A simple and efficient method for the deoxygenation of amine N-oxides to corresponding amines is reported using the green and economical reagent phenylboronic acid. Deoxygenation of N,N-dialkylaniline N-oxides, trialkylamine N-oxides and pyridine N-oxides were achieved in good to excellent yields. The reduction susceptible functional groups such as ketone, amide, ester and nitro groups are well tolerated with phenylboronic acid during the deoxygenation process even at high temperature. In addition, an indirect method for identification and quantification of tert-amine N-oxide is demonstrated using UV–Vis spectrometry which may be useful for drug metabolism studies. 相似文献

One-pot synthesis of novel ferrocene grafted N-methyl-spiropyrrolidines has been accomplished in good yields through a facile 1,3-dipolar cycloaddition reaction of various azomethine ylide derived from ninhydrin and sarcosine with various ferrocene derivatives as dipolarophile. The regiochemical and stereochemical outcome of the cycloaddition reaction is ascertained by X-ray crystallographic studies of one of the cycloadducts. 相似文献

Synthesis of selenoxo peptides by the treatment of N(α)-protected peptide esters with a combination of PCl(5) and LiAlHSeH is delineated. The method is simple, high-yielding, and free from racemization. Thus obtained selenoxo peptides are used as units for N-terminal chain extension through N(α)-deprotection/coupling to yield peptide-selenoxo peptide hybrids. Multiple selenation is demonstrated by conversion of two peptide bonds of tripeptides into selenoxo peptide bonds. Amino acid derived arylamides are also converted into aryl selenoamides. C(6)H(5)-CSeNH-Val-OMe 8f is obtained as single crystal, and its structure was determined through X-ray diffraction study. 相似文献

The time scale for ordering of the polypeptide backbone relative to the side chains is a critical issue in protein folding. The interplay between ordering of the backbone and ordering of the side chains is particularly important for the formation of β-sheet structures, as the polypeptide chain searches for the native stabilizing cross-strand interactions. We have studied these issues in the N-terminal domain of protein L9 (NTL9), a model protein with mixed α/β structure. We have developed a general approach for introducing site-specific IR probes for the side chains (azide) and backbone ((13)C═(18)O) using recombinant protein expression. Temperature-jump time-resolved IR spectroscopy combined with site-specific labeling enables independent measurement of the respective backbone and side-chain dynamics with single residue resolution. We have found that side-chain ordering in a key region of the β-sheet structure occurs on a slower time scale than ordering of the backbone during the folding of NTL9, likely as a result of the transient formation of non-native side-chain interactions. 相似文献

Sufficient dimension reduction methodologies in regressions of Y on a p-variate X aim at obtaining a reduction \(R(X) \in {\mathbb R}^{d}, d \le p\), that retains all the regression information of Y in X. When the predictors fall naturally into a number of known groups or domains, it has been established that exploiting the grouping information often leads to more effective sufficient dimension reduction of the predictors. In this article, we consider group-wise sufficient dimension reduction based on principal fitted components, when the grouping information is unknown. Principal fitted components methodology is coupled with an agglomerative clustering procedure to identify a suitable grouping structure. Simulations and real data analysis demonstrate that the group-wise principal fitted components sufficient dimension reduction is superior to the standard principal fitted components and to general sufficient dimension reduction methods. 相似文献

An experimental study on the synthesis, tautomerism, and acid promoted structural changes of spiro-pyrazolines is described. The target was achieved through a [3+2]-dipolar cycloaddition of an alkene with nitrile imines generated in situ and was isolated in high yield. The synthesized cycloadduct displayed a tendency to exhibit an imine–enamine type of tautomerism as evidenced by X-ray crystal and NMR studies. Furthermore, addition of an acid resulted in the transformation of an imine tautomer to an enamine. The current report constitutes a first formal observation of this kind of tautomerism observed in spiro-indoline pyrazolines. 相似文献

Improvements in the synthesis of carbon-linked glucuronide/glucoside conjugates of cancer chemopreventive retinoids have been achieved starting with 2,3,4,6-tetra-O-benzyl-D-glucopyranose. The revised approach demonstrates better yields, eliminates the use of an expensive, carcinogenic protecting group reagent, and avoids much painstaking chromatography. The new approach should allow synthesis of larger quantities of the agents for detailed animal and mechanistic studies. 相似文献

A new class of S-linked 1,3,4-oxadiazole-tethered Nα-protected peptidomimetics is designed and synthesized by a reaction of Nα-Z/Boc-protected 1,3,4-oxadiazole-2-thiol with α-bromo ester derived from amino acid. The protocol has also been employed for the synthesis of glycosylated amino acids and N,N′-orthogonally protected dipeptidomimetics bearing S-linked 1,3,4-oxadiazole mimetics as well. The intermediate 5-alkyl amino-1,3,4-oxadiazole-2-thiols have been isolated as stable compounds. Further, the chain extension at both N- and C-termini of N-protected S-linked 1,3,4-oxadiazole tethered dipeptidomimetics was also demonstrated. 相似文献

Sequential combination of Ugi-MCR and click chemistry has been employed for the synthesis of triazole linked cyclic glycopeptidomimetics. The protocol employs Poc-amino alkyl isonitriles, sugar-1-amines, azido acids, and simple aldehydes as precursors. The dual nature of the propargyloxycarbonyl (Poc) group was explored for amine protection as well as cycloaddition with an azide. All the cyclic glycopeptidomimetics are isolated and characterized. 相似文献
