土壤根癌杆菌体外转化胡萝卜悬浮培养细胞

本文报道了土壤根癌杆菌C58菌株体外转化胡萝卜悬浮培养单细胞。结果表明,土壤根癌杆菌可以直接转化具有完整细胞壁的高等植物细胞。在25℃下,细菌和植物细胞混合比为100(即100个细菌对1个植物细胞)时,转化频率约为10~4。虽然在转化组织及其再生植株中没有检测到胭脂碱,但被转化细胞具有激素自主性生长的能力,并且很容易再生成完整植株。此外,还比较了土壤根癌杆菌和土壤发根杆菌诱导胡萝卜圆片所形成的瘤组织和发根瘤在生长速率和再生能力方面的差异。     

In vitro TRANSFORMATION OF SUSPENSION CULTURED CELLS FROM Daucus carota BY Agrobacterium tumefaciens STRAIN C58

In vitro transformation of suspension cultured single cells of carrot by Agrobacterium tumefaciens strain C58 was reported here. The results indicate that plant cells with intact cell walls can be directly transformed by A. tumefaciens. At 25℃,and at a ratio of 100 bacteria per plant cell, the transformation frequency was about 10~(-4).The transformed cells had the ability to grow in the absence of plant hormones and very easily regenerated into plants, although nopaline was not detected in the tissues and plants from transformed cells.The differences in the growth rate and regeneration ability between tumors induced by A.tumefaciens strain C58 and hairy roots induced by A. rhizogenes strain A4 on carrot disks were compared.
