Abstract—DNA crosslinks in Escherichia coli cells. exposed to 4.5',8-trimethylpsoralen plus 360 nm light, were measured using a rapid and sensitive new approach. The assay is based on the specificity of S1 nuclease from Aspergillus oryzae to single-stranded DNA. Bacterial cells were lysed and the DNA denatured by alkali. Following acid neutralization. crosslinked DNA undergoes spontaneous renaturation and is rendered S1-nuclease resistant and therefore acid-precipitable. The single-stranded fraction after denaturation by alkali decreases with increasing near UV light exposure in the presence of TMP following first order kinetics. The kinetics were faster when exposure was at 4°C rather than at 20°C. This suggests that excision of crosslinks occurs during exposure at the higher temperature. Indeed. since the rate of DNA crosslinking in a uvr B mutant which is excision-deficient was higher than in wild type bacteria at 4°C, some excision must have occurred even in the cold. DNA from excision-proficient cells incubated at 37°C following exposure to TMP-plus-near UV at 4° showed a greater single stranded fraction than that from non-incubated cells. This indicates repair of DNA crosslinks. which proceeded with a half-time of 8 min at 37°C and was unaffected by substitution of thymine in DNA by 5-bromouracil.  相似文献

Positioning accuracy of 3D ultrasound slice planes can be enhanced by speckle decorrelation of the B-scans. A suitable method requires accurate identification of zones of non-specular reflection in the image, a feature of parenchymal tissue. This has traditionally been achieved with statistics such as signal-to-noise ratios and measures of autocorrelation curves. Co-occurrence matrices and structural approaches have also been applied to ultrasound images. In this paper we assess the performance of these methods when only B-scan display data is available. We also report improved results with in vivo images using a proposed structural algorithm that detects speckle kernels. All detection algorithms yielded detection error rates between 28% and 38%, suggesting that user intervention may be required for clinically useful performance.  相似文献

The paper is concerned with the dynamics of a thin circular plate made of a plastic-rigid material that obeys the yield condition ofTresca and the associated flow rule. The basic equations are established in the case of rotational symmetry, and their complete solution is given for a simply supported plate subjected to a uniformly distributed load which is brought on suddenly and, after a certain time interval, removed suddenly.
Résumé Cet article étudie la dynamique d'une plaque circulaire infiniment mince fabriquée avec un matériau plastique qui obéit à la condition de plasticité deTresca et à la loi d'écoulement associée. Les équations fondamentales sont établies dans le cas où existe une symétrie de révolution. Leur solution complète est donnée pour une plaque reposant sur des appuis simples, soumise à une charge uniformément répartie qui est appliquée rapidement et demeure constante pendant un certain temps, après lequel elle est rapidement retirée.

Fast ions are observed to be very well confined in the Madison Symmetric Torus reversed field pinch despite the presence of stochastic magnetic field. The fast-ion energy loss is consistent with the classical slowing down rate, and their confinement time is longer than expected by stochastic estimates. Fast-ion confinement is measured from the decay of d-d neutrons following a short pulse of a 20 keV atomic deuterium beam. Ion confinement agrees with computation of particle trajectories in the stochastic magnetic field, and is understood through consideration of ion guiding center islands.  相似文献

Magnetic field fluctuations (and the associated current perturbation) have been measured in the core of a high-temperature reversed-field pinch using a newly developed fast-polarimetry system. Radial magnetic field fluctuation levels of approximately 1% are measured in standard-reversed-field pinch discharges which increase to approximately 4% during the sawtooth crash (enhanced dynamo). The fluctuation level is reduced fourfold for high-confinement plasmas where the core-resonant tearing modes are suppressed.  相似文献

Amorphous and crystalline states of As2Se3, (As2Se3)3 : Tl2Se and As2Se3 : Tl2Se have been studied using X-ray diffraction techniques. Structural changes arise during the process of annealing in the temperature range between their softening and melting points are reported and their rates investigated. The crystallization temperatures were found to be 105 ± 5 °C, 135 ± 5 °C and 180 ± 5 °C respectively. The unit cell parameters are identified for each of the three resulting crystalline phases, that for As2Se3 : Tl2Se being orthorhombic while the other two are monoclinic.  相似文献
