Het(aryl)isatin to het(aryl)aminoindoline scaffold hopping: A route to selective inhibitors of matrix metalloproteinases

Matrix metalloproteinases (MMPs) are a large family of zinc-dependent endoproteases known to exert multiple regulatory roles in tumor progression. A variety of chemical classes have been explored for targeting individual MMP isoforms. In the present study, we further developed our isatin based scaffold BB0223107 capable of binding to and inactivating MMP-2 in a zinc-independent manner (Agamennone et al., 2016). Forty four new compounds were synthesized based on the modified BB0223107. All compounds were tested in enzyme inhibition assays against MMP-2, ?8 and ?13. SAR studies demonstrated that 5-het(aryl)-3-aminoindolin-2-ones (37–39) were active toward MMP-2 and MMP-13. The most potent compounds 33 and 37 displayed an IC₅₀ of 3 µM against MMP-13 and showed a negligible activity toward MMP-8; almost all new compounds were inactive toward MMP-8. Replacement of the isatin ring with a biaryl system (compound 33) did not decrease the potency against MMP-13 but reduced the selectivity. Structure-based computational studies were carried out to rationalize the inhibitory activity data. The analysis of binding geometries confirmed that all fragments occupied the S1′ site in the three enzymes while no ligand was able to bind the catalytic zinc ion. To the best of our knowledge, this is the first example of 3-aminoindolin-2-one-based MMP inhibitors that, based on the computer modeling study, do not coordinate the zinc ion. Thus, the het(aryl)-3-aminoindolin-2-one derivatives emerge as a drug-like and promising chemotype that, along with the hetaryl variations, represents an alternative and thrifty tool for chemical space exploration aimed at MMP inhibitor design.

基于不确定控制理论的最优策略研究

本文在不确定理论的框架下,研究一类带背景状态变量的最优控制模型.在乐观值准则下,利用不确定动态规划的方法,证明了不确定最优性原则,得到最优性方程.作为应用,求解一个固定缴费(DC)型养老金的最优投资策略问题,在乐观值准则下,以工资变量为背景状态变量,建立养老金模型.通过求解不确定最优性方程得到最优投资策略和最优支付率.     

Regularity of optimal sets for some functional involving eigenvalues of an operator in divergence form

In this paper we consider minimizers of the functional$\min \{{\lambda }_1(\mathrm{\Omega })+\cdots +{\lambda }_k(\mathrm{\Omega })+\mathrm{\Lambda }|\mathrm{\Omega }|,:\mathrm{\Omega }\subset D\text{ open}\}$ where $D\subset {\mathbb{R}}^d$ is a bounded open set and where $0<{\lambda }_1(\mathrm{\Omega })\le \cdots \le {\lambda }_k(\mathrm{\Omega })$ are the first k eigenvalues on Ω of an operator in divergence form with Dirichlet boundary condition and with Hölder continuous coefficients. We prove that the optimal sets ${\mathrm{\Omega }}^{⁎}$ have finite perimeter and that their free boundary $\partial {\mathrm{\Omega }}^{⁎}\cap D$ is composed of a regular part, which is locally the graph of a $C^{1,\alpha }$-regular function, and a singular part, which is empty if $d<d^{⁎}$, discrete if $d=d^{⁎}$ and of Hausdorff dimension at most $d-d^{⁎}$ if $d>d^{⁎}$, for some $d^{⁎}\in \{5,6,7\}$.     

A Far-Red Emitting Fluorescent Chemogenetic Reporter for In Vivo Molecular Imaging

Far-red emitting fluorescent labels are highly desirable for spectral multiplexing and deep tissue imaging. Here, we describe the generation of frFAST (far-red Fluorescence Activating and absorption Shifting Tag), a 14-kDa monomeric protein that forms a bright far-red fluorescent assembly with (4-hydroxy-3-methoxy-phenyl)allylidene rhodanine (HPAR-3OM). As HPAR-3OM is essentially non-fluorescent in solution and in cells, frFAST can be imaged with high contrast in presence of free HPAR-3OM, which allowed the rapid and efficient imaging of frFAST fusions in live cells, zebrafish embryo/larvae, and chicken embryos. Beyond enabling the genetic encoding of far-red fluorescence, frFAST allowed the design of a far-red chemogenetic reporter of protein–protein interactions, demonstrating its great potential for the design of innovative far-red emitting biosensors.     

钩形弯交叉口信号协同控制优化方法研究

针对车辆在钩形弯交叉口与相邻常规交叉口间的整体运行特性，建立了各条车道的车辆平均延误计算模型。以各交叉口的周期时长和相位绿灯时间为约束，钩形弯交叉口与相邻常规交叉口间的车辆平均延误最小为目标，建立了信号协同配时方案。以位于苏州市高新区内有轨电车沿线的两个相邻交叉口为例，采用实际数据验证所建立的方法，并设置钩形弯渠化方案。将设置的钩形弯交叉口与其相邻常规交叉口的信号协同作为改进方法，在VISSIM软件中对比分析改进方案与现状方案的车辆延误指标。实验结果表明，钩形弯交叉口与相邻常规交叉口的信号协同优化方法可有效提高交叉口的通行能力，尤其是干道直行机动车的通行能力，可降低车辆的平均延误。     

基于LQG控制的整车8-DOF系统振动分析

建立了整车8-DOF系统动力学模型,考虑了主动悬架控制,并增设了主动座椅控制,设计了车辆主动悬架系统的LQG控制器。基于Matlab仿真平台建立了整车8-DOF系统动力学仿真模型,对所得最优控制策略下的动态响应进行了仿真验证。仿真结果表明:为了改善人椅系统质心及车身质心的跳振性能需要在一定程度上弱化各轮轮胎动位移性能。从控制效能上来看,该最优控制器能够满足各行驶状态下对悬架性能的要求,改善了车辆的行驶平顺性。     

基于低碳理念的鄂州市城市土地集约利用评价

结合国内外低碳土地利用的研究背景和实践,分析出不同土地利用方式碳排放效应,并对城市土地低碳化与集约化利用进行定性与定量分析,并采用鄂州市实际构建低碳理念下城市土地集约利用评价指标体系,对鄂州市2000-2013年土地利用状况进行分析评价.由此,总结鄂州市再加入低碳因素后土地集约利用的变化规律,并提出相关建议.     

Wave-based vibration control of large cable net structures

Large cable net structures have been widely applied in aerospace engineering due to the feature of light-weight, high packaging efficiency, and high thermal stability. Structural vibrations induced by a variety of disturbances are inevitable in the space environment, resulting in the requirement of effective vibration control strategies for large cable net structures. Since the large cable net structures have many closely spaced vibrational modes in the range of low frequencies, traditional modal based control may cause modal truncation and spillover problems. In this paper, a wave-based boundary control strategy is adopted and its effectiveness to control the vibration of cable net structures is investigated, by transfer function analysis and numerical methods. It is found that the structural vibration can be absolutely resisted by applying the wave-based boundary controllers onto all the exterior nodes, when disturbances come from the external boundaries of the cable net. Our results in this paper can provide a theoretical basis for the vibration control of large cable net structures.     

地理国情普查地表覆盖内业解译的质量问题研究

地表覆盖内业解译是地理国情普查的一道重要工序。地理国情地表覆盖内业解译现存图斑平面位置精度不足、分类错误多种多样等普遍问题,问题突出典型。文中针对这些问题图文并茂、清晰明了的进行了说明,并从多方面分析问题产生的原因,提出加强对技术指标和采集原则的学习和理解,加强内业与外业的结合,充分发挥解译样本的作用等建议,以有效提高内业解译的质量。
