1 INTRODUCTION Hexamethylenebis[acetamide] (HMBA) is an effective agent in phaseⅡclinical trial[1] that induces differentiation of certain types of tumors to nonmalignant phenotypes. The title compound C20H30N4O6, Mr = 422.48)was synthesized and its crystal structure was determined by X-ray diffraction method. It crystallizes in the triclinic system, space group P ī with cell parameters: a = 8.330(2), b = 8.468(2), c = 16.017(3) A, α = 97.30(3), β = 92.33(3), γ = 103.94(3)o, V = 1084.7(4) A3, Dc = 1.294 g/cm3, Z = 2, F(000) = 452 and μ = 0.096 mm-1. The structure was refined to R = 0.0483 for 3732 observed reflections with I>2(I) and wR = 0.1335 for 4828 unique reflections. The hydantoin rings are planar and the two ring planes of one molecule are paralleled to each other.  相似文献

Summary This paper describes the direct injection analysis of the anti-cancer drug hexamethylene bisacetamide (HMBA) in biological fluids by an HPLC column switching technique. The first chromatographic column, which provides for sample extraction and cleanup, employs a micellar mobile phase with SDS as the modifier. The second column, coupled on-line to the first, utilizes reversed-phase conditions for analysis. UV detection is employed at 210 nm. 282 samples from 12 cancer patients were analysed and good pharmacokinetics curves obtained. The drug gives recoveries of 94.0–100.9% with a relative standard deviation of 1.88%. A sample analysis is completed within 15 minutes. This method should be satisfactory not only for the analysis of HMBA but also, probably for other drugs in biological fluids.  相似文献

Abstract

A rapid, simple and specific gas chromatographic method has been developed to measure the differentiating agent hexamethylene bisacetamide (HMBA) in biological samples. Addition of a homolog as an internal standard, ultrafiltration and then direct packed column GC-FID analysis of the ultrafiltrate gives a detection limit of less than 50 μg/ml (0.25 mM) for HMBA in plasma or urine. Ultrafiltration is quantitative and assay precision is better than 4.3% for the 1–5 mM range. This method has been applied to determine the bolus dose pharmacokinetics and disposition of HMBA in a single small animal such as a rat. The developed assay should be suitable for therapeutic monitoring of human patients undergoing HMBA treatment.  相似文献

α-,β-,γ-CD与抗癌药HMBA相互作用的NMR研究表明HMBA分子大小与α-CD内腔尺寸最为匹配,相互作用力最强。13C-NMR研究表明α-CD与HMBA形成了包结物,而且两者之间还有氢键存在。HMBA上的α-C,在与α-CD作用后产生的较大低场位移,说明在HMBA·α-CD体系中包结作用较强,使HMBA上α-C化学位移变化大。而HMBA上的-C=O,在与α-,β-CD作用后产生高场位移,位移程度HM-BA.β-CD>HMBA.α-CD,则氢键作用在HMBA.β-CD体系中较强些。  相似文献

Fluorescein isothiocyanate (FITC) is an amine reactive derivative of fluorescein dye that has wide ranging application in biochemistry. It has been extensively used to label peptides and proteins. However, its use in solid phase peptide synthesis is restricted. Indeed, in acidic conditions required for linker cleavage, N-ter FITC-labeled peptides undergo a cyclization leading to the formation of a fluorescein with subsequent removal of the last amino acid. This can be avoided when a spacer such as amino hexanoic acid is used or if non-acidic cleavage is operated to release targeted peptide from the resin.  相似文献

Zn-Al-layered double hydroxides (LDHs) containing the organic anion of 2-hydroxy-4-methoxybenzophenone-5-sulfonic acid (HMBA), a highly efficient UV absorber, have been synthesized by reaction in air of HMBA with an LDH-carbonate precursor obtained by a process involving separate nucleation and aging steps. The structural properties, photo- and thermal stability of the resulting Zn-Al-HMBA-LDHs were characterized by PXRD, FT-IR and UV-visible spectroscopy and laser particle-size analysis. The results obtained indicated that Zn-Al-HMBA-LDHs synthesized by this method were relatively highly crystalline and had a narrow distribution of particle size. Intercalation of HMBA in the LDH interlayer galleries resulted in an enhanced photo- and thermal stability without affecting its UV absorption capacity; this shows that the hybrid HMBA-LDH phase has potential applications as a UV absorber.  相似文献

采用两根反相柱串联的多维HPLC,直接进样测定血浆/尿中抗癌药物六亚甲基二乙酰胺(HMBA)。第一根柱使用胶束流动相,作为样品的纯化和富集步骤,第二根柱为分析柱。紫外检测波长为210 nm。药物的回收率在94.0~100.9%之间,相对标准偏差为1.88%。用该法对十二位不同恶性肿瘤患者的282个血浆/尿样品进行了测定,得到了满意的结果并建立了药代动力学曲线。  相似文献
