偶合反应伏安酶联免疫分析测定人血清乙型肝炎E抗体的研究

本文提出用竞争性抑制偶合反应伏安酶联免疫分析法测定人血清乙型肝炎Ｅ抗体（ＨＢｅＡｂ）方法基于酶标ＨＢｅＡｂ辣根过氧化物酶（ＨＲＰ）催化Ｈ２Ｏ２氧化邻联甲苯胺（ＯＴ）的反应与邻联甲苯胺氧化产物在电极上的还原反应相偶合，测定标记在ＨＢｅＡｂ上ＨＲＰ量，以求得抑制免疫反应的乙型肝炎Ｅ抗体含量本法测定酶标ＨＢｅＡｂＨＲＰ及ＨＢｅＡｂ的灵敏度均高于经典的ＥＬＩＳＡ光度法方法用于病人血清样品分析，与ＥＬＩＳＡ光度法对照，其相关性很好

Determination of HBeAb in Serum by Voltammetric Enzyme Immunoassay with Coupling Reaction

In this paper, a new voltammetric enzyme immunoassay with coupling reaction for determining the HBeAb in human serum is developed. It is based on coupling the reaction of OT H 2O 2 catalyzed by HBeAb HRP with the electro reduction reaction of the oxidizing product of OT to measure the activity of HBeAb HRP and the latter reaction exhibits a sensitive second order derivative linear sweep voltammetric wave at -0.58V(SCE) in BR buffer, so HBeAb could be measured. The present method to detect HBeAb HRP and HBeAb in human serum is more sensitive than ELISA spectrophotometric method. The competitive inhibition immunological method is used to determine HBeAb. Some patient serum samples have been measured by this method and ELISA method with satisfactory results.
