利用可见光及紫外分光光度法测定酸浆宿萼总黄酮含量,可见光分光光度法是以芦丁为参照品,硝酸铝作显色剂,在510 nm波长测定总黄酮含量.紫外分光光度法是以芦丁为参照品,在359 nm波长测定总黄酮含量.经过比较,认为紫外分光光度法是一种较理想的测定方法.  相似文献

从锦灯笼中分离得到5个甾体类化合物,分别鉴定为酸浆素A(1,alkekengilin A)、酸浆素B(2,alkekengilin B)、酸浆苦素D(3,physalin D)、酸浆苦素P(4,physalin P)、4,7-二去氢新酸浆苦素B(5,4,7-didehydro-neophysalin B).其中酸浆素A(1,alkekengilin A)和酸浆素B(2,alkekengilin B)为新的天然产物.运用现代波谱技术对上述化合物的结构进行了确证.  相似文献

目的观察人乳头瘤病毒(HPV)和肥大细胞(MC)在正常宫颈组织、宫颈上皮内瘤变(CIN)及宫颈鳞癌(SCC)组织中的表达。方法采用S—P免疫组化法检测60例CIN患者、40例SCC组患者和20例子宫肌瘤患者(对照组)组织中MC的表达,同时采用DNA导流杂交基因芯片技术检测各级组织脱落细胞中HPVDNA分型。结果随着宫颈病变程度的加重.高危型(HR)-HPV感染阳性率逐渐增加,分别为10%、55%、65%、85%、100%,其中最常见的高危型HPV感染的基因型为:16、58、52、18。与CINIII组比较,MC密度在CIN11组、SCC组、对照组中的表达明显升高(P〈0.01或0.05),并且MC与肿瘤临床分期和血管转移有关(均P〈0.05)。MC低表达者与高表达者间HR—HPV阳性率的差异有统计学意义(P〈0.01)。结论HR—HPV感染可以降低MC的表达,癌组织中浸润的MC可以促进肿瘤血管的形成和肿瘤的浸润转移。  相似文献

We demonstrate the capability of differential interference contrast (DIC) microscopy as a simple and useful tool for studying cellular events without fluorescence labeling. By coupling an advanced DIC microscope to a computer-controlled motorized vertical stage and a high-speed, high-resolution CCD camera, real-time three-dimensional monitoring is possible in a high-throughput manner. The performance among three modes of microscopy, bright-field, dark-field and DIC, in terms of horizontal resolving power and vertical sectioning was investigated. As a model, exocytosis of rat peritoneal mast cells was recorded on the subsecond time scale. Three-dimensional tracking of granules during degranulation was achieved and granule–granule fusion before plasma membrane fusion was recorded. An affinity two‐dimensional chromatography method was developed for the recognition, separation, and identification of allergic components from tubeimu saponin extracts, a preparation often injected to treat various conditions as indicated by traditional Chinese medicine. Rat basophilic leukemia‐2H3 cell membranes were used as the stationary phase of a membrane affinity chromatography column to capture components with affinity for mast cells that could be involved in a degranulation reaction. The retained components were enriched and analyzed by membrane affinity chromatography with liquid chromatography and mass spectrometry via a port switch valve. Suitability and reliability of the method was investigated using appropriate standards, and then, the method was applied to identify components retained from tubeimu saponin extracts. Tubeimoside A was identified in this way as a potential allergen, and degranulation assays confirmed that tubeimoside A induces RBL‐2H3 cell degranulation in a dose‐dependent manner. An increase in Ca2+ influx indicated that degranulation induced by tubeimoside A is likely Ca2+ dependent. Coupled with the degranulation assay, RBL‐2H3 cell‐based affinity chromatography coupled with liquid chromatography and mass spectrometry is an effective method for screening and identifying allergic components from tubeimu saponin extracts.  相似文献

通过水提、酸沉法得到锦灯笼果实提取物,其中蛋白质含量为188 mg/g(以提取物干重计),共含有18种氨基酸,其中8种人体必需氨基酸占氨基酸总量的31%。基于鸟枪法蛋白质组学的分析方法,用纳升级反相液相色谱-串联质谱(nano-RPLC-MS/MS)系统分析锦灯笼果实提取物中蛋白质的酶解产物,结合数据库检索,共鉴定得到60种蛋白质;通过生物信息学分析,得到锦灯笼提取物中的蛋白质具有催化活性、抗氧化活性、酶调节活性、养分贮液囊活性、运输活性、结合活性六大生物活性,其中鉴定到与抗氧化相关的蛋白质有3种,为锦灯笼中蛋白质的功能性质的进一步研究奠定了基础。  相似文献
