Autologous bone grafts, used mainly in extensive bone loss, are considered the gold standard treatment in regenerative medicine, but still have limitations mainly in relation to the amount of bone available, donor area, morbidity and creation of additional surgical area. This fact encourages tissue engineering in relation to the need to develop new biomaterials, from sources other than the individual himself. Therefore, the present study aimed to investigate the effects of an elastin and collagen matrix on the bone repair process in critical size defects in rat calvaria. The animals (Wistar rats, n = 30) were submitted to a surgical procedure to create the bone defect and were divided into three groups: Control Group (CG, n = 10), defects filled with blood clot; E24/37 Group (E24/37, n = 10), defects filled with bovine elastin matrix hydrolyzed for 24 h at 37 °C and C24/25 Group (C24/25, n = 10), defects filled with porcine collagen matrix hydrolyzed for 24 h at 25 °C. Macroscopic and radiographic analyses demonstrated the absence of inflammatory signs and infection. Microtomographical 2D and 3D images showed centripetal bone growth and restricted margins of the bone defect. Histologically, the images confirmed the pattern of bone deposition at the margins of the remaining bone and without complete closure by bone tissue. In the morphometric analysis, the groups E24/37 and C24/25 (13.68 ± 1.44; 53.20 ± 4.47, respectively) showed statistically significant differences in relation to the CG (5.86 ± 2.87). It was concluded that the matrices used as scaffolds are biocompatible and increase the formation of new bone in a critical size defect, with greater formation in the polymer derived from the intestinal serous layer of porcine origin (C24/25).  相似文献

Mutant p53 tends to form aggregates with amyloid properties, especially amyloid oligomers inside the nucleus, which are believed to cause oncogenic gain-of-function (GoF). The mechanism of the formation of the aggregates in the nucleus remains uncertain. The present study demonstrated that the DNA-binding domain of p53 (p53C) underwent phase separation (PS) on the pathway to aggregation under various conditions. p53C phase separated in the presence of the crowding agent polyethylene glycol (PEG). Similarly, mutant p53C (M237I and R249S) underwent PS; however, the process evolved to a solid-like phase transition faster than that in the case of wild-type p53C. The data obtained by microscopy of live cells indicated that transfection of mutant full-length p53 into the cells tended to result in PS and phase transition (PT) in the nuclear compartments, which are likely the cause of the GoF effects. Fluorescence recovery after photobleaching (FRAP) experiments revealed liquid characteristics of the condensates in the nucleus. Mutant p53 tended to undergo gel- and solid-like phase transitions in the nucleus and in nuclear bodies demonstrated by slow and incomplete recovery of fluorescence after photobleaching. Polyanions, such as heparin and RNA, were able to modulate PS and PT in vitro. Heparin apparently stabilized the condensates in a gel-like state, and RNA apparently induced a solid-like state of the protein even in the absence of PEG. Conditions that destabilize p53C into a molten globule conformation also produced liquid droplets in the absence of crowding. The disordered transactivation domain (TAD) modulated both phase separation and amyloid aggregation. In summary, our data provide mechanistic insight into the formation of p53 condensates and conditions that may result in the formation of aggregated structures, such as mutant amyloid oligomers, in cancer. The pathway of mutant p53 from liquid droplets to gel-like and solid-like (amyloid) species may be a suitable target for anticancer therapy.

Theoretical aspects of the MS Xα method are analyzed. It is shown that real field self-consistency is not achieved in an MS Xα calculation and some consequences of this fact are discussed. This article reviews different kinds of models for the electric power grid that can be used to understand the modern power system, the smart grid. From the physical network to abstract energy markets, we identify in the literature different aspects that co-determine the spatio-temporal multilayer dynamics of power system. We start our review by showing how the generation, transmission and distribution characteristics of the traditional power grids are already subject to complex behaviour appearing as a result of the the interplay between dynamics of the nodes and topology, namely synchronisation and cascade effects. When dealing with smart grids, the system complexity increases even more: on top of the physical network of power lines and controllable sources of electricity, the modernisation brings information networks, renewable intermittent generation, market liberalisation, prosumers, among other aspects. In this case, we forecast a dynamical co-evolution of the smart grid and other kind of networked systems that cannot be understood isolated. This review compiles recent results that model electric power grids as complex systems, going beyond pure technological aspects. From this perspective, we then indicate possible ways to incorporate the diverse co-evolving systems into the smart grid model using, for example, network theory and multi-agent simulation.  相似文献

Many studies are focused on the development of materials for converting carbon dioxide into multicarbon oxygenates such as methanol and ethanol, because of their higher energy density and wider applicability. In this work, TiO2 nanotubes (NT/TiO2) were modified with CuxO nanoparticles in order to investigate the contribution of different ratio of Cu2O/CuO and its distribution over NT/TiO2 for CO2 photoelectro-conversion to methanol. The photoelectrodes were built by anodization process to obtain NT/TiO2 layer, and the decoration with CuxO hybrid system was carried out by electrodeposition process, using Na2SO4 or acid lactic as electrolyte, followed by annealing at different temperatures. X-ray photoelectron spectroscopy analysis revealed the predominance of Cu+1 and Cu+2 at 150 °C and 300 °C, respectively. X-ray diffraction and scanning electron microscopy indicated that under lactic acid solution, the oxide nanoparticles exhibited small size, cubic shape, and uniform distribution on the nanotube wall. While under Na2SO4 electrolyte, large nanoparticles with two different morphologies, octahedral and cubic shapes, were deposited on the top of the nanotubes. All modified electrodes converted CO2 in methanol in different quantities, identified by gas chromatograph. However, the NT/TiO2 modified with CuO/Cu2O (80:20) nanoparticles using lactic acid as electrolyte showed better performance in the CO2 reduction to methanol (0.11 mmol L−1) in relation to the other electrodes. In all cases, a blend among the structures and nanoparticle morphologies were achieved and essential to create new site of reactions what improved the use of light irradiation, minimization of charge recombination rate and promoted high selectivity of products.

We consider a nearest-neighbor-interaction ±J Ising spin glass in a square lattice. Inspired by natural evolution, we design a dynamic rule that includesselection, randomness, andmultibranch exploration. Following this rule, we succeed in walking along the space of states between local energy maxima and minima alternately. During the walk, we store various information about the spin states corresponding to these minima and maxima for later statistical analysis. In particular, we plot a histogram displaying how many times each minimum (or maximum) energy is visited as a function of the corresponding density value. Through finite-size scaling analysis, we conclude that a nonvanishing fraction of bonds remains unsatisfied (satisfied) at these energy minimum (maximum) states in the thermodynamic limit. This fraction measures the degree of unavoidable frustration of the system. Also in this limit, the width of these histograms vanishes, meaning that almost all metastable states occur at the same energy density value, with no dispersion.  相似文献

Structure and magnetism of iron clusters with up to 641 atoms have been investigated by means of density functional theory calculations including full geometric optimizations. Body-centered cubic (bcc) isomers are found to be lowest in energy when the clusters contain more than about 100 atoms. In addition, another stable conformation has been identified for magic-number clusters, which lies well within the range of thermal energies as compared to the bcc isomers. Its structure is characterized by a close-packed particle core and an icosahedral surface, while intermediate shells are partially transformed along the Mackay path between icosahedral and cuboctahedral geometry. The gradual transformation results in a favorable bcc environment for the subsurface atoms. For Fe55, the shellwise Mackay-transformed morphology is a promising candidate for the ground state.  相似文献

In this work we propose the use of a targeting method applied to chaotic systems in order to reach special trajectories that encode arbitrary sources of messages. One advantage of this procedure is to overcome dynamical constraints which impose limits in the amount of information that the chaotic trajectories can encode. Another advantage is the message decoding, practically instantaneous and independent of any special technique or algorithm. Furthermore, with this procedure, information can be transmitted with no errors due to bounded noise.  相似文献

This work describes the electroanalytical determination of Chlorpyriphos pesticide in natural waters using hanging mercury drop electrode allied to square wave adsorptive cathodic stripping voltammetry. The best responses were obtained in Britton?Robinson buffer solutions at pH 2.0, using a frequency of 100 s–1, a scan increment of 5 mV, a square wave amplitude of 25 mV and an accumulation potential of–0.4 V during 60 s. Therefore, voltammetric responses showed the presence of one well-defined and irreversible reduction peak, at–1.08 V vs. Ag/AgCl/KCl 1.0 M, which involves two electrons in the reduction of carbon?nitrogen bond in the N-heterocyclic system with the participation of protonation equilibrium preceding the electron transfer reaction. Analytical curves were constructed and compared to similar curves performed by gas chromatograph coupled to a selective nitrogen?phosphorus detector, which demonstrates that the proposed methodology is suitable for determining contamination by Chlorpyriphos in complex samples.  相似文献
