HPLC-ELSD测定不同厂家中/长链脂肪乳注射液中的溶血磷脂酰胆碱

采用高效液相色谱-蒸发光散射检测器(HPLC-ELSD)测定不同厂家中/长链脂肪乳注射液中溶血磷脂酰胆碱(LPC)的含量。采用Lichrospher Diol色谱柱(250mm×4.6mm,5μm),以正己烷-异丙醇-水(29.5:59:11.5,V/V/V)为流动相A,正己烷-异丙醇(43:57,V/V)为流动相B,进行梯度洗脱,流速1mL/min,漂移管温度85℃,气体流速2.7L/min。结果溶血磷脂酰胆碱在0.299—2.99mg/mL浓度范围内线性关系良好(r=0.9999),平均回收率为99.62%,RSD为0.89%。不同厂家产品中溶血磷脂酰胆碱的含量为0.92—1.34mg/mL。方法准确、方便,可以准确测定中/长链脂肪乳注射液中溶血磷脂酰胆碱的含量。

Determination of Lyso-Phosphatidylcholine in MCT/LCT Fat Emulsion Injection from Different Manufacturers by HPLC-ELSD

The contents of lyso-phosphatidylcholine(LPC) in MCT/LCT fat emulsion injection from different manufacturers were determined by HPLC-ELSD.The lichrospher Diol (250mm×4.6mm,5μm) column was used.The mobile phase were A:n-hexane-isopropanol-water (29.5:59:11.5,V/V/V) and B:n-hexane-isopropanol(43:57,V/V) with gradient elution.The flow rate was lmL/min.The temperature of drift tube was 85℃and the gas flow rate was 2.7L/min.There was a good linear relationship for LPC in the range of 0.299-2.99mg/mL (r=0.9999).The average recovery was 99.62%and RSD was 0.89%.The concentrations of LPC in MCT/LCT fat emulsion injection from different manufacturers were from 0.92mg/mL to 1.34mg/mL.The method is accurate and convenient,and could be suitable for the determination of the content of LPC in MCT/LCT fat emulsion injection.
