多维色谱分离、串联质谱分析技术已在蛋白质组研究中得到广泛应用。然而生物样品的蛋白质以及全酶切肽段具有高度的复杂性,这严重干扰了蛋白质高通量、规模化的分析。通过标签肽段富集进行样品预分离可以降低体系的复杂程度。本文建立了一种基于共价色谱技术选择性分离富集含半胱氨酸肽的方法,从而降低了样品体系的复杂程度。首先以牛血清白蛋白(BSA)的酶切肽段为模型,对富集条件进行了优化和考察,并在此基础上通过5种蛋白质酶切肽段混合物的富集对该方法进行了验证。结果证明此方法的重现性好,富集效率高,富集特异性好,能有效地富集鉴定含半胱氨酸肽段。所建立的方法在复杂体系的蛋白质组研究中具有广泛的应用前景,为复杂样品的蛋白质高通量、自动化、规模化鉴定和定量研究提供了实用技术。  相似文献

发展了一种大体积循环进样方法,用于富集低丰度蛋白质。在优化的色谱分离条件下,通过增加蛋白质样品的上样体积提高低丰度蛋白质的绝对含量;进一步采用增加样品进样循环次数的方法提高蛋白质的富集效率。以猪肝提取蛋白质为样品,每次上样量500 μL的大体积11次循环进样。根据色谱峰的信号强弱,选择了在原始谱图中看不到色谱峰、有较少小峰和有较多小峰出现的时间段等有代表性的馏分进行研究。在中等极性的组分中,保留时间为11.38 min和12.58 min组分的富集效率分别提高了52倍和61倍,实验结果与理论富集效率相近。所发展的方法为生物蛋白质样品研究提供了一种新的富集制备及检测方法。  相似文献

蛋白质的糖基化是生物体内重要的蛋白质翻译后修饰之一,但其丰度通常较低,糖基化蛋白质酶解肽段中仅有2%~5%为糖基化肽段,因此,为实现糖基化蛋白质组的深度覆盖分析,对糖基化蛋白质/肽段进行富集是非常必要的。该文对糖基化蛋白质组样品不同富集方法的原理、特点以及最新研究进展进行了综述,同时也对N-糖基化蛋白质组学富集策略的发展前景进行了展望。  相似文献

蛋白质的磷酸化是一种可逆性的翻译后修饰,在细胞的增值、分化、信号转导以及转录与翻译调控、蛋白质复合体的形成、蛋白质降解等方面发挥着极为重要的作用.因此磷酸化蛋白的鉴定成为翻译后修饰研究的重要内容.但由于磷酸化蛋白的丰度较低, 难以用质谱直接检测.为了解决这个问题,改善质谱对磷酸肽的信号响应, 需要对磷酸化蛋白质或磷酸肽进行富集.本文系统地介绍了磷酸化蛋白组学研究中应用较为广泛和最新建立的各种分离富集方法的原理、特点、应用研究进展,包括抗体富集法、激酶特异富集法、亲和富集法、化学修饰法、多种色谱分离富集方法以及MALDI靶盘富集法.  相似文献

蛋白质糖基化是一种广泛存在的重要的蛋白质翻译后修饰,糖基化肽在总酶解肽中占的比例不超过5%,这使得糖肽的分离富集成为糖蛋白质组学研究发展的关键技术之一。在诸多糖蛋白质组富集技术中,化学方法是富集技术的主导,本文从化学反应的角度介绍糖基化蛋白质富集的技术进展。富集过程按照连接和释放分别讨论,在连接过程中,重点介绍硼酸化学法、肼化学法、胺化学法和肟点击法;在释放过程中,以N-糖蛋白的酶释放法和O-糖蛋白的β-消除法为主导,一并介绍了最新的氧化断裂释放的化学法。最后,讨论总体富集策略的发展现状。该文以糖蛋白富集的共价反应为核心,分析不同方法的优缺点以及各技术在糖蛋白质组学研究中的应用和贡献。  相似文献

由于生物样本中蛋白质含量的动态范围跨越多个数量级,快速、高效的蛋白质分离富集是低丰度蛋白质成功鉴定的关键.一些具有重要生物学意义的蛋白质的表达量通常很低,预富集就成了这些蛋白质获得成功质谱鉴定和分析的必备条件.富集的主要目的就是有选择性地从复杂生物样本中分离目标分子,从而达到降低样本复杂程度,辅助后续质谱高灵敏鉴定.近些年来,将纳米颗粒引入到这一领域大大促进了富集技术的发展.在本文中,我们集中介绍了近年来发展起来的,利用不同的纳米颗粒,富集低丰度和特定翻译后修饰的蛋白质或多肽并适合质谱鉴定的蛋白质预处理技术.  相似文献

磷酸化是蛋白质翻译后修饰的重要形式之一,其异常往往会导致细胞内信号通路的紊乱和疾病的发生。固定化金属离子亲和色谱(IMAC)是磷酸化肽段的高效富集技术,在磷酸化蛋白质组研究方面应用广泛。该研究以金属钛离子(Ti4+)螯合IMAC材料(Ti4+-IMAC)为载体,进行磷酸化肽段富集。比较了10 μm Ti4+-IMAC通过振荡法和固相萃取法(SPE)富集磷酸肽的效果,发现振荡法可以富集到更多的磷酸肽;对比了两种尺寸(10 μm和30 μm)Ti4+-IMAC在磷酸化肽段富集中的差异,发现小尺寸材料富集效果更佳。进一步采用优化的策略比较了不同转移能力肺癌细胞的磷酸化蛋白质组,免标记定量蛋白质组学结果表明,优化的Ti4+-IMAC方法可以从正常的肺成纤维细胞MRC5、低转移肺癌细胞95C和高转移肺癌细胞95D中分别鉴定到510、863和1108种磷酸化蛋白质,其中317种为3组所共有。该研究共鉴定到1268种磷酸化蛋白质上的7560个磷酸化位点,其中1130个为差异磷酸化位点,文献报道显示部分异常表达的激酶与癌症转移密切相关。通过生信对比分析发现,异常表达的磷酸化蛋白质主要与细胞侵袭、迁移和死亡等细胞迁移方面的功能有关。通过优化磷酸化肽富集策略,初步阐明了磷酸化蛋白质网络的异常与肺癌转移之间的相关性,该方法有望用于肺癌进展相关的磷酸化位点、磷酸化蛋白质及其信号通路研究。  相似文献

为了提高二氧化钛富集磷酸肽法对磷酸肽的富集效率,以6种标准蛋白酶切肽段混合物为研究对象,对二氧化钛富集磷酸肽过程中的乙腈比例、三氟乙酸比例、二氧化钛用量等条件分别进行了优化。结果表明在乙腈含量为80%(v/v),三氟乙酸含量为1%(v/v),二氧化钛用量与需要富集肽段的质量比为40:1的条件下,可以取得较好的富集效果。将优化后的富集方法应用于腾冲嗜热厌氧菌磷酸化蛋白质的分析,初步鉴定到25个磷酸化蛋白质,为进一步研究这种极端环境下生存的低等生物的生命活动提供了参考信息。  相似文献

This overview highlights the possibilities of on- or in-line preconcentration procedures in combination with a CZE separation, focused on the determination of peptides and proteins. The discussed methods, including sample stacking, field-amplified injection, isotachophoresis, solid phase extraction, membrane preconcentration, electroextraction, supported liquid membranes, hollow fibers, immunoaffinity, and molecularly imprinted polymers technology preconcentration are categorized in electrophoresis-based and chromatography-based preconcentration. The chromatography-based preconcentration is subdivided in low-specificity and high-specificity methods. A number of preconcentration methods are available, however, this paper demonstrates that various compounds in different media (aqueous solutions, urine, and plasma) require different preconcentration systems. The preconcentration techniques of first choice in general seem to be solid-phase extraction and membrane preconcentration, because of their high concentration ability, multiapplicability, relative simplicity and clean-up capability. For the future, hollow fibers seem to hold a great potential as preconcentration technique, yielding high concentration factors, using simple designs. New techniques, such as hollow fibers, molecularly imprinted polymers technology and supported liquid membranes may have the potential to supersede the conventional preconcentration techniques in some cases. The larger the arsenal of preconcentration techniques becomes, the more efficiently peptides and proteins may be analyzed in the future. These techniques, in some cases, require pre-cleanup procedures, to ensure the purity of the samples to concentrate.  相似文献

The need for preconcentration of trace and ultratrace amounts of lanthanides from environmental, geological and biological samples is brought out in introductory part. Both on-line and off-line preconcentration procedures developed for lanthanides since 1980 are reviewed. The preconcentration techniques covered in this review include liquid-liquid extraction (LLE), ion-exchange, co-precipitation, and solid phase or solid-liquid extraction. Separate sections are devoted to each of the preconcentration techniques employed for enrichment of individual or mixtures of lanthanides. Preconcentration microfluidic devices are fabricated incorporating straight or convergent-divergent microchannels and hydrogel or Nafion membranes. Sample preconcentration is achieved utilizing concentration-polarization effects. The effects of the microchannel geometry on the preconcentration intensity are systematically examined. It is shown that for the preconcentrator with the straight microchannel, the time required to achieve a satisfactory preconcentration intensity increases with an increasing channel depth. For the convergent-divergent microchannel, the preconcentration intensity increases with a reducing convergent channel width. Comparing the preconcentration performance of the two different microchannel configurations, it is found that for an equivalent width of the main microchannel, the concentration effect in the convergent-divergent microchannel is faster than that in the straight microchannel.  相似文献

Subtractive differential pulse voltammetry following adsorptive preconcentration of organic compounds at solid electrodes is described. Different preconcentration periods are used, and the difference between the oxidation (stripping) currents is recorded. Background currents which are independent of the preconcentration period cancel out. Combining the enhanced peak current, due to the preconcentration step, with the background current correction of the subtractive mode, gives improved sensitivity and/or allows the use of shorter preconcentration periods. Chlorpromazine and dopamine have been used as test systems. A detection limit of around 1 x 10(-9)M has been obtained for chlorpromazine with a 10-min preconcentration period. Applicability to clinical samples is illustrated by the determination of chlorpromazine in whole blood and urine.  相似文献

The miniaturization of the preconcentration methods is one of the fields of contemporary analytical chemistry. A new method of liquid-liquid microextraction preconcentration has been developed for the last 10–15 years; it is highly efficient, rapid, simple in implementation, and ecologically safe. The methodology and principles of the implementation of microextraction preconcentration are considered in the review. A preconcentration method incorporated with neutron activation analysis was developed for the accurate determination of trace metals in biological and environmental samples. Bismuth diethyldithiocarbamate, thallium diethyldithiocarbarnate, and ammonium pyrrolidinedithiocarbamate were used as the preconcentration agents for three groups of metals. Trace metals were enriched and interfering elements such as Na, Br, P. K. etc. were removed simultaneously during the preconcentration process so that the sensitivity for each metal was greatly improved. Optimal conditions for sample digestion, group preconcentration of trace metals and neutron activation analysis of biological and environmental samples were discussed.  相似文献

A preconcentration method of 13 rare earth elements (REES) was studied for neutron activation ananlysis (NAA). Hydrated magnesium oxide was used as the preconcentration agent to absorb the REES ions from aqueous solution onto the solid magnesium oxide, which was separated and analyzed. It was observed that the Langmuir equation for isothermal adsorption was well obeyed by the REES under the condition studied. The efficiency of the preconcentration process using hydrated magnesium oxide was critically examined for each of REE and for mixture of REES by preparing a known volume of solution containing known amount of trace REES. NAA was used to analyze REES recovered by the preconcentration process. It was found that the REES recoveries were satisfactory and the preconcentration process is reliable. There are several resort resort areas in Taiwan where local people are enjoying its hot spring water. It is generally believed that the hot spring water spa would bring about some sorts of therapeutic functions. The preconcentration method developed above, was applied to analyze the trace amounts of REES in hot spring wate in Taiwan.  相似文献

A simple, accurate, and sensitive preconcentration method for determination of esomeprazole magnesium in treated sewage water was developed based on HPLC. A preconcentration method was developed for aqueous solution containing pure drug using solid phase extraction. Macroporous beads of polystyrene divinyl benzene (PSDVB) polymer were used for preconcentration followed by chromatographic determination. Experimental parameters were optimized. This optimized method can detect esomeprazole magnesium concentration up to 0.003 mg L?1 after preconcentration. This method was used for determination of esomeprazole magnesium in water collected from a sewage treatment facility. Esomeprazole magnesium could not be detected in the treated sewage water sample collected for the study.  相似文献
