Shortwave infrared (SWIR) dyes are characterized by their ability to absorb light from 900 to 1400 nm, which is ideal for deep tissue imaging owing to minimized light scattering and interference from endogenous pigments. An approach to access such molecules is to tune the photophysical properties of known near-infrared dyes. Herein, we report the development of a series of easily accessible (three steps) SWIR xanthene dyes based on a dibenzazepine donor conjugated to thiophene ( SCR-1 ), thienothiophene ( SCR-2 ), or bithiophene ( SCR-3 ). We leverage the fact that SCR-1 undergoes a bathochromic shift when aggregated for in vivo studies by developing a ratiometric nanoparticle for NO ( rNP-NO ), which we employed to successfully visualize pathological levels of nitric oxide in a drug-induced liver injury model via deep tissue SWIR photoacoustic (PA) imaging. Our work demonstrates how easily this dye series can be utilized as a component in nanosensor designs for imaging studies.  相似文献

Mo0, W0, Fe0, Ru0, Re0, and Zn0 nanoparticles—essentially base metals—are prepared as a general strategy by a sodium naphthalenide ([NaNaph])‐driven reduction of simple metal chlorides in ethers (1,2‐dimethoxyethane (DME), tetrahydrofuran (THF)). All the nanoparticles have diameters ≤10 nm, and they can be obtained either as powder samples or long‐term stable suspensions. Direct follow‐up reactions (e.g., Mo0+S8, FeCl3+AsCl3, ReCl5+MoCl5), moreover, allow the preparation of MoS2, FeAs2, or Re4Mo nanoparticles of similar size as the pristine metals (≤10 nm).  相似文献

 This contribution presents a survey on the influence of long-chain branching on the linear viscoelastic properties zero shear-rate viscosity and steady-state recoverable compliance of polyethylene melts. The materials chosen are linear and slightly long-chain branched metallocene-catalyzed polyethylenes of narrow molecular mass distribution as well as linear and highly long-chain branched polyethylenes of broad molecular mass distribution. The linear viscoelastic flow properties are determined in shear creep and recovery experiments by means of a magnetic bearing torsional creep apparatus. The analysis of the molecular structure of the polyethylenes is performed by a coupled size exclusion chromatography and multi-angle laser light scattering device. Polyethylenes with a slight degree of long-chain branching exhibit a surprisingly high zero shear-rate viscosity in comparison to linear polyethylenes whereas the highly branched polyethylenes have a much lower viscosity compared to linear samples. Slightly branched polyethylenes have got a higher steady-state compliance in comparison to linear products of similar polydispersity, whereas the highly branched polyethylenes of broad molecular mass distribution exhibit a surprisingly low elasticity in comparison to linear polyethylenes of broad molecular mass distribution. In addition sparse levels of long-chain branching cause a different time dependence in comparison to linear polyethylenes. The experimental findings are interpreted by comparison with rheological results from literature on model branched polymers of different molecular topography and chemical composition. Amphipathic agents are widely used in various fields including biomedical sciences. Micelle-forming detergents are particularly useful for in vitro membrane-protein characterization. As many conventional detergents are limited in their ability to stabilize membrane proteins, it is necessary to develop novel detergents to facilitate membrane-protein research. In the current study, we developed novel trimaltoside detergents with an alkyl pendant-bearing terphenyl unit as a hydrophobic group, designated terphenyl-cored maltosides (TPMs). We found that the geometry of the detergent hydrophobic group substantially impacts detergent self-assembly behavior, as well as detergent efficacy for membrane-protein stabilization. TPM-Vs, with a bent terphenyl group, were superior to the linear counterparts (TPM-Ls) at stabilizing multiple membrane proteins. The favorable protein stabilization efficacy of these bent TPMs is likely associated with a binding mode with membrane proteins distinct from conventional detergents and facial amphiphiles. When compared to n-dodecyl-β-d -maltoside (DDM), most TPMs were superior or comparable to this gold standard detergent at stabilizing membrane proteins. Notably, TPM-L3 was particularly effective at stabilizing the human β2 adrenergic receptor (β2AR), a G-protein coupled receptor, and its complex with Gs protein. Thus, the current study not only provides novel detergent tools that are useful for membrane-protein study, but also suggests a critical role for detergent hydrophobic group geometry in governing detergent efficacy.  相似文献

The effect of formation and modification methods on the physical properties of polymersomes is critical for their use in applications relying on their ability to mimic functional properties of biological membranes. In this study, we compared two formation methods for polymersomes made from polybutadiene‐polyethylene oxide diblock copolymers: detergent‐mediated film rehydration (DFR) and solvent evaporation (SE). DFR‐prepared polymersomes showed a three times higher permeability compared to SE‐prepared polymersomes as revealed by stopped‐flow light scattering. SE‐prepared polymersomes broke down faster to structures <50 nm diameter when processed with extrusion, which was more pronounced at 5 mg mL?1, compared to 10, 20, and 25 mg mL?1. Our results indicate that the bilayer of SE‐prepared polymersomes has a lower apparent fluidity. We also investigated the role of n‐octyl‐β‐d ‐glucopyranoside (OG), a detergent typically used for reconstitution of membrane proteins into lipid bilayers. Specifically, we compared dialysis and biobeads for OG removal to investigate the influence of these methods on bilayer conformation and polymer rearrangement following detergent removal. There was no significant difference found between method, temperature, or time within each method. Desirable components for dye‐sensitzed solar cell (DSC) sensitizers and fluorescent imaging dyes include strong donating building blocks coupled with well‐balanced acceptor functionalities for absorption beyond the visible range. We have evaluated the effects of increasing acceptor strengths and incorporation of dye morphology controlling groups on molar absorptivity and absorption breadth with indolizine donor‐based dyes. Indolizine‐based D –A and D –π–A sensitizers incorporating bis‐rhodanine, tricyanofuran (TCF), and cyanoacrylic acid functionalities were analyzed for performance in DSC devices. The TCF derivatives were also evaluated as near‐infrared (NIR)‐emissive materials with the AH25 emissions extending past 1000 nm.  相似文献
