The applicability of comprehensive two-dimensional gas chromatography (GC×GC) for flavonoids analysis was investigated by separation and identification of flavonoids in standards, and a complex matrix natural sample. The modulation temperature was optimized to achieve the best separation and signal enhancement. The separation pattern of trimethylsilyl (TMS) derivatives of flavonoids was compared on two complementary column sets. Whilst the BPX5/BPX50 (NP/P) column set offers better overall separation, BPX50/BPX5 (P/NP) provides better peak shape and sensitivity. Comparison of the identification power of GC×GC-TOFMS against both the NIST05 MS library and a laboratory (created in-house) TOFMS library was carried out on a flavonoid mixture. The basic retention index information on high-performance capillary columns with a non-polar stationary phase was established and database of mass spectra of trimethylsilyl derivatives of flavonoids was compiled. TOFMS coupled to GC×GC enabled satisfactory identification of flavonoids in complex matrix samples at their LOD over a range of 0.5-10 μg/mL. Detection of all compounds was based on full-scan mass spectra and for each compound a characteristic ion was chosen for further quantification. This study shows that GC×GC-TOFMS yields high specificity for flavonoids derived from real natural samples, dark chocolate, propolis, and chrysanthemum.  相似文献

通过对升温速度、二维补偿温度、调制周期等关键实验参数的优化,建立了全二维气相色谱-飞行时间质谱(GC×GC-TOF MS)分析重馏分油中芳烃组分的方法,得到了重馏分油芳烃组分按环数分布的二维点阵图。根据谱库检索、标准化合物对照及文献报道,对重馏分油芳烃组分中菲、甲基菲及芘、苯并蒽等常见多环芳烃(PAH)进行了准确定性,并将该方法应用到重馏分油加氢处理工艺研究中,对菲、芘的加氢处理产物进行了定性分析。该研究为重馏分油芳烃组分的准确定性提供了新的技术手段,为加深对油品加氢规律的认识提供了技术支持。全二维气相色谱与普通一维色谱对比,在重馏分油的芳烃组分分析上体现了极大优势。  相似文献

Comprehensive gas chromatography (GC) has emerged in recent years as the technique of choice for the analysis of volatile and semivolatile compounds in complex matrices. Coupling it with high‐resolution mass spectrometry (MS) makes a powerful tool for identification and quantification of organic compounds. The results obtained in this study showed a significant improvement by using GC×GC‐EI‐MS in comparison with GC‐EI‐MS; the separation of chromatogram peaks was highly improved, which facilitated detection and identification. However, the limitation of Orbitrap mass analyzer compared with time‐of‐flight analyzer is the data acquisition rate; the frequency average was about 25 Hz at a mass resolving power of 15.000, which is barely sufficient for the proper reconstruction of the narrowest chromatographic peaks. On the other hand, the different spectra obtained in this study showed an average mass accuracy of about 1 ppm. Within this average mass accuracy, some reasonable elemental compositions can be proposed and combined with characteristic fragment ions, and the molecules can be identified with precision. At a mass resolving power of 7.500, the scan rate reaches 43 Hz and the GC×GC‐MS peaks can be represented by more than 10 data points, which should be sufficient for quantification. The GC×GC‐MS was also applied to analyze a cellulose bio‐oil sample. Following this, a highly resolved chromatogram was obtained, allowing EI mass spectra containing molecular and fragment ions of many distinct molecules present in the sample to be identified.  相似文献

Systemic lupus erythematosus (SLE) is an autoimmune disease with heterogeneous organ and system manifestations. In this study, urinary metabolic alterations related to SLE were investigated by performing gas chromatography/mass spectrometry (GC/MS) based metabolomics and multivariate statistical analysis. Patients with SLE and healthy controls could be clearly differentiated in view of the metabolic abnormity in urine. Among 70 identified endogenous metabolites, 23 metabolites were dramatically increased in SLE patients, which involved in several key metabolic pathways including energy metabolism, nucleotide metabolism, oxidative stress and gut‐microbiome‐derived metabolism. This noninvasive and GC/MS‐based metabolomic technique is a promising and potent strategy for identifying novel biomarkers and understanding pathogenesis of SLE. Pyrolysis gas chromatography (P/GC) has been successful in differentiating coal tar pitches which are used in the manufacture of high temperature graphite materials [1,2]. In the present study, coal tar and petroleum pitches received from five suppliers were readily differentiated using three distinct methods of polymer analysis: dynamic headspace and pyrolysis gas chromatography mass spectrometry (DH/GC/MS and P/GC/MS, [3]), and pyrolysis mass spectrometry (P/MS, [4,5]). All of these methods are appropriate for the analysis and pattern recognition study of coal and petroleum products; however, methods which incorporate GC usually necessitate long analysis times and are subject to problems associated with gas chromatographic reproducibility (e.g. active sites [6], and nonconstant retention times). As a fingerprint analysis method, P/MS is rapid and ideally suited to computerized pattern recognition study of pitches due to the simple format of P/MS data (computer tabulation of normalized intensities versus masses); whereas, in the present study, the normalized peak area versus retention time date from DH/GC/MS and P/GC/MS analyses were not suitable for direct input to a computerized pattern recognition analysis program, (due to non-constant retention times and improper data format). One limitation is that P/MS does not provide positive identification of thermally evolved products (as can DH/GC/MS and P/GC/MS) due to the complexity of the spectra of a mixture of compounds produced by P/MS. Within the limits of the commercial instrumentation and computer programs used in this study, a combination of all three techniques provided the greatest capability for the differentiation of the five coal tar and petroleum based pitches studied.  相似文献

The National Institute of Standards and Technology (NIST) Automated Mass Spectral Deconvolution and Identification System (AMDIS) is applied to a selection of data files obtained from the gas chromatography/mass spectrometry (GC/MS) analysis of urinary organic acids. Mass spectra obtained after deconvolution are compared with a special user library containing both the mass spectra and retention indices of ethoxime-trimethylsilyl (EO-TMS) derivatives of a set of organic acids. Efficient identification of components is achieved and the potential of the procedure for automated diagnosis of inborn errors of metabolism and for related research is demonstrated.  相似文献

The aim of this study was to retrospectively diagnose and confirm inherited metabolic diseases (IMD), from a small population of IMD high‐risk patients, with the aid of gas chromatography–tandem mass spectrometry (GC–MS/MS), technologies yet to be popularized in Sichuan, China. Using GC–MS/MS coupled with clinical diagnosis, we retrospectively analyzed samples of dried blood spots and urine specimen from 183 IMD high‐risk infant patients, who visited the West China Second Hospital of Sichuan University between June 2013 and October 2015. Four out of 183 IMD high‐risk infant patients were finally diagnosed to be IMD positive, among which two patients were identified with phenylketonuria, one with maple syrup urine disease, and 1 with methylmalonic academia. Restrictive diets and other symptomatic treatments were employed to treat the confirmed infant patients whose conditions are still under tracking and there are zero cases of death so far. GC–MS/MS was found to be an efficient and reliable way to detect IMD. It is necessary to apply GC–MS/MS, in addition to other clinical approaches, for diagnosing candidate IMD patients so that the confirmed patients can get medical intervention and timely treatment.  相似文献

建立了孔石莼脂肪酸的气相色谱/质谱(GC/MS)测定方法。使用Folch法提取了孔石莼中的总脂,经过2 mol/L HCl-甲醇溶液的甲酯化处理后,采用GC/MS法对其脂肪酸组成进行了分离分析,同时结合有机质谱学规律,分别对饱和脂肪酸甲酯、单不饱和脂肪酸甲酯和多不饱和脂肪酸甲酯的裂解规律和质谱特征进行了分析归纳。通过质谱数据库检索和标准品对照,鉴定出孔石莼中的24种脂肪酸,其中9,12,15-十八碳三烯酸、4,7,10,13-十六碳四烯酸和6,9,2,15-十八碳四烯酸3种主要多不饱和脂肪酸占总脂肪酸含量的45.14%。通过对孔石莼中脂肪酸的分析,表明特征离子在脂肪酸甲酯尤其是多不饱和脂肪酸甲酯的定性方面具有很好的应用价值。  相似文献

GC/MS定性分析中,由于同分异构体的质谱图相似,常常出现结构鉴定错误.同分异构体化合物的保留行为具有较大地差别,采用文献保留指数和保留温度测定值回归分析方法对2篇已发表论文进行了研究,发现了对单萜烯类同分异构体结构鉴定中存在的错误,并提出可能的正确结构.文献保留指数和保留温度测定值回归分析方法可以简化GC/MS色谱峰的定性分析过程,提高定性分析的准确性,在样品定性分析中具很强的使用价值.  相似文献

This study describes the use of high performance liquid chromatography (HPLC) and capillary gas chromatography/mass spectrometry (GC/MS) in the characterization of polar glucuronide conjugates of doxylamine and their subsequent aglycones following deconjugation. Rat urinary extracts which contained doxylamine and both nonconjugated and conjugated doxylamine metabolites, were examined by HPLC before and after incubation with rat intestinal microflora. The subsequent deconjugated urinary metabolites and the nonconjugated products remaining in the urinary extracts were then isolated, acetylated, and assayed by GC/MS. Incubation with the intestinal microflora indicated that anaerobic bacteria were capable of effecting hydrolytic cleavage of these polar O-glucuronide metabolites of doxylamine and its demethylated products to their subsequent aglycones. GC/MS analysis was performed using a fused silica DB-5 GC column and was utilized for the identification of these deconjugated metabolites.  相似文献

建立了一种气相色谱(GC)与单光子电离-飞行时间质谱(SPI-TOF MS)联用(GC/SPI-TOF MS)的分析方法。首先,设计了一种双层套管的传输管用于连接GC与SPI-TOF MS,实现了GC与单光子电离离子源的无缝连接。在此基础上,以n-十五烷标准品和苯/甲苯/二甲苯的标准气为对象,对电离源的重要电压参数进行了优化,得到了纯净的分子离子峰,实现了对各类有机物的快速和准确定性。最后,将该方法用于分析柴油中的挥发性与半挥发性有机物,获得了柴油组分的二维GC×SPI-TOF MS谱图。不需要复杂的谱图解析和数据处理,根据谱图中离子的质荷比(m/z)归纳了柴油的主要成分,包括脂肪烃、芳香烃和含量很低的苯并吡咯等含氮化合物;根据色谱的保留时间将柴油中的同分异构体区分开来。结果表明GC/SPI-TOF MS法是一种简单、有效的分析方法,非常适于柴油及复杂环境样品等的分析表征。  相似文献

采用气相色谱/质谱方法,对渤海海上4个不同区块、5个平台的6口油井原油进行了烷基化多环芳烃系列化合物和美国环保署(EPA)优先控制多环芳烃系列化合物的准确定性定量分析。通过多环芳烃原始指纹谱图、多环芳烃组分分布模式和特征比值的比较对上述原油进行鉴别。结果证明不同区块的原油中多环芳烃指纹信息不尽相同,即使在同一平台不同油井中所产的原油其指纹也存在一定差异。为确保原油鉴别的准确性,分析过程中必须在仪器的稳定性和样品前处理方面实施严格的质量控制措施。  相似文献

Combining qualitative data from the chromatographic structure of 2-D gas chromatography with flame ionization detection (GC×GC-FID) and that from gas chromatography-mass spectrometry (GC/MS) should result in a more accurate assignment of the peak identities than the simple analysis by GC/MS, where coelution of analytes is unavoidable in highly complex samples (rendering spectra unsuitable for qualitative purposes) or for compounds in very low concentrations. Using data from GC×GC-FID combined with GC/MS can reveal coelutions that were not detected by mass spectra deconvolution software. In addition, some compounds can be identified according to the structure of the GC×GC-FID chromatogram. In this article, the volatile fractions of fresh and dehydrated pineapple pulp were evaluated. The extraction of the volatiles was performed by dynamic headspace extraction coupled to solid-phase microextraction (DHS-SPME), a technique appropriate for slurries or solid matrices. Extracted analytes were then analyzed by GC×GC-FID and GC/MS. The results obtained using both techniques were combined to improve compound identifications.  相似文献

Analytical methods that are used for the detection and identification of chemicals related to the Chemical Weapons Convention in complex organic solutions were developed. A matrix modified by the addition of complex diesel oil background compounds and interferences was used for the development of a gas chromatography-mass spectrometry or gas chromatography-tandem mass spectrometry (GC-MS or GC-MS/MS) method for unambiguous identification of the scheduled chemicals. Co-elution of diethyl N,N-dimethylphosphoramidate and ethyl N,N-dimethylphosphoramidocyanidate, which are not separated by GC, was identified by GC-MS. Although GC-MS can identify the compounds, GC-MS/MS identification has greater accuracy and sensitivity in the case of complex matrixes. Four scheduled chemicals were accurately and selectively identified against numerous interfering substances by GC-MS/MS. The fragmentation chemistry of these compounds using MS/MS analysis was investigated.  相似文献

The separation provided by conventional gas chromatography (1D-GC) can be significantly enhanced by using comprehensive two-dimensional GC (GC X GC) instead. Combination with mass spectrometric detection is desirable for unambiguous confirmation of target compounds and the provisional identification of unknowns. A GC X GC system using a cryogenic modulator was coupled to a time-of-flight mass spectrometric (TOF MS) detector. With the determination of pesticides in vegetable extracts as an example, it was demonstrated that GC X GC improves the separation dramatically. All 58 pesticides of interest could be identified using their full-scan mass spectra, which was not possible when using ID-GC-TOF MS. In addition, the high scan speed of the TOF MS allowed the deconvolution of compounds partly co-eluting in GC X GC.  相似文献

采用顶空固相微萃取(HS-SPME)-气相色谱/质谱法(GC/MS)测定了牛粪、猪粪以及鸡粪中的挥发性有机化合物(VOCs)。在优化HS-SPME条件的基础上,通过GC/MS分析,从牛粪中分离鉴定出44种VOCs,从猪粪中分离鉴定出40种VOCs,从鸡粪中分离鉴定出41种VOCs。HS-SPME-GC/MS具有简单、快速、无需有机溶剂等特点,适合于畜禽粪便中挥发性有机化合物的分析与鉴定。同时,该研究揭示了畜禽养殖场臭味物质的来源,为治理其环境污染提供了科学依据。  相似文献
