A review of sample preparation methods for organic acids in biological fluids, in particular serum and urine, is presented. It covers techniques on organic acid determination without sample preparation, release of organic acids from binding locations, removal of proteins by protein precipitation and ultrafiltration, isolation of the organic acids by liquid-liquid and liquid-solid extraction, purification of the extract, derivatization and pre-fractionation. The various alternative sample preparation steps are compared and critically discussed. Examples of applications including profile analysis of organic acids by gas chromatography (GC), determination of particular organic acids by GC or liquid chromatography and determination of fatty acids as a distinct chemical class of acids demonstrate that the kind of sample preparation chosen depends strongly on the analytical aims.  相似文献

A novel and reliable high-performance liquid chromatographic method for trace short-chain organic acids measurement in wheat root exudates under aluminum stress has been optimized and validated. The chromatographic separation of the short-chain organic acids (citric, oxalic, malonic, succinic, tartaric, malic, and acetic acids) was achieved with Bio-rad Aminex HPX-87H cation exchange resin column. These seven organic acids were identified and quantified in 25 min. Well-shaped peaks were obtained for trace organic acids using dilute sulfuric acid as mobile phase. Under optimum conditions, Bio-rad Aminex HPX-87H column showed distinct advantages of the ability to well separate different short-chain organic acids (especially for tartaric and malic acids, as well as malonic and acetic acids) in wheat root exudates under aluminum stress, and offered accurate and precise results for the analysis of these organic acids. This HPLC method can efficiently eliminate the aluminum’s interference and is quite suitable to the trace detection of trace organic acids in wheat root exudates under aluminum stress.

利用减压蒸溜法分离废液中的低沸点有机酸,结合红外光谱、气相色谱、质谱,确定了废液中以乙酸、丙酸为主的10种有机酸的结构,其含量占分析废液的2.64%。其中丙酸含量最高,其次为乙酸。同时被检测到的还有乙醇,乙醛等。另外通过柱层析分离高级脂肪酸,并进行衍生化处理,经GC、GC-MS检测,确定了四种高沸点有机酸的结构,含量为1.71%。9,12-十八二烯酸含量相对较高。因此有机酸占废液总量的质量分数为4.35%。  相似文献

Altogether 143 of the organic acids regularly occurring in urine of healthy individuals are identified as methyl esters by gas chromatography-mass spectrometry with respect to their complete chemical structures. They are classified as dicarboxylic acids, oxocarboxylic acids, hydroxycarboxylic acids, aromatic acids, furancarboxylic acids, nitrogen-containing acids and acid conjugates. By pre-fractionating the complex mixture of the total organic acids, peak overlap is minimized, and substances in low concentrations can also be detected and identified. The qualitative patterns of the urinary organic acids in the fractions are constant and reproducible, and in many cases a reliable identification of organic acids is possible by gas chromatography alone, using methylene units and separation on OV-1701 capillary columns.  相似文献

This article proposes a simple and sensitive HPLC method with photo‐diode array detection for the analysis of organic acids, monomeric polyphenols and furanic compounds in wine samples by direct injection. The chromatographic separation of 8 organic acids, 2 furans and 22 phenolic compounds was carried out with a buffered solution (pH 2.70) and acetonitrile as mobile phases and a difunctionally bonded C18 stationary phase, Atlantis dC18 (250×4.6 mm, 5 μm) column. The elution was performed in 12 min for the organic acids and in 60 min for the phenolic compounds, including phenolic acids, stilbenes and flavonoids. Target compounds were detected at 210 nm (organic acids, flavan‐3‐ols and benzoic acids), 254 nm (ellagic acid), 280 nm (furans and cinnamic acid), 315 nm (hydroxycinnamic acids and trans‐resveratrol) and 360 nm (flavonoids). The RSD for the repeatability test (n=5) of peak area and retention times were below 3.1 and 0.3%, respectively, for phenolics and below 1.0 and 0.2% for organic acids. The RSDs expressing the reproducibility of the method were higher than for the repeatability results but all below 9.0%. Method accuracy was evaluated by the recovery results, with averaged values between 80 and 104% for polyphenols and 97–105% for organic acids. The calibration curves, obtained by triplicate injection of standard solutions, showed good linearity with regression coefficients higher than 0.9982 for polyphenols and 0.9997 for organic acids. The LOD was in the range of 0.07–0.49 mg/L for polyphenols (cinnamic and gallic acids, respectively) and 0.001–0.046 g/L for organic acids (oxalic and lactic acids, respectively). The method was successfully used to measure and assess the polyphenolic fingerprint and organic acids profile of red, white, rosé and fortified wines.  相似文献

End-product identification of organic pollutants during oxidation water treatments is of environmental concern due to their potential toxicity. In this work the coupling of ion chromatography (IC), equipped with a membrane ion suppressor, and electrospray ionization mass spectrometry (ESI-MS) has been successfully exploited for the identification of novel low molecular weight organic acids formed as final by-products of 2,4-dichloro-phenol (DCP) degradation by Fenton's reagent, an oxidation method widely employed to degrade recalcitrant organic pollutants in industrial wastewater. The IC-ESI-MS analytical set-up not only allowed the detection of known organic acids but also the identification of 13 new low molecular weight organic acids as a result of extensive oxidation of the parent organic pollutant. Some of the novel organic acids were shown to be chlorine containing by-products and, consequently, of environmental concern. The analytical set-up could be used for characterizing the unknown organic carbon fraction arising from oxidation water treatments.  相似文献

本文研究了气相色谱分析羊水中痕量有机酸的各种预分离方法。对各种结构类型的标准有机酸水溶液进行回收实验以及实际的羊水样品分析表明,硅酸吸附法具有比较全面的优越性,是研究羊水中痕量有机酸较为理想的预分离方法。离子交换法对大多数有机酸也具有较高的回收率,但它的操作过程比较冗长。溶剂萃取法对柠檬酸、甘油酸等某些多元酸或羟基酸的回收不很理想,但对其它许多有机酸也可以取得较好的结果。所研究的方法均能分离除去羊水中大量的磷酸盐和硫酸盐,消除它们对有机酸气相色谱分析的干扰。  相似文献

测定了25种成品卷烟的15种有机酸含量,采用逐步回归分析法建立了卷烟感官风格品质与有机酸之间的关系模型。根据逐步回归模型选择的变量,评价影响卷烟香韵表现与感官品质的关键有机酸成分。将回归模型成功地应用于试制卷烟香韵表现和感官品质得分预测,证明了模型的有效性及有机酸对卷烟香韵表现和感官品质的解释能力。研究结果表明,异戊酸、丙二酸、苯乙酸和柠檬酸等有机酸对卷烟香韵表现或感官品质存在正面或负面的不同影响。  相似文献

Chromatography has played a pivotal role in the advances made during the last 30 years in our knowledge of inborn errors of metabolism. This review discusses the application of some of these techniques to the analysis of organic acids and acylcarnitines. The separation of organic acids needed a comprehensive approach that would permit all of the many organic acids present in urine or other complex mixtures to be extracted, analysed and identified in a single run. This required analytical methods of great resolving power, wide linear range and universal detectors such as gas chromatography (GC), or GC coupled with mass spectrometry. Sample preparation was another problem that has been tackled by a variety of approaches. Organic solvents have been employed widely for the extraction of organic acids from physiological fluids. Unfortunately, recoveries of the different organic acids by this method are sometimes less than quantitative and variable depending on the compound. Other methods, such as the use of DEAE-Sephadex columns, have the advantage of resulting in close to 100% recoveries, but are more tedious. Liquid partition chromatography on short silicic acid columns has also been recommended as a useful clean-up step prior to GC, permitting both the identification and quantitation of organic acids in urine, plasma or amniotic fluid. Although many derivatization procedure have been used to prepare organic acids for gas chromatography, the most common is trimethylsilylation. Oxo acids are usually reacted with one of several commonly used reagents to form oximes. Based on differences in acidity, organic acids and phenols were enriched by pH gradient extraction method from walnut shell pyroligneous acid. Contents of organic acids and phenols were measured by acid-base titration method and Folin colorimetric assay, respectively to assess the effectiveness of the extraction. In addition, the antimicrobial activity of the organic acids and antioxidant activity of phenols extracted were investigated. Chemical components of the extracts that were from the optimal concentrations of NaHCO3 and NaOH were measured by GC-MS. The results showed that 5% NaHCO3 could enrich the highest amount of organic acids, whereas 4% NaOH could enrich the highest amount of phenols. The enriched organic acids exhibited high antimicrobial activity, and the enriched phenols exhibited antioxidant activity under low concentrations, and demonstrated dosage dependency.  相似文献

Two well known derivatization methods, butylation and methylation, are compared by gas chromatography (GC)-mass spectrometry (MS) to identify organic acids in Bayer liquors. These two derivatization methods should be combined together for the determination of the carboxylic acids. Twenty-four organic acids are identified by GC-MS and 13 organic acids are firstly found in Bayer liquors. The retention times and the carbon number of these seven n-dicarboxylic acids (C(4)-C(10)) are fit into the linear relationship in Microsoft Excel.  相似文献

We describe a simple and sensitive liquid chromatographic method for the analysis of organic acids using on-line complexation with copper(II) ion. Organic acids complexed with copper(II) ion were separated on a reversed-phase C18 column and detected by UV absorption at 240 nm. The copper(II) ion concentration in the mobile phase had a great influence on separation and sensitivity. A mobile phase consisting of 10 mM copper(II) sulfate in 5 mM sulfuric acid (pH 2.3) was used to separate nine organic acids (tartaric, malic, malonic, lactic, acetic, citric, maleic, succinic and fumaric acids). The detection limits of the examined organic acids calculated at S/N = 3 ranged from 0.6 to 100 μM. The detector signal was linear over three orders of magnitude of organic acid concentration. The method successfully measured organic acids in juice and vinegar samples.  相似文献

烟草中的多元酸对抽吸品质有重要影响,它们能与生物碱结合成盐,调节烟草中碱性成分的挥发性。高级脂肪酸可分成饱和与不饱和两类,饱和脂肪酸能增加烟气的脂肪味、腊味并使之圆和;不饱和脂肪酸增加烟气的丰满度和粗糙感。卷烟在烘丝过程中由于温度、湿度的变化而引起烟草内部化学成分的变化。本文应用甲酯衍生化和GC-MS法,对卷烟加工过程中烘丝工序前后的多元酸和高级脂肪酸进行了定性定量分析。为卷烟配方和加工工艺参数的优化提供理论依据。  相似文献
