Specifications of a neutron exposure accelerator system for biological effects experiments (NASBEE) in NIRS

We developed a neutron irradiation facility, neutron exposure accelerator system for biological effect experiments (NASBEE) for biological studies in National Institute of Radiological Sciences, Japan. Irradiation field of 2 MeV average neutrons generated by a Be(d–n)B reaction is established. Dose uniformity of 240 mm in diameter irradiation field is producible within ±2.5% with a dose rate of 0.87 Gy/h at sample target distance of 1170 mm. Two irradiation rooms, a specific pathogen-free (SPF) conditioned one and a conventional, are now available. Irradiation protocols for in vitro experiments are now established and demonstrated by obtaining a relative biological effectiveness (RBE) of cell inactivation measured to be 3.54 with 10% survival dose (D₁₀).     

Evolution of groups with a hierarchical structure

The universal occurrence of a hierarchical structure and its dynamic behavior in various types of group, living or abstract, are discussed. Here the word “group” refers not only to tangible aggregation but also to invisible aggregation of social psychological and of geopolitical meaning. The evolution of these groups is simulated using a model of agents distributed on the lattices of cellular grids. It is assumed that agents, fearing isolation, interact asymmetrically with each other with regard to exchange of “power”. As an indicator of hierarchy, the Gini coefficient is introduced. Example calculations are made for the aggregation, fusion and fission of animal groups, and for the appearance of a powerful empire and the rise and fall of supremacy. It is shown that such abstract objects evolve with time in accordance with the universal rules of groups common to birds and fish.     

A partial order in the knot table II

A partial order on the set of the prime knots can be defined by the existence of a surjective homomorphism between knot groups. In the previous paper, we determined the partial order in the knot table. In this paper, we prove that 31 and 41 are minimal elements. Further, we study which surjection a pair of a periodic knot and its quotient knot induces, and which surjection a degree one map can induce.     

Enantioseparation of nicotine alkaloids in cigarettes by CE using sulfated β‐CD as a chiral selector and a capillary coated with amino groups

Nicotine (NC) and its related compounds (cotinine (CN), nornicotine (NN), anatabine (AT) and anabasine (AB)) were simultaneously enantioseparated by CE using a capillary with amino groups and sulfated β‐CD as a chiral selector. The optimum running conditions were found to be 30 mM acetate buffer (pH 5.0) containing 8% sulfated β‐CD with an applied voltage of +15 kV at 30°C using direct detection at 260 nm. Using a capillary coated with amino groups, the EOF migrates toward the positive pole. However, when sulfated β‐CD was added to the BGE, it was found that the EOF migrated toward the negative pole due to ionic adsorption of sulfated β‐CD to amino groups on the capillary inner wall. All the cationic analytes migrated as anions, suggesting that they formed stable anionic complexes with sulfated β‐CD. With this system and a simple pretreatment with mini‐cartridges, NC alkaloids in five cigarette samples were enantioseparated. As a result, each of the compounds except for CN was detected. In the case of NC, only (S)‐NC was detected (more than 99.9%), but in the case of NN, AT and AB, the ratios of (S)‐isomer to total isomers were in the ranges 58–70, 81–85 and 59–65%, respectively. On the other hand, only NC was detected in cigarette smoke and the ratio of (S)‐ and (R)‐NCs was 96:4. The amounts of NC alkaloids in cigarettes suggest that the production of (R)‐NC resulted from racemization due to the high temperature/burning of the cigarette.     

Effects of loading conditions and temperature on environmental stress cracking of low-density polyethylene

Fracture mechanics was used to investigate the environmental stress cracking (ESC) of low density polyethylene (LDPE). Annealed and quenched samples were prepared; a single edge notch was made and the samples were fractured under constant load in a liquid methanol environment. The relation between the stress intensity factor K and the crack speed ? has been investigated. There is a large difference between annealed and quenched samples in the variation of this relation with temperature and applied load. The cause of this difference is discussed in detail. We propose that thermally activated molecular motion is essential to ESC of the annealed LDPE while stress concentration contributes markedly to ESC of the quenched LDPE.     

Transformation of arctiin to estrogenic and antiestrogenic substances by human intestinal bacteria

After anaerobic incubation of arctiin (1) from the seeds of Arctium lappa with a human fecal suspension, six metabolites were formed, and their structures were identified as (-)-arctigenin (2), (2R,3R)-2-(3',4'-dihydroxybenzyl)-3-(3",4"-dimethoxybenzyl)butyrolactone (3), (2R,3R)-2-(3'-hydroxybenzyl)-3-(3",4"-dimethoxybenzyl)butyrolactone (4), (2R,3R)-2-(3'-hydroxybenzyl)-3-(3"-hydroxy-4"-methoxybenzyl)butyrolactone (5), (2R,3R)-2-(3'-hydroxybenzyl)-3-(3",4"-dihydroxybenzyl)butyrolactone (6), and (-)-enterolactone (7) by various spectroscopic means including two dimensional (2D)-NMR, mass spectrometry, and circular dichroism. A possible metabolic pathway was proposed on the basis of their structures and the time course of the transformation. Enterolactones obtained from the biotransformation of arctiin and secoisolariciresinol diglucoside (SDG, from the seeds of Linum usitatissium) by human intestinal bacteria were proved to be enantiomers, with the (-)-(2R,3R) and (+)-(2S,3S) configurations, respectively. Compound 6 showed the most potent proliferative effect on the growth of MCF-7 human breast cancer cells in culture among 1 and six metabolites, while it showed inhibitory activity on estradiol-mediated proliferation of MCF-7 cells at a concentration of 10 microM. These results indicate that the transformation of 1 by intestinal flora might be essential for the manifestation of the estrogenic and antiestrogenic activity of 1.
