以1个常规六通阀直接连接两支常规尺寸的色谱柱(250 mm×4.6 mm i.d.),构建简单的SCX/RP在线二维液相色谱系统,对中成药六味地黄丸组分进行了优化分离.样品经过第一维阳离子交换色谱(Hypersil SCX),洗脱产物分离后通过六通阀直接富集到反相分析柱(C18)顶端,被转移到第二维色谱柱上继续进行分离.经过11步不连续的线性梯度洗脱,二维分离系统出峰数量达到550多个,峰容量达到2266.构建的二维液相色谱系统结构简单,与一维色谱相比,具有分辨率高、峰容量大的特点.  相似文献

以GFC/RP模式构建全二维液相色谱系统,第一维凝胶过滤色谱柱使用ShodexProteinKW 802. 5(300mm×8mmi.d. ),以0. 2mol/LNaH2PO4 (pH7. 0)的流动相在0. 15mL/min的流速下等度洗脱,第二维反相色谱柱使用HypersilBDSC18 (35mm×4. 6mmi.d. ),在3mL/min的流速下梯度洗脱。采用平行柱交替分析的形式作切换接口, 2. 5min切换一次,两个反相柱交替富集、分析第一维洗脱产物。以5个标准蛋白混合物的分离评价该系统,在单独一维模式中不能分离的样品在全二维液相色谱中得到了较好的分离,二维系统的总峰容量为225。与一维色谱相比,系统的总峰容量、分辨率得到较大提高。并用于山羊血清的纯化分析,对一维分离中的重合谱峰进行验证,对制备纯化有一定的实际意义。  相似文献

1 多维分离技术新进展 一维色谱是目前最常用的分离分析方法,然而对于复杂体系如蛋白质组,采用一维分离模式其分离度远远不能满足要求.Giddings理论告诉我们:对于分离机理相互正交的二维分离系统(如色谱),峰的容量应该为两个色谱柱峰容量的乘积.因此,多维分离系统是解决复杂分离体系的一个最佳选择.在多维色谱中二维气相色谱发展较快,目前全二维气相色谱仪业已商品化,其峰的容量达到104以上.而二维液相色谱,尤其是正相/反相二维液相色谱技术发展较为缓慢,其主要的技术瓶颈在于第一维色谱(正相)分离后的流动相严重干扰第二维色谱(反相)的分离.  相似文献

以十通阀为切换接口, 构建了SCX/RP常规柱二维液相色谱系统, 并以珠蛋白水解产物的分析对其加以评价. 样品首先由第一维阳离子交换色谱(Hypersil SCX, 100 mm×4.6 mm I.D.)在pH 4.0的磷酸盐缓冲体系中分离, 洗脱产物进入切换接口, 样品组分被富集在捕集柱(Hypersil BDS C18, 15 mm×4.6 mm I.D.)中, 进一步脱盐后被导入第二维反相色谱(Hypersil BDS C18, 250 mm×4.6 mm I.D.)分离分析. 阳离子交换色谱采用逐步增加盐浓度的12步台阶等度间断方式洗脱, 每次将洗脱产物捕集在捕集柱中进而由反相色谱分析, 实现对第一维洗脱产物的切割转移及第二维分析. 与一维色谱相比, 二维液相色谱系统的分辨率、峰容量也得到提高, 系统峰容量达到2280.  相似文献

对毛细管高效液相色谱-毛细管电泳二维分离的动态脉冲接触式接口系统进行了最优化研究。讨论了它的设计原理,优化了操作条件,并对其性能进行了评价。利用这种接口,使样品在第二维的进样浓度达到第一维流出浓度的81%,100次连续进样峰高及迁移时间的RSD平均值分别为3.0%、1.8%,进样时间也得以缩短,并对甘草的水提取物实现了高效二维分离。,  相似文献

毛细管电色谱(CEC)以内含色谱固定相的毛细管为分离柱,以电渗流为驱动力,既可以分离带电物质也可以分离中性物质。它结合了毛细管电泳和高效液相色谱两者的优点,兼具高柱效、高分辨率、高选择性和高峰容量的特点,同时具有色谱和电泳的双重分离机理。然而,“纯粹”的电色谱在实际应用中有着天然的弱点,即: 在电流通过毛细管柱中的流动相时容易产生气泡(焦耳热作用),从而使电流中断和电渗流停止,毛细管柱必须被重新用流动相润湿后方能再次使用。加压毛细管电色谱(pCEC)将液相色谱中的压力流引入CEC系统中,不仅解决了气泡、干柱等问题,而且实现了定量阀进样和二元梯度洗脱。CEC和pCEC作为微分离领域的两种前沿技术,满足了当前复杂样品分析和分析仪器微型化的需求,近年来获得了广泛的关注。本文综述了这两种技术近来的发展,包括仪器、色谱固定相的发展,总结了其在生命科学、药物分析、食品安全以及环保样品分析等方面的应用进展,评述了各方法的特点,并展望了CEC和pCEC今后的发展和应用前景。  相似文献

建立了二维液相色谱-质谱联用方法分离中药复方葛根芩连汤的成分。以CN柱作第一维色谱柱,水和甲醇梯度洗脱分离;以ODS柱作第二维色谱柱,20 mmol/L乙酸铵缓冲液和乙腈梯度洗脱分离;质谱检测采用电喷雾电离/大气压化学电离(ESI/APCI)复合离子源,正负离子扫描。实验结果表明搭建的二维液相色谱的峰容量显著高于一维色谱,分离效率得到了明显的提高。以第一维色谱的第3个流分为例,对其二维分离进行仔细分析,发现质谱比紫外光谱检测到的组分多,质谱中采用负离子模式比正离子模式检测到的组分多。表明搭建的二维液相色谱-质谱分离平台分离效果好,提高了液相色谱的峰容量和分离效率。该方法操作简便,可作为中药等复杂体系分离分析的有效手段。  相似文献

搭建了一个纳升级的二维液相色谱分离平台(nano-2D-LC),该平台可以自动完成进样、除盐、分离及鉴定。以离子交换色谱(SCX)为第一维,反相液相色谱(RPLC)为第二维,对鼠肝组织的蛋白质组进行了研究。SCX采用阶梯式洗脱,RPLC运用线性梯度洗脱,以200 nL/min的速度进行分离,峰容量可达620。  相似文献

A 2-D separation platform was constructed using micro strong cation-exchange liquid chromatography (μ-SCXLC) and reversed-phase pressurized capillary electrochromatography (RP-pCEC) for the analysis of complex samples. Samples were fractionated by the first-dimension μ-SCXLC with a linear solvent gradient and then injected into the second-dimension RP-pCEC for further separation. The μ-SCXLC/RP-pCEC 2-D system with three separation mechanisms, namely strong cation-exchange, reversed-phase chromatography and electrophoresis, provided high selectivity, high resolution and high peak capacity compared to one-dimensional chromatographic approaches. Separation effectiveness of this 2-D system was demonstrated by the analysis of different kinds of complex samples, such as traditional Chinese medicine Cortex Phellodendri, bovine serum albumin (BSA) tryptic digest and real serum tryptic digest. A theoretical peak capacity of approximately 1200 was achieved, which proves its promising potential for the separation and analysis of complex samples.  相似文献

A comparison of different separation methods (high-performance liquid chromatography (HPLC), capillary HPLC (CHPLC) and pressurized capillary electrochromatography (pCEC)) coupled on-line with mass spectrometry (MS) is undertaken using the separation of a crude extract of ergot fungus (secalis cornuti) as an example. New and simple setups for a two-dimensional CHPLC coupled on-line with electrospray ionization (ESI)-MS (2D-CHPLC-MS) as well as for capillary size-exclusion chromatography performed under pCEC conditions and coupled on-line with ESI-MS (CSEC-pCEC-MS) are shown. 以加压毛细管电色谱(pCEC)为技术平台,对其在建立中药黄柏指纹图谱中的方法学进行了研究.通过对提取溶剂、流动相中有机相种类、盐溶液等条件的优化,发现1%盐酸的甲醇溶液为提取溶液,20mmol/LNH4C1溶液.乙腈为流动相梯度洗脱,紫外检测波长为230nm时对其分离效果最好.并通过在色谱柱上施加不同的电压,详细地阐明了pCEC的双重分离机制对分离选择性的影响,发现黄柏中的主要成分药根碱、巴马汀和小檗碱在pCEC模式中随电压的不同,有不同的出峰顺序.当电压为0—4kV时出峰顺序为药根碱、巴马汀和小檗碱,当电压为8—14kV时出峰顺序为药根碱、小檗碱和巴马汀.对此原因进行了详细讨论,同时与微径液相色谱模式进行了比较,说明pCEC可以为复杂样品的分离提供更多更好的分离途径.  相似文献

A novel gradient pressurized capillary electrochromatography (pCEC) instrument was developed to separate peptides.Two gradient elution modes,hydrophobic and hydrophilic interaction mode in pCEC,were performed on this instrument.Baseline separation of six peptides was obtained on two gradient modes with C18 column and strong cationic exchange column respectively.The effects of mixer volume and total frow rate of pumps on resolution were also discussed.  相似文献

A pressurized electrochromatography (pCEC) instrument with gradient capability was used in this work for separation of peptides. Three separation modes, namely, pCEC, high-performance liquid chromatography and capillary electrophoresiscan be carried out with the instrument. In pCEC mode, the mobile phase is driven by both electroosmotic flow and pressurized flow, facilitating fine-tuning in selectivity of neutral and charged species. A continuous gradient elution can be carried out conveniently on this instrument, which demonstrates that it is more powerful than isocratic pCEC for separation of complicated samples. The effects of applied voltage, supplementary pressure and ion-pairing agents on separation of peptides in gradient pCEC were investigated. The effects of flow-rate of the pump and the volume of the mixer on resolution were also evaluated.  相似文献

A mixture of unsaturated fatty acid methyl esters was separated with a new splitless capillary set-up. With the employed apparatus configuration different capillary separation techniques such as capillary high-performance liquid chromatography (cHPLC), capillary electrochromatography (CEC) and pressurized capillary electrochromatography (pCEC) could be applied. The detection and identification of the sample compounds were accomplished by hyphenating these capillary separation techniques with nuclear magnetic resonance (NMR) spectroscopy using a novel configuration of the detection capillary set-up. Using modified electrokinetically driven separation techniques, the electric field was applied solely across the separation column. With this improved interface for capillary liquid chromatography-NMR on-line coupling, the stereochemical assignment of the cis and trans configuration of unsaturated fatty acids could be easily accomplished. Microwave irradiation can provide a viable alternative to the traditional means such as ultraviolet light and thermal initiation for the preparation of monolithic capillary columns. Polystyrene-based monolithic stationary phases were prepared in situ in fused-silica capillaries and simultaneously in vials. The column permeability, electrophoretic and chromatographic behavior were evaluated using pressure-assisted capillary electrochromatography (pCEC), capillary electrochromatography (CEC) and low pressure liquid chromatography (LPLC). With an optimal monolithic material, the largest theoretical plates for preparing the column could be close to 18,000 plates/m for thiourea in the mode of pCEC. Furthermore, the influence of the composition of the porogenic solvents (toluene/isooctane) on the morphology of organic-based monoliths [poly(styrene-divinylbenzene-methacrylic acid)] was systematically studied with mercury intrusion porosimetry and scanning electron microscopy. The monoliths which were prepared with a high content of isooctane had a bigger pore size and better permeability, and hence resulted in a faster separation.  相似文献

Submicron, non‐porous, chiral silica stationary phase has been prepared by the immobilization of functionalized β‐CD derivatives to isocyanate‐modified silica via chemical reaction and applied to the pressurized capillary electrochromatography (pCEC) enantio‐separation of various chiral compounds. The submicron, non‐porous, cyclodextrin‐based chiral stationary phases (sub_μm‐CSP2) exhibited excellent chiral recognition of a wide range of analytes including clenbuterol hydrochloride, mexiletine hydrochloride, chlorpheniramine maleate, esmolol hydrochloride, and metoprolol tartrate. The synthesized submicron particles were regularly spherical and uniformly non‐porous with an average diameter of around 800 nm and a mean pore size of less than 2 nm. The synthesized chiral stationary phase was packed into 10 cm × 100 μm id capillary columns. The sub_μm‐CSP2 column used in the pCEC system showed better separation of the racemates and at a higher rate compared to those used in the capillary liquid chromatography mode (cLC) system. The sub_μm‐CSP2 possessed high mechanical strength, high stereoselectivity, and long lifespan, demonstrating rapid enantio‐separation and good resolution of samples. The column provided an efficiency of up to 170 000 plates/m for n‐propylbenzene.  相似文献
