Two combined strategies are reported for improving the sensitivity of organohalide detection by redox catalysis. These are, improvement of the second order rate constant (k) for catalytic reduction of the organohalide, and improvement of the rate of substrate diffusion. Values of k are calculated for both alkyl and aryl halides, from slow scan rate cyclic voltammograms in homogeneous solution. It is shown that a Zn(ii) porphyrin exhibits higher catalytic rates than the previously used Co(ii) porphyrin or Co(i) salen. Amperometric and rotating disk electrode studies of electropolymerised films of the Zn(ii) porphyrin, reveal that at optimum thickness, mediator-substrate reaction and substrate diffusion are the rate limiting steps. Hence, immobilisation of the Zn(ii) porphyrin within the more open structure of a cubic phase liquid crystal produces an increase in sensitivity of approx. 10 times, and lowers the limit of detection by one order of magnitude. The optimised sensor responds linearly to seven organohalides in the range 0.1 microM to 1.0 microM with a sensitivity of 6.95 A M(-1) cm(-2). Chronoamperometric experiments with a microdisk electrode show that the rate of charge transport in the cubic phase films (apparent diffusion coefficient, D(E)= 5.65 x 10(-10)+/- 0.11 x 10(-10) cm(2) s(-1)) is faster than in the electropolymerised films (D(E)= 3.64 x 10(-11)+/- 0.02 x 10(-11) cm(2) s(-1)). The variation of D(E) with the concentration of Zn(ii) in the cubic phase suggests that diffusion of charge is predominantly by electron self-exchange, rather than by physical movement.  相似文献

Formalin, formaldehyde in the presence of methanol, was determined by pulsed amperometric detection (PAD). A triple waveform using Edet=−0.3 V (tdet=30 ms), Eoxd=+0.8 V (toxd=200 ms), and Ered=−0.8 V (tred=350 ms) versus Ag/AgCl was applied at a Au electrode for detection in a flow injection (FI) system. The approach was rapid and yielded a sub-millimolar detection limit (0.0129 mM) with a dynamic range up to 100 mM. A precision of 8.8% R.S.D. at 1.0 mM for two hundred repetitive injections by the FI-PAD was obtained, whereas holding at a constant potential (−0.3 V versus Ag/AgCl) for anodic oxidation of formaldehyde caused the response to decrease dramatically after a few measurements. The method developed was used to analyze the formalin contents of water from rinsed samples of vegetables and fruit and ice-melt from seafood, and the method showed good agreement with the liquid chromatography (LC) method.  相似文献

A facile and simple paper-based scanometric assay was developed to detect Pb2+ using GR5-DNAzyme. Magnetic beads (MBs) and gold nanoparticles (AuNPs) were used as a signal collector and a signal indicator, respectively. They were linked together by GR5-DNAzyme, comprising an enzyme and a substrate strand pairing up with each other. In the presence of Pb2+, the substrate strand is cut into two pieces, resulting in the disassembly of AuNPs from the MBs. These AuNPs were spotted on predefined areas on a chromatography paper, where signal is amplified through silver reduction. This sensing platform exhibits high sensitivity and selectivity toward Pb2+, giving a detection limit of 0.3 nM and a linear fitting range from 0.1 to 1000 nM. Testing of this biosensor in river water and synthetic urine samples also showed satisfying results. Besides offering simultaneous and multi-sample analysis, this paper-based sensing platform presented here could be potentially applied and served as a general platform for on-site, naked eyes, and low-cost monitoring of other heavy metal ions in environmental and body fluid samples.  相似文献

We report a highly sensitive immunoassay protocol based on the use of redox-modified multi-walled carbon nanotubes (MWNTs) as electrochemical labels. The MWNTs were coated with methylene blue (MB) at an optically-determined loading of 3.41 × 10(-3) mol g(-1), and were then attached to secondary antibodies (Ab(2)) by adsorption. As a model analyte mouse IgG was collected by primary antibody (Ab(1))-coated magnetic beads. Following binding of the MB-MWNT-Ab(2) conjugates, IgG could be measured by MB reduction. Using differential pulse voltammetry for quantification, IgG was calibrated with a dynamic range of 0.1 pg mL(-1) to 100 pg mL(-1). Given the different possible Ab(1)-MB-MWNT-Ab(2) orientations on the magnetic beads, it was likely that not all the MB communicated with the electrode. A greater quantity of MB could be accessed by using the Fe(CN)(6)(3-/4-) redox couple as a solution phase mediator. This enabled us to lower the dynamic range down to 5 fg mL(-1) to 100 fg mL(-1).  相似文献

We have examined the anodic stripping voltammetry (ASV) of Cd and Pb at carbon screen printed electrodes modified by an in situ deposited Bi film, and have demonstrated significant cross talk between the stripping peaks of the two metals. A simple and generally applicable method for dealing with this problem is described, based on curve-fitting three-dimensional calibration plots using MATLAB. Non-linear fitting to the calibrations produced coefficients of determination R2 > 0.99 for both metals. We have illustrated use of the plots in conjunction with Bi-plated electrodes by measuring 15 randomly selected mixtures of Cd and Pb of known concentration.  相似文献

A new strategy of three‐electrode system fabrication in polymer‐based microfluidic systems is described here. Standard lithography, hot embossing and UV‐assisted thermal bonding were employed for fabrication and assembly of the microfluidic chip. For the electrode design the gold working (WE) and counter electrodes (CE) are placed inside a main channel through which the sample solution passes. A silver reference electrode (RE) is embedded in a small side channel containing KCl solution that is continuously pushed into the main channel. In the present work, the overall electrochemical set up and its microfabrication is described. Conditions including silver ion concentration, cyclic voltammetry (CV) settings, and the flow rate of KCl solution in the RE channel were optimized. The electrochemical performance of the three‐electrode system was evaluated by CV and also by amperometric oxidation of ferro hexacyanide ([Fe(CN)6]4?) and ruthenium bipyridyl ([Ru(bipy)3]2+) at 400 mV and 1200 mV, respectively. CV analysis using ferri/ferro hexacyanide showed a stable, quasi‐reversible redox reaction at the electrodes with 96 mV peak separation and an anodic/cathodic peak ratio of 1. Amperometric analysis of the electrochemical species resulted in linear correlation between analyte concentration and current response in the range of 0.5–15 µM for [Fe(CN)6]4?, and 0–1000 µM for [Ru(bipy)3]2+. Upon the given experimental conditions, the limit of detection was found to be 3.15 µM and 24.83 µM for [Fe(CN)6]4? and [Ru(bipy)3]2+, respectively. As a fully integrated three‐electrode system that is fabricated on polymer substrates, it has great applications in microfluidic‐based systems requiring stable electrochemical detection.  相似文献

A novel multi-channel poly(methyl methacrylate) (PMMA) microfluidic biosensor with interdigitated ultramicroelectrode arrays (IDUAs) for electrochemical detection was developed. The focus of the development was a simple fabrication procedure and the realization of a reliable large IDUA that can provide detection simultaneously to several microchannels. As proof of concept, five microchannels are positioned over a large single IDUA where the channels are parallel with the length of the electrode finger. The IDUAs were fabricated on the PMMA cover piece and bonded to a PMMA substrate containing the microfluidic channels using UV/ozone-assisted thermal bonding. Conditions of device fabrication were optimized realizing a rugged large IDUA within a bonded PMMA device. Gold adhesion to the PMMA, protective coatings, and pressure during bonding were optimized. Its electrochemical performance was studied using amperometric detection of potassium ferri and ferro hexacyanide. Cumulative signals within the same chip showed very good linearity over a range of 0–38 μM (R 2?=?0.98) and a limit of detection of 3.48 μM. The bonding of the device was optimized so that no cross talk between the channels was observed which otherwise would have resulted in unreliable electrochemical responses. The highly reproducible signals achieved were comparable to those obtained with separate single-channel devices. Subsequently, the multi-channel microfluidic chip was applied to a model bioanalytical detection strategy, i.e., the quantification of specific nucleic acid sequences using a sandwich approach. Here, probe-coated paramagnetic beads and probe-tagged liposomes entrapping ferri/ferro hexacyanide as the redox marker were used to bind to a single-stranded DNA sequence. Flow rates of the non-ionic detergent n-octyl-β-d-glucopyranoside for liposome lysis were optimized, and the detection of the target sequences was carried out coulometrically within 250 s and with a limit of detection of 12.5 μM. The robustness of the design and the reliability of the results obtained in comparison to previously published single-channel designs suggest that the multi-channel device offers an excellent opportunity for bioanalytical applications that require multianalyte detection and high-throughput assays.

A lipase-based assay for detection of specific DNA sequences has been developed. Lipase from Candida antarctica was conjugated to DNA and captured on magnetic beads in a sandwich assay, in which the binding was dependent on the presence of a specific target DNA. For amplification and to generate a detectable readout the captured lipase was applied to an optical assay that takes advantage of the enzymatic activity of lipase. The assay applies p-nitrophenol octanoate (NPO) as the substrate and in the presence of lipase the ester is hydrolyzed to p-nitrophenolate which has a strong absorbance at 405 nm. The method provides detection a detection limit of 200 fmol target DNA and it was able to distinguish single base mismatches from the fully complementary target.  相似文献

Polycrystalline gold electrodes of the kind that are routinely used in analysis and catalysis in aqueous media are often regarded as exhibiting relatively simple double-layer charging/discharging and monolayer oxide formation/removal in the positive potential region. Application of the large amplitude Fourier transformed alternating current (FT-ac) voltammetric technique that allows the faradaic current contribution of fast electron-transfer processes to be emphasized in the higher harmonic components has revealed the presence of well-defined faradaic (premonolayer oxidation) processes at positive potentials in the double-layer region in acidic and basic media which are enhanced by electrochemical activation. These underlying quasi-reversible interfacial electron-transfer processes may mediate the course of electrocatalytic oxidation reactions of hydrazine, ethylene glycol, and glucose on gold electrodes in aqueous media. The observed responses support key assumptions associated with the incipient hydrous oxide adatom mediator (IHOAM) model of electrocatalysis.  相似文献
