Isolation and purification of honokiol and magnolol from cortex Magnoliae officinalis by high-speed counter-current chromatography 总被引:5,自引:0,他引:5
High-speed counter-current chromatography was used to isolate and purify honokiol and magnolol from cortex Magnoliae Officinalis (Magnolia officinalis Rehd. et Wils.), a plant used in the traditional Chinese medicine. A crude sample, 150 mg, was successfully separated with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (1:0.4:1:0.4, v/v), and the fractions were analyzed by high-performance liquid chromatography. The separation produced 80 and 45 mg of honokiol and magnolol with purities of 99.2 and 98.2%, respectively, in 2.5 h. 相似文献

We report on an ultrasensitive fluorescence immunoassay for human chorionic gonadotrophin antigen (hCG). It is based on the use of silica nanoparticles coated with a copolymer (prepared from a fluorene, a phenylenediamine, and divinylbenzene; PF@SiO2) that acts as a fluorescent label for the secondary monoclonal antibody to β-hCG antigen. In parallel, Fe3O4 nanoparticles were coated with polyaniline, and these magnetic particles (Fe3O4@PANI) served as a solid support for the primary monoclonal antibody to β-hCG antigen. The PF@SiO2 exhibited strong fluorescence and good dispersibility in water. A fluorescence sandwich immunoassay was developed that enables hCG concentrations to be determined in the 0.01–100 ng·mL?1 concentration range, with a detection limit of 3 pg·mL?1. Figure

Colloidal carbon microspheres (CMS) are dispersed in chitosan (CHIT) solution to form an organic-inorganic hybrid with excellent micro-environment for the immobilization of biomolecules. A novel amperometric biosensor for the determination of hydrogen peroxide (H(2)O(2)) has been constructed by entrapping horseradish peroxidase (HRP) in as-synthesized CMS/CHIT hybrid. The modification of glassy carbon electrode is made by a simple solution-evaporation method. The electrochemical properties of the biosensor are characterized in electrochemical methods. The proposed biosensor shows high sensitive determination and fast response to H(2)O(2) at -0.15 V. The constructed HRP/CHIT/CMS/GC electrode also exhibits a fine linear correlation with H(2)O(2) concentration. The calculated value of the apparent Michaelis-Menten constant, 2.33 mM, suggests that the HRP in CMS/CHIT hybrid keeps its native bioactivity and has high affinity for H(2)O(2). 相似文献

Mn-doped (Bi0.5Na0.5)TiO3-BaTiO3 (BNBMT) thin film with the composition near the morphotropic phase boundary was grown on (La0.6Sr0.4)CoO3-electroded SrTiO3 single-crystal substrate by using pulsed laser deposition method. Ascribed to the crystal structure and lattice similarity, (100)-oriented BNBMT film with pure single-phase perovskite structure was obtained through carefully controlling the growth conditions. Enhanced ferroelectric and dielectric properties were obtained with large remanent polarization P r of ~21 μC/cm2, coercive field E c of ~7.4 kV/mm and dielectric constant $\varepsilon_{33}^{T}/\varepsilon_{0}$ of ~750 at 1 kHz. The excellent global electrical properties of the BNBMT film are promising for environmentally friendly ferroelectric devices. 相似文献

Background
Phosphatase of regenerating liver-3 (PRL-3 or PTP4A3) has been implicated in controlling cancer cell proliferation, motility, metastasis, and angiogenesis. Deregulated expression of PRL-3 is highly correlated with cancer progression and predicts poor survival. Although PRL-3 was categorized as a tyrosine phosphatase, its cellular substrates remain largely unknown.Results
We demonstrated that PRL-3 interacts with integrin β1 in cancer cells. Recombinant PRL-3 associates with the intracellular domain of integrin β1 in vitro. Silencing of integrin α1 enhances PRL-3-integrin β1 interaction. Furthermore, PRL-3 diminishes tyrosine phosphorylation of integrin β1 in vitro and in vivo. With site-specific anti-phosphotyrosine antibodies against residues in the intracellular domain of integrin β1, tyrosine-783, but not tyrosine-795, is shown to be dephosphorylated by PRL-3 in a catalytic activity-dependant manner. Phosphorylation of Y783 is potentiated by ablation of PRL-3 or by treatment with a chemical inhibitor of PRL-3. Conversely, depletion of integrin α1 decreases the phosphorylation of this site.Conclusions
Our results revealed a direct interaction between PRL-3 and integrin β1 and characterized Y783 of integrin β1 as a bona fide substrate of PRL-3, which is negatively regulated by integrin α1.10.
