Herein, we report ultrasound-propelled graphene-oxide coated gold nanowire motors, functionalized with fluorescein-labeled DNA aptamers (FAM-AIB1-apt), for qualitative detection of overexpressed AIB1 oncoproteins in MCF-7 breast cancer cells. The movement of nanomotors under the ultrasound field facilitated intracellular uptake and resulted in a faster aptamer binding with the target protein and thus faster fluorescence recovery. The propulsion behavior of the aptamer functionalized nanomotors greatly enhanced the fluorescence intensity compared to static conditions. The new aptamer@nanomotor-based strategy offers considerable potential for further development of sensing methodologies towards diagnosis of breast cancer.  相似文献

Model studies of prebiotic chemistry have revealed compelling routes for the formation of the building blocks of proteins and RNA, but not DNA. Today, deoxynucleotides required for the construction of DNA are produced by reduction of nucleotides catalysed by ribonucleotide reductases, which are radical enzymes. This study considers potential non‐enzymatic routes via intermediate radicals for the ancient formation of deoxynucleotides. In this context, several mechanisms for ribonucleotide reduction, in a putative H2S/HS. environment, are characterized using computational chemistry. A bio‐inspired mechanistic cycle involving a keto intermediate and HSSH production is found to be potentially viable. An alternative pathway, proceeding through an enol intermediate is found to exhibit similar energetic requirements. Non‐cyclical pathways, in which HSS. is generated in the final step instead of HS., show a markedly increased thermodynamic driving force (ca. 70 kJ mol?1) and thus warrant serious consideration in the context of the prebiotic ribonucleotide reduction.  相似文献

Investigation of photopolymerization kinetics of 4-(4-methacryloyloxyphenyl)-butan-2-one (1) in comparison with 2-phenoxyethyl methacrylate (2) and phenyl methacrylate (3) using a UV-LED emitting at 395 nm shows significantly faster polymerization of 1 compared to both 2 and 3 at 40°C. Vitrification affects photopolymerization kinetics of all methacrylates under investigation. Interestingly, quantitative final conversion is observed during photoinitiated polymerization of 1 and 2 whereas 3 shows limited conversion at about 80%. Furthermore, higher degree of polymerization is obtained by photoinitiated polymerization of 1 compared to 2 and 3. This shows that the 3-oxobutyl substituent at the phenyl ring of 1 significantly affects both polymerization kinetics and final conversion of the photoinitiated polymerization. Moreover, an additional higher molecular weight fraction is observed in case of polymerization of 1 at 85°C that is above the glass transition temperature of the polymer formed during photoinitiated polymerization. As a thermal polymerization at 85°C in the absence of light results in a high molecular weight polymer as well, an additional thermal process may be discussed as reason for the higher molecular weight polymer fraction in case of the photopolymer made at 85°C.  相似文献

NIR exposure at 790 nm activated photopolymerization of monomers comprising UV-absorbing moieties by using [CuII/(TPMA)]Br2 (TPMA=tris(2-pyridylmethyl)amine) in the ppm range and an alkyl bromide as initiator. Some of them comprised structural elements selected either from those showing proton transfer or photocycloaddition upon UV excitation. Polymers obtained comprise living end groups serving as macroinitiator for controlled synthesis of block copolymers with relatively narrow molecular weight distributions. Chromatographic results indicated formation of block copolymers produced by this synthetic approach. Free-radical polymerization of monomers pursued for comparison exhibited the expected broader dispersity of molecular weight compared to photo-ATRP. Polymerization of these monomers by UV photo-ATRP failed on the contrary to NIR photo-ATRP demonstrating the UV-filter function of the monomers. This work conclusively provides a new approach for the polymerization of monomers comprising UV-absorbing moieties through photo-ATRP in the NIR region. This occurred in a simple and efficient pathway. However, studies also showed that not all monomers chosen successfully proceeded in the NIR photo-ATRP protocol.  相似文献

Improving the germination of economically important crops and the condition of young plants is a major challenge currently facing agricultural practice. Pea (Pisum sativum L.) is one of the four most important cultivated legumes, along with groundnut (Arachis hypogaea L.), soybean (Glycine max L.) and beans (Phaseolus vulgaris L.). Due to the high protein content (23–33%), there is an interest in growing this crop as a source of protein for humans and animals. In this study, we focused on the effect of Cold Atmospheric Pressure Plasma (CAPP) on the decontamination and germination of pea seeds, on young seedling growth and production parameters, and on increasing their resistance and mechanical strength. We can state that germination increased by 10 to 25% after plasma treatment, and the most significant decontamination effect was detected when using non-thermal plasma generated in the ambient air (A-variants) and in the nitrogen atmosphere (N-variants). The increased in situ activity of peroxidases (POX) in the cell walls of A-variants and N-variants is also closely related to the increase in the mechanical strength of the cell walls and thus contributes to the higher resistance of these seedlings. This is also illustrated by the differences in lignin deposition among the different variants after CAPP treatment. To our knowledge, this is the first study concerning the influence of CAPP on the lignification of root tissues and on increasing the strength and resistance of plants.

Novel electrochemical DNA‐sensor based on glassy carbon electrode (GCE) modified with Ag nanoparticles, Neutral red covalently attached to its surface and native DNA adsorbed on modifier coating was developed for the estimation of DNA damage on example of model system based on Fenton reagent. As was shown, the oxidation process resulted in synchronous increase of electron transfer resistance and capacitance measured by electrochemical impedance spectroscopy (EIS). The contribution of each sensor component on the signal was specified and sensitivity estimated against similar surface coatings. The shift of EIS parameters was found to be higher than that of similar biosensors reported. The DNA sensor was tested on the estimation of antioxidant capacity of green tea infusions again the results of coulometric titration with electrogenerated bromine.  相似文献

Light as an external trigger is a valuable and easily controllable tool for directing chemical reactions with high spatial and temporal accuracy. Two o‐nitrobenzyl derivatives, benzoyl‐ and thiophenyl‐NPPOC, undergo photo‐deprotection with significantly improved efficiency over that of the commonly used NPPOC group. The two‐ and twelvefold increase in photo‐deprotection efficiency was proven using photolithograph synthesis of microarrays.  相似文献
