Two kinds of thio-heterocyclic fused naphthalene carboxamides, 3a-b, 4a-b, were designed, synthesized and quantitatively evaluated as efficient antitumor and DNA photocleaving agents. Compound 3a or 3b, having the thiophene ring, intercalated into DNA more strongly than compound 4a or 4b, having the thioxanthene ring. Compound 4a or 4b, photocleaved DNA more efficiently than 3a or 3b via superoxide anion. Compound 4a was the strongest inhibitor for P388 (murine leukemia cell), while 3a was the most cytotoxic one against A549 (human lung cancer cell). Each new compound showed stronger DNA photocleaving activity than corresponding naphthalimide.  相似文献

The reaction of copolymer of N,N-dimethylacrylamide (DMAA) and bromoethyl methacrylate with potassium cinnamate produced water-soluble photosensitive polymers. Photosensitive polyDMAA films were irradiated with a 400 W high-pressure mercury lamp (λ > 280 nm) to produce crosslinked polymers, which were swollen in water. The degree of swelling was controlled by the irradiation time and content of cinnamate moieties in copolymers. Higher cinnamoylation and longer irradiation time resulted in higher yield of crosslinked polymers and less swellability. Partial degelation upon irradiation at λ ~ 254 nm was observed. The advantage of gelation via photodimerization over conventional chemical crosslinking methods is discussed in conjunction with biomedical applications. © 1992 John Wiley & Sons, Inc.  相似文献

Two ruthenium complexes containing a new ligand phipz (phipz = 2‐(1,10‐phenanthroline)‐1H‐imidazo[4,5‐b]phenazine) were designed and synthesized. These complexes were found to inhibit the DNA supercoiled relaxation mediated by topoisomerase I (topo I), cleave DNA under irradiation and bind to calf thymus DNA through intercalative mode. Furthermore, complex 2 shows higher photocleavage activity, topo I inhibition activity and DNA affinity than complex 1 . Additionally, introduction of phenazine unit may be the reason that two complexes exhibit DNA ‘light switch’ behavior. The present work shows that two complexes might be potential as new DNA ‘light switches’, DNA photocleavers and topo I inhibitors.  相似文献

A series of Co(II), Ni(II), Cu(II), and Zn(II) complexes of a tridentate hydrazone were prepared and characterized by various spectro‐analytical techniques and magnetic moment studies. The complexes were found to be monomeric and non‐electrolytes. The copper complex is electrochemically active in the applied potential range. The compounds synthesized in the present study have shown promising antiproliferative activity when screened using the in vitro method against two human cancer cell lines: HeLa and HepG2. The Escherichia coli DNA‐binding properties of all the compounds were investigated with UV–visible absorption spectrophotometric titrations, viscosity measurements, DNA melting experiments and gel electrophoreses measurements. The compounds were demonstrated to act as DNA intercalators with appreciable DNA‐binding constant values. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

合成了两种三齿多吡啶钴(II)配合物 (A)和[Co(H2Bzimpy)2]Cl2 (B), 用元素分析、IR对配合物的组成和结构进行了表征, 测定了配合物A的晶体结构. 用电子吸收光谱、荧光光谱、循环伏安法及凝胶电泳实验等方法研究了配合物与DNA的相互作用. 结果表明配合物AB与小牛胸腺(CTDNA)的作用属部分插入和静电结合, 凝胶电泳实验表明配合物A在310 nm光辐射15 min, 可使超螺旋pBR322DNA断裂为开环缺口型和线型DNA.  相似文献

The effective guidance of mesenchymal stem cell (MSC) differentiation on a substrate by near‐infrared (NIR) light is particularly attractive for tissue engineering and regenerative medicine. However, most of current substrates cannot control multidirectional differentiation of MSCs like natural tissues. Herein, a photocontrolled upconversion‐based substrate was designed and constructed for guiding multidirectional differentiation of MSCs. The substrate enables MSCs to maintain their stem‐cell characteristics due to the anti‐adhesive effect of 4‐(hydroxymethyl)‐3‐nitrobenzoic acid modified poly(ethylene glycol) (P1) attached on the upconversion substrate. Upon NIR irradiation, the P1 is released from the substrate by photocleavage. The detachment of P1 can change cell–matrix interactions dynamically. Moreover, MSCs cultured on the upconversion substrate can be specifically induced to differentiate to adipocytes or osteoblasts by adjusting the NIR laser. Our work provides a new way of using NIR‐based upconversion substrate to modulate the multidirectional differentiation of MSCs.  相似文献

以五氟苯甲醛和吡咯经缩合反应合成了五氟苯基二吡咯烷烃(1); 1与3-氯-4-羟基苯甲醛经缩合反应合成了一个新的10-位苯环被氯原子取代的单羟基自由咔咯--10-(3-氯-4-羟苯基)-5,15-二(五氟苯基)咔咯(〖STHZ〗2〖STBZ〗),收率12.0%,其结构经UV-Vis, 1H NMR, 19F NMR, HR-ESI-MS和X-射线单晶衍射表征。琼脂糖凝胶电泳实验发现:2在光照下能够有效引起超螺旋质粒pBR 322 DNA发生断裂,当浓度为100 μmol·L-1时,活性最强,断裂百分比为68%。  相似文献

A cationic porphyrin–anthraquinone hybrid and its Zn(II), Cu(II), Co(III), and Mn(III) complexes were synthesized and their interactions with duplex DNA were studied using a combination of absorption, fluorescence titration, circular dichroism spectroscopy, thermal DNA denaturation, viscosity measurements, gel electrophoresis, and gas chromatography. Metal coordination induces intermolecular steric hindrance and thus the metal hybrids have smaller DNA binding affinities than the free base hybrid; the steric hindrance could relax the intermolecular aggregation and increase 1O2 generation of the metal hybrids. The DNA photocleavage abilities of these hybrids were investigated.  相似文献

Four photoreactive coumarin derivatives were successfully synthesized from 7-hydroxy-coumarin and 7-hydroxy-4-methylcoumarin, i.e., 7-propionyloxy-4-methylcoumarin (M1), 7-palmitoyloxy-4-methylcoumarin (M2), 7-propionyloxycoumarin (M3), and 7-palmitoy-loxycoumarin (M4). Reversible photodimerization (350 or 300 nm) and photocleavage (254 nm) of these coumarin derivatives dispersed in poly(vinyl acetate) (PVAc) were investigated by tracing their UV absorbance variations at 310 nm. The M2 and M4 with long palmitoyl chain show much better photoreaction reversibility than M1 and M3 with short propionyl chain. Moreover, photodimerization rate (under 350 nm) of M2 is greater than 200 times of that of M1. This has been explained by the formation of suitable conformation for revers-ible photodimerization due to the hydrophobic interactions. Photodimerization of M2 is ca. 3 times quicker than that of M4, indicating 4-methyl substitution enhances pho-todimerization. The influence of photodimerization wavelength (350 and 300 nm) and photosensitizer (benzophenone) have also been investigated in detail. © 1995 John Wiley & Sons. Inc.  相似文献

The quinoxaline moiety in antitumor quinoxaline antibiotics cleaved double stranded DNA at the 5′ side guanine of the 5′-GG-3′ site upon irradiation with UV light with a long wavelength and without any additive. The quinoxaline-carbohydrate hybrid system was very effective for the DNA cleavage. Furthermore, the quinoxaline-carbohydrate hybrids exhibited strong and selective cytotoxicity against cancer cells with photoirradiation.  相似文献
