Eight novel amidothiophosphates have been successfully achieved using cycloiminium salts of pyridine as the starting material. Phosphorylation was done using phosphorus trichloride, and the resulting functionalized iminohalophosphines in situ were further oxidized and substituted through sulfur and a suitable secondary amine (alkyl, alicyclic, and aryl). The reactions were carried out in a dry nitrogen atmosphere. The synthesized products were characterized using different spectroscopic techniques (1H and 31P NMR). The pyridine nucleus incorporating a phosphoryl group is highly bioactive. Hence, the synthesized organothiophosphates should be potential pesticidal molecules.  相似文献

The mammalian visual dim‐light photoreceptor rhodopsin is considered a prototype G protein‐coupled receptor. Here, we characterize the kinetics of its light‐activation process. Milligram quantities of α,ε‐15N‐labeled tryptophan rhodopsin were produced in stably transfected HEK293 cells. Assignment of the chemical shifts of the indole signals was achieved by generating the single‐point‐tryptophan to phenylalanine mutants, and the kinetics of each of the five tryptophan residues were recorded. We find kinetic partitioning in rhodopsin decay, including three half‐lives, that reveal two parallel processes subsequent to rhodopsin activation that are related to the photocycle. The meta II and meta III states emerge in parallel with a relative ratio of about 3:1. Transient formation of the meta III state was confirmed by flash photolysis experiments. From analysis of the site‐resolved kinetic data we propose the involvement of the E2‐loop in the formation of the meta III state.  相似文献

The orthorhombic holmium oxisulphate orders as a two-sublattice antiferromagnet atT=3.5 K. In external fields along the crystallographica- andc-directions with large and medium-sized magnetic moment, respectively, a ferrimagnetic phase with 1/3 of the saturation magnetization is passed before the paramagnetic phase is reached. Calculations in mean-field theory reveal that for thec-direction the ferrimagnetic phase is not stable atT=0, it only exists for finite temperatures. Magnetization and susceptibility contain large contributions of van Vleck paramagnetism which at any rate have to be taken into consideration. The phase diagram for the two field directions and the magnetic structures of the different phases are established.  相似文献

Swing bridge: The triplet species ethenedithione has been generated within the coordination sphere of cobalt, leading to a dinuclear μ-η(2)-η(2)-C(2)S(2) complex (see picture: C?gray, Co?blue, P?purple, S?yellow). Depending on the solvent, the C(2)S(2) moiety displays a transoid or a cisoid geometry. This isomerization step changes the sign of the magnetic coupling between the cobalt centers.  相似文献

Growing high-quality crystals is a bottleneck in the determination of protein structures by x-ray diffraction. Experiments find that materials with a disordered pitted surface seed the growth of protein crystals. Here we report computer simulations of rapid crystal nucleation in nanoscale pits. Nucleation is rapid, as the crystal forms in pits that have filled with liquid via capillary condensation. Surprisingly, we find that pits whose surfaces are rough are better than pits with crystalline surfaces; the roughness prevents the growing crystal from trying to conform to the pit surface and becoming strained.  相似文献

We report the magnetic field effect on the linear and nonlinear optical properties of pulse laser ablated Ti-incorporated Cr2O3 nanostructured thin film. Optical properties have been experimentally analyzed under Voigt geometry by performing ultraviolet-visible spectroscopy and closed aperture Z-scan technique using a continuous wave He–Ne laser source. Nonlinear optical response reveals a single peak-valley feature in the far field diffraction pattern in absence of magnetic field (B=0) confirming self-defocussing effect. This feature switches to a valley-peak configuration for B=5000G, suggesting self-focusing effect. For B750G, oscillations were observed revealing the occurrence of higher order nonlinearity. Origin of nonlinearity is attributed to the near resonant d-d transitions observed from the broad peak occurring around 2 eV. These transitions are of magnetic origin and get modified under the application of external magnetic field. Our results suggest that magnetic field can be used as an effective tool to monitor the sign of optical nonlinearity and hence the thermal expansion in Ti-incorporated Cr2O3 nanostructured thin film.  相似文献

Drug purity and affinity are essential attributes during development and production of therapeutic proteins. In this work, capillary electrophoresis (CE) was used to determine both the affinity and composition of the biotechnologically produced “nanobody” EGa1, the binding fragment of a heavy-chain-only antibody. EGa1 is an antagonist of the epidermal growth factor receptor (EGFR), which is overexpressed on the surface of tumor cells. Using a background electrolyte (BGE) of 50 mM sodium phosphate (pH 8.0) in combination with a polybrene-poly(vinylsulfonic acid) capillary coating, CE analysis of EGa1 showed the presence of at least three components. Affinity of the EGa1 components towards the extracellular domain of EGFR was assessed by adding different concentrations (0–12 nM) of the receptor to the BGE while measuring the effective electrophoretic mobility of the respective EGa1 components. Binding curves obtained by plotting electrophoretic mobility shifts as a function of receptor concentration, yielded dissociation constants (Kd) of 1.65, 1.67, and 1.75 nM for the three components, respectively; these values were comparable to the Kd of 2.1 nM obtained for the bulk EGa1 product using a cellular assay. CE with mass spectrometry (MS) detection using a BGE of 25 mM ammonium acetate (pH 8.0) revealed that the EGa1 sample comprised of significant amounts of deamidated, bisdeamidated and N-terminal pyroglutamic acid products. CE–MS using a BGE of 100 mM acetic acid (pH 2.8) in combination with a polybrene–dextran sulfate–polybrene capillary coating demonstrated the additional presence of minor products related to incomplete removal of the signal peptide from the produced nanobody. Combining the results obtained from affinity CE and CE–MS, it is concluded that the EGa1 nanobody product is heterogeneous, comprising highly-related proteins that exhibit very similar affinity towards EGFR.  相似文献

For the application in organic electronics as well as pH meters, a series of novel 2,4,6-trisubstituted pyridines have been synthesized by the reaction of substituted aromatic aldehydes and substituted 3-acetylcoumarin with ammonium acetate. The structures of all the new compounds were characterized by IR, NMR, and GC–MS analysis. The important photo physical prerequisites for organoelectronic such as optical absorption and thermal stability were determined for the synthesized molecules. Optical properties were studied by UV–visible absorption and fluorescence spectroscopy. Optical band gaps of the 2,4,6-trisubstituted pyridines were found to be around 3.01–3.06?eV as calculated from their onset absorption edge. The pH-dependent changes in the fluorescence intensity suggest that 2,4,6-trisubstituted pyridines are useful applicants in intracellular pH meters.  相似文献
