小麦胚性非再生细胞中游离氨基酸分析

摘    要:利用毛细管电泳技术分析了小麦胚性非再生细胞中游离氨基酸的组成与含量。与已经发表的胚性再生细胞游离氨基酸浓度比较发现,两类胚性细胞中游离氨基酸的含量差异明显:在9种可以分开的游离氨基酸中,胚性再生细胞中的游离精氨酸、赖氨酸、谷氨酸和天冬氨酸分别是胚性非再生细胞中的10、3、2和2倍,其它氨基酸的浓度在两类细胞中没有明显的变化。本研究不但为检测植物细胞氨基酸成分提供了方便快捷的方法,而且为进一步探索小麦两类胚性细胞在分子水平上的差异和形成机理提供了生化基础。

Abstract:The contents of free amino acids(AAs) in wheat embryonic cells without regenerated ability were investigated by capillary electrophoresis(CE).It is found that these AAs of wheat embryonic cells without(regenerated) ability have significant difference from that of wheat embryonic cells with regenerated ability reported before.Among the nine determined AAs in the two types of cells,the content of arginine,lysine,glutamic acid and aspartic acid in the embryonic cells with regenerated ability is 10,3,2 and 2 times more than that of in embryonic cells without regenerated ability respectively.Others have no obvious difference in density.This study could not only provide a convenient and speedy method for detecting AAs in plant cells,but also provide a biochemical basement for the research of embryonic formation mechanism and differentiation of the two types of embryonic cells in molecular.
