Five new representatives of the cembrane class of marine natural products have been isolated as minor metabolites from an undescribed species of Eunicea, a sea whip collected near the Colombian Southwestern Caribbean Sea. The structure of the crystalline metabolite 1 was solved by single-crystal X-ray diffraction analysis. Structures could then be proposed for cembradienes 2-5 by comprehensive spectral analyses involving 2D NMR, IR, UV, and high-resolution mass spectrometry, as well as chemical interconversion studies. Compound 1 exhibited weak cytotoxicity in the NCI 3-cell line panel human cancer screening program, whereas compounds 1, 2 and 4 displayed significant antiplasmodial activity against Plasmodium falciparum.  相似文献

A novel flavone, penimethavone A (1), possessing a rare unique methyl group at ring-B, was isolated from the fungus Penicillium chrysogenum cultured from a gorgonian Carijoa sp. collected from the South China Sea. The structure was elucidated by extensive spectroscopic analysis and by comparison with related known compound. Compound 1 showed selective and moderate cytotoxicity against cervical cancer (HeLa) and rhabdomyosarcoma cell lines with IC50 values of 8.41 and 8.18 μM, respectively.  相似文献

Three new polyoxygenated diterpenoids with a rare 4-isopropyl-1,5,8a-trimethylperhydrophenanthrane structure of the klysimplexane skeleton, briarols A‒C (1‒3), and one eunicellin-based diterpenoid, briarol D (4), were isolated from Briareum violaceum, a gorgonian inhabiting Taiwanese waters. The chemical structures of these compounds were determined by employing extensive analyses of NMR and high-resolution electrospray ionization mass spectrometry (HRESIMS) data. Metabolites 1‒3 were found to possess the rarely found skeleton of the diterpenoid klysimplexin T. All isolated compounds showed very weak cytotoxic activity against the growth of three cancer cell lines. A plausible biosynthetic pathway for briarols A‒C from the coexisting eunicellin diterpenoid briarol D (4) was postulated.  相似文献

A new 19-oxygenated steroid, 25-acetate-nebrosteroid K (1), and five known steroids (26), along with one known 19-hydroxy steroidal glycoside (7) were obtained from the gorgonian Dichotella gemmacea collected from the South China Sea. The structure and relative configuration of 1 were elucidated by using comprehensive spectroscopic data including NOESY spectra. Compound 1 represents the first example of 19-oxygenated steroid with a 19-oic acid methyl ester group isolated from gorgonians. All of the isolated compounds were evaluated for lethal activity to brine shrimp Artemia salina and cytotoxicity against A549 and HL-60 cell lines. Among them, 1 showed strong lethality towards A. salina.  相似文献

Chemical examination of a Chinese gorgonian Anthogorgia sp. resulted in the isolation of seven terpenoids, including two new compounds, an rearranged serrulatane‐type diterpenoid anthogorgiene P ( 1 ) and a guaiazuene‐based terpenoid anthogorgiene Q ( 2 ). Anthogorgiene P ( 1 ) contains an unprecedented cubebane nucleus which is rarely found from nature. The structures of new compounds were elucidated on the basis of extensive spectroscopic methods (IR, UV, MS, CD and NMR). Compound 7 showed potent antifouling activity against the larval settlement of Balanus amphitrite, while 1 and 4 possessed moderate inhibition.  相似文献

Ileabethoxazole (1), a new perhydroacenaphthene-type diterpene alkaloid containing the uncommon benzoxazole moiety, was isolated from the Caribbean sea whip Pseudopterogorgia elisabethae. The structure of 1 was elucidated by extensive spectroscopic data interpretation. Ileabethoxazole showed 92% inhibition of Mycobacterium tuberculosis (H37Rv) at the concentration range of 128-64 μg/mL.  相似文献

A new pregnane, 3α-hydroxy-7-ene-6,20-dione (1), and five known steroids (26), along with one known steroidal glycoside (7) were obtained from the fungus Cladosporium sp. WZ-2008-0042 cultured from a gorgonian Dichotella gemmacea collected from the South China Sea. The structure and absolute configuration of the new compound (1) were elucidated by comprehensive spectroscopic data and X-ray diffraction data. The compound has a rare configuration of 3α-OH that is different from most of pregnanes. All of the isolated compounds were evaluated for their antiviral activities against respiratory syncytial virus (RSV). Among them, 1 exhibited potential antiviral activity with the IC50 value of 0.12 mM.  相似文献

Due to sedentary lifestyle and harsh environmental conditions, gorgonian coral extracts are recognized as a rich source of novel compounds with various biological activities, of interest to the pharmaceutical and cosmetic industries. The presented study aimed to perform chemical screening of organic extracts and semi-purified fractions obtained from the common Adriatic gorgonian, sea fan, Eunicella cavolini (Koch, 1887) and explore its abilities to exert different biological effects in vitro. Qualitative chemical evaluation revealed the presence of several classes of secondary metabolites extended with mass spectrometry analysis and tentative dereplication by using Global Natural Product Social Molecular Networking online platform (GNPS). Furthermore, fractions F4 and F3 showed the highest phenolic (3.28 ± 0.04 mg GAE/g sample) and carotene (23.11 ± 2.48 mg β-CA/g sample) content, respectively. The fraction F3 inhibited 50% of DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) and ABTS (2,2′-azino-bis (3-ethylbenzthiazolin-6-yl) sulfonic acid) radicals at the concentrations of 767.09 ± 11.57 and 157.16 ± 10.83 µg/mL, respectively. The highest anti-inflammatory potential was exhibited by F2 (IC50 = 198.70 ± 28.77 µg/mL) regarding the inhibition of albumin denaturation and F1 (IC50 = 254.49 ± 49.17 µg/mL) in terms of soybean lipoxygenase inhibition. In addition, the most pronounced antiproliferative effects were observed for all samples (IC50 ranging from 0.82 ± 0.14–231.18 ± 46.13 µg/mL) against several carcinoma cell lines, but also towards non-transformed human fibroblasts pointing to a generally cytotoxic effect. In addition, the antibacterial activity was tested by broth microdilution assay against three human pathogenic bacteria: Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. The latter was the most affected by fractions F2 and F3. Finally, further purification, isolation and characterization of pure compounds from the most active fractions are under investigation.  相似文献

A systematic re-study on gorgonian Muricella sibogae from South China Sea yielded 10 eunicellin-based diterpenoids including two new ones, sibogins C and D (1 and 2). Their structures were elucidated by extensive spectroscopic analyses (1D and 2D NMR, IR and MS) and by comparison with data reported in literatures. All the isolates were tested for cytotoxic and antifouling activities. Compounds 3 and 5 showed significant antifouling activity against the green mussel Perna viridis, and especially 3 was suggested as a potent low-toxic antifouling agent with a large LC50/EC50 value of 18.6. This was the first report on the antifouling activity of the eunicellin-type diterpenoids against the green mussel.  相似文献
