Authentic samples of whisky produced in Scotland and USA and counterfeit whisky samples commercialized in Brazil have been directly submitted to electrospray ionization mass spectrometry (ESI-MS) analysis in both the negative and positive ion modes to assess the potential of this technique for simple and rapid quality control and proof of authenticity of whisky samples. ESI in the negative ion mode yields the most characteristic whisky fingerprinting mass spectra in just a few seconds by direct infusion of the samples, detecting the most polar or acidic components of each sample in their deprotonated anionic forms. No pre-treatment of the sample, such as extraction or derivatization or even dilution, is required. The analysis of the ESI(-)-MS data both by simple visual inspection but more particularly by chemometric data treatment enables separation of the whisky samples into three unequivocally distinct groups: Scotch, American and counterfeit whisky, whereas single malt and blended Scotch whiskies are also distinguished to some extent. As indicated by ESI-MS/MS analysis, the diagnostic anions are simple sugars, disaccharides and phenolic compounds. Direct infusion ESI-MS therefore provides immediate chemical fingerprinting of whisky samples for type, origin and quality control, as demonstrated herein for American, Scottish and counterfeit samples, whereas ESI-MS/MS analysis of diagnostic ions adds a second dimension of fingerprinting characterization when improved selectivity is desired. 相似文献

Schistosomiasis is a neglected disease with large geographic distribution worldwide. Among the several different species of this parasite, S. mansoni is the most common and relevant one; its pathogenesis is also known to vary according to the worms' strain. High parasitical virulence is directly related to granulomatous reactions in the host's liver, and might be influenced by one or more molecules involved in a specific metabolic pathway. Therefore, better understanding the metabolic profile of these organisms is necessary, especially for an increased potential of unraveling strain virulence mechanisms and resistance to existing treatments. In this report, MALDI‐MSI and the metabolomic platform were employed to characterize and differentiate two Brazilian S. mansoni strains: males and females from Belo Horizonte (BH) and from Sergipe (SE). By performing direct analysis, it is possible to distinguish the sex of adult worms, as well as identify the spatial distribution of chemical markers. Phospholipids, diacylglycerols and triacylglycerols were located in specific structures of the worms' bodies, such as tegument, suckers, reproductive and digestive systems. Lipid profiles were found to be different both between strains and males or females, giving specific metabolic fingerprints for each group. This indicates that biochemical characterization of adult S. mansoni may help narrowing‐down the investigation of new therapeutic targets according to worm composition, molecule distribution and, therefore, aggressiveness of disease. Neutron Activation Analysis, INAA, is an analytical technique of choice for homogeneity testing. An interesting possibility
is the use of complementary gamma ray photopeaks to confirm INAA homogeneity test results. In this study, the use of the complementary
gamma ray photopeaks of 110mAg, 82Br, 60Co, 134Cs, 152Eu, 59Fe, 140La, 233Pa (for Th determination), 46Sc and 75Se radionuclides was investigated in the between bottle homogeneity study of a mussel candidate RM under preparation at IPEN–CNEN/SP.
Although some photopeaks led to biased element content results, the use of complementary gamma ray photopeaks proved to be

The analysis of impurities and degradation products in pharmaceutical preparations are usually performed by chromatographic techniques such as high-performance liquid chromatography (HPLC). This approach demands extensive analysis time, mostly due to extraction and separation phases. These steps must be carried out in samples in order to adapt them to the requirements of the analytical method of choice. In the present contribution, matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) was employed to quantify an important degradation product in atorvastatin calcium 80 mg tablets: the atorvastatin lactone. Through the standard of the impurity, it was possible to perform quantitative analysis directly on the drug tablet, using a quick and novel approach, suitable for quality control processes in the pharmaceutical industry. 相似文献

Samples of must derived from six different varieties of grapes taken during the fermentation process, as well as the respective wine samples directly after the end of the malolactic fermentation, were analyzed by direct infusion negative ion mode electrospray ionization mass spectrometry (ESI-MS). Diagnostic ions for must were different from those of wine samples, although small variations for each of the grape varieties were also detected. The addition of unfermented must or sugar to wine could also be clearly detected. The spectra were acquired in a few minutes per sample, indicating that ESI-MS can be used for high-throughput analysis of samples and should prove useful for quality control during and after the fermentation process. 相似文献

We hypothesize that the energy strategy of a cell is a key factor for determining how, or if, the immune system interacts
with that cell. Cells have a limited number of metabolic states, in part, depending on the type of fuels the cell consumes.
Cellular fuels include glucose (carbohydrates), lipids (fats), and proteins. We propose that the cell's ability to switch
to, and efficiently use, fat for fuel confers immune privilege. Additionally, because uncoupling proteins are involved in
the fat burning process and reportedly in protection from free radicals, we hypothesize that uncoupling proteins play an important
role in immune privilege. Thus, changes in metabolism (caused by oxidative stresses, fuel availability, age, hormones, radiation,
or drugs) will dictate and initiate changes in immune recognition and in the nature of the immune response. This has profound
implications for controlling the symptoms of autoimmune diseases, for preventing graft rejection, and for targeting tumor

The degradation of the dye Indigo Carmine by hypochlorite in aqueous solution was monitored by electrospray ionization mass spectrometry in the negative ion mode (ESI(—)‐MS). Hypochlorite was highly efficient in removing the color of aqueous solutions of the dye. ESI(—)‐MS monitoring showed that concomitant with the Indigo Carmine consumption two transient species appeared (detected as doubly charged anions) probably formed via a net insertion of two hydroxyl groups at the exocyclic C?C bond followed by the incorporation of two (mainly) or one oxygen atoms at the indolic rings of the dye. Structures of these products were proposed based on the ESI(—)‐MS/MS data and high accuracy mass measurements. These two transient intermediates quickly decomposed, both in the condensed and in the gas phase, to yield mono‐charged anions. Based on these results, a route for the Indigo Carmine degradation by hypochlorite in aqueous solution has been proposed. Copyright © 2007 John Wiley & Sons, Ltd. 相似文献
