Evoking Ferroptosis by Synergistic Enhancement of a Cyclopentadienyl Iridium-Betulin Immune Agonist

Ferroptosis is a form of programmed cell death driven by iron-dependent lipid peroxidation (LPO) with the potential for antitumor immunity activation. In this study, a nonferrous cyclopentadienyl metal-based ferroptosis inducer [Ir(Cp*)(Bet)Cl]Cl ( Ir-Bet ) was developed by a metal-ligand synergistic enhancement (MLSE) strategy involving the reaction of [Ir(Cp*)Cl]₂Cl₂ with the natural product Betulin. The fusion of Betulin with iridium cyclopentadienyl (Ir-Cp*) species as Ir-Bet not only tremendously enhanced the antiproliferative activity toward cancer cells, but also activated ferritinophagy for iron homeostasis regulation by PI3K/Akt/mTOR cascade inhibition with a lower dosage of Betulin, and then evoked an immune response by nuclear factor kappa-B (NF-κB) activation of Ir-Cp* species. Further immunogenic cell death (ICD) occurred by remarkable ferroptosis through glutathione (GSH) depletion, glutathione peroxidase 4 (GPX4) deactivation and ferritinophagy. An in vivo vaccination experiment demonstrated desirable antitumor and immunogenic effects of Ir-Bet by increasing the ratio of cytotoxic T cells (CTLs)/regulatory T cells (Tregs).     

反式阿魏酸钌配合物的合成、表征、荧光光谱及其与DNA/蛋白作用

对天然产物反式阿魏酸进行化学修饰,设计、合成了2种单核钌配合物[Ru(cym)(L)Cl]Cl (Ru-1)与[Ru(bpy)2(L)]Cl2(Ru-2)(cym=对伞花烃,bpy=2,2′-联吡啶,L=(E)-3-(4-hydroxy-3-methoxyphenyl)-N-((4′-methyl-(2,2′-bipyridin)-4-yl)methyl)acrylamide),通过核磁共振氢谱和碳谱、电喷雾质谱对配合物进行了结构表征。利用荧光、紫外-可见光谱等方法研究了配合物的溶解性和光学性质。配合物均具有良好的亲水性。Ru-2有良好的荧光性能,其发射波长达到近红外的631 nm,且可以在pH值为8～10之间实现响应。Ru-2还具有较高的单线态氧量子产率(Φ=0.70),有望成为一类高效的光敏剂。2种配合物都与CT-DNA发生嵌入作用(Kb分别为1.210×104和1.233×103L·mol-1);均与BSA有一个结合位点,使其荧光发生静态淬灭(Ka值分别为1.94×104,2.45×104)。     

柔性双咪唑配体环状金（Ⅰ）配合物的合成、表征及荧光性质

采用扩散法制备了2个基于柔性双咪唑配体的Au(Ⅰ)配合物,[Au_2(m-bitmb)_2][AuCl_2]Cl·2CH_3OH (1)和[Au_2(m-bitmb)_2][AuCl_2]_2·2CH_3CN (2)(m-bitmb=1,3-bis(imidazol-1-ylmethyl)-2,4,6-trimethylbenzene),并使用X射线单晶衍射、元素分析、XRD等测试手段进行了表征,研究了配合物的荧光性质。结构分析结果表明配合物1和2均具有M_2L_2的环状结构单元,其结构中双齿配体mbitmb均采用顺式构型参与配位,M_2L_2结构单元通过π-π堆积相互作用排列而形成一维链状结构。配合物1和2具有更强的π-π堆积作用,但亲金性稍弱。通过对比2种配合物和配体的荧光发射,发现配合物中存在的亲金相互作用对其荧光性质具有很重要的影响和作用。     

Note on Generalized Quantum Gates and Quantum Operations

Recently, Gudder proved that the set of all generalized quantum gates coincides the set of all contractions in a finite-dimensional Hilbert space (S. Gudder, Int. J. Theor. Phys. 47:268–279, 2008). In this note, we proved that the set of all generalized quantum gates is a proper subset of the set of all contractions on an infinite dimensional separable Hilbert space ℋ. Meanwhile, we proved that the quantum operation deduced by an isometry is an extreme point of the set of all quantum operations on ℋ. This subject is supported by NSF of China (10571113).     

单核锇配合物的合成、晶体结构及其与DNA的作用

使用溶剂热合成法,以p-bitmb配体(1,4-二(1-咪唑基-亚甲基)-2,3,5,6-四甲基苯)与[(η6-cymene)Os(μ-Cl)Cl]2或[(η6-bip)Os(μ-Cl)Cl]2为原料,合成了2种单核芳基锇配合物,并利用核磁、质谱、元素分析和X射线单晶衍射等手段对配合物进行了表征。配合物1属于单斜晶系,P21/c空间群,为一个单核锇的结构。中心锇原子与2个配体p-bitmb上的氮原子以及氯原子进行配位,2个配体的另一个咪唑基团通过一个亚甲基碳原子进行连接形成咪唑嗡离子,形成一个类似"碗"状的结构。一个氯离子通过氢键装载在结构的空腔内。利用核磁共振氢谱研究了结构中亚甲基的来源,并研究了配合物在缓冲溶液中的稳定性。用紫外吸收光谱、圆二色谱以及粘度法研究了配合物与DNA的相互作用,结果表明,配合物中的亚甲基来自于溶剂二氯甲烷。配合物以嵌入的方式与CT-DNA相互作用,结合常数分别为3.222×10~4 L·mol-1 (1)和1.53×10~4 L·mol-1 (2),同时配合物会减弱DNA的碱基堆积作用并可以使DNA发生解旋。
