Remarkable synergistic effect between MonBI and MonBII on epoxide opening reaction in ionophore polyether monensin biosynthesis

Enzymatic epoxide-opening cascade is one of the key biosynthetic processes for constructing structurally diverse polyethers. Here we report the first in vitro analysis of the cyclization catalyzed by two epoxide hydrolases, MonBI and MonBII involved in monensin biosynthesis, using simple epoxy-alcohols and the unprecedented synergistic effect on the epoxide-opening activity between these epoxide hydrolases.     

A straightforward approach to antibodies recognising cancer specific glycopeptidic neoepitopes

Aberrantly truncated immature O-glycosylation in proteins occurs in essentially all types of epithelial cancer cells, which was demonstrated to be a common feature of most adenocarcinomas and strongly associated with cancer proliferation and metastasis. Although extensive efforts have been made toward the development of anticancer antibodies targeting MUC1, one of the most studied mucins having cancer-relevant immature O-glycans, no anti-MUC1 antibody recognises carbohydrates and the proximal MUC1 peptide region, concurrently. Here we present a general strategy that allows for the creation of antibodies interacting specifically with glycopeptidic neoepitopes by using homogeneous synthetic MUC1 glycopeptides designed for the streamlined process of immunization, antibody screening, three-dimensional structure analysis, epitope mapping and biochemical analysis. The X-ray crystal structure of the anti-MUC1 monoclonal antibody SN-101 complexed with the antigenic glycopeptide provides for the first time evidence that SN-101 recognises specifically the essential epitope by forming multiple hydrogen bonds both with the proximal peptide and GalNAc linked to the threonine residue, concurrently. Remarkably, the structure of the MUC1 glycopeptide in complex with SN-101 is identical to its solution NMR structure, an extended conformation induced by site-specific glycosylation. We demonstrate that this method accelerates dramatically the development of a new class of designated antibodies targeting a variety of “dynamic neoepitopes” elaborated by disease-specific O-glycosylation in the immunodominant mucin domains and mucin-like sequences found in intrinsically disordered regions of many proteins.

Sondenmessung der Plasmaparameter und deren Fluktuation in Argon-Niederdruck-Schwachstrom-Säulen im transversalen Magnetfeld

This paper reports on measurements of probe noise and double probe characteristics in argon-low-pressure low-current discharges under the influence of a local transversal magnetic field. Above a critical value of magnetic field intensity B_c there are no moving striations but partially correlated fluctuations. The space distribution of probe noise (ion saturation current and floating potential) and the measured values of electron temperature and density are discussed. At B_c a characteristic value of electron temperature T_e,c ≈ 1,4 eV independent of filling pressure has been found. A first opinion on the character of the fluctuations is given.     

Phase unwrapping method along radial coordinates for speckle image reconstruction

We describe a phase estimating technique for a stellar speckle image reconstruction, which is based on the phase unwrapping method in polar coordinates. The phase unwrapping along radial lines is favorable to reduce accumulating errors in the phase estimation. This method is combined with Bruck-Sodin method which gives the estimate of the modulus of an object transform without a point reference source. Computer simulations of the image reconstruction are presented.     

Nine new secoiridoid glucosides from Jasminum nudiflorum

Phytochemical study of the leaves of Jasminum nudiflorum has led to the isolation of nine new secoiridoid glucosides, jasnudiflosides F-L (1-7), nudifloside D (8) and isooleoacteoside (9). The structures of these compounds were elucidated on the basis of chemical and spectroscopic evidence.     

Far infrared reflectance of a mixed phase Y-Ba-Cu-O superconductor

We have studied the far infrared reflectance of the mixed phase superconductor Y_1.2Ba_0.8CuO_4–y. The reflectance spectrum shows sharp phonon resonances and an electronic contribution that we attribute to reflection at a mesh-like structure of the conducting phase embedded in nonconducting material; in comparison, the reflectance of single phase superconducting material is mainly determined by the dynamical electronic conductivity.     

A quantum field theory of lattice dynamics at finite temperature

A quantum field theoretical treatment of three-dimensional cubic crystals at finite temperature is presented from the view-point of the spontaneous breakdown of the spatial translational invariance using thermo field theory. The effective interaction Hamiltonian is constructed by taking into account the dynamical map of the molecular density operator which is obtained from the Ward-Takahashi relations. The acoustic phonons are expected to be the excitation of particle-hole pairs. The conventional secular equation for the lattice vibrations is obtained by neglecting some quantum effects in the Bethe-Salpeter equation for the molecular density fluctuations. The phonon spectra, the phonon propagators and the dynamical map of the molecular density operator are calculated at finite temperature.     

Development of Water-Repellent Glass Improved Water-Sliding Property and Durability

A water-repellent glass with high water-sliding property using FAS for the hydrophobic material has been developed. It had been thought that the sliding property of FAS was inferior to PDMS. But it was found that the sliding angles of the surfaces coated with FAS were affected by coating condition. And the glass on which fluorinated alkyl groups oriented toward normal direction of the surface had almost equal water-sliding property to the one coated with PDMS. Furthermore its durability against wiper abrasion and outside weathering was much higher than that with PDMS. And XPS analysis was carried out to confirm the orientation of fluorinated alkyl groups on the developed water-repellent glass. This kind of water-repellent glass was adopted as commercial automotive windows.
