reversed‐phase high‐performance liquid chromatography method with ultraviolet detection using on‐line complexation with Cu(II) ion for analysis of five alcohols including diols and triol (methanol, ethanol, 1,2‐propanediol, 1,3‐propanediol, and glycerol). The Cu(II) ion concentration in the mobile phase had a great influence on the peak areas of these alcohols, but not on their retention times. Column temperature (25−40°C) and pH of the mobile phase did not affect the separation of analytes. The optimum separation conditions were determined as 5 mM CuSO4, 3 mM H2SO4, and 3 mM NaOH at 30°C. The ratio of the peak areas for three alcohols (methanol, 1,2‐propanediol, and glycerol) was in good agreement with that calculated from the obtained stability constants, molar absorption coefficients for the 1:1 Cu(II) complexes with the three alcohols, and the injected molar quantities. This fact strongly suggests that the observed high‐performance liquid chromatography signals resulted from formation of the 1:1 Cu(II)–alcohol complexes. Using the proposed method, these five alcohols in spirit, liquid for electronic cigarette, mouthwash, and nail enamel remover samples were successfully analyzed with only a simple pretreatment.  相似文献

A gas chromatography/mass spectrometry (GC/MS) method for determination of nine N-nitrosamines (NAs) in water is described. Two ionization modes, electron impact (EI) and chemical ionization (CI) with methanol, as well as different ion analysis techniques, i.e. full scan, selected ion storage (SIS) and tandem mass spectrometry (MS/MS) were tested. Chemical ionization followed by SIS resulted the mass spectrometric method of choice, with detection limits in the range of 1-2ng/L. Solid Phase Extraction (SPE) with coconut charcoal cartridges was applied to extract NAs from real samples, according EPA Method 521. Drinking water samples were collected from seven surface- and two groundwater treatment plants. Three surface water treatment plants were sampled before and after addition of O(3)/ClO(2) to observe the effect of disinfection on NAs' formation. N-nitrosodiethylamine (NDEA), n-nitrosodipropylamine (NDPA), n-nitrosomorpholine (NMOR) and n-nitrosodibutylamine (NDBA) were found up to concentrations exceeding three times the risk level of 10ng/L set by the California Department of Public Health. Because dermal adsorption has been recently indicated as a new contamination route of exposure to NAs for people who practice swimming activity, water samples from five swimming pools in the Bologna (Italy) area were collected. N-nitrosopyrrolidine (NPYR) was detected in all samples at concentrations larger than 50ng/L, likely as a disinfection by-product from the amino acid precursor proline, a main constituent of skin collagen.  相似文献

A liquid chromatography-mass spectrometry (LC-MS) method was developed for the determination of five macrolides in natural water samples, using kitasamycin as surrogate. The macrolides were extracted from water samples using Oasis HLB cartridges. Pre-concentration factors up to 250 were obtained. Separation was carried out in an end-capped silica-based C18 column and mobile phases consisting of water/acetonitrile mixtures containing ammonium acetate. Detection was performed by mass spectrometry with a single quadrupole and a triple quadrupole using an electrospray interface. The quality parameters obtained with these two approaches were compared. The detection limits of the whole process were about 1 ng l(-1). The recoveries from 250 ml of water samples spiked at 25-125 ng l(-1) level were in the range 85-115%, except for azithromycin levels, which were around 70%. Erythromycin-H2O, clarithromycin and azithromyzin were found, at the sub ng l(-1) level, in the studied rivers.  相似文献

A simple and sensitive method for the determination of polar pesticides in water and wine samples was developed by coupling automated in-tube solid-phase microextraction (SPME) to high-performance liquid chromatography-electrospray ionization mass spectrometry (HPLC-ESI-MS). To achieve optimum performance, the conditions for both the in-tube SPME and the ESI-MS detection were investigated. In-tube SPME conditions were optimized by selecting the appropriate extraction parameters, especially the stationary phases used for SPME. For the compounds studied, a custom-made polypyrrole (PPY)-coated capillary showed superior extraction efficiency as compared to several commercial capillaries tested, and therefore, it was selected for in-tube SPME. The influence of the ethanol content on the performance of in-tube SPME was also investigated. It was found that the amount of pesticides extracted decreased with the increase of ethanol content in the solutions. The ESI-MS detection conditions were optimized as follows: nebulizer gas, N2 (30 p.s.i.; 1 p.s.i.=6894.76 Pa); drying gas, N2 (10 l/min, 350 degrees C); capillary voltage, 4500 V; ionization mode, positive; mass scan range, 50-350 amu; fragmentor voltage, variable depending on the ions selected. Due to the high extraction efficiency of the PPY coating and the high sensitive mass detection, the detection limits (S/N = 3) of this method for the compounds studied are in the range of 0.01 to 1.2 ng/ml, which are more than one order of magnitude lower than those of the previous in-tube SPME-HPLC-UV method. A linear relationship was obtained for each analyte in the concentration range of 0.5 to 200 ng/ml with MS detection. This method was applied to the analysis of phenylurea and carbamate pesticides in spiked water and wine samples.  相似文献

The use of methanol or ethanol as the injection solvent for the gas chromatographic-mass spectral (GC-MS) analysis of low molecular weight amine drugs of abuse results in the formation of additional components in the sample. Primary amines, such as amphetamine, 3,4-methylenedioxyamphetamine, and phenethylamine, yield imines upon injection as methanol or ethanol solutions. In methanol, the imine formed has a mass that is 12 mass units higher than the parent compound. In ethanol, the products formed have 26 additional mass units. Secondary amines appear to undergo methylation under similar conditions with methanol as the injection solvent. These products are absent from the analysis of equivalent amine samples dissolved in chloroform.  相似文献

Bimodally porous titania powders were made by hydrolysis of titanium tetraisopropoxide (TTIP) dissolved in various alcohols (methanol, ethanol, isopropanol, and sec-butanol). The specific surface area (SSA) of the powders dried at 150 degrees C ranged from 332 to 624 m(2)/g as determined by nitrogen adsorption. At excess alcohol concentration, the SSA of the dried powders decreased in the order of sec-butanol, iso-propanol, ethanol, and methanol at a constant alcohol/TTIP molar ratio. The pore size distribution was bimodal with fine intraparticle pore diameters at 1-6 nm and larger interparticle pore diameters at 30-120 nm as determined by nitrogen adsorption isotherms. The average intraparticle pore diameter decreased with increasing alcohol concentration for methanol and ethanol, while it was rather constant at 3.3 nm, irrespective of alcohol concentration for iso-propanol and sec-butanol. The evolution of particle phase composition was determined by X-ray diffraction ranging from amorphous to crystalline anatase and rutile largely proportional to the calcination temperature and to a lesser extent on the type and concentration of alcohols. Fragrance materials such as synthetic musks in aqueous samples, are normally determined by gas chromatography-mass spectrometry in the selected ion monitoring (SIM) mode to provide maximum sensitivity after liquid-liquid extraction of 1-1 samples. Full-scan mass spectra are required to verify that a target analyte has been found by comparison with the mass spectra of fragrance compounds in the National Institute of Standards and Technology (NIST) mass spectral library. A 1-1 sample usually provides insufficient analyte for full scan data acquisition. This paper describes an on-site extraction method developed at the US Environmental Protection Agency (Las Vegas, NV, USA) for synthetic musks from 60 l of wastewater effluent. Such a large sample volume permits high-quality, full-scan mass spectra to be obtained for a wide array of synthetic musks. Quantification of these compounds was achieved from the full-scan data directly, without the need to acquire SIM data. The detection limits obtained with this method are an order of magnitude lower than those obtained from liquid-liquid and other solid-phase extraction methods. This method is highly reproducible, and recoveries ranged from 80 to 97% in spiked sewage treatment plant effluent. The high rate of sorbent-sample mass transfer eliminated the need for a methanolic activation step, which reduced extraction time, labor, and solvent use. More samples could be extracted in the field at lower cost. After sample extraction, the light-mass cartridges are easily transported and stored.  相似文献
