Direct Observation of Non-covalent Complexes for Phosphorylated Flavonoid-protein Interaction by ESI

Diethyl flavon-7-yl phosphate was synthesized by modified Atheron-Todd reaction. The result of ESI shows that the phosphated flavonoids possess stronger binding affinities toward proteins such as myoglobin, insulin and lysozyme and are easier to form the non-covalent complexes with them.

高效液相色谱-电喷雾质谱(HPLC-ESI-MS/MS)测定新型抗癌铂配合物

采用高效液相色谱-电喷雾-多级质谱法(HPLC-ESI-MS/MS), 研究了cis-3,5-diisopropylsalylic cyclohexanodiaminoplatinum(Ⅱ)的电喷雾质谱行为, 鉴定了其杂质, 并测定了其纯度.     

Synthesis and characterization of poly(amidoamine)-platinum(II) complexes. Detailed speciation by Matrix-Assisted Laser Desorption Ionization Mass Spectrometry

MALDI and ESI-MS have been applied to the characterization of the reaction products between the labile cis-[Pt(DMSO)₂Cl₂] (1) and trans-[Pt(DMSO)₂Cl(CH₃)] (2) complexes with the simplest poly(amidoamine) ligand (PAMAM, G = 0, 1,2-diaminoethane as core). The comparison of the mass spectra of the starting G₀ and those of the metallo-dendrimers formed upon mixing of the reagents in an equimolecular ratio, and the analysis of the isotopic distribution in the ESI spectra, have revealed the formation of cationic and neutral mononuclear complexes with PAMAM as ligand, e.g., cis-[Pt(DMSO)(PAMAM)Cl]Cl or trans-(C,N)[Pt(DMSO)(PAMAM)Cl(CH₃)], together with various minor components, which have been identified as derivatives from defective structures of PAMAM. The geometry of the main products has been deduced from the values of the protons coupling constants with the isotopically abundant ¹⁹⁵Pt. The metal-to-ligand bond is restricted to the peripheral amino groups of PAMAM which shows sufficient flexibility to involve either one or two branches in the coordination bonding.     

四种N-杂环卡宾钯配合物的电喷雾质谱研究

利用电喷雾质谱技术研究了四种N-杂环钯卡宾配合物,优化出较适宜检测的电喷雾质谱条件极性较高的溶剂(如乙腈),较低的离子源温度,样品浓度以1.0×10-4mol/L左右为宜,使用较低锥孔电压(5~35V).在此实验条件下,在全扫描电喷雾正离子谱中都会出现[M-I CH3CN] 及[M-I] 的质谱峰.通过源内碰撞诱导电离(CID)技术进一步分析样品在溶液中的性质.实验说明ESI不仅可作为一种分析工具,也可作为获得更多信息的一种方法,来研究有机金属基团在溶液中的性质.     

含有机配体的活性双过氧钒配合物与组氨酸的相互作用研究

在合成和表征了4种具有较强生物活性双过氧钒配合物K_3[VO(O_2)_2(ox)]· 2H_2O，Na[VO(O_2)_2(bipy)]·5H_2O，K[VO(O_2)_2(phen)]·3H_2O和K_2[VO (O_2)_2(pic)]·2H_2O[分别缩写为pV(ox)，pV(bipy)，pV(phen)和pV(pic)，其中 ox为草酸根，bipy为2,2'-联吡啶，phen为邻菲咯啉，pic为2-羧酸吡啶负离子]的 基础上，利用多种NMR技术和电喷雾质谱（ESI-MS）研究了这4种含有机配体双过氧 钒酸合物与组氨酸（His）在溶液中的相互作用以及反应物浓度、时间、pH等对相 互作用体系的影响。~(51)V NMR跟踪监测的结果表明：双过氧钒配合物pV(ox)和 pV(pic)与His在中性水溶液中存在强配位相互作用，而pV(bipy)和pV(phen)与His 在中性水溶液中无明显作用。我们还利用谱学方法确定了pV(ox)和His相互作用后 所生成产物是pV(ox)分别与His在中性水溶液中无明显作用。我们还利用谱学方法 确定了pV(ox)和His相互作用后所生成产物是pV(ox)分别与His咪唑基上的3-N和1- N配位的一对异构体。     

Electrochemical studies of the oxidation mechanism of polyamide on glassy carbon electrode

Polyamides containing N-methyl pyrroles and N-methyl imidazoles are a type of small molecule that can bind and recognize the bases of DNA with high affinity and specificity. Five polyamides were studied at glassy carbon electrode in acetate buffer by cyclic and differential pulse voltammetry to clarify their redox pathways. The polyamide electrochemical responses are compared by peak currents and peak potentials. The slopes of the three anodic E_p vs. pH plots of a typical polyamide are linear and show 0.059, 0.057, 0.056 V per pH in acid media, respectively, which correspond to a mechanism involving the equal number of electrons and protons. A possible mechanism for the redox pathway of various polyamides is proposed: the oxidation product of imidazole ring is acylamide and the results of in situ UV–Vis spectroscopy at Pt web electrode support the proposed mechanism. electrospray ionization mass spectroscopy (ESI-MS) indicates that one or two oxygen atoms are added into polyamide molecule after electrochemical oxidation.     

Synthesis and Electrospray Ionization Mass Spectra of N-（1,3, 2-Dioxaphosphorinan-2-ylmethyl）thiophosphoramidates

N-（1,3,2-Dioxaphosphorinan-2-ylmethyl）thiophosphoramidates were synthesized and determined by NMR spectra and positive ion electrospray ionization mass spectrometry （ESI-MS） in conjunction with tandem mass spectrometry （MS/MS）. The fragmentation pathways were investigated. The results show that these characteristic ions in ESI mass spectra are useful in the structural determination of N-（1,3,2-dioxaphosphorinan-2-ylmethyl）- thiophosphoramidates.     

Construction and Analysis of N-phosphoryl Peptide Libraries

N-Phosphoryl peptide libraries were constructed by transformation from homo-oligopeptide libraries, which was synthesized by self-assembly of amino acids with the assistance of phosphorus oxychloride. Electrospray ionization mass spectrometry (ESI-MS) was used to monitor the reaction.     

Comparative study of the phenolic profile,antioxidant and antimicrobial activities of leaf extracts of five Juniperus L. (Cupressaceae) taxa growing in Turkey

Abstract

The purpose of this study was to conduct a comparative evaluation of some biological properties of methanol and water extracts of leaves of five Juniperus taxa growing in Turkey: J. communis L. var. communis (Jcc), J. communis L. var. saxatilis Pall. (Jcs), J. drupacea Labill. (Jd), J. oxycedrus L. subsp. oxycedrus (Joo), J. oxycedrus L. subsp. macrocarpa (Sibth. & Sm.) Ball. (Jom). The antioxidant properties were examined in vitro; both in the DPPH and in the reducing power tests, Joo methanol extract resulted the most active (IC₅₀?=?0.09?±?0.01mg/mL and ASE/mL?=?2.56?±?0.06). In the TBA assay, Jcs methanol extract exhibited the highest activity (IC₅₀?=?4.39?±?0.47?μg/mL). The extracts displayed bacteriostatic activity against Staphylococcus aureus, and Jd methanol extract resulted the most effective (MIC?=?19.53?μg/mL); no effect on the S. aureus biofilm formation was observed. The extracts resulted non-toxic in the Artemia salina lethality bioassay. Finally, the phenolic profile of the methanol extracts was characterized by HPLC-PDA/ESI-MS.
