The incorporation of small molecules into lipid bilayers is a process of biological importance and clinical relevance that can change the material properties of cell membranes and cause deleterious side effects for certain drugs. Here we report the direct observation, using surface-enhanced Raman and IR spectroscopies (SERS, SEIRA), of the insertion of ibuprofen molecules into hybrid lipid bilayers. The alkanethiol-phospholipid hybrid bilayers were formed onto gold nanoshells by self-assembly, where the underlying nanoshell substrates provided the necessary enhancements for SERS and SEIRA. The spectroscopic data reveal specific interactions between ibuprofen and phospholipid moieties and indicate that the overall hydrophobicity of ibuprofen plays an important role in its intercalation in these membrane mimics.  相似文献

Thermogravimetry (TG), thermogravimetry/mass spectrometry (TG/MS), and loss-on-drying methodology are used to provide residual moisture results for freeze-dried biological products regulated by the US Food and Drug Administration. Residual moisture specifications must be met in order to ensure freeze-dried biological product potency and stability throughout the licensed product's shelf life. TG, TG/MS, loss-on-drying and vapor pressure moisture measurements are compared for a BCG Vaccine. Comparisons are made between residual moisture data for the freeze-dried cake and vapor pressure moisture determinations in the space above the freeze-dried cake in the final container. Vapor pressure moisture precision data is presented for α-interferon and BCG vaccine. Impact of residual moisture and vapor pressure moisture upon product stability is presented.  相似文献

Advances in scattering-based optical imaging technologies offer a new approach to noninvasive point-of-care detection, diagnosis, and monitoring of cancer. Emerging photonics technologies provide a cost-effective means to image tissue in vivo with high resolution in real time. Advancing the clinical potential of these imaging strategies requires the development of optical contrast agents targeted to specific molecular signatures of disease. We describe the use of a novel class of contrast agents based on nanoshell bioconjugates for molecular imaging in living cells. Nanoshells offer significant advantages over conventional imaging probes including continuous and broad wavelength tunability, far greater scattering and absorption coefficients, increased chemical stability, and improved biocompatibility. We show that nanoshell bioconjugates can be used to effectively target and image human epidermal growth factor receptor 2 (HER2), a clinically relevant biomarker, in live human breast carcinoma cells.  相似文献

A slice of black shale rock cut by various metal sulphide veins of different generations from the Kupferschiefer deposits of Lubin, Poland was subjected to bombardment in a Laser Microprobe Combustion Reactor to produce SO2 for S-isotope analyses. The delta34S values ranged from-22 to-29 per thousand consistent with previous findings using conventional IRMS and attributable to primary generation of H2S by bacterial sulphate reduction. Systematic trends in delta34S values of a few per mil over distances of the order of mm attest to low temperatures of mineralization with accompanying change in the isotope composition of the fluids due to kinetic or equilibrium isotope fractionation.  相似文献

Measurements of synchrotron radiation emitted by 30-MeV runaway electrons in the TEXTOR-94 tokamak show that the runaway population decays after switching on neutral beam injection (NBI). The decay starts only with a significant delay, which decreases with increasing NBI heating power. This delay provides direct evidence of the energy dependence of runaway confinement, which is expected if magnetic modes govern the loss of runaways. Application of the theory by Mynick and Strachan [Phys. Fluids 24, 695 (1981)] yields estimates for the "mode width" (delta) of magnetic perturbations: delta<0.5 cm in Ohmic discharges, increasing to delta = 4.4 cm for 0. 6 MW NBI.  相似文献

Abstract

A modified technique for the conversion of sulphates and sulphides to SO2 with the mixture of V2O5—SiO2 for sulphur isotopic analyses is described. This technique is more suitable for routine analysis of large number of samples. Modification of the reaction vessel and using manifold inlet system allows to analyse up to 24 samples every day. The modified technique assures the complete yield of SO2, consistent oxygen isotope composition of the SO2 gas and reproducibility of δ34S measurements being within 0.10‰. It is observed, however, oxygen in SO2 produced from sulphides differs in δ18O with respect to that produced from sulphates.  相似文献

Abstract

The paper presents a whole-year study (1990) of an unique area in S-E Poland with numerous small rivers and streams carrying clean waters. We report the results of δ18O of waters and δ34S of the sulphates sampled 4 times in 1990 from 20 rivers of the study area. The observations clearly show the impact of biological activity on the oxygen and sulphur isotopic compositions in sulphates. Attempts have been made to interpret the correlation between δ34S and δ18O in sulphates. The highest correlation coefficient has been noticed for samples collected in April, whereas the lowest in August. The conclusion of this study is that the river sulphates are predominantly produced outside the river environment. We have distinguished three major sources of sulphates: (1) ones produced in the aquifer from which waters are discharged, (2) those produced in soils and marshes of forest environment, and (3) ones on anthropogenic origin.  相似文献
