A gas diffusion-flow injection system was developed for the determination of carbon dioxide in gaseous samples. The calibration was based on the use of either gaseous carbon dioxide or aqueous sodium carbonate standards. Gaseous carbon dioxide samples and gaseous or aqueous standards were injected directly into a donor stream of 1.0×10−4 M H2SO4. In the gas diffusion unit, carbon dioxide diffused through a PTFE membrane into an acceptor stream containing a mixed acid/base indicator. The absorbance of the acceptor stream was monitored spectrophotometrically at 554 nm. The calibration plot was linear over the range of 5.00×102 to 1.27×104 μl l−1 with a sample throughput of 28 h−1 and 3.2% R.S.D. ([CO2]=2.37×103 μl l−1, n=12). The detection limit was determined as 2.50×102 μl l−1. The flow system was successfully applied to the analysis of several natural gaseous samples and the headspace of milk containers during storage. The flow injection results were found to be statistically indistinguishable at the 95% confidence level from those obtained by gas chromatography using thermal conductivity detection.  相似文献

Abstract

Rhodomyrtone was isolated from the leaves of Rhodomyrtus tomentosa (Aiton) Hassk grown in Vietnam using chromatographic methods. Its chemical structure was confirmed by means of spectroscopic data analysis. The pH drop measurement, enzyme activity assays and fluorescence stain were used to examine rhodomyrtone anticaries activity. It was found that rhodomyrtone suppressed acid production by Streptococcus mutans, a major cariogenic agent in human by inhibiting enzyme activities responsible for acid production and tolerance, including membrane bound enzymes F-ATPase and phosphotransferase system (PTS), as well as glycolysis enzymes glyceraldehyphosphate dehydrogenase (GAPDH) and pyruvate kinase (PK) in cytoplasm with the IC50 values of 24?μM, 19?μM, 23?μM and 28?μM, respectively. Moreover, 50?μM rhodomyrtone reduced biofilm biomass formed by S. mutans up to 59% (p?<?0.05). Fluorescent images indicated that cells on the biofilms were significantly killed. Thus, rhodomyrtone is a new and potential anticaries agent against S. mutans.  相似文献

Norwalk and Norwalk-like viruses (NLV) are major causes of food- and water-related disease in the United States. There is no host cell line in which the NLV can be tested for infectivity. Feline calicivirus (FCV) and NLV both belong to the family Caliciviridae. FCV can be assayed for infectivity in the Crandell Reese feline kidney cell line, so FCV serves as a surrogate for NLV. This study is the first report of UV inactivation of FCV and also of using the plaque technique, in contrast to the 50% tissue culture infectious dose end point technique, to determine the FCV infectivity titer. The infectivity titers (log10 plaque-forming units/mL) of UV-inactivated FCV, hepatitis A virus (HAV), poliovirus type 1 (PV1) and two small, round coliphages were plotted as a function of UV dose and analyzed by regression analysis and analysis of variance. These fitted straight-line curves represent exponential inactivation, so UV inactivation can be said to show "one-hit kinetics." The decimal inactivation doses of UV for FCV, HAV, PV1, MS2 and phiX174 were 47.85, 36.50, 24.10, 23.04 and 15.48 mW s/cm2, respectively. FCV appears to be the most UV resistant among the tested viruses.  相似文献
