Previous work has shown that glucose oxidase can be immobilized on platinum to give an electrode that responds potentiometrically to glucose over the clinically useful range of about 10-250 mg glucose/100 mL. The present studies were carried out with electrochemically pretreated platinum and with gold or porous graphite substituted for the platinum support. The presence of the enzyme gave a significantly enhanced potentiometric response over that obtained with the bare support for both the pretreated platinum and the porous graphite, but not with gold. However, with platinum the potentiometric response became more negative with increasing glucose concentration. With porous graphite, the potential changed in the positive direction as the glucose concentration was increased. Hysteresis was demonstrated for the platinum-enzyme electrode. Mass transfer measurements with a rotating ring-disc electrode (RRDE) showed measurable diffusional resistances to the transport of a model electroactive compound (potassium ferrocyanide) through a matrix of immobilized enzyme attached to the disc of the RRDE. These results are part of a larger study to define the source of the potentiometric response by examining the roles of the support and the mass transfer resistances through the immobilized enzyme matrix.  相似文献

Literature reports have described the covalent coupling of the primary amine-containing anticancer drug, adriamycin, to polymeric supports through the amine group on the drug. These reports also have described drug mechanism studies with the immobilized adriamycin, where the release of the drug would undermine the validity of the conclusions. In the present paper, detailed experimental conditions are given for preparation of nonwater-soluble particles of polyvinyl alcohol by crosslinking water-soluble polyvinyl alcohol with 1,4-benzenedicarboxaldehyde, and for activation with cyanuric chloride and covalent attachment of adriamycin. The expected stability of this drug-support linkage against hydrolytic cleavage is compared mechanistically to that expected for less stable coupling through a carbamate linkage or for less stable coupling via an azomethine link.  相似文献

A Mannich-type reaction was used to attach flavin adenine dinucleotide (FAD) covalently to aminosilane derivatized indium/tin oxide-coated glass plates. The aminosilane was activated with formaldehyde to give an intermediate that attached specifically to the adenine amino group of FAD. The presence of the intermediate also was demonstrated by coupling hydroquinone to the formaldehyde activated support. The immobilized FAD and hydroquinone were characterized by cyclic or differential pulse voltammetry. The immobilized FAD was shown to reduce the overpotential for NADH oxidation by 180 mV. In keeping with results for FAD on glassy carbon, FAD attached to indium/tin oxide at the adenine amino group did not lead to reconstitution of activity with apoglucose oxidase. Four spherically symmetric but non-asymptotically flat black hole solutions surrounded with spherical dark matter distribution perceived under the minimal length scale effect is derived via the generalized uncertainty principle. Here, the effect of this quantum correction, described by the parameter γ $\gamma$ , is considered on a toy model galaxy with dark matter and the three well-known dark matter distributions: the cold dark matter, scalar field dark matter, and the universal rotation curve. The aim is to find constraints to γ $\gamma$ by applying these solutions to the known supermassive black holes: Sagittarius A (Sgr. A*) and Messier 87* (M87*), in conjunction with the available Event Horizon telescope. The effect of γ $\gamma$ is then examined on the event horizon, photonsphere, and shadow radii, where unique deviations from the Schwarzschild case are observed. As for the shadow radii, bounds are obtained for the values of γ $\gamma$ on each black hole solution at 3 σ $3\sigma$ confidence level. The results revealed that under minimal length scale effect, black holes can give positive (larger shadow) and negative values (smaller shadow) of γ $\gamma$ , which are supported indirectly by laboratory experiments and astrophysical or cosmological observations, respectively.  相似文献

Studies are underway to see if flavin adenine dinucleotide or nicotinamide adenine dinucleotide cofactors can be attached to electrode surfaces to give rapid rates of electron transfer and in some cases reconstitution of enzyme activity with appropriate apoenzymes. The range of properties available in the lanthanide series has inspired research into the use of lanthanide nanoparticles for numerous applications. We aim to use NaLnF(4) nanoparticles for isotopic tags in mass cytometry. This application requires nanoparticles of narrow size distribution, diameters preferably less than 15 nm, and robust surface chemistry to avoid nonspecific interactions and to facilitate bioconjugation. Nanoparticles (NaHoF(4), NaEuF(4), NaGdF(4), and NaTbF(4)) were synthesized with diameters from 9 to 11 nm with oleic acid surface stabilization. The surface ligands were replaced by a series of mono-, di-, and tetraphosphonate PEG ligands, whose synthesis is reported here. The colloidal stability of the resulting particles was monitored over a range of pH values and in phosphate containing solutions. All of the PEG-phosphonate ligands were found to produce non-aggregated colloidally stable suspensions of the nanoparticles in water as judged by DLS and TEM measurements. However, in more aggressive solutions, at high pH and in phosphate buffers, the mono- and diphosphonate PEG ligands did not stabilize the particles and aggregation as well as flocculation was observed. However, the tetraphosphonate ligand was able to stabilize the particles at high pH and in phosphate buffers for extended periods of time.  相似文献

This article reports on the fabrication of oriented composite fibers between polylactide (PLA) and multiwall carbon nanotube (MWNT). The fibers were fabricated using a custom‐built melt fiber‐drawing setup. The influence of processing parameters on the final fiber diameter and on the orientation were characterized and optimized. Composite fibers were fabricated at various MWNT contents. Addition of low amounts of MWNT (0.25–1 wt %) to PLA did not have a significant effect on the diameters of the fibers. Observations of the composite morphology under STEM indicated preferential orientation of the MWNTs along the draw direction of the fibers. Increasing amounts of MWNTs was found to increase crystallization kinetics and content. The crystalline content had a direct and profound implication on the mechanical properties with 0.5‐wt % MWNT fibers having the highest crystalline content and also the highest Young's modulus. © 2014 Wiley Periodicals, Inc. Two chain-shortened analogs of lipoic (1,2-dithiolane-3-pentanoic) acid, viz. bisnor-lipoic (1,2-dithiolane-3-propanoic) and tetranorlipoic (1,2-dithiolane-3-carboxylic) acids, have been chemically synthesized.
