Bryostatins are a class of naturally occurring macrocyclic lactones with a unique fast developing portfolio of clinical applications, including treatment of AIDS, Alzheimer's disease, and cancer. This comprehensive account summarizes the recent progress (2014–present) in the development of bryostatins, including their total synthesis and biomedical applications. An emphasis is placed on the discussion of bryostatin 1 , the most-studied analogue to date. This review highlights the synthetic and biological challenges of bryostatins and provides an outlook on their future development.  相似文献

The electronic structures of N,N′-bis[(1-phenyl-3-methyl-5-oxo-4-pyrazolinyl) α-furylmethylidyne]ethylendii- mine (and -o-phenylendiimine), and their complexes of transition metals M (M=Ni, Cu, Co, Pd, Pt) have been calculated by B3LYP method of DFT. Their geometries and bonding characters are discussed, and the relationship between their bioactivities and electronic structures are also discussed from the calculated results of electronic populations and frontier orbital energies.  相似文献

Several novel N′-[(1-ferrocenylethylidene)amino]-N′-β-D-glycopyranosylthioureas have been efficiently synthesized via a six-step procedure, utilizing three monosaccharides (D-glucose, D-galactose and D-xylose) as starting materials, respectively. They have been preliminarily determined to display biological activity against certain fungi. The target compounds have been unambiguously characterized by spectroscopic method and elemental analysis. An initially alternative synthetic route, which was seemingly reasonable, has been excluded due to its inefficiency.  相似文献

在氢氧化钠和四丁基溴化铵存在下,将化合物2-N-茄呢基胺基苯甲酸(3)和N-茄呢基己二酰胺酸(6)分别与O-乙酰基溴代葡萄糖、O-乙酰基溴代半乳糖、O-乙酰基溴代乳糖和O-乙酰基溴代麦芽糖反应制得对应的糖酯4a~4d和7a~7d,由元素分析,IR,1H NMR和MS确证了8个新化合物的结构,并对其中6个化合物(4a,4d和7a~7d)在三种癌细胞模型上进行了一些初步体外生理活性的测试.  相似文献

Metallic implants are widely used in orthopaedic surgery but metal release has been reported by several authors. High metallic ion concentration in surrounding tissues may play a major role in therapeutic failure. We have investigated in vivo and in vitro two biological glasses (BVA and BVH) used as coatings of metal implants. Physicochemical characterization was made by several complementary methods, particularly particle induced x‐ray emission (PIXE) and energy‐dispersive x‐ray spectroscopy (EDXS) associated with electronic microscopy. Analyses clearly show the differences of behaviour between both glasses. The BVA glass is bioactive, i.e. it develops a direct chemical bond between prosthesis and bone by the formation of a calcium phosphate layer at its surface. These structural and compositional modifications are caused by hydrolysis of the glass. After its dissolution BVA glass is replaced by bone, which ensures better integration of the implant in the bone site. The BVH glass is not bioactive and is used as a cement to isolate the metal implant from the biological environment, but the coating disaggregates with implantation time and glass grains migrate through the bony lacuna network. 以取代苯甲醛、取代吡啶和取代苯甲酸酯等为原料合成了 10种含硫杂环化合物 (其中 2a~ 2g化合物为国内外未见报道 ) .在合成过程中采用了超声波技术 ,极大地缩短了反应时间 ,提高了收率 .所得化合物的结构经IR ,1 HNMR ,MS和元素分析证实 .对其生物活性进行了初步测试 ,结果证明一些化合物具有良好的抑菌或杀菌作用 ,其中 2b ,2c ,2e ,2h ,2i和2j对大肠杆菌 ( 10 40 7号 )的抗菌效果与CET和SMZ相当 ;2b ,2c ,2f对草绿色链球菌 ( 112 3号 )的抗菌效果与CET和SMZ相当 .  相似文献

Design, synthesis, and bioactivity evaluation of novel mannich bases ( 2a-2j ) and triazole-chalcone derivatives ( 7a-7k ) of Eugenol 1 were reported. Among all the derivatives tested for antiproliferative activity, di-amine manich derivative 2b (32.92 μM), and 4-methoxy chalcone triazole derivative 7d (33.05 μM) significantly inhibited HepG2 cell lines when compared to the standard doxorubicin (37.29 μM). Whereas most of the compounds such as diethylamine 2a (17.75 μM), (aminomethyl) methane diamine 2b (17.02 μM), and bis (chloromethyl) amine 2c (20.12 μM) showed moderate to better inhibition towards MCF-7 cell lines. The synthesized analogues were also tested for antidiabetic and antiobesity potentials. Compounds 2f (55.50%), 2c (54.34%), 7g (55.5%), and 2a (55.5%) have shown moderate inhibitory potentials toward intestinal α-glucosidase enzyme when compared to the standard Acarbose (72.86%). Likewise, compounds 7d (82.95%), 7f (76.19%), 7g (74.81%), 7e (74.81%), and 2g (72.50%) have shown significant to moderate inhibitory potentials toward Pancreatic lipase enzyme when compared to the standard orlistat (91.10%). ROS induces life-threatening diseases like diabetes, cancer, etc., and antioxidants play a major role in controlling their production. Compounds 2c (99.81%), 2i (99.80%), 2d (99.26%), 2g (98.79%), and 2f (98.42%) have shown significant antioxidant profiles in ABTS assay when compared to the standard Trolox (99.07%). Further, In silico Molecular docking and pharmacokinetic screening of the eugenol derivatives complemented the in vitro results indicating the drug likeness of the obtained active compounds.  相似文献
