Molecular dynamics simulations were used to characterize the binding of the chiral drugs chlorthalidone and lorazepam to the molecular micelle poly-(sodium undecyl-(L)-leucine-valine). The project’s goal was to characterize the nature of chiral recognition in capillary electrophoresis separations that use molecular micelles as the chiral selector. The shapes and charge distributions of the chiral molecules investigated, their orientations within the molecular micelle chiral binding pockets, and the formation of stereoselective intermolecular hydrogen bonds with the molecular micelle were all found to play key roles in determining where and how lorazepam and chlorthalidone enantiomers interacted with the molecular micelle.  相似文献

A chiral amino acid-based monomeric and polymeric surfactant, sodium oleyl-L-leucylvalinate) (L-SOLV) and poly(sodium oleyl-L-leucylvalinate) (poly-L-SOLV) were synthesized and used for chiral separations in micellar electrokinetic chromatography (MEKC). Poly-L-SOLV was used successfully in the separation of various enantiomers of neutral, acidic, and basic analytes such as 1,1'-bi-2-napthol, 1,1'-binaphthyl-2,2'-diamine, benzoin, hydrobenzoin, benzoin methylether, warfarin, and coumachlor obtaining well-resolved peaks but with only partial separation of temazepam. In addition, the atropisomer 1,1'-binaphthyl-2, 2'-dihydrogen phosphate was chosen to study the applicability of the polymeric surfactant over a wide range of parameters such as concentration, temperature, voltage, and pH. The most striking characteristic of this new surfactant is its high hydrophobicity. It is favorable to interactions with hydrophobic chiral analytes, and thus may provide better chiral recognition for the compounds.  相似文献

Metal complexes-catalyzed hydrolysis and alcoholysis of organic substrates such as alkenyl esters, alkenyl ethers, and azlactones (oxazol-5(4H)-ones) are described. These reactions were applied for kinetic resolution of chiral compounds and high selectivities were achieved with vinyl ethers of 2-substituted cyclohexanols, 1,1′-bi-2-naphthols, 1,1′-bi-2-phenols, and 4,4-disubstituted azlactones. Oxidative carbon–carbon bond cleavage reactions, which were found in the course of the study of asymmetric hydrolysis were also described.  相似文献

We show by quantum mechanical/molecular mechanical (QM/MM) simulations that phenylbenzothiazoles undergoing an excited‐state proton transfer (ESPT) can be used to probe protein binding sites. For 2‐(2′‐hydroxy‐4′‐aminophenyl)benzothiazole (HABT) bound to a tyrosine kinase, the absolute and relative intensities of the fluorescence bands arising from the enol and keto forms are found to be strongly dependent on the active‐site conformation. The emission properties are tuned by hydrogen‐bonding interactions of HABT with the neighboring amino acid T766 and with active‐site water. The use of ESPT tuners opens the possibility of creating two‐color fluorescent markers for protein binding sites, with potential applications in the detection of mutations in cancer cell lines.  相似文献

NMR spectroscopy was used to characterize the binding of the chiral compound 1,1′‐binaphthyl‐2,2′‐diyl hydrogen phosphate (BNP) to five molecular micelles with chiral dipeptide headgroups. Molecular micelles have covalent linkages between the surfactant monomers and are used as chiral mobile phase modifiers in electrokinetic chromatography. Nuclear overhauser enhancement spectroscopy (NOESY) analyses of (S)‐BNP:molecular micelle mixtures showed that in each solution the (S)‐BNP interacted predominately with the N‐terminal amino acid of the molecular micelle's dipeptide headgroup. NOESY spectra were also used to generate group binding maps for (S)‐BNP:molecular micelle mixtures. In these maps, percentages are assigned to the (S)‐BNP protons to represent the relative strengths of their interactions with a specified molecular micelle proton. All maps showed that (S)‐BNP inserted into a previously reported chiral groove formed between the molecular micelle's dipeptide headgroup and hydrocarbon chain. In the resulting intermolecular complexes, the (S)‐BNP protons nearest to the analyte phosphate group were found to point toward the N‐terminal Hα proton of the molecular micelle headgroup. Finally, pulsed field gradient NMR diffusion experiments were used to measure association constants for (R) and (S)‐BNP binding to each molecular micelle. These K values were then used to calculate the differences in the enantiomers' free energies of binding, Δ(ΔG). The NMR‐derived Δ(ΔG) values were found to scale linearly with electrokinetic chromatography (EKC) chiral selectivities from the literature. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Four novel derivatives of 1,1′-bi-2-naphthol have been prepared and the structures of these compounds characterised by IR, MS, 1H and 13C NMR spectroscopy and elemental analysis. The enantioselective recognition of these sensors has been studied by fluorescence titration and 1H NMR spectroscopy. The sensors exhibited different chiral recognition abilities towards N-Boc-protected amino acid anions and formed 1:1 complexes between the host and the guest. Sensors exhibit excellent enantioselective fluorescent recognition ability towards the amino acid derivatives.  相似文献

NMR spectroscopy was used to investigate the association of four chiral molecules with the molecular micelle poly(sodium N-undecanoyl-l-leucylvalinate) (poly(SULV)). Adding poly(SULV) to the background electrolyte in electrokinetic chromatography (EKC) allows enantiomeric resolution to be achieved because enantiomers interact differentially with the chiral centers on the micelle headgroups as they both move in the electric field. Pulsed field gradient diffusion experiments were used to measure molecular micelle association constants for enantiomers of each analyte. These association constants were consistent with EKC elution order for the compounds 1,1'-binaphthyl-2,2'-diyl hydrogen phosphate (BNP), 1,1'-bi-2-naphthol (BOH), and Troger's base. In addition, nuclear Overhauser enhancement spectroscopy, nuclear Overhauser effect difference, and intermolecular cross relaxation diffusion experiments were used to generate binding interaction maps for each chiral analyte. These maps showed that BNP and BOH inserted into the surfactant headgroup's major chiral groove and interacted predominately with the leucine chiral center. (+)-Troger's base was also found to insert into the major chiral groove. However, this compound instead interacted with the valine chiral atom. In diffusion experiments with long diffusion times, the linearized diffusion plots for each analyte-molecular micelle mixture showed curvature characteristic of intermolecular cross relaxation. The magnitude of this effect scaled linearly with the analytes' free energies of binding.  相似文献

Two chiral phosphoramidates,(R)-(-)-1,1'-binaphthyl-2,2'-dihydroxy-N-[α-(S)-methylbenzyl] phosphoramidate and (-)-1,1'-biphenyl-2,2'-dihydroxy-N-[α-(S)-methylbenzyl]-phosphoramidate were synthesized.Their crystal structures were determined by X-ray single crystal diffraction analysis.The phosphoramidate molecules are self-associated by inter-molecular N-H...O = P hydrogen bonds and aromatic edge to face interactions.  相似文献

A new preparative method for enantiomerically pure 1,1′-bi-2-naphthols is described. 1,1′-Bi-2-naphtholboric anhydride generated from the reaction of racemic 1,1′-bi-2-naphthol and boric acid in toluene is reacted with (S)-proline to produce 1,1′-bi-2-naphtholboric proline anhydride. Its two diastereomers were efficiently separated in THF. After treating successively with NaOH, HCl, and recrystallizing from benzene, enantiomerically pure (S)- and (R)-1,1′-bi-2-naphthol were obtained in 71–79% yield and in 62–74% yield, respectively.  相似文献

The chiral recognition mechanism of a cinchona alkaloid based chiral selector for N-protected peptide enantiomers was investigated. A chiral stationary phase derived from this selector was employed for liquid chromatographic enantiomer separations. It showed exceptionally high enantiomer discrimination for the (all-R)- and (all-S)-enantiomers of dialanine (alpha = 20), while a pronounced loss of chiral recognition occurred upon the insertion of an additional alanine residue into the peptide backbone. This reduction of enantioselectivity was investigated in great detail by NMR spectroscopy of complexes of the chiral selector and the analyte enantiomers accompanied by molecular modeling studies. Investigation of intramolecular NOEs provided the conformational states of the free and complexed forms of the selector. The analysis of complexation-induced shifts yielded information on intermolecular interactions and allowed us to propose binding models, which were further supported by the observation of intermolecular NOEs, indicating the relative arrangements of selector and analytes. Stochastic molecular dynamics simulations were able to reproduce the chromatographic retention orders and energy differences, as well as the intermolecular NOEs. The computational data were used to evaluate the intermolecular forces responsible for analyte binding. In addition, the relative contributions of the fragments of the chiral selector to the enantioselective binding event were assessed. A spatial arrangement of the chiral selector and the analyte allowing the primary ionic interaction as well as hydrogen bonding and pi-pi-stacking to take place simultaneously was found to be essential to obtain very high enantioselectivities.  相似文献

(+)‐(1S)‐1,1′‐Binaphthalene‐2,2′‐diyl hydrogen phosphate (bnppa) is one of the useful optical selectors. To disclose the molecular mechanism by which bnppa recognizes aliphatic L ‐α‐amino acids and separates them by fractional crystallization, X‐ray analyses of bnppa and of its salts with L ‐alanine, L ‐valine, L ‐norvaline, and L ‐norleucine have been undertaken. All the amino acids adopt energetically favorable conformations in the crystal structures. The conformations and the packing patterns of bnppa in these crystal structures are very similar. The bnppa molecules are packed in a specific way to form hydrophobic and hydrophilic layers that are well separated. Between bnppa molecules, at the interface of these hydrophobic and hydrophilic layers, a space with chirality is formed. This space, designated as chiral space, recognizes the optically active amino acids. The packing of bnppa is mainly governed by intermolecular CH⋅⋅⋅π interactions between naphthalene moieties. The chiral space is responsible for the molecular recognition by bnppa allowing fractional crystallization of the L ‐α‐amino acids.  相似文献

Mixed mode (MM) separation using a combination of MEKC and polyelectrolyte multilayer (PEM) coatings is herein reported for the separation of achiral and chiral analytes. Many analytes are difficult to separate by MEKC and PEM coatings alone. Therefore, the implementation of a MM separation provides several advantages for overcoming the limitations of these well‐established methods. In this study, it was observed that achiral separations using MEKC and PEM coatings individually resulted in partial resolution of eight very similar aryl ketones when the molecular micelle (sodium poly(N‐undecanoyl‐L ‐glycinate)) concentration was varied from 0.25 to 1.00% w/v and the bilayer number varied from 2 to 4. However, when MM separation was introduced, baseline resolution was achieved for all eight analytes. In the case of chiral separations, temazepam, aminoglutethimide, benzoin, benzoin methyl ether, and coumachlor were separated using the three separation techniques. For chiral separations, the chiral molecular micelle, sodium poly(N‐undecanoyl‐L ‐leucylvalinate), was employed at concentrations of 0.25–1.50% w/v for both MEKC and PEM coatings. Overall, the results revealed partial separation with MEKC and PEM coatings individually. However, MM separation enabled baseline separation of each chiral mixture. The separation of achiral and chiral compounds from different compound classes demonstrates the versatility of this MM approach.  相似文献

用差紫外光谱滴定法考察了以脱氧胆酸作spacer的手性分子钳1~3对一系列α-氨基酸甲酯的对映选择性识别性能。结果表明,分子钳1和2与客体氨基酸甲酯形成1:1型超分子配合物,并显示较好的手性识别能力。分钳3对所考察的氨基酸甲酯均没有明显的识别作用。讨论了主-客体间尺寸/形状匹配、几何互补等因素对形成超分子配合物的影响,并利用计算机模拟作辅助手段对实验结果和现象进行了解释。  相似文献

Lysozyme is a well‐studied enzyme that hydrolyzes the β‐(1,4)‐glycosidic linkage of N‐acetyl‐β‐glucosamine (NAG)n oligomers. The active site of hen egg‐white lysozyme (HEWL) is believed to consist of six subsites, A‐F that can accommodate six sugar residues. We present studies exploring the use of polarizable force fields in conjunction with all‐atom molecular dynamics (MD) simulations to analyze binding structures of complexes of lysozyme and NAG trisaccharide, (NAG)3. MD trajectories are applied to analyze structures and conformation of the complex as well as protein–ligand interactions, including the hydrogen‐bonding network in the binding pocket. Two binding modes (ABC and BCD) of (NAG)3 are investigated independently based on a fixed‐charge model and a polarizable model. We also apply molecular mechanics with generalized born and surface area (MM‐GBSA) methods based on MD using both nonpolarizable and polarizable force fields to compute binding free energies. We also study the correlation between root‐mean‐squared deviation and binding free energies of the wildtype and W62Y mutant; we find that for this prototypical system, approaches using the MD trajectories coupled with implicit solvent models are equivalent for polarizable and fixed‐charge models. © 2012 Wiley Periodicals, Inc.  相似文献

Chirality discrimination at a binary toluene (organic)/water(aqueous) interface between R- or S-Tol-BINAP (2,2′-Bis(di-p-tolylphosphino)-1,1′-binaphthyl) molecules and the water-soluble serine chiral specie is examined for the first time, using a combination of interfacial tension measurements and molecular dynamic simulations. Experimental interfacial measurements exhibit a clear chirality-controlled difference when a homochiral versus a heterochiral enantiomeric pairs are introduced at the interfaces. The related molecular dynamics simulations support the experimental results and provide further molecular insight of intermolecular interactions at the interfaces. The results indicate that interfacial tension measurements can capture the preferential interactions which exist between different pairs of enantiomers at the binary interfaces, opening up a new way for probing chirality discrimination at liquid-liquid interfaces.  相似文献

Optically active 2,2′-bis(2-trifluoro-4-aminophenoxy)-1,1′-binaphthyl and its corresponding racemate were prepared by a nucleophilic substitution reaction of 1,1′-bi-2-naphthol with 2-chloro-5-nitrotrifluorotoluene and subsequently by the reduction of the resulting dinitro compounds. A series of optically active and optically inactive aromatic polyimides also were prepared therefrom. These polymers readily were soluble in common organic solvents such as pyridine, N,N′-dimethylacetamide, and m-cresol and had glass-transition temperatures of 256 ∼ 278 °C. The specific rotations of the chiral polymers ranged from 167 ∼ 258°, and their chiroptical properties also were studied. © 1999 John Wiley & Sons, Inc. Two novel amino acid based surfactants sodium N-(4-n-decyloxybenzoyl)-l-valinate (SDeBV) and sodium N-(4-n-octyloxybenzoyl)-l-valinate (SOBV) have been synthesized and used as chiral selectors for enantiomeric separations by micellar electrokinetic chromatography (MEKC). The aggregation behavior of the surfactants was studied in buffered aqueous solution using surface tension and fluorescence probe techniques. The microenvironment of the aggregates was studied using pyrene, and 1,6-diphenyl-1,3,5-hexatriene (DPH) as probe molecules. Results of these studies indicate that these two surfactants form micelles in buffered aqueous solution. Successful enentioseparation has been achieved for 1,1′-bi-2-naphthol (BOH), 1,1′-binaphthyl-2,2′-diylhydrogenphosphate (BNP), 2,8-dimethyl-6H-5,11-methanodibenzo[b,f] [1,5]diazocine (Tröger's base, TB), and benzoin (BZN) using the two chiral selectors SDeBV and SOBV. The separations were optimized with respect to surfactant concentration, pH, and buffer concentration. The results are discussed in light of the aggregation behavior of the surfactants. A comparison of the results of this study has been made with the data from literature to investigate the effect of self-assembly morphology on enantiomeric separations.  相似文献
