Self‐assembly of amphiphilic ABA random triblock copolymers in water serves as a novel approach to create unique structure micelles connected with flexible linkages. The ABA triblock copolymers consist of amphiphilic random copolymers bearing hydrophilic poly(ethylene glycol) and hydrophobic dodecyl pendants as the A segments and a hydrophilic poly(ethylene oxide) (PEO) as the middle B segment. The A block is varied in dodecyl methacrylate content of 20%–50% and degree of polymerization (DP) of 100‐200. By controlling the composition and DP of the A block, various architectures can be tailor‐made as micelles in water: PEO‐linked double core unimer micelles, PEO‐looped unimer or dimer micelles, and multichain micelles. Abstract

A series of tertiary amine containing PHMEMA-PEG-PHMEMA ABA triblock copolymers were synthesized by atom transfer radical polymerization (ATRP) using bromine-capped poly(ethylene glycol) (Br-PEG-Br) and 2-(hexamethyleneimino)ethyl methacrylate (HMEMA) as macro-initiator and monomers, respectively. The chemical structures and molecular weights of triblock copolymers were characterized by 1H NMR and gel permeation chromatography (GPC). The self-assembly behaviors of copolymers in different pH conditions were studied by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Triblock copolymers self-assembled into micelles in water (pH 7.4) and the micelles disassembled at acidic pH (pH 5.0). Anticancer drug doxorubicin (DOX) was used as a drug model and physically encapsulated into polymeric micelles. The drug release of DOX-loaded polymeric micelles was pH-responsive; the drug-loaded micelles that had higher contents of tertiary amine in polymer pendant groups showed faster release speed. In addition, the drug-loaded micelles showed excellent inhibition efficacy against HeLa cells in vitro.  相似文献

This study reports the synthesis of poly(ethylene glycol)methyl ether‐block‐poly(glycidyl methacrylate) (MPEG‐b‐PGMA) diblock, and poly(ethylene glycol)methyl ether‐block‐poly(glycidyl methacrylate)‐block‐poly(methyl methacrylate) (MPEG‐b‐PGMA‐b‐PMMA) triblock copolymers via atom transfer radical polymerization and their self‐assembly behaviors in aqueous media by using acetone as cosolvent. These block copolymers formed near monodisperse core–shell micelles having cross‐linkable cores. Two types of cross‐linked micelles, namely spherical MPEG‐b‐PGMA core cross‐linked (CCL) micelles and MPEG‐b‐PGMA‐b‐PMMA interlayer cross‐linked (ILCL) micelles, were also successfully prepared from these block copolymers by using various bifunctional cross‐linkers such as hexamethylenediamine (HMDA), ethylenediamine (EDA), and 2‐aminoethanethiol (AET). Cross‐linking was successfully carried out via ring‐opening reactions of epoxy residues of hydrophobic‐cores with primary amine or thiol groups of bifunctional cross‐linkers. Finally, these cross‐linked micelles were successfully used as nanoreactors in the synthesis of gold nanoparticles (AuNPs) in aqueous media. Both CCL and ILCL micelles were found to be good stabilizers for AuNPs in aqueous media. Both CCL‐ and ILCL‐stabilized AuNP dispersions were stable for a long time without any size changes and flocculation at room temperature. 2-Dimethylaminoethyl methacrylate (DMAEMA) and 2-diethylaminoethyl methacrylate (DEAEMA) block copolymers have been synthesized by using poly(ethylene glycol), poly(tetrahydrofuran) (PTHF) and poly(ethylene butylenes) macroinitiators with copper mediated living radical polymerization. The use of difunctional macroinitiator gave ABA block copolymers with narrow polydispersities (PDI) and controlled number average molecular weights (Mn’s). By using DMAEMA, polymerizations proceed with excellent first order kinetics indicative of well-controlled living polymerization. Online 1H NMR monitoring has been used to investigate the polymerization of DEAEMA. The first order kinetic plots for the polymerization of DEAMA showed two different rate regimes ascribed to an induction period which is not observed for DMAEMA. ABA triblock copolymers with DMAEMA as the A blocks and PTHF or PBD as B blocks leads to amphiphilic block copolymers with Mn’s between 22 and 24 K (PDI 1.24-1.32) which form aggregates/micelles in solution. The critical aggregation concentrations, as determined by pyrene fluorimetry, are 0.07 and 0.03 g dm−1 for PTHF- and PBD-containing triblocks respectively.  相似文献

Soft tissues, such as fat and skin, present high flexibility and are capable of withstanding large deformation in various functions. Hydrogels that can resemble the mechanical performance of soft tissue are unique and widely demanded. In this study, micellar hydrogels based on biocompatible poly(l ‐glutamic acid) (PLGA) were designed with the enhanced capacity to bear large deformation. Amphipathic triblock copolymer poly(ethylene glycol) acrylate‐co‐poly(ε‐caprolactone)‐co‐poly (ethylene glycol) acrylate (APEG‐PCL‐APEG) with two terminal double bonds was synthesized and self‐assembled into micelles. At the same time, graft copolymers, poly(l ‐glutamic acid)‐g‐hydroxyethyl methacrylate (PLGA‐g‐HEMA) with double bonds were synthesized. APEG‐PCL‐APEG micelles and PLGA‐g‐HEMA were mixed to construct micellar hydrogel via radical polymerization. The crystalline structure and hydrophobic aggregation of copolymers (APEG‐PCL‐APEG) were found to associate with PCL molecular weight. Due to the hydrophobic stress dissipation and crystalline structure of the micelles, the softness and toughness of hydrogels were promoted, exhibiting a 25% increase in ultimate strain. Moreover, the micellar hydrogels were able to load proteins with long‐term retention. In addition, under dynamic mechanical stimulation, the release of proteins could be accelerated. Besides, the micellar hydrogels also supported rabbit adipose‐derived stem cells (rASCs) growth, thus exhibiting the potential toward soft tissue engineering. © 2019 Wiley Periodicals, Inc. J. Polym. A series of amphiphilic triblock copolymers, poly[oligo(ethylene glycol) methacrylate]xblock‐poly(ε‐caprolactone)‐block‐poly[oligo(ethylene glycol) methacrylate]x, POEGMACo(x), were synthesized. Formation of hydrophobic domains as cores of the micelles was studied by fluorescence spectroscopy. The critical micelle concentrations in aqueous solution were found to be in the range of circa 10?6 M. A novel methodology by modulated temperature differential scanning calorimetry was developed to determine critical micelle temperature. A significant concentration dependence of cmt was found. Dynamic light scattering measurements showed a bidispersed size distribution. The micelles showed reversible dispersion/aggregation in response to temperature cycles with lower critical solution temperature between 75 and 85 °C. The interplay of the two hydrophobic and one thermoresponsive macromolecular chains offers the chance to more complex morphologies. © 2010 Wiley Periodicals, Inc. ABA‐type amphiphilic tri‐block copolymers were successfully synthesized from poly(ethylene oxide) derivatives through anionic polymerization. When poly(styrene) anions were reacted with telechelic bromine‐terminated poly(ethylene oxide) ( 1 ) in 2:1 mole ratio, poly(styrene)‐b‐poly(ethylene oxide)‐b‐poly(styrene) tri‐block copolymers were formed. Similarly, stable telechelic carbanion‐terminated poly(ethylene oxide), prepared from 1,1‐diphenylethylene‐terminated poly (ethylene oxide) ( 2 ) and sec‐BuLi, was also used to polymerize styrene and methyl methacrylate separately, as a result, poly (styrene)‐b‐poly(ethylene oxide)‐b‐poly(styrene) and poly (methyl methacrylate)‐b‐poly(ethylene oxide)‐b‐poly(methyl methacrylate) tri‐block copolymers were formed respectively. All these tri‐block copolymers and poly(ethylene oxide) derivatives, 1 and 2 , were characterized by spectroscopic, calorimetric, and chromatographic techniques. Theoretical molecular weights of the tri‐block copolymers were found to be similar to the experimental molecular weights, and narrow polydispersity index was observed for all the tri‐block copolymers. Differential scanning calorimetric studies confirmed the presence of glass transition temperatures of poly(ethylene oxide), poly(styrene), and poly(methyl methacrylate) blocks in the tri‐block copolymers. Poly(styrene)‐b‐poly(ethylene oxide)‐b‐poly(styrene) tri‐block copolymers, prepared from polystyryl anion and 1 , were successfully used to prepare micelles, and according to the transmission electron microscopy and dynamic light scattering results, the micelles were spherical in shape with mean average diameter of 106 ± 5 nm. © 2011 Wiley Periodicals, Inc. This paper reports the studies on micelle formation of new biodegradable amphiphilic poly(ethylene oxide)-poly[(R)-3-hydroxybutyrate]-poly(ethylene oxide) (PEO-PHB-PEO) triblock copolymer with various PHB and PEO block lengths in aqueous solution. Transmission electron microscopy showed that the micelles took an approximately spherical shape with the surrounding diffuse outer shell formed by hydrophilic PEO blocks. The size distribution of the micelles formed by one triblock copolymer was demonstrated by dynamic light scattering technique. The critical micellization phenomena of the copolymers were extensively studied using the pyrene fluorescence dye absorption technique, and the (0,0) band changes of pyrene excitation spectra were used as a probe for the studies. For the copolymers studied in this report, the critical micelle concentrations ranged from 1.3 x 10(-5) to 1.1 x 10(-3) g/mL. For the same PEO block length of 5000, the critical micelle concentrations decreased with an increase in PHB block length, and the change was more significant in the short PHB range. It was found that the micelle formation of the biodegradable amphiphilic triblock copolymers consisting of poly(beta-hydroxyalkanoic acid) and PEO was relatively temperature-insensitive, which is quite different from their counterparts consisting of poly(alpha-hydroxyalkanoic acid) and PEO.  相似文献

Two brush-type amphiphilic diblock copolymers, poly(poly(ethylene glycol)methyl ether methacrylate-block-polystyrene) (P(PEGMA)-b-PS) and poly(glycidyl methacrylate)-block-poly(poly(ethylene glycol)methyl ether methacrylate) (P(GMA)-b-P(PEGMA)) were synthesized, respectively, via consecutive atom-transfer radical polymerizations (ATRPs) and reversible addition-fragmentation chain-transfer (RAFT) polymerizations. The diblock copolymers were characterized by gel permeation chromatography (GPC), (1)H nuclear magnetic resonance (NMR) spectroscopy, and FT-IR spectroscopy. The aggregation behavior of the two amphiphilic diblock copolymers in water was also studied. Scanning electron and transmission electron microscopic images revealed that spherical micelles (40-80 nm in diameter) from self-assembly of the P(PEGMA)-b-PS copolymers and wormlike micelles (60-120 nm in length and 20-30 nm in diameter) from self-assembly of the P(GMA)-b-P(PEGMA) copolymers were prevalent. The spherical P(PEGMA)-b-PS micelles could self-assemble gradually into giant aggregates of several micrometers in diameter.  相似文献

A series of novel side‐chain liquid crystalline ABC triblock copolymers composed of poly(ethylene oxide) (PEO), polystyrene (PS), and poly[6‐(4‐methoxy‐4′‐oxy‐azobenzene) hexyl methacrylate] (PMMAZO) were synthesized by atom transfer radical polymerization (ATRP) using CuBr/1,1,4,7,7‐pentamethyldiethylenetriamine (PMDETA) as a catalyst system. First, the bromine‐terminated diblock copolymer poly(ethylene oxide)‐block‐polystyrene (PEO‐PS‐Br) was prepared by the ATRP of styrene initiated with the macro‐initiator PEO‐Br, which was obtained from the esterification of PEO and 2‐bromo‐2‐methylpropionyl bromide. An azobenzene‐containing block of PMMAZO with different molecular weights was then introduced into the diblock copolymer by a second ATRP to synthesize the novel side‐chain liquid crystalline ABC triblock copolymer poly(ethylene oxide)‐block‐polystyrene‐block‐poly[6‐(4‐methoxy‐4′‐oxy‐azobenzene) hexyl methacrylate] (PEO‐PS‐PMMAZO). These block copolymers were characterized using proton nuclear magnetic resonance (1H NMR) and gel permeation chromatograph (GPC). Their thermotropic phase behaviors were investigated using differential scanning calorimetry (DSC) and polarized optical microscope (POM). These triblock copolymers exhibited a smectic phase and a nematic phase over a relatively wide temperature range. Langmuir monolayers and Langmuir–Blodgett (LB) film morphology of amphiphilic triblock copolymers are studied using surface pressure-area measurements and atomic force microscopy (AFM), respectively. The triblock copolymers are composed of long water-soluble poly(ethylene oxide) (PEO) chains as middle block with very short poly(perfluorohexylethyl methacrylate) (PFMA) end blocks. The surface pressure-area isotherms show phase transitions in the brush regime. This phase transition is due to a rearrangement of PFMA block at the air–water interface. It becomes more significant with increasing PFMA content in the copolymer. LB films transferred at low surface pressures from the air–water interface to hydrophilic silicon substrates show surface micelles in the size range of 50–100 nm. A typical crystalline morphology of the corresponding PEO homopolymer is observed in LB films of copolymers with very short PFMA blocks, transferred in the brush region at high surface pressure. This crystallization is hindered with increasing PFMA content in the copolymer.  相似文献

A new class of supramolecular and biomimetic glycopolymer/poly(epsilon-caprolactone)-based polypseudorotaxane/glycopolymer triblock copolymers (poly(D-gluconamidoethyl methacrylate)-PPR-poly(D-gluconamidoethyl methacrylate), PGAMA-PPR-PGAMA), exhibiting controlled molecular weights and low polydispersities, was synthesized by the combination of ring-opening polymerization of epsilon-caprolactone, supramolecular inclusion reaction, and direct atom transfer radical polymerization (ATRP) of unprotected D-gluconamidoethyl methacrylate (GAMA) glycomonomer. The PPR macroinitiator for ATRP was prepared by the inclusion complexation of biodegradable poly(epsilon-caprolactone) (PCL) with alpha-cyclodextrin (alpha-CD), in which the crystalline PCL segments were included into the hydrophobic alpha-CD cavities and their crystallization was completely suppressed. Moreover, the self-assembled aggregates from these triblock copolymers have a hydrophilic glycopolymer shell and an oligosaccharide threaded polypseudorotaxane core, which changed from spherical micelles to vesicles with the decreasing weight fraction of glycopolymer segments. Furthermore, it was demonstrated that these triblock copolymers had specific biomolecular recognition with concanavalin A (Con A) in comparison with bovine serum albumin (BSA). To the best of our knowledge, this is the first report that describes the synthesis of supramolecular and biomimetic polypseudorotaxane/glycopolymer biohybrids and the fabrication of glucose-shelled and oligosaccharide-threaded polypseudorotaxane-cored aggregates. This hopefully provides a platform for targeted drug delivery and for studying the biomolecular recognition between sugar and lectin.  相似文献

Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3-hydroxy-butyrate)-poly(ethyl ethylene phosphate) (PEEP-b-PHB-b-PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers self-assemble into biodegradable nanoparticles with a core-shell micellar structure in aqueous solution, verified by the probe-based fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3-hydroxybutyrate) (PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles as a model drug and a sustained drug release from the micelles is observed. MTT assay also demonstrates that the block copolymers are biocompatible, rendering these copolymers attractive for drug delivery. 以基于亚胺键的嵌段共聚物为构筑单元的温度/pH响应性共聚物复合胶束(CMs), 由于具有亚胺键和核-壳-冠结构, 表现出较高的灵敏度和稳定性. 以聚乙二醇单甲醚(MPEG)、 N-乙烯基己内酰胺(NVCL)和ε-己内酯(ε-CL)为原料, 分别制备了端醛基聚乙二醇单甲醚(MPEG-CHO)、 端醛基聚N-乙烯基己内酰胺(PNVCL-CHO)和端氨基聚己内酯(H2N-PCL), 利用希夫碱反应, 进一步制备了基于亚胺键的聚乙二醇单甲醚-b-聚己内酯(MPEG-b-PCL)和聚N-乙烯基己内酰胺-b-聚己内酯(PNVCL-b-PCL)嵌段共聚物, 对共聚物结构进行了确认. 以MPEG-b-PCL和PNVCL-b-PCL为构筑单元, 制备了共聚物复合胶束, 研究了复合胶束对阿霉素的包载、 释放性质和细胞毒性等. 研究结果表明, 室温下MPEG-b-PCL和PNVCL-b-PCL能够在水中自组装形成以PCL为核、 MPEG和PNVCL为混合壳的共聚物复合胶束, 在生理温度下, 温敏性PNVCL链段发生相变塌缩在PCL核表面, 能够防止药物扩散释放, 亲水性MPEG链段形成可控通道. 药物体外释放结果表明, 在弱酸性环境中, 亚胺键能够断裂, 胶束被破坏, 促进药物的释放, 噻唑蓝(MTT)实验表明, 复合胶束的细胞毒性较低.  相似文献

Amphiphilic triblock copolymer of poly(2,2-dimethyl-trimethylene carbonate)–poly(ethylene glycol)–poly(2,2-dimethyl trimethylene carbonate) (PDTC–PEG–PDTC) was synthesized by dihydroxyl capped PEG with molecular weight of 1,000, 4,000, and 6,000 in the presence of rare earth tris(2,6-di-tert-butyl-4-methylphenolate)s. The rare earth phenolates/PEG system could prepare triblock copolymer with predictable molecular weights with narrow molecular weight distribution. The polymers were characterized by nuclear magnetic resonance spectroscopy, gel permeation chromotography, and differential scanning calorimetry to confirm the structure. The micelles formed from the amphiphilic triblock copolymer were determined by fluorescence spectrophotometer and dynamic light scattering. The critical micelle concentrations fell in the range of 1.67∼5.25 mg/L. Transmission electron microscopy pictures showed that the micelles possess spherical morphology, and the diameters of micelles in number averaged scale ranged from 20–70 nm. The micelles formed from triblock amphiphilic copolymers were explored as carrier for indomethacin (IND), and they could enhance IND solubility in water dramatically.  相似文献

Copper(I)-mediated living radical polymerization was used to synthesize a series of self-crosslinkable ABA triblock copolymers in which the side blocks are formed by a monomer supporting a reactive functional group, as allyl methacrylate (AMA). The copolymers were prepared according with a two steps synthetic methodology. In the first step, ,ω-dibromo homopolymers of polystyrene (PS), poly(methyl methacrylate) (PMMA) and poly(butyl acrylate) (PBA) were synthesized by atom transfer radical polymerization (ATRP). In the second step, these telechelic polymers were employed as macroinitiators for the ATRP of AMA in benzonitrile solution at 70 °C with CuCl/N,N,N′,N″,N″-pentamethyldiethylenetriamine (PMDETA) as catalyst system in order to obtain well-defined functionalized triblock copolymers. The living nature of the block copolymerizations involved was investigated in each case and a similar general behaviour was found. Thus, the molecular weights increased fairly linearly with the conversion degree with first-order kinetics in respect of monomer until moderate conversions, where secondary reactions become more relevant. Finally, intermacromolecular crosslinking were observed giving macrogels as a unique reaction product. The polymers were characterized by different characterization techniques, such as size exclusion chromatography (SEC), 1H NMR spectroscopy and differential scanning calorimetry (DSC). In addition, the facile thermal crosslinking of these block copolymers was evaluated from rheological measurements.  相似文献

Multisegmented poly(methacrylate)s were synthesized using one pot reversible addition fragmentation chain transfer polymerization. Initially, a series of triblock copolymers were synthesized with different ratios of trimethylsilyl methacrylate, di(ethylene oxide) methacrylate, and oligo(ethylene oxide) methacrylate, and different total polymer molecular weights. Additionally, a polymer containing seven distinct blocks of methacrylic monomers was synthesized in one pot. Polystyrene‐block‐poly(2‐vinyl pyridine)‐block‐poly(methyl methacrylate) ABC triblock copolymers were synthesized by sequential living anionic polymerization. Their solution properties were investigated in toluene, which is a bad solvent for the middle block. Spherical micelles are formed, which consist of a poly(2‐vinyl pyridine) dense core bearing polystyrene and poly(methyl methacrylate) soluble chains at the corona. These micelles exhibit the architecture of heteroarm star copolymers obtained by “living” polymerization methods. The aggregation numbers strongly depend on the length of the insoluble P2VP middle block, thus remarkably affecting the size of the micelles.  相似文献

Amphiphilic triblock copolymers of poly(methyl methacrylate)-b-poly(ethylene oxide)-b-poly(methyl methacrylate) (PMMA-b-PEO-b-PMMA) with well-defined structure were synthesized via atom transfer radical polymerization (ATRP) of methyl methacrylate (MMA) initiated by the PEO macroinitiator. The macroinitiator and triblock copolymer with different PMMA and/or PEO block lengths were characterized with 1H and 13C NMR and gel permeation chromatography (GPC). The micelle formed by these triblock copolymers in aqueous solutions was detected by fluorescence excitation and emission spectra of pyrene probe. The critical micelle concentration (CMC) ranged from 0.0019 to 0.016 mg/mL and increased with increasing PMMA block length, while the PEO block length had less effect on the CMC. The partition constant Kv for pyrene in the micelle and in aqueous solution was about 105. The triblock copolymer appeared to form the micelles with hydrophobic PMMA core and hydrophilic PEO loop chain corona. The hydrodynamic radius Rh,app of the micelle measured with dynamic light scattering (DLS) ranged from 17.3 to 24.0 nm and increased with increasing PEO block length to form thicker corona. The spherical shape of the micelle of the triblock copolymers was observed with an atomic force microscope (AFM). Increasing hydrophobic PMMA block length effectively promoted the micelle formation in aqueous solutions, but the micelles were stable even only with short PMMA blocks.  相似文献

Miktoarm star triblock copolymers mu-[poly(ethylethylene)][poly(ethylene oxide)][poly(perfluoropropylene oxide)] self-assemble in dilute aqueous solution to give multicompartment micelles with the cores consisting of discrete poly(ethylethylene) and poly(perfluoropropylene oxide) domains. Tetrahydrofuran is a selective solvent for both the poly(ethylethylene) and poly(ethylene oxide) blocks, and thus in tetrahydrofuran mixed corona micelles are favored with poly(perfluoropropylene oxide) cores. The introduction of tetrahydrofuran into water induces an evolution from multicompartment micelles to mixed corona [poly(ethylethylene) + poly(ethylene oxide)] micelles, as verified by dynamic light scattering and nuclear magnetic resonance spectroscopy. A mixed solvent containing 60 wt % tetrahydrofuran corresponds to the transition point, as verified by analysis of a poly(ethylethylene)-poly(ethylene oxide) diblock copolymer in the same solvent mixtures. Furthermore, cryogenic transmission electron microscopy suggests that, as the poly(ethylethylene) block transitions from the core to the corona, the micelle morphologies evolve from disks to oblate ellipsoid micelles (with some vesicles), with worms and spheres evident at intermediate compositions.  相似文献
