Summary Two static headspace selective methods for residual solvents belonging to Class I, II and III have been developed, optimized and validated for drug products, which are insoluble in water. The methods give very good sensitivities (detection limits under 10 ppm) and precision (under 5.5% RSD) for all solvents. The detection limit for benzene was 0.1 ppm, in concordance with Pharmacopoeia requirements. During method optimization we found that sample volume and water content have a critical influence on the sensitivity. From our data, it is beneficial to choose low sample volume. If sample solubility in the organic solvent allows it, the optimum sample volume is between 0.1 and 0.3 mL. For drug products with water content greater than 7%, the increase in sensitivity produced by water presence should be taken into consideration, otherwise inconsistent recovery data and underestimation of residual solvent content will happen. Excessive and uncontrolled exposures of the workers to benzene, toluene, ethylbenzene and xylene (BTEX) have currently raised great concerns among industrial hygienist all over the world. Therefore, the effective monitoring of such exposures is assumed to be of prime importance. A cold fiber solid-phase microextraction device based on a cooling capsule as a cooling unit and CO2 as a coolant was applied to quantitatively analyze BTEX in aqueous samples. A gas chromatography with flame ionization detection was recruited to analyze the target analytes, which had been identified according to their retention times. Several factors such as coating temperature, extraction time and temperature, sample volume and sodium content were optimized. Two modes of extraction, i.e., headspace (HS) and headspace cold fiber (HS-CF) in SPME, were investigated and compared under optimized conditions. The results revealed that HS-CF-SPME has the most appropriate outcome for the extraction of BTEX from aqueous samples. Under the optimized conditions, the calibration curves were linear within the range of 0.2–500 ng ml?1 and the detection limits were between 0.02 and 0.07 ng ml?1.The intraday relative standard deviations was lower than about 10%. The method was successfully applied to the determination of BTEX in urine samples with good recovery.  相似文献

In this work, portable gas chromatography-microflame ionization detection (portable GC-μFID) coupled to headspace solid-phase microextraction (HS-SPME) was developed for the field analysis of benzene, toluene, ethylbenzene and xylene (BTEX) in water samples. The HS-SPME parameters such as fiber coating, extraction times, stirring rate, the ratio of headspace volume to sample volume, and sodium chloride concentration were studied. A 65 μm poly(dimethylsiloxane)-divinylbenzene (PDMS-DVB) SPME fiber, 900 rpm, 3.0 ml of headspace (1.0 ml water sample in 4.0 ml vial), and 35% sodium chloride concentration (w/v) were respectively chosen for the best extraction response. An extraction time of 1.0 min was enough to extract BTEX in water samples. The relative standard deviation (R.S.D.) for the procedure varied from 5.4% to 8.3%. The method detection limits (MDLs) found were lower than 1.5 μg/l, which was enough sensitive to detect the BTEX in water samples. The optimized method was applied to the field analysis of BTEX in wastewater samples. These experiment results show that portable GC-μFID combined with HS-SPME is a rapid, simple and effective tool for field analysis of BTEX in water samples.  相似文献

A new cloud vapor zone (CVZ)-based headspace solid-phase microextraction (HS-SPME) technique has been demonstrated with the capability of heating the sample matrix and simultaneously cooling the sampling zone. A bi-temperature-controlled (BTC) system, allowing 10 mL of test sample heating and headspace external-cooling, was employed for the CVZ formation around the SPME-fiber sampling area. In the CVZ procedure, the heated headspace vapor undergoes a sudden cooling near the SPME to form a dense cloud of analyte–water vapor, which is helpful for adsorption or absorption of the analyte. The device was evaluated for the quantitative analysis of aqueous chlorothalonil. Parameters influencing sampling efficiency, e.g., SPME fiber coating, SPME sampling temperature and time, solution modifier, addition of salt, sample pH, and temperature, were investigated and optimized thoroughly. The proposed BTC-HS-SPME method afforded a best extraction efficiency of above 94% accuracy (less than 4.1% RSD, n = 7) by using the PDMS fiber to collect chlorothalonil in the headspace at 5 °C under the optimized condition, i.e., heating sample solution (added as 10% ethylene glycol and 30% NaCl, at pH 7.0) at 130 °C for 15 min. The detection was linear from 0.01 to 80 μg L−1 with a regression coefficient of 0.9998 and had a detection limit of 3.0 ng L−1 based on S/N = 3. Practical application was demonstrated by analyzing chlorothalonil in farm water samples with promising results and recoveries. The approach provided a very simple, fast, sensitive, and solvent-free procedure to collect analytes from aqueous solution. The approach can provide a new platform for other sensitive HS-SPME assays.  相似文献

Solid-phase microextraction (SPME) is a miniaturized and solvent-free sample preparation technique for chromatographic–spectrometric analysis by which the analytes are extracted from a gaseous or liquid sample by absorption in, or adsorption on, a thin polymer coating fixed to the solid surface of a fiber, inside an injection needle or inside a capillary. In this paper, the present state of practical performance and of applications of SPME to the analysis of blood, urine, oral fluid and hair in clinical and forensic toxicology is reviewed. The commercial coatings for fibers or needles have not essentially changed for many years, but there are interesting laboratory developments, such as conductive polypyrrole coatings for electrochemically controlled SPME of anions or cations and coatings with restricted-access properties for direct extraction from whole blood or immunoaffinity SPME. In-tube SPME uses segments of commercial gas chromatography (GC) capillaries for highly efficient extraction by repeated aspiration–ejection cycles of the liquid sample. It can be easily automated in combination with liquid chromatography but, as it is very sensitive to capillary plugging, it requires completely homogeneous liquid samples. In contrast, fiber-based SPME has not yet been performed automatically in combination with high-performance liquid chromatography. The headspace extractions on fibers or needles (solid-phase dynamic extraction) combined with GC methods are the most advantageous versions of SPME because of very pure extracts and the availability of automatic samplers. Surprisingly, substances with quite high boiling points, such as tricyclic antidepressants or phenothiazines, can be measured by headspace SPME from aqueous samples. The applicability and sensitivity of SPME was essentially extended by in-sample or on-fiber derivatization. The different modes of SPME were applied to analysis of solvents and inhalation narcotics, amphetamines, cocaine and metabolites, cannabinoids, methadone and other opioids, fatty acid ethyl esters as alcohol markers, γ-hydroxybutyric acid, benzodiazepines, various other therapeutic drugs, pesticides, chemical warfare agents, cyanide, sulfide and metal ions. In general, SPME is routinely used in optimized methods for specific analytes. However, it was shown that it also has some capacity for a general screening by direct immersion into urine samples and for pesticides and other semivolatile substance in the headspace mode.  相似文献

建立了顶空固相微萃取联结气相色谱-电子捕获检测器(HS-SPME-GC-ECD)测定水中多溴联苯醚的方法。制作了多壁碳纳米管涂层固相微萃取探头。优化了萃取时间,萃取温度,搅拌速度,顶空体积,溶液的pH,离子强度及有机溶剂等影响萃取效率的各种因素。比较了室温和100 ℃顶空萃取和直接萃取的效率。结果表明,室温下直接萃取比顶空萃取的效率高2-4倍,而在100 ℃时顶空萃取比直接萃取的效率高1-8倍。除BDE-154外,无论直接萃取还是顶空萃取,100 ℃时的萃取效率均高于室温。方法的线性范围50-1600 ng/L,相关系数为0.995-0.998,5种多溴联苯醚的最低检出限(S/N=3)为1.14-16.25 ng/L,相对标准偏差(RSD%,n=5)小于10%。本方法用于真实水样的测定,回收率为74.2%-98.7%。  相似文献

A solid-phase microextraction method (SPME) followed by gas chromatography with micro electron capture detection for determining trace levels of nitro musk fragrances in residual waters was optimized. Four nitro musks, musk xylene, musk moskene, musk tibetene and musk ketone, were selected for the optimization of the method. Factors affecting the extraction process were studied using a multivariate approach. Two extraction modes (direct SPME and headspace SPME) were tried at different extraction temperatures using two fiber coatings [Carboxen–polydimethylsiloxane (CAR/PDMS) and polydimethylsiloxane–divinylbenzene (PDMS/DVB)] selected among five commercial tested fibers. Sample agitation and the salting-out effect were also factors studied. The main effects and interactions between the factors were studied for all the target compounds. An extraction temperature of 100 °C and sampling the headspace over the sample, using either CAR/PDMS or PDMS/DVB as fiber coatings, were found to be the experimental conditions that led to a more effective extraction. High sensitivity, with detection limits in the low nanogram per liter range, and good linearity and repeatability were achieved for all nitro musks. Since the method proposed performed well for real samples, it was applied to different water samples, including wastewater and sewage, in which some of the target compounds (musk xylene and musk ketone) were detected and quantified. A new solid phase microextraction (SPME) fibre using carbon nanotubes as fibre coating incorporated into a groove of a stainless steel rod is suggested. It is mechanically stable and exhibits relatively high thermal stability (up to 280 °C). The coating showed especially good extraction efficiency for aromatic hydrocarbons. The extraction properties of the fibre to benzene, toluene, ethylbenzene and o-xylene were examined using both direct and headspace SPME modes coupled to gas chromatography-flame ionization detection. The parameters affecting the extraction efficiency (extraction temperature and time, salt addition, desorption temperature and time) were investigated and quality parameters were measured under the optimized conditions. For both headspace and direct SPME the calibration graphs were linear up to 100 mg L−1 (R 2 > 0.996) and detection limits ranged from 0.09 to 0.39 μg L−1. The repeatabilities were 5.9–13.3%. The proposed coating was applied for aromatic hydrocarbons determination in petrol station waste waters.  相似文献

Medical devices sterilized by ethylene oxide (EtO) retain trace quantities of EtO residuals, which may irritate patients' tissue. Reliably quantifying trace level EtO residuals in small medical devices requires an extremely sensitive analytical method. In this research, a Doehlert uniform shell design was utilized in obtaining a response surface to optimize a novel headspace–solid‐phase microextraction–gas chromatographic (HS‐SPME‐GC) method developed for analyzing trace levels of EtO residuals in sterilized medical devices, by evaluating sterilized, polymer‐coated, drug‐eluting cardiovascular stents. The effects of four independent experimental variables (HS‐SPME desorption time, extraction temperature, GC inlet temperature and extraction time) on GC peak area response of EtO were investigated simultaneously and the most influential experimental variables determined were extraction temperature and GC inlet temperature, with the fitted model showing no evidence of lack‐of‐fit. The optimized HS‐SPME‐GC method demonstrated overall good linearity/linear range, accuracy, repeatability, reproducibility, absolute recovery and high sensitivity. This novel method was successfully applied to analysis of trace levels of EtO residuals in sterilized/aerated cardiovascular stents of various lengths and internal diameter, where, upon heating, trace EtO residuals fully volatilized into HS for extraction, thereby nullifying matrix effects. As an alternative, this novel HS‐SPME‐GC method can offer higher sensitivity compared with conventional headspace analyzer‐based sampling. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

Solid-phase microextraction (SPME) was used for the analysis of some pesticides (bromoxynil, chlorotoluron, diuron, isoproturon, 2,4-MCPA, MCPP and 2,4-D) in rainwater after derivatisation with PFBBr and gas chromatography-ion trap mass spectrometry. The derivatisation procedure was optimized by testing different methods: direct derivatisation in the aqueous phase followed by SPME extraction, on-fibre derivatisation and derivatisation in the injector. The best result was obtained by headspace coating the PDMS/DVB fibre with PFBBr for 10 min followed by direct SPME extraction for 60 min at 68 °C (pH 2 and 75% NaCl). Good detection limits were obtained for all the compounds: these ranged between 10 and 1,000 ng L−1 with a relatively high uncertainty due to the combination of derivatisation and SPME extraction steps. The optimized procedure was applied to the analysis of pesticides in rainwater and results obtained shows that this method is a fast and simple technique to assess the spatial and temporal variations of concentrations of pesticides in rainwater.  相似文献

This paper proposed a multiple headspace single-drop microextraction (MHS-SDME) method coupled to gas chromatography with flame-ionization detection (GC-FID) for direct determination of residual solvents in solid drug product. The MHS-SDME technique is based on extrapolation to an exhaustive extraction of consecutive extractions from the same sample which eliminates the matrix effect on the quantitative analysis of solid samples. The total peak area of analyte is calculated with a beta constant which can be obtained from the slope of the linear regression that related to the peak area of each extraction and the number of extraction times. In this work, a model drug powder was chosen and the amounts of residues of two solvents, methanol and ethanol, were investigated. The factors influencing the extraction process including extraction solvent, microdrop volume, extraction time, sample amount, thermostatting temperature and incubation time were studied. 10 mg of drug powder was incubated for 3 h at 140 °C prior to the first extraction and thermostatted for 15 min at 140 °C between each extraction. Extraction was carried out with 2 μL of dimethyl sulfoxide (DMSO) as the microdrop for 5 min. The features of the method were established using standard solutions. Validation of the proposed method showed good agreement with the traditional dissolution method for analysis of residual solvents in drug product. The results indicated that MHS-SDME has a great potential for the quantitative determination of residual solvents directly from the solid drug products due to its low cost, ease of operation, sensitivity, reliability and environmental protection.  相似文献

Microwave-assisted thermal desorption (MAD) coupled to headspace solid-phase microextraction (HS-SPME) has been studied for in-situ, one-step, sample preparation for PAHs collected on XAD-2 adsorbent, before gas chromatography with mass spectrometric detection. The PAHs on XAD-2 were desorbed into the extraction solution, evaporated into the headspace by use of microwave irradiation, and absorbed directly on a solid-phase microextraction fiber in the headspace. After desorption from the SPME fiber in the hot GC injection port, PAHs were analyzed by GC–MS. Conditions affecting extraction efficiency, for example extraction solution, addition of salt, stirring speed, SPME fiber coating, sampling temperature, microwave power and irradiation time, and desorption conditions were investigated. Experimental results indicated that extraction of 275 mg XAD-2, containing 10–200 ng PAHs, with 10-mL ethylene glycol–1 mol L−1 NaCl solution, 7:3, by irradiation with 120 W for 40 min (the same as the extraction time), and collection with a PDMS–DVB fiber at 35 °C, resulted in the best extraction efficiency. Recovery was more than 80% and RSD was less than 14%. Optimum desorption was achieved by heating at 290 °C for 5 min. Detection limits varied from 0.02 to 1.0 ng for different PAHs. A real sample was obtained by using XAD-2 to collect smoke from indoor burning of joss sticks. The amounts of PAHs measured varied from 0.795 to 2.53 ng. The method is a simple and rapid procedure for determination of PAHs on XAD-2 absorbent, and is free from toxic organic solvents.  相似文献

A new procedure for determining eleven organochlorine pesticides in soils using microwave-assisted extraction (MAE) and headspace solid phase microextraction (HS-SPME) is described. The studied pesticides consisted of mirex, α- and γ-chlordane, p,p′-DDT, heptachlor, heptachlor epoxide isomer A, γ-hexachlorocyclohexane, dieldrin, endrin, aldrine and hexachlorobenzene. The HS-SPME was optimized for the most important parameters such as extraction time, sample volume and temperature. The present analytical procedure requires a reduced volume of organic solvents and avoids the need for extract clean-up steps. For optimized conditions the limits of detection for the method ranged from 0.02 to 3.6 ng/g, intermediate precision ranged from 14 to 36% (as CV%), and the recovery from 8 up to 51%. The proposed methodology can be used in the rapid screening of soil for the presence of the selected pesticides, and was applied to landfill soil samples.  相似文献

A solid-phase microextraction (SPME) procedure for the determination of four haloanisoles (2,4,6-trichloroanisole, 2,3,4,6-tetrachloroanisole, pentachloroanisole and 2,4,6-tribromoanisole), as well as their precursor halophenols (2,4,6-trichlorophenol, 2,3,4,6-tetrachlorophenol, pentachlorophenol and 2,4,6-tribromophenol), involved in the presence of cork taint in wine, was developed. Firstly, analytes were concentrated on a SPME fiber, and then halophenols were derivatised using N-methyl-N-trimethylsilyltrifluoroacetamide (MSTFA). The compounds were desorbed for 5 min in the gas chromatography injector port and then determined with an electron capture detector. The influence of different parameters on the efficiency of extraction (volume of sample, type of fibre coating and time) and derivatisation (time, temperature and volume of MSTFA) steps was evaluated. Polyacrylate (PA) was selected as the extraction fiber, optimised parameters for SPME were 10 ml of wine, temperature 70 degrees C and extraction time 60 min. The optimal conditions identified for the derivatisation step were temperature 25 degrees C, reagent volume 50 microl and extraction time 25 min. Under optimal conditions, the proposed method showed satisfactory linearity, precision and detection limits. The method was applied successfully to the analysis of red wine samples. To our knowledge, this is the first time that headspace (HS) SPME combined with on-fiber derivatisation has been applied to determine cork taint responsible compounds in wine.  相似文献

A headspace liquid phase microextraction (HS-LPME) method has been developed and optimized for the residual solvent determination in pharmaceutical products. A microdrop of n-hexanol containing isopropanol (as internal standard) was suspended at the tip of a gas chromatographic syringe and exposed to the headspace of the sample solution. After extraction for an optimized time, the microdrop was retracted into the syringe and injected directly into a GC injection port. Critical experimental factors, including extraction solvent, temperature, ionic strength, stirring rate, extraction time, equilibrium time, drop volume, and sample volume were investigated and optimized. Compared with the static headspace technique, HS-LPME method showed superior results, being compatible with the pharmaceutical samples.  相似文献

Solid-phase microextraction (SPME) with a disposable ionic liquid (IL) coating was developed for headspace extraction of benzene, toluene, ethylbenzene, and xylenes (BTEX) in paints. The SPME fiber was coated with IL prior to every extraction, then the analytes were extracted and desorbed on the injection port of gas chromatography, and finally the IL coating on the fiber was washed out with solvents. The coating and washing out of IL from the fiber can be finished in a few minutes. This disposable IL-coated fiber was applied to determine BTEX in water-soluble paints with results in good agreement with that obtained by using commercially available SPME fibers. For all the four studied paints samples, the benzene contents were under the detection limits, but relatively high contents of toluene, ethylbenzene and xylenes (56-271 microg g(-1)) were detected with spiked recoveries in the range of 70-114%. Compared to the widely used commercially available SPME fibers, this proposed disposable IL-coated fiber has much lower cost per determination, comparable reproducibility (RSD < 11%), and no carryover between each determination. Considering that IL possess good extractability for various organic compounds and metals ions, and that task-specific IL can be designed and synthesized for selective extraction of target analytes, this disposable IL coating SPME might has great potential in sample preparation.  相似文献

采用电化学沉淀法,成功地制备了多孔、高效聚苯胺固相微萃取涂层,并建立了顶空固相微萃取-气相色谱(HS-SPME-GC)快速测定水体和牛奶中的痕量多溴联苯醚的方法。详细研究了萃取模式、萃取温度、萃取时间、顶空体积及离子强度对萃取效率的影响。在优化实验条件下,本法测定的6种多溴联苯醚的线性范围为1~4000 ng/L(除BDE-154和BDE-153分别为1~3000 ng/L、1~2500 ng/L外),相关系数大于0.99,检出限(S/N=3)在0.08~0.20 ng/L之间,相对偏差小于8.5%(n=7)。自制聚苯胺涂层对多溴联苯醚的萃取效率优于商品化100μm-PDMS纤维。将本法用于河水和牛奶中痕量多溴联苯醚的测定,实际样品回收率分别在90%和80%以上。  相似文献

A direct solid-phase microextraction (SPME) procedure has been developed and applied for the simultaneous determination of nonylphenol, nonylphenol mono- and diethoxylates and their brominated derivatives in raw and treated water at low microg l(-1) concentrations. Several parameters affecting the SPME procedure, such as extraction mode (headspace or direct-SPME), selection of the SPME coating, extraction time, addition of organic modifiers such as methanol and temperature were optimized. The divinylbenzene-carboxen-polydimethylsiloxane fiber was the most appropriate one for the determination of nonylphenol ethoxylates (NPEOs) and bromononylphenol ethoxylates (BrNPEOs) by SPME-GC-MS. The optimized method was linear over the range studied (0.11-2.5 microg l(-1)) and showed good precision, with RSD values between 4 and 15% and detection limits ranging from 30 to 150 ng l(-1) depending on the compound. The SPME procedure was compared with a solid-phase extraction-GC-MS method (C18 cartridge) for the analysis of NPEO and BrNPEOs in water samples. There was good agreement between the results from both methods but the SPME procedure showed some advantages such as lower detection limits, a shorter analysis time and the avoidance of organic solvents. The optimized SPME method was applied to determine nonylphenol and brominated metabolites in raw and treated water of Barcelona (NE Spain).  相似文献

This work compares two miniaturised sample preparation methods, solid phase microextraction (SPME) and hollow fiber liquid phase microextraction (HF-LPME), in combination with gas chromatography coupled to tandem mass spectrometry with a triple quadrupole analyzer for the determination of 77 pesticides in drinking water. In the case of SPME, extraction temperature and time were optimized by experimental design, although other parameters, as desorption time, pH, and ionic strength, were also evaluated. The extraction and desorption solvents [octanol/dihexyl ether (75:25, v/v) and cyclohexane, respectively], as well as the extraction and desorption time, ionic strength, and pH, were studied for the HF-LPME procedure. Under the optimal conditions, recoveries (70.2–113.5% for SPME and 70.0–119.5% for HF-LPME), intra-day precision (2.1–19.4% for SPME and 4.3–22.5% for HF-LPME), inter-day precision (5.2–21.5% for SPME and 8.4–27.3% for HF-LPME), and limits of detection, between 0.1 and 28.8 ng/L for SPME and 0.2 and 47.1 ng/L for HF-LPME and overall uncertainty (9.6–25.2% for SPME and 13.3–27.5% for HF-LPME) were established for both extraction procedures. Finally, the proposed methods were successfully applied to the analysis of 41 drinking water samples, and similar results were obtained with both extraction approaches.  相似文献

A headspace solid-phase microextraction (HS-SPME) procedure has been developed and applied for the determination of cyanogen halides in treated water samples at microg/L concentrations. Several SPME coatings were tested, the divinylbenzene-Carboxen-polydimethylsiloxane fiber being the most appropriate coating. GC-electron-capture detection was used for separation and quantitation. Experimental parameters such as sample volume, addition of a salt, extraction time and desorption conditions were studied. The optimized method has an acceptable linearity, good precision, with RSD values <10% for both compounds, and it is sufficiently sensitive to detect ng/L levels. HS-SPME was compared with liquid-liquid microextraction (US Environmental Protection Agency Method 551.1) for the analysis of spiked ultrapure and granular activated carbon filtered water samples. There was good agreement between the results from both methods. Finally, the optimized procedure was applied to determine both compounds at the Barcelona water treatment plant (N.E. Spain). Cyanogen chloride in treated water was <1.0 microg/L and cyanogen bromide ranged from 3.2 to 6.4 microg/L.  相似文献
