心血管疾病(Cardiovascular disease, CVD)是全球疾病致死的主要原因之一. 动脉粥样硬化(Atherosclerosis, AS)是引发各种CVD的首要危险因素, 其发生发展通常经历持续的慢性炎症过程. 因此, 及时高效地检测AS, 对于早期评估、 诊断和治疗CVD具有重要临床意义. 光学探针成像拥有极高的灵敏度和空间分辨率以及超快的信号采集处理速度, 被广泛应用于生物医学检测与成像. 本文综合评述了6种常见用于AS成像的光学探针, 涉及小分子荧光探针、 聚集诱导发光(Aggregate-induced emission, AIE)纳米探针、 量子点探针、 上转换纳米探针、 光声探针和多模态探针等; 并对各种探针的优缺点进行了分析比较. 在此基础上, 展望了光学探针在AS成像领域的发展前景, 并提出了相应建议.  相似文献

In this review, we described the design strategies of SNAP-tag fl uorogenic probes with turn-on fl uorescence responses, which minimized the fl uorescence background and allowed for direct imaging in living cells without wash-out steps. These probes can apply in real-time analysis of protein localization, dynamics, and protein– protein interactions in living cells. Furthermore, the excellent fl uorescent properties made it possible to apply some of the probes in super-resolution fl uorescence imaging.  相似文献

Novel fluorescent molecular probes possessing both a hydroxystyryl and a cyanopyranyl moieties were designed and synthesized to detect the proteins via noncovalent bonding. These fluorescent probes indicated very weak fluorescence emission in the absence of protein. On the other hand, the fluorescence spectra of these probes showed a large Stokes shift and dramatic increase of fluorescence intensity, and red emission was observed after addition of BSA. These fluorescence spectral changes upon binding proteins were caused by the ICT process. Fluorescence intensities of the probes were plotted as a function of protein concentrations. A good linear relationship was observed up to 1000 microg/mL of protein, and the detection limit was found to be 100 ng/mL at the given assay conditions. Similar results were observed for the measurements of not only BSA but also other proteins (BGG, etc.). The responses of these probes to various nonprotein substances (inorganic salts, chelating agents, etc.) were observed, the fluorescence intensity did not change before and after the addition of foreign substances, and correct protein monitoring was successful using these fluorescent probes. To demonstrate the application of these probes, proteins after the separation using SDS-PAGE were stained in the medium containing 1, and the imaging of the proteins in the gel was successful. The experimental results clearly showed that these probes are good protein indicators for easy and highly sensitive detection.  相似文献

TEMPO and 4-nitro-TEMPO spin probes were used to monitor dose-dependent changes in the EPR spectra of irradiated wheat and rice embryos and sunflower embryo parts. Rice embryos were studied in the 233–293 K temperature range using 4-nitro-TEMPO. TEMPAMINE, TEMPYO and DTBN spin probes were also studied for their applicability in the determination of irradiated seeds. All the recorded spectra were simulated, and spectral parameters and partition of the probes among various domains were determined. Despite the contribution of the signal from extracellular regions, it was possible to detect the changes in the water/lipid ratios with dose. The hydrophilic character of the probe alone was not sufficient to distinguish the different doses of irradiation. Line widths and rotational correlation times of various domains within embryo also play an important role. Partition after dehydration was another measure in the selection of the suitable probes for irradiation studies. Better results were obtained in dehydrated embryos for the probes preferring lipid bodies.  相似文献

Silica-polypyrrole particles have been used as a composite stationary phase for liquid chromatography. Determination of capacity factors (k') of a wide number of polycyclic aromatic hydrocarbon (PAH) molecular probes allows the characterisation of the chromatographic properties of the silica-polypyrrole stationary phase. Capacity factors in the range of 0.10 up to 6.1 were determined, thus demonstrating the high affinity of the PAH probes towards the stationary phase. The selectivity of the composite stationary phase was also evaluated as a function of the planarity of the molecular probes injected. Capacity factors determined for PAHs (two-dimensional molecular probes) are higher than those measured for phenyl-substituted PAHs (phenyl-PAHs, three-dimensional molecular probes). Determination of capacity factors, dependence on the composition of the mobile phase demonstrates the reverse alpha-phase properties of the composite stationary phase. The acid-base properties of the composite silica-polypyrrole stationary phase were investigated using benzene derivative molecular probes (i.e., toluene, phenol, benzoic acid and aniline). Capacity factors in the range of 0.45 to 1.0 were determined. This study clearly demonstrates that this composite stationary phase exhibits selective interactions towards PAHs and phenyl-substituted PAHs and strong acid-base properties depending on the structure, the geometry and the acid-base properties of the molecular probes eluted.  相似文献

We report on the application of a new class of oligonucleotide reporter probes, QUAL probes, that "light up" when a nucleophilic phosphorothioate probe binds adjacent to a dabsyl-quenched probe. These self-ligating DNA probes were used for sequence-specific detection of 16S rRNA in Escherichia coli cells. Strong fluorescence was observed only when the phosphorothioate and quenched dabsyl probes bind side-by-side on a 16S rRNA target. The results demonstrate the use of QUAL probes to detect specific RNA sequences in bacterial cells without enzymes and without washing steps.  相似文献

研究了基因芯片相关的DNA探针在芯片表面最佳固定化方法。用两种不同的双功能试剂1,4-苯二异硫氰酸酯和戊二醛分别把5'-端氨基衍生的21-mer寡脱氧核苷酸探针直接共价固定到玻片表面,固定化的寡脱氧核苷酸探针与5'-端FITC标记的互补靶序列进行分子杂交,杂交后用配有CCD的IX70型荧光倒置显微镜成像检测。结果表明,两种固定化方法的效果都比较好,能检测到靶序列的最低终浓度为1.5×10^-9mol/L,优化了探针固定化时间、杂交时间、杂交温度等对DNA芯片分析性能的影响,为构建高灵敏度基因芯片打下良好基础。  相似文献

Fluorobenzene probes for protein profiling through selective cysteine labeling have been developed by rational reactivity tuning. Tuning was achieved by selecting an electron‐withdrawing para substituent in combination with variation of the number of fluorine substituents. Optimized probes chemoselectively arylated cysteine residues in proteins under aqueous conditions. Probes linked to azide, biotin, or a fluorophore were applicable to labeling of eGFP and albumin. Selective inhibition of cysteine proteases was also demonstrated with the probes. Additionally, probes were tuned for site‐selective labeling of cysteine residues and for activity‐based protein profiling in cell lysates.  相似文献

采用含时密度泛函理论与响应函数理论相结合的方法,研究了两种实验室新合成的水溶性双光子荧光次氯酸根(ClO-)探针分子HCH和HCM的单光子吸收、双光子吸收和荧光发射性质。计算结果表明,HCH和HCM分子与ClO-反应后,生成物的光吸收和光发射性质均发生明显变化,相应的吸收和发射峰位都发生了明显蓝移,荧光强度展现出明显的增强。另外,两探针分子都具有较大的双光子吸收截面,且与ClO-反应后,生成物的双光子吸收截面值显著增加,因此两分子均可作为性能优良的双光子荧光探针分子。此外,通过分析HCH和HCM分子与ClO-反应前后的Mulliken电荷布居情况,从理论上证实了该系列荧光探针分子的识别机理是C=N异构化。  相似文献

The pH detection in the alkaline range is particularly important in many fields such as leather processing, waste water treatment, paper industry, and metal mining and finishing. Compared with traditional analysis methods such as colorimetric sensors and electrochemical sensors, the fluorescence and colorimetric probes for pH measurements have attracted much more attention due to their advantages of high sensitivity, excellent selectivity, noninvasiveness, low cost, fast response time, the possibility of continuously measurement, etc. However, there are few fluorescent probes fiting for alkaline pH monitoring. Acturally, the design and synthesis of them were more significant for new probes producing. In this study, the design, synthesis, and practical application of two novel fluorescent pH probes for alkaline pH assay were discussed. Both of the two probes were derived from perylene tetra-(alkoxycarbonyl). The red or blue shift of the absorption/fluorescence spectrum was caused by the introduction of electron donor amino or oxygen ring in the bay region. Due to electronic separation of the OH group from the electron-withdrawing core, the probes have high pKa values and cover the pH range from 8 to 12. They exist in either fluorescent acid form or non-fluorescent basic form. It was investigated that the amino substituent at bay region had a higher pKa value than O-heterocyclic annulated perylene, which showed that the adjustable pKa value could be achieved by the modification of electron withdrawing groups. The probes would have a wide use for testing strips measurements and monitoring pH changes in concrete.  相似文献

Molecular beacons (MBs) are sensitive probes for many DNA sequence-specific applications, such as DNA damage detection, but suffer from technical and cost limitations. We have designed smart probes with self-quenching properties as an alternative to molecular beacons to monitor sequence-specific UV-induced photodamage of oligonucleotides. These probes have similar stem-loop structural characteristics as molecular beacons, but quenching is achieved instead via photoinduced intramolecular electron transfer by neighboring guanosine residues. Our results indicate that the probes are sensitive enough to detect nanomolar target concentrations and are specific enough to discriminate single-base damage. When the probes were used to monitor UV-induced photodamage in oligonucleotide sequences that differ by a single-base mismatch, the photodamage time constant was higher for the perfectly complementary target sequences than for the mismatch sequences, indicating that these probes are specific for each target sequence. In addition, time constants obtained for oligonucleotide target sequences with both stem and loop base mismatches are lower than those with only loop mismatches, suggesting that these sequences are also specifically distinguished by the smart probes. These probes thus constitute robust, sensitive, specific, and cheaper alternatives to MBs for sequence-specific DNA damage detection.  相似文献

We have monitored the reaction dynamics of the DNA hybridization process on a liquid/solid interface at the single-molecule level by using a hairpin-type molecular beacon DNA probe. Fluorescence images of single DNA probes were recorded by using total internal reflection fluorescence microscopy. The fluorescence signal of single DNA probes during the hybridization to individual complementary DNA probes was monitored over time. Among 400 molecular beacon DNA probes that we tracked, 349 molecular beacons (87.5 %) were hybridized quickly and showed an abrupt fluorescence increase, while 51 probes (12.5 %) reacted slowly, resulting in a gradual fluorescence increase. This ratio stayed about the same when varying the concentrations of cDNA in MB hybridization on the liquid/surface interface. Statistical data of the 51 single-molecule hybridization images showed that there was a multistep hybridization process. Our results also showed that photostability for the dye molecules associated with the double-stranded hybrids was better than that for those with the single-stranded molecular beacon DNA probes. Our results demonstrate the ability to obtain a better understanding of DNA hybridization processes using single-molecule techniques, which will improve biosensor and biochip development where surface-immobilized molecular beacon DNA probes provide unique advantages in signal transduction.  相似文献

New ruthenium(II) diimine complexes are presented which are useful as luminescent oxygen probes. Because their luminescence excitation maxima are between 535 and 570 nm, they can be photo-excited by green LEDs which are much brighter than the blue LEDs used so far for existing Ru diimines. The spectral and photophysical properties as well as the solubility properties of the new probes are investigated and discussed in terms of quenching, photostability, and lifetimes. The probes were incorporated into organic polymers by three different methods, to obtain oxygen-sensitive materials for use in optical oxygen sensing. The membranes were characterized with respect to oxygen sensitivity, luminescence intensity, response times, and stability. Notwithstanding the poor luminescence of the new ruthenium(II) probes, their stability, LED compatibility and efficient quenching by oxygen makes them an interesting alternative to existing luminescent oxygen probes.  相似文献

A post-PCR nucleic acid work by comparing experimental data, from electrochemical genosensors, and bioinformatics data, derived from the simulation of the secondary structure folding and prediction of hybridisation reaction, was carried out in order to rationalize the selection of ssDNA probes for the detection of two Bonamia species, B. exitiosa and B. ostreae, parasites of Ostrea edulis.Six ssDNA probes (from 11 to 25 bases in length, 2 thiolated and 4 biotinylated) were selected within different regions of B. ostreae and B. exitiosa PCR amplicons (300 and 304 bases, respectively) with the aim to discriminate between these parasite species. ssDNA amplicons and probes were analyzed separately using the “Mfold Web Server” simulating the secondary structure folding behaviour. The hybridisation of amplicon-probe was predicted by means of “Dinamelt Web Server”. The results were evaluated considering the number of hydrogen bonds broken and formed in the simulated folding and hybridisation process, variance in gaps for each sequence and number of available bases. In the experimental part, thermally denatured PCR products were captured at the sensor interface via sandwich hybridisation with surface-tethered probes (thiolated probes) and biotinylated signalling probes. A convergence between analytical signals and simulated results was observed, indicating the possibility to use bioinformatic data for ssDNA probes selection to be incorporated in genosensors.  相似文献

Developing improved fluorescent probes for imaging the endoplasmic reticulum (ER) is necessary for structure‐activity studies of this dynamic organelle. Two coumarin‐based compounds with sulfonamide side groups were synthesized and characterized as ER‐targeting probes. Their selectivity to target the ER in HeLa and GM07373 mammalian cells was shown with co‐localization experiments using commercially available probes that localize in the ER, mitochondria, or lysozymes. The hydrophobicity of the coumarin‐based probes was comparable to known probes that partition into the ER membrane. Their cytotoxicity in mammalian cells was low with IC50 values that range from 205 to 252 μm . The fluorescent quantum yields of the coumarin‐based probes when excited with 400 nm light were 0.60, and they have a much narrower emission spectrum (from 435 to 525 nm in methanol) than that of the only commercially available ER probe that is exited with 400 nm light (ER‐Tracker? Blue‐White DPX). Thus, the coumarin‐based probes are more useful for multicolor imaging with yellow and red emitting fluorophores. In addition to the above benefits, ER labeling was achieved with the coumarin‐based probes in both live cells and fixed cells, revealing their versatility for a wide range of cellular imaging applications.  相似文献

The ditopic fluorescent photoinduced electron transfer (PET) amino acid sensory probes and were designed and synthesized from cholic acid. To confer the probes with specific binding ability, an amidothiourea moiety and a cyclic diamino-chiral receptive site were introduced on the C17 side chain and the C7 and C12 hydroxyl pendants, respectively. In acetonitrile, the probes demonstrated differential binding toward trifunctional amino acids like serine, lysine, threonine and tyrosine against other simple amino acids. Enantioselectivities (KD/KL) of up to 8.9 and sensitivities in the micromolar range with the probes were observed for trifunctional amino acids.  相似文献

The vicinal dithiol motif is widely present in proteins, and is critical for proteins' structures and functions.In recent years, a variety of fluorescent probes with high specificity and outstanding optical properties for sensing protein vicinal dithiols have been developed. In this review, we summarized the fluorescent probes of protein vicinal dithiols in literature. These probes are classified into four types based on their acceptor sites, i.e., biarsenical probes, monoarsenical probes, dimaleimide probes and diacrylate probes.Through analyzing the properties of different probes, we expect that this review would help readers further understand the structural factors of these probes and provide the design strategy for novel fluorescent probes with improved properties.  相似文献

The DOSY analysis of dilute mixtures can be a challenge because a high signal-to-noise ratio is required for the best DOSY results. The sensitivity increase gained from new probe technologies (e.g. cold and nano probes) could enable one to acquire good DOSY spectra on sample amounts too low for conventional probes. In this article, we investigated the performance of cold and nano probes for qualitative DOSY analysis of concentrated and sample-limited mixtures, and compared the results with those of the conventional probe. We first measured the fluid flow for each probe. All three probes exhibited only relatively small levels of flow; consequently, a double-stimulated echo pulse sequence was not employed in the subsequent DOSY experiments. This decision was based on three facts: (1) flow-induced phase distortions were not observed, (2) our intentions are only to perform qualitative mixture analysis, and (3) discarding 50% of the already limited signal cannot be afforded. Although the cold and nano probes produced DOSY results for the concentrated mixture that were inferior to the conventional probe, the increase in the signal-to-noise ratio observed with these probes proved to be advantageous for the dilute three-component mixture. Furthermore, the cold probe showed slightly superior performance over the nano probe; thus, we conclude that among the probes examined the cold probe is best suited for qualitative DOSY analysis of sample-limited mixtures.  相似文献
