Superbase-Promoted Acylation of Hindered Alcohols

The commercially available nonionic superbase P(MeNCH(2)CH(2))(3)N (1a) is very useful for the acylation of unreactive hindered alcohols as well as acid-sensitive alcohols. The reactions proceed in high yields using an acid anhydride, and 1a can be regenerated in a single step. The relative rates for benzoylation of (+/-)-menthol in C(6)D(6) using conventional acylation reagents and strong nonionic bases are compared. In general, acetylation with 1a is accelerated in the polar solvent CH(3)CN whereas benzoylation is faster in the nonpolar solvent C(6)H(6). The benzoylation intermediate RC(O)P(MeNCH(2)CH(2))(3)N(+) was found to be in equilibrium with 1a, with lower temperatures favoring the intermediate. The relative stabilities of several known acylating intermediates are compared.     

P(i-PrNCH2CH2)3N: An efficient nonionic catalyst for the regioselective Michael addition of a β,γ-unsaturated ester and a nitrile

Michael additions of a β,γ-unsaturated ester and a nitrile to a variety of α,β-unsaturated ketones can be effected with high regioselectivity and in good yields using P(i-PrNCH₂CH₂)₃N as a catalyst.     

(RO)(2)Ta[tris(2-oxy-3,5-dimethylbenzyl)amine]: structure and lactide polymerization activities

The novel atrane-like six-coordinate (RO)(2)TaL complexes [where R = Me or Et and L = tris(2-oxy-3,5-dimethylbenzyl)amine] containing three six-membered rings have been synthesized and characterized. The R = Me complex is the first group 5 representative of this class of compounds structurally characterized by X-ray means. Somewhat surprisingly, these compounds failed to function as single-site initiators for the polymerization of l-LA to isotactic PLA and rac-LA to atactic PLA, whereas Ta(OEt)(5) and two titanium analogues ROTiL (where R = 2,6-di-i-PrC(6)H(3) and i-Pr) as well as Ti(O-i-Pr)(4) were effective catalysts for both polymerizations.     

Mild and efficient deprotection of the amine protecting p-methoxyphenyl (PMP) group

Mild and efficient procedures for deprotection of the amine nitrogen protecting p-methoxyphenyl (PMP) group are described. Periodic acid and trichloroisocyanuric acid (TCCA) were found to be particularly effective in realizing amine liberation using 1 and 0.5 equiv of the oxidant, respectively. Extension of the periodic acid-mediated conditions to simultaneous alcohol oxidation by combination with a catalytic amount of sodium dichromate led to smooth conversion of PMP-protected Mannich products into the corresponding β-amino acids in a one-pot procedure.     

Phase-vanishing methodology for efficient bromination, alkylation, epoxidation, and oxidation reactions of organic substrates

[reaction: see text] In cases where both reactants in a phase-vanishing reaction are less dense than the fluorous phase, an alternative to the U-tube method is to employ a solvent with greater density than the fluorous phase, such as 1,2-dibromoethane. This modification has been successfully applied to the methylation of a phenol derivative with dimethyl sulfate and to the m-CPBA-induced epoxidation of alkenes, N-oxide formation from nitrogen-containing compounds, and S-oxide or sulfone formation from organic sulfides.     

P(i-BuNCH2CH2)3N: an efficient promoter for the nucleophilic aromatic substitution reaction of aryl fluorides with aryl TBDMS (or TMS) ethers

The nucleophilic aromatic substitution reaction between electron-deficient aryl fluorides and aryl TBDMS (or TMS) ethers has been shown to be efficiently promoted by proazaphosphatranes such as P(i-BuNCH(2)CH(2))(3)N (3). Excellent yields of diaryl ether products were obtained under unusually mild conditions.     

P(i-BuNCH2CH2)3N: an efficient ligand for the direct alpha-arylation of nitriles with aryl bromides

A new catalyst system for the synthesis of alpha-aryl-substituted nitriles is reported. The bicyclic triaminophosphine P(i-BuNCH(2)CH(2))(3)N (1b) serves as an efficient and versatile ligand for the palladium-catalyzed direct alpha-arylation of nitriles with aryl bromides. Using ligand 1b, ethyl cyanoacetate and primary as well as secondary nitriles are efficiently coupled with a wide variety of aryl bromides possessing electron-rich, electron-poor, electron-neutral, and sterically hindered groups.     

CRYSTAL AND MOLECULAR STRUCTURE OF THE SPIRODIPHOSPHITE ESTER 3,9-DIMETHOXY-2,4,8,10-TETRAOXA-3,9-DIPHOSPHASPIRO[5.5]UNDECANE

Abstract

The crystal and molecular structure of the strainless phosphite ester MeOP(OCH₂)₂C(CH₂O)₂POMe determined by x-ray means is reported. The methoxy groups are in axial positions on the chair-form-six-membered rings.     

Lactose as a “Trojan Horse” for Quantum Dot Cell Transport

A series of glycan‐coated quantum dots were prepared to probe the effect of glycan presentation in intracellular localization in HeLa and SV40 epithelial cells. We show that glycan density mostly impacts on cell toxicity, whereas glycan type affects the cell uptake and intracellular localization. Moreover, we show that lactose can act as a “Trojan horse” on bi‐functionalized QDs to help intracellular delivery of other non‐internalizable glycan moieties and largely avoid the endosomal/lysosomal degradative pathway.
