Two different human mammary carcinoma cell lines were xenotransplanted into nude mice. Serum samples were obtained prior to and after transplantation and investigated by two-dimensional electrophoresis (2-DE). By comparison of these silver-stained patterns additional protein spots were detected resulting either from proteins secreted or shed by the tumor itself or from mouse proteins induced by the tumor or the transplantation procedure. One group of spots detectable in post-transplantation serum as well as in control serum after mock-transplantation but not in pretransplantation serum was microsequenced and identified as mouse beta-haptoglobin. The carbohydrate structures of beta-haptoglobin were characterized by two different immunochemical glycoprotein staining procedures to detect differential terminal glycan modifications.  相似文献

Electrodynamical model of a classical distributed Bragg reflector (DBR) consisting of alternating quarter-wave layers of high and low permittivity is considered at the plane wave normal incidence. Reflective characteristics of DBR possessing absorption loss in constituting layers are analysed via correct wavelength-scale boundary problem solution by the method of single expression (MSE). Analysis of optical field and power flow density distributions within the lossy DBR structures explained the peculiarities of their reflective characteristics. Optimal configurations of lossless and lossy DBRs are revealed. Specific DBR structures possessing full transparency at definite number of layers are also analysed.  相似文献

Preselection of compounds that are more likely to induce a phenotype can increase the efficiency and reduce the costs for model organism screening. To identify such molecules, we screened ~81,000 compounds in Saccharomyces cerevisiae and identified ~7500 that inhibit cell growth. Screening these growth-inhibitory molecules across a diverse panel of model organisms resulted in an increased phenotypic hit-rate. These data were used to build a model to predict compounds that inhibit yeast growth. Empirical and in silico application of the model enriched the discovery of bioactive compounds in diverse model organisms. To demonstrate the potential of these molecules as lead chemical probes, we used chemogenomic profiling in yeast and identified specific inhibitors of lanosterol synthase and of stearoyl-CoA 9-desaturase. As community resources, the ~7500 growth-inhibitory molecules have been made commercially available and the computational model and filter used are provided.  相似文献

We propose procedures to address product design and manufacturing process configurations concurrently in environments characterized by large degrees of product proliferation. Exploiting the intrinsic flexibility of product and process design, we present two approaches that synchronize production flows through the manufacturing system. These approaches integrate product and manufacturing system design decisions with operational concerns and provide powerful means for managing production in environments characterized by a proliferation of products. Experimental results show that the proposed methods can substantially reduce manufacturing lead times, work in process (WIP), and overall system complexity.  相似文献

This paper proposes operational frameworks for structuring product development processes. The primary objective of this research is to develop procedures to minimize iterations during the development process which adversely affect development time and costs. Several procedures are introduced to restructure the development process. The computation of the corresponding product development times is facilitated by two Markov models addressing different types of learning. The methodologies are employed to identify a set of managerial concerns in restructuring the product development processes.The developed framework has become an integral part of a re-engineering project for the development of rocket engines at Rocketdyne Division of Rockwell International.
