Uptake of trace elements into fish otoliths is governed by several factors such as life histories and environment in addition to stock and species differences. In an attempt to elucidate the elemental signatures of rare earth elements (REEs) in otoliths, a solid phase extraction (SPE) protocol was used in combination with electrothermal vaporization (ETV) as a sample introduction procedure for the determinations by inductively coupled plasma quadrupole mass spectrometry (ICP-MS). Effects of various parameters, such as carrier gas flow rate, atomization temperature and chemical modification, were examined for optimization of the conditions by ETV-ICP-MS. Atomization was achieved at 2800 °C. Lower temperatures (i.e. 2600 °C) resulted in severe memory problems due to incomplete atomization. Palladium was used as a chemical modifier. It was found that an increase in Pd concentration up to 0.5 μg in the injection volume (70 μl) led up to four-fold enhancement in the integrated signals. This phenomenon is attributed to the carrier effect of Pd rather than the stabilization since no significant losses were observed for high temperature drying around 700 °C even in the absence of Pd. Preconcentration was performed on-line at pH 5 by using a mini-column of Toyopearl AF-Chelate 650M chelating resin, which also eliminated the calcium matrix of otolith solutions. After preconcentration of 6.4 ml of solution, the concentrate was collected in 0.65 ml of 0.5% (v/v) HNO3 in autosampler cups, and then analyzed by ETV-ICP-MS. The method was validated with the analysis of a fish otolith certified reference material (CRM) of emperor snapper, and then applied to samples. Results obtained from otoliths of fish captured in the same habitat indicated that otolith rare earth element concentrations are more dependent on environmental conditions of the habitat than on species differences.  相似文献

The performance of matrix solid-phase dispersion (MSPD) for the extraction of polycyclic aromatic hydrocarbons (PAHs) in fish tissue is described. The suitability of different solid supports was tested as well as the influence on the extraction efficiency of the natural fat content in samples. Under optimal conditions 0.6-0.8 g of tissue sample, are dispersed with 2 g of octadecylsiloxane (C18) and 0.5 g of anhydrous sodium sulphate and transferred to the top of a polyethylene solid-phase extraction cartridge which already contains 2 g of florisil and 1 g of C18. Cartridges were eluted with acetonitrile. The analysis of the extracts was carried out by high-performance liquid chromatography (HPLC) coupled with fluorescence detection. The proposed method provides detection limits between 0.04 and 0.32 ng/g for the different considered PAHs, below the maximum levels established by the some regulatory bodies for the six PAHs after recent oil spill episodes and European Union regulations. Recoveries over 80% were obtained for all compounds. Accuracy validation was carried out using the US National Institute of Standards and Technology (NIST) SRM 2977 reference material.  相似文献

The assessment of hazard from selenium (Se) and mercury (Hg) contaminants in the food chain based on their potential bioaccessibility and on estimate of their actual content in fish is reported under an in vitro model. Atomic fluorescence spectroscopy (AFS) was applied for total selenium and mercury quantification. Selenium and mercury bioaccessibility varied depending on the type of fish analyzed. Se solubility in the gastrointestinal supernatants was higher in swordfish and sardine (76 and 83%, respectively) than in tuna (50%). A low Hg bioaccessibility (9-17%) was found for all the samples.Simulated human gastric and intestinal digestion led to the identification of selenomethionine (SeMet) and organic mercury in the three digested fish. Furthermore, these species were not modified during the digestion. Speciation of selenium compounds was done by liquid chromatography in conjunction with inductively coupled plasma mass spectrometry (LC-ICP-MS).Sardine had the most favorable Se:Hg, [Se:Hg]bioaccessible and [SeMet:Hg]bioaccessible molar ratios, making it preferable to tuna and swordfish.The effect of cooking was also evaluated.  相似文献

The development of a slurry sampling method for the determination of calcium, copper, iron, magnesium and zinc in fish tissue samples by flame atomic absorption spectrometry is described. In comparison with microwave-assisted digestion, the proposed method is simple, requires short time and eliminates total sample dissolution before analysis. Suspension medium was optimized for each analyte to obtain quantitative recoveries from fish tissue samples without matrix interferences. Nevertheless, iron recoveries higher than 46% were not found. Treatment of samples slurried in nitric acid by microwave irradiation for 15-30 s at 75-285 W permitted to achieve efficient recoveries for calcium, iron, magnesium and zinc. Further improvement in the matrix effects for iron determination was accomplished by the use of an additional step of short microwave-assisted suspension treatment. However, standard addition method was required for calcium and copper determination, being necessary hydrochloric acid as suspension medium for the last one. Although copper could not be determined in the certified reference material using microwave-assisted digestion, the accuracy of the slurry sampling method was verified for all the investigated analytes. Detection limits were 22.8 ± 8.0, 0.884 ± 0.092, 5.07 ± 0.76, 35.5 ± 0.7 and 1.17 ± 0.04 μg g−1 for calcium, copper, iron, magnesium and zinc, respectively. The standard deviations obtained using slurry sampling method and microwave-assisted digestion were not significantly different, and the mean relative standard deviation of the over-all method (n = 3) of the slurry sampling method for different concentration levels was below 12%.  相似文献

An analytical methodology was proposed and validated to be applied to the determination of p,p′-DDT and its metabolites p,p′-DDE and p,p′-DDD in fish oil. The analytical procedure presented in this paper involves a single-step clean up process prior to the analysis. A solution of 1,2,3,4-tetrachloronaphtalene was used as internal standard.The analytical technique used was gas chromatography coupled to an electron capture detector. Details on the validation process are provided.The limits of detection ranged from 2.6 to 4.7 pg μL− 1. The BCR 598 standard reference material (cod liver oil) was used to evaluate the performance of the methodology with satisfactory recoveries for all the compounds.The analytes were determined in three different fish oil pills sold in Spain as a supplementary vitamin support. The sum of p,p′-DDT and metabolites was from 13.2 to 51.3 ng g− 1, the dominant compound being p,p′-DDE.  相似文献

The simultaneous analysis of α-HCH, β-HCH, γ-HCH, HCB, p,p′-DDD, p,p′-DDT, p,p′-DDE, o,p′-DDT, mirex, dieldrin and 62 chlorobiphenyl congeners on two parallel capillary GC columns of different polarity is described for nine Mediterranean fish species. Ten commercially available columns with stationary phases completely characterized in respect of their PCB elution patterns were considered for dual-column GC-ECD analysis. The combination of a 60 m × 0.25 mm i.d. column coated with a 0.25 μm film of 50% diphenyl dimethylsiloxane and a series combination of a 25 m × 0.25 mm i.d. column coated with a 0.25 μm film of 5% diphenyl dimethylsiloxane with a 25 m × 0.22 mm i.d. column coated with a 0.10 μm film of 1, 10-dicarba-closo-dodecarborane dimethylpolysiloxane furnished the highest number of separated chlorobiphenyl congeners (104). The dual-column GC system performed with high stability and reproducibility over a broad concentration range (1–3000 ng/g lipid) of the organochlorine compounds in the investigated fish.  相似文献

The misuses of veterinary drugs can result in the accumulation of residues in food of animal origin that can make its way to the final consumer. Herein we describe a simple method for the accurate determination of beta-lactams, quinolones, sulphonamides, and tetracyclines in fish, poultry, and red meat. No extraction cartridges were used; instead, the extraction process consisted of the addition of an organic solvents, shaking, centrifugation, and dilution. An extensive validation process demonstrated an excellent linearity (R2 ≥ 0.99) for 23-drug residues. The recovery of drugs in different matrices at two concentration levels (n = 6) was in the range of 82–119%. The method was proved to be repeatable and reproducible with intra/inter-day measurements (RSDs lower than 20%). The quantification limits of drug residues were in the range of 0.8 to 45.3 ug/kg, which is well below the maximum residue limits set by most regulatory authorities. This method was successfully applied to the routine analysis of 20 fish, poultry, and red meat samples (n = 60).  相似文献

An amperometric enzyme electrode for the determination of hypoxanthine in fish meat is described. The hypoxanthine sensor was prepared from xanthine oxidase immobilized by covalent binding to cellulose triacetate and a carbon paste electrode containing hydroxymethylferrocene. The xanthine oxidase membrane was retained behind a dialysis membrane at a carbon paste electrode. The sensor showed a current response to hypoxanthine due to the bioelectrocatalytic oxidation of hypoxanthine, in which hydroxymethyiferrocene served as an electron-transfer mediator. The limit of detection is 6 × 10?7 M, the relative standard deviation is 2.8% (n=28) and the response is linear up to 7 × 10?4 M. The sensor responded rapidly to a low hypoxanthine concentration (7 × 10?4 M), the steady-state current response being achieved in less than 1 min, and was stable for more than 30 days at 5 ° C. Results for tuna samples showed good agreement with the value determined by the conventional method.  相似文献

A novel flow-injection system is proposed for the rapid measurement of the fish freshness indices K1 and K2: K1=[([HxR+[Hx])×100/([IMP]+[HxR]+[Hx])] and K2=[[Hx]×100/([HxR]+[Hx])], where [IMP], [HxR] and [Hx] are inosine-5′-monophosphate, inosine and hypoxanthine concentrations, respectively. For the estimation of index K1, 5′-nucleotidase immobilized reactor and nucleoside phosphorylase (NP)/xanthine oxidase (XO) coimmobilized reactor were incorporated in series in the flow-injection line made up by a 16-way switching valve with two sample loops. For the estimation of index K2, NP and XO immobilized reactors were also incorporated in the similar flow-line. Two sample portions passed through the flow-line with different residence times so that two peaks were obtained. The first and second peaks obtained in the K1-determining system corresponded to the total of HxR and Hx and the total of Hx, HxR and IMP, respectively. Similarly, the first and second peaks obtained in the K2-determining system corresponded to Hx and the total of Hx and HxR, respectively. Therefore, the indices K1 and K2 can be estimated by
where i1 and i2 present the peak current of the first and second peaks, respectively, and f the ratio of the peak currents of the first and second peaks for a Hx standard solution. A sea bream was selected as a model fish and it was stored at 4 °C after death. A good correlation was found between the index K1 and the storage time over a period of 400 h, with a correlation coefficient of 0.992, but no correlation between the index K2 and the storage time. The measurements could be performed at a rate of 22 samples per hour with satisfactory precision (0.6–1.2% R.S.D.), without calibration for each species, accurate weighing of fish meat and any interferences in fish meat.  相似文献

A method is developed for determination of trace elements, including Ag, As, Cd, Co, Cr, Cu, Mn, Ni, Se, Tl and Zn, in fish otoliths by electrothermal vaporization inductively coupled plasma mass spectrometry (ETV-ICP-MS). Hydrofluoric acid was used to precipitate calcium resulting from acid dissolution of otolith calcium carbonate. Initial acidity of the sample solution influenced the precipitation efficiency of calcium fluoride. Up to 99.5% of Ca was precipitated in solutions that contained less than 2% (v/v) HNO3. Recoveries of the elements obtained from spiked artificial otolith solutions were between 90 and 103%. Stabilization of the elements within the ETV cell was achieved with 0.3 μg Pd/0.2 μg Rh chemical modifier that also afforded optimum sensitivity for multielement determination. The method was validated by the analysis of a fish otolith reference material (CRM) of emperor snapper, and then applied to the determination of the trace elements in otoliths of several fish species captured in Raritan Bay, New Jersey. Results indicated that fish physiology and biological processes could influence the levels of Cu, Mn, Se and Zn in the otoliths of fish inhabiting a similar aqueous environment. Otolith concentrations of Cr and Ni did not show any significant differences among different species. Concentrations for Ag, As, Cd, Co and Tl were also not significantly different, but were very low indicating low affinity of otolith calcium carbonate to these elements.  相似文献
