The quinolones are derivatives of oxoquinolines and mostly known for their antibacterial and antiviral activities. Many quinolones are chiral compounds having asymmetric centers and important due to their enantioselective biological activities. In order to study the biological activities of quinolone enantiomers, to control the manufacturing of homochiral drugs and to prepare necessary quantities of pure enantiomers for preclinical or clinical trials, respective chiral separation methods are urgently needed. In this context, the present review discusses chromatographic and electrophoretic methods for the enantioseparation of chiral quinolones and provides some useful information on their physical and pharmaceutical properties. The drawbacks of currently used techniques are revealed and ways to overcome them are outlined. Moreover, recommendations for an optimal choice of a separation protocol are given.  相似文献

Abstract

The total energy, dipole moment and electron densities for each possible rotational conformation about the Cpy-S bonds of di-2-pyridyl disulfide were evaluated by using the semi-quantitative CNDO/2 method. The conformations in which the pyridine rings are coplanar with the valency plane of the bonded sulfur atom (cis-cis, cis-trans and trans-trans) were predicted to be the most favored ones.

Results of the theoretical study, when compared to some experimental determinations such as dipole moment and variable temperature pmr spectra, provided evidence that easy interconversion between these conformations can occur.  相似文献

This study presents a simple and green dispersive micro‐solid phase extraction method for preconcentration of acidic quinolones from honey prior to high performance liquid chromatography determination. A two‐dimensional nanostructured zinc‐aluminum layered double hydroxide was synthesized and used as the sorbent for dispersive micro‐solid phase extraction. Its different characteristics from conventional sorbents is that it is dissolvable in acidic solution (pH < 4). After the extraction, the analyte elution step was omitted and thus the use of organic solvents was avoided. The key parameters influencing the extraction efficiency such as the amount of sorbent, pH of sample solution, vortex time, type and volume of acidic solution were investigated and optimized. The method exhibited low limits of detection (3.0?5.0 ng/g), good linearity (10?2000 ng/g) with coefficients of determinations higher than 0.9991, acceptable precision (RSD<9.1%) and accuracy (RE<5.8%). The proposed method is fast, efficient, eco‐friendly, and suitable for the determination of acidic quinolones in honey samples.  相似文献

The paper gives a brief account of the recently introduced Szeged index (Sz). Using this index antitubercular activities of N-2,4-difluorophenyl quinolones are subjected to quantitative structure-activity relationship analysis. The potential of Sz related to the Wiener index (W) is critically discussed. In addition, Huckel molecular orbital energies:E HOMO,E LUMO andE total were also used for comparing and modelling antitubercular activities of the quinolones. The results, based on univariate as well as multivariate regressions, have shown that W, SZ andE total give better results and that the correlations improve in multivariate regression analyses.  相似文献

The synthetic utility of trifluoromethanesulphonic anhydride (triflic anhydride, TA) without additive or base for the high-yielding synthesis of a wide variety of 1,2-benzisoxazoles from 2-hydroxyaryl aldoximes and ketoximes under mild conditions has been carried out for the first time. As a continuation of our study, syntheses of isoxazolo and isothiazolo quinolones have also been demonstrated using triflic anhydride under similar conditions. This method is simple and has benefits from the easy way to isolate the products in excellent yields.

Functional polymers with a metal–coordination interaction have been fabricated for sample pretreatment. Poly(N‐4‐vinyl‐benzyl iminodiacetic acid‐co‐methacrylic acid‐co‐styrene)‐modified magnetic nanoparticles were prepared and used as solid‐phase extraction adsorbents for the analysis of quinolones by tuning the metal–coordination interaction. In the construction of the polymer‐based adsorbents, functional monomer (N‐(4‐vinyl)‐benzyl iminodiacetic acid) and comonomers (methacrylic acid and styrene) were fabricated onto the magnetic nanoparticles by free radical polymerization. Factors affecting the performance of the adsorbents were investigated, and the results revealed that Fe3+ played a vital role in the formation of metal–coordination adsorbents. Compared with other compounds, the resultant adsorbents displayed good selectivity to quinolones due to the metal–coordination complex (N‐4‐vinyl‐benzyl iminodiacetic acid‐Fe3+quinolones). Interestingly, the captured quinolones could be rapidly released by manipulating the metal–coordination interaction with Cu2+. The linearity range for analysis of the test quinolones was 0.025–2.0 μg/mL (R2 > 0.999), and the recovery varied from 80.0 to 100.7%. Further, the proposed adsorbents were combined with high‐performance liquid chromatography for the analysis of quinolones in real urine samples. The results demonstrated that the prepared adsorbents have good selectivity and sensitivity for quinolones, showing great potential for drug analysis in real samples.  相似文献

建立并优化了同时测定牛奶中10种磺胺类、6种喹诺酮类、8种甾体激素类以及1种四环素类药物共25种兽药残留的超高效液相色谱-串联质谱检测方法。样品中目标药物经5%乙酸乙腈提取,HLB固相萃取小柱净化后,通过UPLC-MS测定,外标法定量。25种兽药在不同加标浓度下的回收率为61.6%~119.2%,组内相对标准偏差(RSD)为2.5%~13.4%,组间RSD为5.8%~14.2%,方法检出限为0.5~2.0μg/kg。  相似文献

The thermal behaviour of Mg(II), Zn(II) and Co(II) compounds of ciprofloxacin was studied by thermogravimetry (TG) and differential thermal analysis (DTA) in order to determine or to confirm some structural characteristics of substances. The complexes decompose in two steps: dehydration and pyrolytic decomposition of the anhydrous complexes to form metal oxide or metal fluoride. The dehydration process of one magnesium(II) compound takes place in two steps suggesting a marked difference in the bonding of water molecules. The different bonding mode of the ciprofloxacin molecules in both magnesium compounds leads to different residues of the thermal decompositions. Wittig olefination of 3-aminoquinoline-2,4(1H,3H)-diones 1 with ethyl (triphenylphosphoranylidene)acetate (Ph3PCHCO2Et) afforded (E)-3-amino-4-ethoxycarbonylmethylene-1,2,3,4-tetrahydro-2-quinolones (E)-2 and pyrrolo[2,3-c]quinoline-2,4(3aH,5H)-diones 3. An alternative approach for the synthesis of 3 via 3-bromoacetamidoquinoline-2,4(1H,3H)-diones 7, their corresponding triphenylphosphonium salts 8, and ylides A that undergo intramolecular Wittig reaction, was investigated. Under the applied reaction conditions, the phosphonium salts 8 and ylides A are so unstable that they partly decompose to 3-acetamidoquinoline-2,4(1H,3H)-diones 9 during the synthesis of 3.  相似文献
