De novo site-specific 13C and 15N backbone and sidechain resonance assignments are presented for uniformly enriched E. coli thioredoxin, established using two-dimensional homo- and heteronuclear solid-state magic angle spinning NMR correlation spectroscopy. Backbone dihedral angles and secondary structure were derived from the statistical analysis of the secondary chemical shifts, and are in good agreement with solution values for the intact full-length thioredoxin, with the exception of a small number of residues located at the termini of the individual secondary structure elements. A large number of cross-peaks observed in the DARR spectra with long mixing times correspond to the pairs of carbon atoms separated by 4-6 angstroms, suggesting that DARR could be efficiently employed for observation of medium- and long-range correlations. The 108 amino acid residue E. coli thioredoxin is the largest uniformly enriched protein assigned to this degree of completeness by solid-state NMR spectroscopy to date. It is anticipated that with a combination of two-dimensional correlation experiments and high magnetic fields, resonance assignments and secondary structure can be generally derived for other noncrystalline proteins.  相似文献

Thermooxidative decomposition (TOD) of seven coal samples from different deposits (Bulgaria, Russia, Ukraine) was studied with the aim to determine characteristics of the process and the differences related to the origin of the coal samples studied. The experiments with a Setaram Setsys 1750 or Labsys Evo 1600 thermoanalyzers coupled to a Nicolet 380 FTIR spectrometer or Pfeiffer mass spectrometer, respectively, were carried out under non-isothermal heating conditions up to 1,000 °C at the heating rates of 1, 2, 5, 10, and 20 °C min?1 in an oxidizing atmosphere. A model-free kinetic analysis approach based on the differential isoconversional method of Friedman was used to calculate the kinetic parameters. The combined TG-FTIR and TG-MS study of TOD of the coal samples made it possible to identify a number of gaseous species formed and evolved at that as well as to determine the differences in the thermal behavior of the coal samples and in the emission profiles of these species depending on their origin. The value of activation energy E along the reaction progress α varied more for the samples with higher content of organic matter and, especially, for the samples having at that also quite high content of mineral matter, indicating to the close association of mineral matter with organic matter and fixed carbon.  相似文献

In this study graphite electrodes modified by a thin DNA‐imprinted polypyrrole layer, which was able to bind specific target‐DNA, are reported. For this aim, electrochemical synthesis of polypyrrole was performed on a pencil graphite electrode by cyclic voltammetry (CV) or by potential pulse sequences (PPS). The modified electrode surface was used for electrochemical determination of target‐DNA by differential pulse voltammetry. According to our best knowledge this is a first report on the application of DNA‐imprinted polymer for the determination of target‐DNA. The results showed that the molecularly imprinted polypyrrole (MIPPy) layer that formed on the carbon electrode surface was sensitive for target‐DNA, while the nonimprinted polypyrrole layer was not sensitive to the same target‐DNA. Comparison of electrodes modified using PPS and CV techniques is presented.  相似文献

The reaction of RH (1) with Hg(OAc)(2), in EtOH, gave the acetate RHgOAc (2) [R = 2,6-[O(CH(2)CH(2))(2)NCH(2)](2)C(6)H(3)]. The corresponding RHgCl (3) was obtained from 2 and LiCl. The reaction of 3 with TeCl(4) (1:1 molar ratio), in anhydrous 1,4-dioxane, resulted in the transfer of the organic ligand from mercury to tellurium and the isolation of the unexpected ionic compounds [RTe](2)[Hg(2)Cl(6)] (4) and [RH(3)][HgCl(4)] (5). The molecular structures of 1-4 and 5·H(2)O were established by single-crystal X-ray diffraction. The acetate 2 and the chloride 3 are monomeric in solid state. In both mercury and tellurium organometallic compounds the organic group acts as an (N,C,N) "pincer" ligand. This coordination pattern provided stability for the rare [RTe](+) cation. Weak cation-anion interactions [Te···Cl 3.869(3) ?] are present between [RTe](+) and the dinuclear anion [Hg(2)Cl(6)](2-) in the crystal of 4. Theoretical calculations with DFT methods were performed for models of 3 and 4. The results show that in the cation of 4 the coordination of the nitrogen atoms play an important role for the stabilization of the structure found in the crystal whereas in 3 the coordination of the nitrogen atoms to the metal centre stabilizes to a less extent the structure found in solid state.  相似文献

The potential of 1.7 μm ethylene bridged hybrid silica phase was investigated for the separation of twelve imidazolium-based ionic liquid cations. U-shaped retention profile was observed for all solutes with an increase in retention at both low and high acetonitrile content. Chromatographic behaviour of imidazolium cations in both hydrophilic interaction chromatography (HILIC) and per aqueous liquid chromatography (PALC) modes was studied by varying key parameters such as buffer concentration and pH, acid additive, organic modifier and column temperature. Experimental data provided some evidences that under PALC conditions cationic solutes are retained predominantly by mixed hydrophobic/ion-exchange interactions. In the HILIC mode, both partitioning and ion-exchange interactions are responsible for the retention of solutes. Compared to PALC, HILIC provided significantly higher efficiencies with less or even no peak tailing, better separation selectivity and greater resistance to overload. In PALC mode gradient elution was required to achieve adequate retentivity of all solutes but selectivity was not sufficient to distinguish between solutes with very similar hydrophobicity. In contrast, under HILIC conditions twelve solutes were almost completely resolved in less than 4 min by using isocratic elution. Summarizing, it could be concluded that ethylene bridged hybrid silica column providing a dual retention mechanism offers the possibility of selecting between the two retention modes with opposite separation selectivity, just by changing the composition of the mobile phase.  相似文献

Summary The free energy of the acidic ammonium sulfate is a good precondition its use as an additive or reagent for decomposition of natural phosphates on the way to obtain NPS or NPKS complex fertilizers. During our previous studies it was confirmed that as a result of thermo-mechanical treatment new solid phases are formed as a result of the phosphates decomposition. The aim of this study is to find out appropriate conditions for thermal treatment of Tunisia phosphorite with ammonium sulfate where the content of P2O5 soluble forms has its maximum. The process was investigated under dynamic thermal conditions. Structure and phase transformations of the mixtures to intermediate and final solid products are confirmed by different techniques. X-ray powder diffraction, infrared spectroscopy and electron microscopy have been applied successfully and relationship found between phase structure and thermal treatment applied. As a result of the complex studies optimal temperatures are determined. The solid products under optimal conditions contain phosphorous in soluble forms available for plants in the soil. As a final it is concluded that the final products could be used as complex mineral fertilizers.  相似文献
